Snakehead fish has been sources that have high protein content and can be used as antioxidant and anti-diabetes. To increase the level of protein content and amino acid in snakehead fish, the treatment of hydrolysis and fermentation were chosen in this study. Therefore, the purpose of this study was to evaluate the characteristics of snakehead fish protein and its potential as antihyperglycemic. Three samples were used in this study, i.e., hydrolysate, fermented and non-fermented isolates. The experimental design used was completely randomized design. Data were analyzed by analysis of variance (ANOVA) and continued by Duncan multiple range test (DMRT) (α = 5%).  The current study reported that the hydrolysate had higher<br />protein content 90.43%, as compared to the fermented and non-fermented isolates were 84.43% and 78.69%, respectively. At a concentration of 10.000 ppm, hydrolysate showed highest inhibition activity (74%), as compared to fermented isolate inhibited 59% and none-fermentation isolate inhibited 56%. Hydrolysate also had higher amino acid content than fermented and non-fermented isolates of 51.15, 44.34, and 32.00 %w/w, respectively. Hydrolysate had the lowest molecular weight (&lt;10 kDa), while fermented and non-fermented isolates were &lt;10 kDa. This probably due to the hydrolysis process using an enzyme was capable to break the peptide fractions of snakehead fish protein. Hence, it increased the levels of protein, amino acids, there by protein hydrolysate had high inhibitory potential than fermented and non-fermented isolates.

Fish protein hydrolysate (FPH) containing small protein or peptides and amino acids has a great attention related to the provision of high protein foods to overcome the problem of malnutrition. This research was purposed to prepare FPH from yellowstripe scad (Selaroides leptolepis) by using a local microbial protease from Bacillus subtilis BII-1. Hydrolysis process was done in a laboratory scale (500 g minced fish) at 55oC for 6 h. The liquid hydrolysate was then spray dried using whey protein and maltodextrin at a concentration of 20 and 30% for each filler. The treatment of whey protein powder produced FPHs with higher protein content (31.71-33.97% db) and slightly yellowish in color compared to maltodextrin (11.88-16.66% db). Their foaming capacity and stability were 20-100% and 15% in 5-10 min, respectively. However, FPHs prepared with maltodextrin had no foaming capacity. The hydrolysates from both treatments had low water and oil absorption with the value less than 3 mL/g hydrolysate. A trial on scaling up production using 30 kg fish, showed that optimization or adjustment should be carried out due to the high amount and high protein content of the residual products.

 Hidrolisat protein ikan (HPI) dihasilkan dari proses penguraian protein ikan menjadi peptida sederhana maupun asam amino melalui proses hidrolisis oleh enzim, asam, atau basa. Enzim yang digunakan adalah enzim protease, baik yang berasal dari tanaman, hewan maupun mikroba. Tujuan dari penelitian ini adalah mengetahui pengaruh penambahan enzim papain terhadap karakteristik HPI dan menentukan optimum terbaik hidrolisat protein dari jenis ikan yang berbeda dengan penambahan enzim papain. Metode penelitian ini adalah experimental laboratories dengan menggunakan Rancangan Acak Lengkap (RAL). Perlakuan yang diterapkan adalah perbedaan jenis ikan yang meliputi ikan nila, ikan bandeng, dan ikan cucut. Data yang diperoleh dianalisis dengan menggunakan sidik ragam ANOVA dengan uji lanjut untuk menentukan nilai yang berpengaruh maupun tidak dengan uji BNJ (Beda Nyata Jujur). Hasil penelitian dari ketiga perlakuan didapatkan HPI terbaik yaitu rendemen  HPI nila 5,64% dengan kadar protein 30,17%, kadar air 9,06%, dan viskositas 1,91 cP sedangkan profil asam amino terbanyak adalah asam glutamat terutama pada HPI ikan cucut sebesar 4,183%. Fish protein hydrolysate (FPH) is produced from the decomposition of fish protein become peptides and amino acids through hydrolysis by enzymes, acids or bases. The enzyme used are protease enzymes, both derived from plants, animals, and microbes. The purpose of this study was to determine the effect of papain enzyme addition on FPH characteristics and to determine the characteristics of the FPH and determine the best optimum of FPH from different fish species with the addition of the papain enzyme. This research method is experimental laboratories by using a Completely Randomized Design (CRD). The applied treatments applied are different fish species including tilapia, milkfish, and shark. The data obtained were analyzed by using ANOVA with further test to determine the value that influencd or not with BNJ test. The results of the research from the three treatments are yield FPH tilapia 5,64% with protein content 30,17%, water content 9,06%, and  viscosity 1,91 cP, while the highest amino acid profile is glutamic acid, especially in FPH shark 4,183%.

