Abstract BackgroundThe esthetic repair of a removable partial appliance is a critical function that determines the success of the therapy. The most challenging challenge is to achieve While maintaining stability, retention, and protect teeth's health, optimal esthetics is achieved. Removable partial prosthesis is an odious therapeutic option that we still have to use in some circumstances and is a common procedure. However, these patients anticipate receiving a prosthesis. ObjectiveTo determine wrought wire clasp for removable partial dentures for esthetic. Materials and MethodsAfter taking impressions (primary and final) for the maxilla or the mandible of many patients, stone castings were pouring in class III (dental stone) using a 30 ml water to 100 g powder ratio. For the installation of prosthetic teeth, a uniform denture base plate wax template was made on each stone cast, and lingual clasps were inserted around the abutment teeth. Dental plaster is used to flake the lower piece of the traditional brass flasks (class II). The denture foundation was meticulously deflasked, after the curing cycle, the surplus is removed and the denture surface is polished. ResultsDentures that have been treated using this method have a high level of retention. Excellent aesthetics, make it more sanitary and dentures with the lingual clasps method are more comfortable for the patient. ConclusionDentures made with this procedure have better retention, a better esthetic, are good hygienic, and are high comfortable for the patient than dentures made using labial clasps.

Inlay-retained dental bridges can be a viable minimally invasive alternative when patients reject the idea of implant therapy or conventional retained full-coverage fixed dental prostheses, which require more tooth preparation. Inlay-retained dental bridges are indicated in patients with good oral hygiene, low susceptibility to caries, and a minimum coronal tooth height of 5 mm. The present study aims to evaluate, through the finite element method (FEM), the stability of these types of dental bridges and the stresses on the supporting teeth, under the action of masticatory forces. The analysis revealed the distribution of the load on the bridge elements and on the retainers, highlighting the areas of maximum pressure. The results of our study demonstrate that the stress determined by the loading force cannot cause damage to the prosthetic device or to abutment teeth. Thus, it can be considered an optimal economical solution for treating class III Kennedy edentation in young patients or as a provisional pre-implant rehabilitation option. However, special attention must be paid to its design, especially in the connection area between the bridge elements, because the connectors and the retainers represent the weakest parts.

Abstract Background Implant-supported removable partial dentures (ISRPDs) are an effective treatment for partially edentulous patients. ISRPDs improve patients’ satisfaction and oral function to a greater extent than RPDs by improving denture stability and enhancing support. However, the effect of a type of direct retainer on displacement of the abutment teeth and dentures in ISRPDs remains unclear. Therefore, we made a resin mandibular model of unilateral mandibular distal-extension partial edentulism for mechanical simulation and compared the dynamic behavior of the abutment teeth and the denture base among different tooth-borne retainers with various rigidities for RPDs and ISRPDs. Methods A resin mandibular model for mechanical simulation that had unilateral mandibular distal-extension edentulism and was missing the first molar, second molar, first premolar, and second premolar, and a denture fabricated from the patient’s computed tomography images were used. Three types of direct retainers with different connecting rigidities were evaluated. The vertical displacement of the denture base and buccal and lingual sides and the mesial displacement of the abutment teeth were measured. Results Regardless of the rigidity of the direct retainers and loading positions, the displacement of the denture bases in the ISRPDs was significantly smaller than that in the RPDs (P < 0.001). There was no significant difference in vertical displacement of the denture bases among direct retainers with various connecting rigidities in the ISRPDs. Conversely, horizontal displacement of the abutment teeth in both the RPDs and ISRPDs tended to be larger with the cone crown telescope, which has high rigidity, than with the cast cingulum rest and wire clasp, which have much lower rigidities. Conclusion Our results suggested that cast cingulum rest and wire clasps as direct retainers are appropriate ISRPDs to minimize denture movement and suppress displacement of the remaining teeth in patients with unilateral mandibular distal-extension partial edentulism.

