Colon cancer is the third most common cause of death from cancer worldwide. Recently, natural products have been widely used as an alternative therapy for colon cancer. Previous studies have reported that Nigella sativa has chemopreventive activity in vitro and in vivo.This study aimed to evaluate the effect of Nigella sativa seed (NSS) on rat-colon cell after initiation of 7,12-dimethylbenz [a] anthracene. Rats were divided into five groups, 12 rats in each group: Group I was given 7,12dimetilbenz [a] anthracene (DMBA) orally 20 mg/kgBW twice a week for five weeks, group V is the solvent control group was given corn oil. The other three groups were given DMBA + NSS, at the dosage of 250 mg/kgBW, 500 mg/kgBW and 750 mg/kgBW. NSS extract was dissolved in corn oil and administered daily per oral during the next two weeks before and during the initiation of DMBA. After 16 weeks, all rats were sacrificed. H&E staining showed that necrosis activity was lower in treated groups compared to DMBA group. AgNOR staining showed mAgNOR was significantly decrease following the increasing dose of NSS (250 mg/kgBW, 500 mg/kgBW and 750 mg/kgBW) were subsequently 1.62 ± 0.086, 1.60 ± 0.101 and 1.39 ± 0.049 (p<0.05). The results showed that NNS reduce the damage of colon cells and inhibit colon cell proliferation in DMBA induced rats.

Aim.To investigate the correlation between Sirtuin 3 (SIRT3) expression and the clinical outcome of patients with colon cancer.Methods. The tumor specimens from 127 patients with colon cancer were obtained for SIRT3 immunohistochemical staining. Patients were followed up. Inin vitrostudy, SIRT3 gene was inhibited to observe the effects of SIRT3 on the biological behavior of cultured colon cells.Results. The SIRT3 expression level was found to be significantly associated with the lymph node metastasis (P<0.001) and tumor stages (P<0.001). The colon cancer-specific survival was 64.6% among patients with high SIRT3 expressions and 88.6% among patients with low SIRT3 expressions (log-rankP=0.016). The overall survival was 80.2% among patients with low SIRT3 expressions and 55.9% among patients with high SIRT3 expressions (log-rankP=0.002).In vitrostudy showed that silencing of SIRT3 gene inhibited the proliferation, invasion, and cells migration but increased the apoptosis in the cultured colon cell lines.Conclusion. This study provides evidence supporting that SIRT3 is closely associated with the clinical outcomes of colon cancer. SIRT3 may be considered as a marker for colon cancer.

Previous study reported that Nigella sativa has in vitro and in vivo cancer activity.This study was conducted to observe the effect of chloroform extract of Nigella sativa seed (NCE) on 7,12-Dimethylbenz[a]anthracene (DMBA)-induced female rats’ splenocyte. The experiment consisted of five groups, corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE. Extract was dissolved in corn oil and oral administered daily for 2 weeks before and during the DMBA induction. Observation of cell proliferation was performed using haematoxylin and eosin (H&E) and AgNOR stainings. H&E staining showed decreased necrocis activity extract groups compared to DMBA group. From AgNOR staining results, mean AgNOR (mAgNOR) of extract groups was less in number compared to DMBA group. The mAgNOR in corn oil solvent control group, DMBA group, DMBA+250 mg/kgBW NCE, DMBA+500 mg/kgBW NCE and DMBA+750 mg/kgBW NCE were 1.22, 1.91, 1.29, 1.36 and 1.33, respectively. Our current results showed that NCE reduces the proliferation of DMBA-induced rat spleenocytes  Thus, NCE has potency to be developed as a chemopreventive agent.Keywords: Nigella sativa, spleen, DMBA, anti-proliferative