Silver bellies, Leiognathus splendens were studied for their spatial (south-west and south-east coasts of India), annual (2008–2011) and seasonal (pre-monsoon, monsoon and post-monsoon) variations of protein, amino acids, vitamins and minerals. The monthly mean Sea Viewing Wide Field-of-view Sensor data for the period from January 2008 to December 2011 were taken into account to indicate the distribution of the photosynthetic pigment chlorophyll-a to test the hypothesis that surface productivity might be related to nutritional biochemistry of this species. The four year average total protein content and chlorophyll-a showed good correlation during monsoon on the south-west coast and monsoon/post-monsoon on the south-east coast, suggesting that the protein content is prejudiced by the chlorophyll-a concentration. Amino acid scores observed monsoon maxima along the south-west and south-east coasts. Significant seasonal variations in vitamin content were observed at the study locations with high content of vitamins D3, E, K1 and C on the south-west coast. Na content was maximal during pre-monsoon on the south-west coast, while post-monsoon maxima of Ca and K content were observed. The Fe, Mn and Zn were abundant in the samples collected from the south-west coast. The concentration of Se exhibited maximum values post-monsoon along the south-west and south-east coasts. The present study demonstrated L. splendens as a valuable source of the protein, amino acids, minerals and vitamins, showing that this low-value species is a good source of well balanced proteins with high biological value to be qualified as a preferred healthy food for human consumption.

Abstract Three methods (Kjeldahl, sulfuric acid-hydrogen peroxide, and summation of amino acid content) for determining and calculating the protein content of apple flower buds were compared. Quantitation of protein content based on summation of amino acids appears to be the most accurate method. A new nitrogen:protein conversion factor (5.51) was calculated based on total amino acid analysis. This new conversion factor could replace the conventional 6.25 factor for estimating total protein content of apple flower bud by the Kjeldahl method. However, Kjeldahl is not an accurate method for estimating protein content in apple flower bud tissue, regardless of the conversion factor, and probably would not be a good method for estimation of protein in other plant species.

Abstract The oocytes of the marine goby Pomatoschistus minutus were analyzed for their amino acid content. Most of the amino acids exist as protein, only a little part is free or peptide-bound. Among the protein-bound amino acids, high levels of glutamic acid, proline, alanine, aspartic acid, valine and leucine were detected. These represent more than 60% of the protein amino acids. Among the free acids, glutamic acid, serine and alanine, are dominant. There are no certain proofs of the occurrence of peptide pools in the oocytes of Pomatoschistus minutus.