ABSTRACT Aim The aim of this study is to evaluate the dimensional accuracy of heat polymerized acrylic resin denture base clamped by the conventional method and by new-press technique and cured by long curing cycle. Materials and methods In this study, a total of 60 standardized maxillary record bases were fabricated with seven reference points as follows: • Point A Incisive papilla, • Point B and C Canine region on either side • Point E and G Midpoint of tuberosities on either side • Point F Midpoint of the line joining the two tuberosities • Point D Midpoint between the line joining A and F Group A Ten maxillary record bases were fabricated by conventional clamping method and cured by long curing cycle. Group A’ Ten maxillary record bases were fabricated by New Press or RS tension clamping method and cured by long curing cycle. The distances between the reference points, i.e. A-B, A-C, A-D, D-F, B-E, C-G, E-F, F-G, B-D, D-G, C-D, D-E of all three thermoplastic denture base plates were measured and recorded with the help of travelling microscope and were used for comparison with the measured and recorded readings of processed acrylic denture bases. The data obtained was analyzed by using the one-way analysis of variance and HSD Multiple Comparison Test. Results The overall results of the study indicate that among all the denture bases cured by the two clamping systems and the long curing cycle, group A‘ were the most dimensionally stable, followed by control group A. Conclusion The study concluded that the denture bases fabricated by the New Press method using the long curing cycle would produce the most dimensionally stable denture bases. An attempt was made in the present investigation Mathematical Abilities among Class-III Students. OBJECTIVES: To study the differences between boys and girls on mathematical abilities of class –III students and to examine the differences between government and private school children on mathematical abilities of class-III students. SAMPLE: The sample of the study consists of 120class-III children were purposefully selected form primary schools of Rangareddy district of Andhra Pradesh. Tool: Mathematical abilities test was developed by the researcher. CONCLUSION: There is no signicant difference in their mathematical abilities including all areas. Private school children are high level of mathematical abilities in areas of subtraction, division, basic geometry than government school children. There is no signicant difference in their mathematical abilities in areas of addition and multiplication

Cardiovascular disease (CVD) is the leading cause of death in both men and women and has largely been attributed to genetic makeup and lifestyle factors. However, genetic regulation does not fully explain the pathophysiology. Recently, epigenetic regulation, the regulation of the genetic code by modifications that affect the transcription and translation of target genes, has been shown to be important. Silent information regulator-2 proteins or sirtuins are an epigenetic regulator family of class III histone deacetylases (HDACs), unique in their dependency on coenzyme NAD+, that are postulated to mediate the beneficial effects of calorie restriction, thus promoting longevity by reducing the incidence of chronic diseases such as cancer, diabetes, and CVD. Emerging evidence shows that SIRT1 is ubiquitously expressed throughout the body. Resveratrol, a plant polyphenol, has cardioprotective effects and its mechanism of action is attributed to regulation of SIRT1. Incoproation of resveratrol into the diet may be a powerful therapeutic option for the prevention and treatment of CVD.

Leukemias that harbor translocations involving the mixed lineage leukemia gene (MLL) possess unique biologic characteristics and often have an unfavorable prognosis. Gene expression analyses demonstrate a distinct profile for MLL-rearranged leukemias with consistent high-level expression of select Homeobox genes, including HOXA9. Here, we investigated the effects of HOXA9 suppression in MLL-rearranged and MLL-germline leukemias using RNA interference. Gene expression profiling after HOXA9 suppression demonstrated co–down-regulation of a program highly expressed in human MLL-AML and murine MLL-leukemia stem cells, including HOXA10, MEIS1, PBX3, and MEF2C. We demonstrate that HOXA9 depletion in 17 human AML/ALL cell lines (7 MLL-rearranged, 10 MLL-germline) induces proliferation arrest and apoptosis specifically in MLL-rearranged cells (P = .007). Similarly, assessment of primary AMLs demonstrated that HOXA9 suppression induces apoptosis to a greater extent in MLL-rearranged samples (P = .01). Moreover, mice transplanted with HOXA9-depleted t(4;11) SEMK2 cells revealed a significantly lower leukemia burden, thus identifying a role for HOXA9 in leukemia survival in vivo. Our data indicate an important role for HOXA9 in human MLL-rearranged leukemias and suggest that targeting HOXA9 or downstream programs may be a novel therapeutic option.

ABSTRACTMultidrug-resistantPseudomonas aeruginosapresents a global medical challenge, and polymyxins are a key last-resort therapeutic option. Unfortunately, polymyxin resistance inP. aeruginosahas been increasingly reported. The present study was designed to define metabolic differences between paired polymyxin-susceptible and -resistantP. aeruginosastrains using untargeted metabolomics and lipidomics analyses. The metabolomes of wild-typeP. aeruginosastrain K ([PAK] polymyxin B MIC, 1 mg/liter) and its pairedpmrBmutant strains, PAKpmrB6and PAKpmrB12(polymyxin B MICs of 16 mg/liter and 64 mg/liter, respectively) were characterized using liquid chromatography-mass spectrometry, and metabolic differences were identified through multivariate and univariate statistics. PAKpmrB6and PAKpmrB12, which displayed lipid A modifications with 4-amino-4-deoxy-l-arabinose, showed significant perturbations in amino acid and carbohydrate metabolism, particularly the intermediate metabolites from 4-amino-4-deoxy-l-arabinose synthesis and the methionine salvage cycle pathways. The genomics result showed a premature termination (Y275stop) inspeE(encoding spermidine synthase) in PAKpmrB6, and metabolomics data revealed a decreased intracellular level of spermidine in PAKpmrB6compared to that in PAKpmrB12. Our results indicate that spermidine may play an important role in high-level polymyxin resistance inP. aeruginosa. Interestingly, bothpmrBmutants had decreased levels of phospholipids, fatty acids, and acyl-coenzyme A compared to those in the wild-type PAK. Moreover, the more resistant PAKpmrB12mutant exhibited much lower levels of phospholipids than the PAKpmrB6mutant, suggesting that the decreased phospholipid level was associated with polymyxin resistance. In summary, this study provides novel mechanistic information on polymyxin resistance inP. aeruginosaand highlights its impacts on bacterial metabolism.