The proportion of patients with multiple unsuccessful attempts of assisted reproductive technology (ART) is about 30% of all patients treated with the use of ART. Women with history of unsuccessful ART attempts - a special category of patients who require emergency attention and a thorough examination at the stage of preparation for superovulation stimulation,the selection of embryos and endometrium preparation for embryo transfer. The objective: to distinguish high-risk group of unsuccessful attempts based on a detailed analysis of anamnestic and clinical data of infertile women with repeated unsuccessful ART attempts that requires more in-depth study of hormonal features, ovarian reserve and condition of the endometrium. Materials and methods. For better understanding of the problem of repeated unsuccessful ART attempts and сreation of efficient infertility treatment algorithms for these couples we conducted a thorough analysis of anamnestic data of three groups of infertile women (105 patients), which were distributed by age: group I – younger than 35, the II group – from 35 to 40, the III group - over 40 years. These groups of patients were compared with each other and with the control group of healthy women (30 persons). Results. Leading stress factors in the percentage three times prevailed in the group of infertile women and had a direct connection with the fact of procedure «fertilization in vitro» and chronic stressors caused by prolonged infertility. Primary infertility was observed significantly more frequent in patients younger than 35 years (p <0.05), secondary infertility - mostly in the second and third experimental groups (p <0.05). Noteworthy significant percentage of wellknown causes of infertility and idiopathic factor in all groups, and the prevalence of tubal-peritoneal factor in the second and third experimental groups, and endocrine dysfunction in the I experimental group. The most common disorder among this category of woman was polycystic ovary syndrome. Frequency of usual miscarriage among patients of I ana II groups was two times higher than in the third group (p <0.05). Among the experimental groups the leading place belongs urinary tract infection, respiratory tract diseases, pathologies of the cardiovascular system. Data of the stratified analysis show an increase likelihood of repeated unsuccessful ART attempts under the influence of constant chronic stress (odds ratio OR=2.06; 95% CI: 0.95–3.17; p<0.05). Conclusions. Among infertile patients with repeated unsuccessful ART attempts must be separated a high risk group of failures. The identity depends on the duration of infertility, female age and leading combination of factors. Key words: repeated unsuccessful ART attempts, anamnesis, infertility, high risk.

AbstractFracture resistance of endodontically treated teeth restored with post. Aims: This study aims to compare the fracture resistance of endodontically treated teeth restored with resin fiber and stainless steel post. Commercially available prefabricated resin fiber post(Dentsply Maillefer Easy Post), prefabricated stainless steel post(Coltene/Whaledent Parapost) were used. Methods and Material: Forty five maxillary central incisors were obturated and divided into 3 groups: Control Group (Group I) without any post (n = 15), Resin Fiber Post Group (Group II) (n = 15) and Stainless Steel Post Group (Group III) (n = 15). In all Groups except control group, post space was prepared; a post was cemented, and a core build-up was provided. All the specimens were subjected to compressive force under a universal testing machine until fracture. Statistical analysis used: The results were analyzed using the variable analysis test (ANOVA). Results: One-way analysis of variance revealed significant difference among test groups. The control group demonstrated highest fracture resistance (925.2183 N), followed by the resin fiber post group (486.7265 N) and stainless steel post group (423.539N). Conclusions: Teeth restored with resin fiber post showed higher fracture resistance values than prefabricated stainless steel post.

Abstract Our work aimed to evaluate the possible effect of Annona muricata (Graviola) leaf extract on Trichinella spiralis in in vitro and in vivo studies. Trichinella spiralis worms were isolated from infected mice and transferred to three culture media – group I (with no drugs), group II (contained Graviola) and group III (contained albendazole) – then they were examined using the electron microscope. In the in vivo study, mice were divided into five groups: GI (infected untreated), GII (prophylactically treated with Graviola for seven days before infection), GIII (infected and treated with Graviola), GIV (infected and treated with albendazole) and GV (infected and treated with a combination of Graviola plus albendazole in half doses). Drug effects were assessed by adults and larvae load beside the histopathological small intestinal and muscular changes. A significant reduction of adult and larval counts occurred in treated groups in comparison to the control group. Histopathologically, marked improvement in the small intestinal and muscular changes was observed in treated groups. Also, massive destruction of the cultured adults’ cuticle was detected in both drugs. This study revealed that Graviola leaves have potential activity against trichinellosis, especially in combination with albendazole, and could serve as an adjuvant to anti-trichinellosis drug therapy.