ABSTRAK   Ikan Bandeng berpotensi menjadi bahan baku hidrolisat protein ikan karena kandungan protein tinggi dan potensi produksi cukup melimpah. Enzim Bromelin merupakan salah satu enzim protease yang dapat membantu dalam mempercepat reaksi hidrolisa protein. Penelitian ini bertujuan untuk mengetahui pengaruh konsentrasi enzim bromelin terhadap kualitas hidrolisat protein Ikan Bandeng (Chanos chanos Forks) segar. Rancangan percobaan yang digunakan pada penelitian ini adalah rancangan acak lengkap dengan perlakuan konsentrasi enzim yang berbeda (4%, 5% dan 6%). Parameter yang diamati adalah nilai kadar protein, lemak, air, karbohidrat, serat kasar dan rendemen.  Konsenterasi enzim bromelin berpengaruh nyata pada nilai kadar protein, lemak, air, abu, serat kasar dan rendemen (P<0,05), tetapi tidak berpengaruh nyata pada kadar karbohidrat (P>0,05). Kondisi optimum untuk menghidrolisis daging Ikan Bandeng menjadi hidrolisat protein adalah konsentrasi enzim bromelin 6% dengan waktu hidrolisis selama 6 jam.   Kata kunci: Ikan Bandeng (Chanos chanos), hidrolisat protein ikan, enzim bromelin     ABSTRACT   Milkfish is potentially become the raw material of fish protein hydrolyzate because of the high protein content and relatively abundant production. Bromelin is a protease enzyme that can help on improving protein hydrolyzate quality. This research aimed to determine the effect of bromelain enzyme concentration on the quality of the protein hydrolyzate of milkfish (Chanos chanos Forsk) fresh. The experimental design used in this study was completely randomized design with different concentrations of the enzyme (4%, 5% and 6%) as a treatment. The parameters measured were the contents of protein, fat, water, carbohydrates, crude fiber and yield. The concentration of enzyme bromelin significant effect on contents of protein, fat, water, ash, crude fiber and yield (P<0,05), but no effect on carbohydrates content (P>0,05). The optimum conditions to hydrolyze the milkfish meat into a protein hydrolyzate used 6% bromelin enzyme at the time of hydrolysis for 6 hours.  Keywords: Milkfish (Chanos chanos), fish protein hydrolisate, bromelin enzym

