In blood of children underwent to the long-term isolated exposure of manganese and stable strontium with drinking water exceeding the acceptable level by up to 1.4 times the elevated concentrations of mentioned metals in blood were shown to be formed in relation to reference level by 2,0-4,4 times. The developing at that mutagenic effect was proved to have the dependence on the level of manganese and stable strontium concentration in blood and is pronounced in the increased rate of cells with cytogenetic anomalies on the background of the enhancement of the proliferative activity, destructive changes in the cell population and oxidative responses at the cell DNA level. The activity of genetic instability and processes of DNA oxidation in the somatic cells in children under the exposure to manganese has more pronounced character in comparison to given indices under the exposure to the stable strontium.

There was made a study of the features of apoptosis in children drinking potable water with an increased content of stable strontium of natural origin. The percentage of non-standard samples of strontium content in the water on the observation territory was 16.7%, which corresponded to the exceedance of the maximum permissible concentration of strontium in drinking water at the level of 1.2 MPC (up to 8.4 mg/l). In children living in the strontium geochemical province, the blood content of strontium (0.1 mg/dm3) was established statistically significant higher (p = 0.02) in comparison with the results obtained from children living in the territory of relative sanitary and hygienic well-being (0.028 mg/dm3). There was made an estimation of parameters of the dependence between the concentration of strontium in drinking water and the concentration of strontium in the blood was estimated, there was obtained a reliable dependence (b0 = 0.04, b1 = 0.012, R2 = 0.44, F = 310, p = 0.00001) of the increase in the blood content of strontium by the 1.3 times relative to the reference level from the concentration of strontium in water. In the exposure range of 7.84 ± 0.62 mg/L per 1 mg/L increase in strontium in water, its concentration in the blood was established to increase by 0.012 mg/dm3. In children living in the strontium geochemical province (1.5-0.8 times) by the method of flow cytometry, there were established to statistically significant decline the (relative and absolute values) number of CD95+ lymphocytes (p = 0.04-0.001) by 3.8 times, the expression of p53, bcl-2, Bax and the amount of Annexin V-FITC+PI- cells dying via the path of apoptosis, as well as the content of Annexin V-FITC+PI+ cells, dying via necrosis was statistically significantly (p = 0.001) elevated, in comparison with the control group. The obtained results prove the prolonged excessive effect on the children’s organism of stable strontium supplied with drinking water to lead to changes in the functioning of the immune system, characterized by a violation of the regulation of cell death - inhibition of apoptosis and activation of necrosis. The ranges of values of strontium in the blood corresponding to the ranges at the level of 0.1 [0.04-0.11] mg/dm3 are given in the ranges of apoptotic regulation (relative content,%): CD95+ - 17.50 [15.48; 22.03], p53 - 0.53 [0.22; 0.98], bcl-2 0.24 [0.21; 0.36], bax = 5.66 [4.67; 7.84], AnnexinV-FITC+PI- cells: 0.67 [0.56; 0.88], AnnexinV-FITC+ PI+ cells: 10.58 [9.54; 16.11]. The indices of apoptotic regulation of CD95+, p53, bcl-2, Annexin V-FITC+ PI- are recommended to be used as indices of early disorders of the immune status, and also for the monitoring the effectiveness of preventive measures in the territories of strontium geochemical provinces.

The article analyze the long-term morbidity, spread of typhoid fever in the southern regions of the Kyrgyz republic, and remains a permanent epidemic focus in the Jalal-Abad region, where against the low availability of the population to high-quality drinking water, an additional factor on the body for more than two generations and radiation factor, which we confirmed by the spread among the inhabitants of Mailuu-Suu of nosological forms of the syndrome of immunological deficiency, as a predictor of risk groups for infectious diseases, including typhoid fever.

While lack of access to consistent safe drinking water is estimated to affect nearly 2 billion people worldwide, many of the efforts to solve this crisis have proven to be unsustainable. This paper discusses some of the reasons for these challenges and suggests interdisciplinary practices that could be integrated from the very beginning of a water intervention to achieve long-term success. Of key importance for sustainable water implementation is an enabling environment that incorporates aspects such as funding, potential for market development, and supportive governance. While this enabling environment is acknowledged, the focus of this work is on the integration of three key areas: (i) social and cultural assessment of behavior and preferences; (ii) market-based implementation approaches that draw on this knowledge; and (iii) technology development for these markets.

