Antidiabetic and Hypolipidemic Effects of Extracts of Gymnema Sylvestre in Streptozotocin Induced Type I Diabetes in Rats

Gymnema sylvestre of the family Asclepiadaceae is one of the most important medicinal plants of the central eco-region. It is popularly known as Gurmar, which means “sugar killer”. Extract of leaves is reported to have tannins, gum, flavonoids, proteins and saponins. It has displayed a wide array of pharmacological activities. This study was aimed to investigate the antidiabetic and hypolipidemic effects of Gymnema sylvestre extract in experimentally induced diabetes in rats. Diabetes was produced in adult Wistar rats with single dose of streptozotocin (STZ) @ 60 mg/kg b.wt. intraperitoneally. After the confirmation of diabetes on 7th day (sugar >200 mg/dl), alcoholic and aqueous extracts of G. sylvestre (400 mg/kg) were administered orally to the experimental rats from 8th day and continued for 42 days thereafter. The antidiabetic and hypolipidemic activity was estimated by measuring blood glucose, lipid profile and histopathological examination of various tissues from all the groups. Administration of STZ resulted in a significant (p less than 0.01) increase in blood glucose and lipid profile and histopathological alterations in Diabetic control group as compared to healthy control group. Gymnema treatment demonstrated significant (p less than 0.01) antidiabetic effect indicated by restoration of blood glucose compared to STZ control group. The study concluded that extracts of Gymnema sylvestre improved the altered glucose and lipid profile in diabetic rats, suggesting that the Gymnema Sylvestre extracts exhibit the antidiabetic and hypolipidemic activity.

The study was undertaken to investigate the effects of Spirulina platensis on body weight, blood glucose and lipid profile on alloxan induced diabetic rats. Fifteen Long Evans male rats at six weeks of age were used for the experiment. Diabetes was induced by intraperitoneal injection of alloxan@ 150 mg/kg and experiment was carried out for a period of 6 weeks. Rats were divided into three groups of 5 rats each. The groups were i)healthy control, ii) diabetic control with both receiving rats’ pellet as supplement, and iii) diabetic treated with oral administration of Spirulina platensis  @ 400 mg/kg bwt. Results from the study showed that Spirulina platensis reduced blood glucose significantly (P<0.01) and increased body weight significantly (P<0.05) in contrast to diabetic control group after 6 weeks. Lipid profile analysis with Spirulina platensis @ 400 mg/kg showed that total cholesterol, plasma triglycerides, and LDL were lowered significantly   compared to diabetic groups in contrast to HDL levels which were increased following the use of Spirulina platensis. Our findings showed that oral administration of Spirulina platensis could reduce the adverse effect of alloxan induced diabetes in rats. Based on this research it can be concluded that Spirulina platensis has antihyperglycemic and hypolipidimic effects in alloxan induced diabetic rats.Progressive Agriculture 29 (2): 139-146, 2018

Introduction: The number of diabetes patients are increasing. If the blood glucose is not controlled in diabetics, patients will face with complications. Self-care is important to control the blood glucose of diabetic patients and prevention of complications such as neurological complications, heart, eye disease, amputations and so on. Therefore aim of this study is to identify the effect of Continuous blood Glucose Monitoring or CGM on the blood glucose control in type I diabetes.Materials and methods: The study was done on 20 patients chosen from an online data set available in http://www.jaeb-diabetes.net/ . These patients used CGM to test blood glucose and the result of three month test was measured. To analyze the data, the software SPSS 21, independent Mann- Whitney test was used.Results: The results showed that blood glucose in patients of the experimental group reduced significantly rather than patient of the control group in the second and third month. Also, HbA1C of the patients has reduced.Conclusion: The CGM has an effective role in controlling blood glucose and prevention of complications. Therefore it is recommended that educational programs on the use of monitoring blood glucose, such as CGM as a requirement to be developed.

Background: Therapies for hyperglycemic treatment, including insulin and oral diabetes medications, have been confirmed to cause several side effects. Thus, finding new drugs with fewer side effects is of high importance. Salung leaf herb (Psychotria malayana Jack) reported used in traditional societies as a treatment for diabetes. However, the scientific proof of this plant for diabetes treatment is still lacking.Objective: To evaluate the antidiabetic effect of the P. malayana jack in induced type 1 diabetic rats by assessing blood glucose level and pancreatic cells in white rats.Methods: Alloxan used to induce type I diabetes. Rats randomly divided into six groups. A Group P1 received 250 mg/kg BW; group P2 received 500 mg/kg BW, group P3 received 1000 mg/kg BW. While group 4 basal received no treatment, group 5 received distilled water as a negative control, and group 6 received glibenclamide as a positive control. Medications are given for six days. Glucose levels were measured, and observation of pancreatic Langerhans cell damages.Results:  A decrease in blood glucose levels observed in all treatment groups. The most significant reduction (49.76%; 1000 mg/kg BW) occurred in the P3 group. Morphological features of pancreatic Langerhans cell damage were slightly high in the P1 group.Conclusion: P. malayana Jack can consider having an antidiabetic effect in a type 1 diabetic rat by reducing blood glucose levels.