ABSTRACT Introduction: Patilo is one of traditional various snacks from Gunung Kidul that is made of fermented cassava waste and cassava starch. Fermentation is intended to reduce or eliminate the HCN content of cassava and to establish special flavour. The protein content of patilo is very low, therefore it can be enriched with animal protein from fresh water fish in form of fish flour or fish protein hydrolysate (FPH). Enrichment with fish protein hydrolysate has an advantage since it will be easier to digest by human because FPH better functional properties and higher solubility.Method: The study consisted of four activities that were 1) making/producing mujair FPH, 2) producing and enriched patilo with mujair FPH, 3) analyzed the nutritive values and protein digestibility of enriched and unenriched patilo and 4) analyzed the consumers acceptability of the products. The experiments were carried out in the 1) Food Engineering and Chemistry Laboratorium of Faculty of Agriculture Technology Semarang University, 2) Pilot Plan PAU and GMSK Laboratory of Bogor Institute of Agriculture, 3) Sidoardjo Village, Tepus Gunung Kidul. Variables being studied were nutritive value and protein digestibility of unenriched and enriched patilo with mujair FPH in several consumer acceptability in terms of flavour, taste, colour and texture. The study design was a complete random design of one factor in which the FPH was added in 5%, 10% and 15% concentration, each treatment repeated 3 times. Statistical analyses used for nutritive value and protein digestibility were Anova followed by Least Significant Difference test with 0,05 level of significancy Friedman and Wilcoxon Sign Rank test were used to test the organoleptic values both in the laboratory and in the field. Result:1) The nutritive value changes of raw enriched patilo were : a) significant decrease in water and carbohydrate content, b) significant increase in ash, fat and protein content compared to the unenriched patilo, 2) The nutritive value change of fried enriched patilo were : a) significant decrease in water and carbohydrate content, b) significant increase in fat, protein and energy content compared to the unenriched patilo, 3) The protein digestibility in raw and fried enriched patilo was significant higher than that without enrichement, 4) Patilo enriched with 10% mujair FPH was mostly liked by consumer either in the laboratory or in the field in term of taste, flavour and texture as for colour, all consumers prefered the unenriched patilo, 5) The changes of the nutritive values and protein digestibility were statistically significant in the enriched patilo, however from the nutrition point of view, only the increased of protein content (six folds increase) was meaningful. Conclusion: Patilo enriched with 10% mujair FPH is the most/well accepted by laboratory panelists and field consumers, and there was six folds increase in the protein content.   Key Word: Patilo, enrichment and fish protein hydrolysate.   ABSTRAK   Latar Belakang: Patilo adalah salah satu makanan jajanan tradisional Gunung Kidul yang diolah dari ampas singkong yang difermentasi dan dicampur dengan pati singkong. Kandungan protein dalam patilo sangat rendah, untuk itu perlu diperkaya antara lain dengan sumber protein hewani dari ikan air tawar baik dalam bentuk tepung ikan atau hidrolisat protein ikan (HPI). Tujuan: Penelitian ini bertujuan untuk 1). menganalisis nilai gizi patilo 2). menganalisis daya cerna protein patilo 3). menganalisis daya terima patilo goreng tanpa dan dengan penambahan HPI mujair Metode: Penelitian ini terdiri dari empat kegiatan yaitu: 1). pembuatan HPI mujair, 2). pembuatan dan pengkayaan patilo dengan HPI mujair, 3). pengujian nilai gizi dan daya cerna protein patilo tanpa dan dengan penambahan HPI mujair dan 4). uji penerimaan patilo baik di laboratorium maupun di lapangan. Variabel yang diamati adalah kandungan gizi patilo dan daya cerna protein patilo. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap dengan 1 faktor yaitu HPI0=tanpa penambahan HPI, HPI1= penambahan HPI 5% dari berat ampas (b/b), HPI2=penambahan HPI 10 % dari berat ampas (b/b) dan HPI3=penambahan HPI 15% dari berat ampas (b/b), masing-masing perlakuan diulang sebanyak tiga kali. Analisis kandungan gizi dan daya cerna patilo dengan Anova, uji dilanjutkan dengan uji Least Significant Difference (LSD) pada taraf dan 5%. Untuk analisis uji organoleptik baik di laboratorium maupun di lapangan dengan uji Friedman. Perbedaan hasil akan dilanjutkan dengan uji lanjut Wilcoxon Sign Rank Test. Hasil: 1) Nilai gizi patilo mentah dengan penambahan HPI mujair dibandingkan tanpa penambahan HPI mujair adalah : a) terjadi penurunan kadar air dan kadar karbohidrat secara signifikan (ρ<0,05) dan b) terjadi peningkatan kadar abu, kadar lemak dan kadar protein secara signifikan (ρ<0,05), 2) Nilai gizi patilo goreng dengan penambahan HPI mujair dibandingkan tanpa penambahan HPI mujair adalah : a). terjadi penurunan kadar air dan kadar karbohidrat secara signifikan (ρ<0,05) dan b).terjadi peningkatan kadar lemak, kadar protein dan energi secara signifikan (ρ<0,05) 3). Daya cerna protein patilo mentah dan goreng dengan penambahan HPI mujair meningkat secara signifikan (ρ=0,000) dibandingkan tanpa penambahan HPI mujair pada semua konsentrasi, 4). Patilo dengan penambahan HPI mujair 10% paling disukai oleh panelis agak terlatih dan panelis konsumen anak sekolah berdasarkan skor rasa, bau dan tekstur. Untuk warna, panelis agak terlatih dan panelis konsumen anak sekolah memilih patilo tanpa penambahan HPI mujair dan 5). Secara statistik perubahan nilai gizi dan daya cerna protein bermakna kecuali untuk kadar abu patilo goreng, namun dari sudut gizi perubahan yang berarti hanya pada kadar protein yang mencapai 6 kali lipat. Simpulan: Patilo yang diperkaya dengan HPI mujair 10% paling disukai oleh panelis agak terlatih maupun panelis konsumen anak sekolah dan terjadi kenaikan secara signifikan pada kadar protein yang mencapai 6 kali lipat. The metabolism of lipid and protein fractions in developing and germinating flaxseed was investigated. Fatty acid concentrations in the early-development stages of field- and greenhouse-grown plants showed similar increases, but the fatty acid composition of the mature seed appeared to be influenced by environment. The pattern of fatty acid interconversions during lipid synthesis suggested that more-unsaturated acids were formed from less-unsaturated ones. Studies at later stages of seed development indicated that the lipid was in a state of dynamic equilibrium. The relative proportions of each fatty acid during germination remained constant, suggesting that fatty acids were being broken down at a rate proportional to the amount originally present. During flaxseed development protein content increased steadily. Until 4 weeks after the plants had bloomed, the total content of each amino acid increased, but the free amino acid portion and amide nitrogen decreased. Subsequently all remained at a constant level. During germination the protein content and the proportions of the various amino acids of flaxseed remained constant but the free amino acid content rose. The radioisotope studies gave no clear indication of amino acid interconversions.