Anaphylaxis is the most common allergic reaction triggered by allergens such as insect poisons, food, and drugs through skin contact, injection, or inhalation. In vitro previous research showed that strawberries fruit have activity as antioxidant, anticancer, anti-inflammation, and anti-allergic. The research aimed to determine the antianaphylaxis strawberry fruit extract in mice (Balb/C strain) with ovalbumin-induced.  Twenty-four Balb/C strain mice were divided into six groups (n=4). Group I and II as a normal and control group. Group III till VI as a treatment group was given cetirizine dose 0.042 mg/20 g BW and strawberry extract doses 0,68; 1,36; and 2,72 mg/20 g BW, respectively. This research showed that 70 % of ethanol extract of strawberries fruit have antiallergic activity in response to active cutaneous anaphylaxis. 70% ethanol extract of strawberries doses 2.72 mg/20 g BW had similar antiallergic activity compare with cetirizine. The conclusion of this study showed that strawberries fruit extract could be developed as an alternative medicine to anti-anaphylaxis or anti-allergic.

Background: It is well known that almond and tea is best known to prevent Diabetes mellitus due to its abundant source of polyphenols. Also, probiotics also have been used in the treatment of Diabetes. This study is focused on the combined effect of all these three ingredients through the process of fermentation. Objective: The aim of this present study is to develop, analyse sensory parameters in human volunteers for optimisation and evaluate the antidiabetic efficiency of Fermented Almond milk tea (FAMT) both in vitro and in vivo analysis. Study Design: Development of FAMTàOptimisation of FAMT based on sensory analysis from 25 human participantsà In vitro antidiabetic analysis of FAMT extract à Animal studies. Place and Duration: The research work was conducted during November, 2019 to March, 2020 at the Department of Biotechnology, Sri Venkateswara College of Engineering, Post Bag No.1, Pennalur, Sriperumbudur Tk, Kancheepuram Dt, TN-602117, India. Materials and Methods: FAMT was prepared by optimisation of different formulation based on sensory analysis recorded from 25 healthy human volunteers. The FAMT extract was prepared and was used for the in vitro analysis and phytochemical screening. The animal study was performed with 30 Albino Wistar rats which were divided into 5 groups under preventive regimen. Group I was healthy normoglycemic control group. Group II served as positive control. Group III received metformin (350 mg/kg bw, p.o) for 28 days. Group IV received 5% Fermented almond milk for 28 days. Group V received 5% FAMT for 28thday. All groups except Group I received single dose of STZ (50 mg/kg bw, i.p) on the 29th day for the induction of Diabetes mellitus. After 7 days from induction, animals were anaesthetized and blood was drawn for the evaluation of plasma glucose and serum TG, cholesterol & insulin.   Results: It was observed that FAMT (8:2) was favoured by the participants more than other formulations. FAMT was found to contain Saponins, flavonoids and phenol. The total poly phenol of FAMT (373 ± 3.0 μg/ml) was high than Fermented almond milk (232.5 ± 2.50 μg/ml). The DPPH scavenging, α-amylase and α-glucosidase inhibiting percentage of FAMT (59 ± 4%,52 ± 3%, 50 ± 4% respectively) was high when compared to fermented almond milk (32 ± 2%,34 ± 2% and 45 ± 2% respectively). From animal studies it was significantly observed that plasma glucose (P<0.0001) was reduced, serum insulin (P<0.001) was increased, serum TG (P<0.0.001) and cholesterol (P<0.01) were reduced when compared to Positive control Group- II. Conclusion: Thus, FAMT was proved to act as a prophylactic anti-diabetic drink and was more potent than normal fermented almond milk.