Abstract Background Under the EU chemicals regulation REACH (Registration, Evaluation, Authorisation and Restriction of Chemicals EC 1907/2006), registrants are not obliged to provide information related to intrinsic substance properties for substances that pose a threat to the drinking water resources. In 2019, perfluorobutane sulfonic acid (PFBS) and 2,3,3,3-tetrafluoro-2-(heptafluoropropoxy)-propanoic acid (HFPO-DA trade name GenX) were demonstrated to have an equivalent level of concern (ELoC) to persistent, bioaccumulative and toxic or very persistent and very bioaccumulative (PBT/vPvB) substances owing to their persistent, mobile and toxic (PMT) substance properties and very persistent and very mobile (vPvM) substance properties, respectively. They were both subsequently identified as substances of very high concern (SVHC) applying Article 57(f) in REACH. This work follows up on this regulatory decision by presenting a science based, conceptual level comparison that all PMT/vPvM substances pose an ELoC to PBT/vPvB substances. Using the two cases named above, as well as 1,4-dioxane, 16 categories were developed to evaluate a) serious effects on human health, b) serious effects on the environment and c) additional effects. 1,4-dioxane has recently been proposed to be classified as Carcinogenic 1B by the Committee for Risk Assessment (RAC). The aim was to enable an objective and scientifically justified conclusion that these classes of substances have an equivalent level of concern for the environment and human health. Results In all of the categories related to human health, the environment and other effects, the PMT/vPvM case study substances exhibited comparable effects to PBT/vPvB substances. A difference in the human and environmental exposure pathways of PMT/vPvM and PBT/vPvB substances exists as they vary temporally and spatially. However, effects and impacts are similar, with PMT/vPvM substances potentially accumulating in (semi-)closed drinking water cycles and pristine aquatic environments, and PBT/vPvB substances accumulating in humans and the food chain. Both PMT/vPvM and PBT/vPvB substances share the common difficulty that long term and long-range transport and risk of exposure is very difficult to determine in advance and with sufficient accuracy. Conclusion The registration process of substances under REACH should reflect that PMT/vPvM substances pose an equivalent level of concern to PBT/vPvB substances.

The most difficult issue when using tissue engineering products is enabling the ability to store them without losing their restorative capacity. The numbers and viability of mesenchymal stem cells encapsulated in a hydrogel scaffold after cryostorage at −80 °C (by using, individually, two kinds of cryoprotectors—Bambanker and 10% DMSO (Dimethyl sulfoxide) solution) for 3, 6, 9, and 12 months were determined, with subsequent assessment of cell proliferation after 96 h. The analysis of the cellular component was performed using fluorescence microscopy and the two fluorochromes—Hoechst 3334 and NucGreenTM Dead 488. The experimental protocol ensured the preservation of cells in the scaffold structure, retaining both high viability and proliferative activity during storage for 3 months. Longer storage of scaffolds led to their significant changes. Therefore, after 6 months, the proliferative activity of cells decreased. Cryostorage of scaffolds for 9 months led to a decrease in cells’ viability and proliferative activity. As a result of cryostorage of scaffolds for 12 months, a decrease in viability and proliferative activity of cells was observed, as well as pronounced changes in the structure of the hydrogel. The described scaffold cryostorage protocol could become the basis for the development of storage protocols for such tissue engineering products, and for helping to extend the possibilities of their clinical use while accelerating their commercialization.

Two critical windows in mammary development have been proposed. The first arises from observations in rodents that nutrition during fetal and neonatal periods can affect mammary ductular outgrowth, subsequent proliferative activity and, eventually, tumorigenesis, that is, potentially it could have a long-term effect on pathological outcome (breast cancer) in women. The second similarly involves early diet, but in this case the outcome is phenotypic, in that dairy heifers reared too quickly during the peripubertal period subsequently show impaired udder development and reduced milk yield persisting throughout life. Most mammary development occurs during pregnancy, but this period is usually thought of only in terms of the immediate outcome for the subsequent lactation; it is not believed to be a critical window, at least in terms of lifetime mammary productivity. This review examines the evidence underlying these various claims and attempts to define the mechanisms involved, and also considers whether derangements occurring earlier in life (prenatally) could also have long-term consequences for physiological or pathological mammary development.