Removable dentures aim to improve masticatory function, esthetics and phonetics. Different materials have been used as denture base materials but poly(methyl methacrylate) (PMMA) is most frequently used in most countries. Surface related properties of denture base materials (roughness, surface free energy, wettability, hydrophobicity, etc.) are of clinical importance since they affect plaque accumulation and staining. Surface roughness specifically influences the adhesion and retention of Candida Albicaans, which has a particular importance in denture-induced stomatitis pathogenesis. Bacterial plaque retention directly affects oral hygiene even if the patient achieves an appropriate cleaning of the denture. Ideally, denture base materials should be smooth so plaque adherence is reduced or even avoided. Test specimens of four heat polymerized materials were obtained by a classical press-pack dough molding technique of 1.5 mm thick wax-plate flasked in class III Moldano followed by the recommended polymerization regime. One of the materials was processed through a fast polymerization cycle. Injection-type materials were automatically produced using a dedicated injection unit. All plates were sectioned in 10 mm x 10 mm samples. No finishing protocol was applied. The selected specimens were examined under field emission scanning electron microscope (SEM). A large variation of surface topography was recorded between materials. There were visual differences between the four heat polymerized resins but also between the two injection-type materials. Surface topography was also influenced by the polymerization regime. Within the limitations of our study, we found that surface topography is influenced by the chemical composition of each material, the heat cure cycle and the processing method.

ABSTRACT Intermediate-filament Nestin and group B1 SOX transcription factors (SOX1/2/3) are often employed as markers for neural primordium, suggesting their regulatory link. We have identified adjacent and essential SOX and POU factor binding sites in the Nestin neural enhancer. The 30-bp sequence of the enhancer including these sites (Nes30) showed a nervous system-specific and SOX-POU-dependent enhancer activity in multimeric forms in transfection assays and was utilized in assessing the specificity of the synergism; combinations of either group B1 or group C SOX (SOX11) with class III POU proved effective. In embryonic day 13.5 mouse spinal cord, Nestin was expressed in the cells with nuclei in the ventricular and subventricular zones. SOX1/2/3 expression was confined to the nuclei of the ventricular zone; SOX11 localized to the nuclei of both subventricular (high-level expression) and intermediate (low-level expression) zones. Class III POU (Brn2) was expressed at high levels, localizing to the nucleus in the ventricular and subventricular zones; moderate expression was observed in the intermediate zone, distributed in the cytoplasm. These data support the model that synergic interactions between group B1/C SOX and class III POU within the nucleus determine Nestin expression. Evidence also suggests that such interactions are involved in the regulation of neural primordial cells.

Education in Indonesia is currently undergoing a significant improvement. Particularly in welcoming the revolution era of the 4.0, which is required learning model with high level-thingking  or commonly known as HOTS (Higher Order Thinking Skills). In addition to thinking critically and creatively, problem solving is also included as thinking characters in high-level skills. Solving problems in the learning process trains learners to resolve problems in real life. In fact, not all teachers can simply apply it. Therefore, HOTS learning using the problem based learning model requires strategy for more effective and efficient study.  The research methods used in this study are qualitative descriptive, data collection techniques used by conducting observations, interviews in-depth and document research. The key informant in the study was a class III teacher at SDN Lengkong Bojonegoro, while the informant was a class III student. The data obtained is analyzed using interactive models (data collection, data presentation , reduction, drawing conclusions). The results of this study show that the teacher's strategy in implementing HOTS learning is a good problem based learning model. Visible from the indicators used through deep interview to the key informant and the informant. The obstacles experienced are differences in understanding, characteristics, learning style of students when learning, teachers are required to always be creative and innovative packing learning and the limitations of school facilities and infrastructure.