Taurine (2-aminoethane-sulfonic acid) is a type of amino acids and has numerous physiological and therapeutic functions, including anti-inflammation. However, there are few studies on the anticancer action of taurine. Our previous studies have demonstrated that taurine exhibits an apoptosis-inducing effect on human nasopharyngeal carcinoma cells in vitro. In this study, we have investigated whether taurine has an anticancer effect, using azoxymethane (AOM)/sulfate sodium (DSS)- induced mouse model for colon carcinogenesis. All mice, except those in control group, received a single intraperitoneal injection of AOM and DSS in the drinking water for 7 days twice, with 1-week interval. After the first DSS treatment, mice were given distilled water (model group) or taurine in the drinking water (taurine group) ad libitum. No tumor was observed in the control group. Taurine significantly suppressed AOM+DSS-induced tumor formation. Histopathological examination revealed AOM/DSS treatment induced colon cancer in all mice (8/8, 100%), and taurine significantly inhibited the progression of colon cancer (4/9, 44.4%). Taurine significantly attenuated cell proliferation in cancer tissues detected by Ki-67 staining. Taurine significantly increased the levels of an apoptosis marker cleaved caspase-9 and tumor suppressor protein PTEN. This is the first study that demonstrated that taurine significantly reduced carcinogenicity in vivo using AOM/DSS-induced colon cancer mouse model.

Background and objectives: Arabinoxylans (AX) can gel and exhibit antioxidant capacity. Previous studies have demonstrated the potential application of AX microspheres as colon-targeted drug carriers. However, the cytotoxicity of AX gels has not been investigated so far. Therefore, the aim of the present study was to prepare AX-based particles (AXM) by coaxial electrospraying method and to investigate their antioxidant potential and cytotoxicity on human colon cells. Materials and Methods: The gelation of AX was studied by monitoring the storage (G′) and loss (G′′) moduli. The morphology of AXM was evaluated using optical and scanning electron microscopy (SEM). The in vitro antioxidant activity of AX before and after gelation was measured using the 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS+), 2,2-diphenyl-1-picrylhydrazyl (DPPH) and ferric reducing antioxidant power (FRAP) methods. In addition, the effect of AX and AXM on the proliferation of human colon cells (CCD 841 CoN) was evaluated using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. Results: The final G′ and G′′ values for AX gels were 293 and 0.31 Pa, respectively. AXM presented spherical shape and rough surface with a three-dimensional and porous network. The swelling ratio and mesh size of AXM were 35 g water/g AX and 27 nm, respectively. Gelation decreased the antioxidant activity of AX by 61–64 %. AX and AXM did not affect proliferation or show any toxic effect on the normal human colon cell line CCD 841 CoN. Conclusion: The results indicate that AXM could be promising biocompatible materials with antioxidant activity.

ABSTRACT Aim This study aimed to assess and compare the fracture resistance of simulated immature teeth reinforced with Biodentine (BD) and mineral trioxide aggregate (MTA) as apical barriers and two root canal backfilling combination (gutta-percha/AH26, MetaSeal). Materials and methods A total of 70 extracted human maxillary incisors were randomly divided into seven groups (n = 10). The positive control group was not instrumented. For the other groups, coronal access was made and root canals were instrumented using the ProTaper, up to F5 followed by six Peeso reamers which were allowed to pass 1 mm beyond the apex to size 6 (1.7 mm) to simulate immature teeth. The apical 4 mm of their root canals was filled with either MTA or BD apical barrier, then backfilled with gutta-percha/AH26 or MetaSeal obturation combination. The negative control group was left unfilled. Composite resin was used to restore the coronal access cavities. The maximum load for fracture of each tooth was recorded utilizing a universal testing machine. Data were analyzed using two-way analysis of variance. Results The noninstrumented group I had the highest fracture resistance and differed significantly (p < 0.05) from the negative control groups. On the contrary, no significant difference was found between BD and MTA groups, regardless of the backfilling combination (p > 0.05). Conclusion There was no difference between MTA and BD apical barriers and the backfilling combination regarding their resistance to root fracture.