The prevalence of DM in Riskesdas in 2018 according to the Perkeni consensus in 2015 is higher than according to the Perkeni consensus in 2011, the prevalence was10.9%. The disease can develop into diabetes nephropathy, Increased prevalence of diabetic nephropathy directly proportional with an increase in diabetes prevalence. Diabetic nephropathy is a microvascular complication in diabetics that develops around 30% in patients with type I DM and about 40% in patients with type II DM. Turmeric extract has antioxidant and anti-inflammatory effects to prevent the bad development of diabetes nephropathy. This study looked at the effect of giving a combination of noni and turmeric extract on histopathology of alloxan-induced renal rats. A total of 25 mice were divided into 5 treatment groups, namely the PI group (250 mg / kgBB extract dose), PII group (500 mg / kgBB extract dose), PIII group (750 mg / kgBB extract dose), positive control group (glibenklamid) and negative control group (without extract and glibenklamid). The study used Post Test Only Group. The highest percentage decrease in blood glucose in the PI group was 56.11% and the lowest decrease in the PIII group was 24.12% with p = 0.012. The results of the study were not based on the number of extract doses. The measurement results of rat body weight and glomerular diameter were not affected by blood glucose level with p = 0.700 for body weight and p = 0.187 for glomerular measurement results.

Introduction. More and more researches dedicated to the communication of diseases of the oral cavity of pregnant women with diabetes. It is proved that the intensity of caries and inflammatory periodontal diseases (gingivitis and periodontitis) increase significantly during pregnancy, while the presence of comorbidities, such as diabetes, increase these indexes.Aim. The aim of the work was to study the dental status of pregnant women with diabetes.Materials and methods. The study compared women with gestational diabetes mellitus, type 1 diabetes, and type 2 diabetes, to a control group of pregnant women without diabetes. In addition to clinical research methods, liquid-based cytology of the contents of the gingival sulcus was performed.Results. The results of clinical and laboratory studies have shown that inflammatory diseases of periodontium and teeth within pregnant women with diabetes are more common than within the pregnant women without this disease. It should be noted that the frequency of occurrence and severity of these diseases in the pregnant women with type I diabetes is higher than in the other groups.Conclusion. Pregnant women with diabetes are at risk for dental disease and require more attention from dentists, endocrinologists and obstetricians. The use of liquid-based cytology method helps in the diagnosis of inflammatory periodontal diseases.

Knowledges of certain key moments in the clinical course of diabetic polyneuropathy (DP) combined with varied comorbidity will allow the disease to be identified more effectively and treated comprehensively at different stages of onset. The aim of the research: was to investigate and summarize the features of the clinical picture, electroneuromyographic parameters in patients with DP in the presence of comorbidity. Materials and methods. 111 patients aged 19 to 69 years with DP were examined. The patients were divided into two groups: DP due to type I diabetes mellitus (DM) (group A; n = 61) and type II (group B; n = 50). According to the detected comorbidity, the following subgroups were identified: persons with DP as the result of type I, II DM with only one pathology (subgroup 1; n = 53) and the presence of multimorbidity (two or more pathologies) (subgroup 2; n = 21). The control group – 30 healthy persons representative by age and gender, 37 patients with DP without comorbidity. The patients were examined for neurological status, laboratory tests, instrumental examination methods. Results and discussion. In general, among the studied groups, the lowest nerve conduction velocity in the motor fibers was in abductor hallucis, tibialis on the left, extensor digitorum brevis, peroneus on the left and right, in sensory fibers – peroneus superficialis on the left and right, n. suralis on the left and right. Such changes primarily reflect the lesion of the distal extremities, which clinically looks like a distal symmetrical DP. Was dominated axonal and demyelinating type of nerve fiber damage. Conclusions. Comorbidity contributes to the progression of DP and deterioration of its clinical picture, electroneuromyographic rates, even in the presence of a single pathology, low duration of DM and HbA1c level.

The effect of Protein isolate from fermented melon seeds (Ogiri Protei Isolates; OPI) of Cucumeropsis manii on blood glucose, lipid profile, and antioxidant enzyme activities in streptozotocin (STZ)-induced diabetic rats was investigated. Thirty Male Wistar rats were divided into five equal groups. GThe first control group with no exposure. The second group of rats with Streptozotocin-induced non-treated diabetes. The 3rd and 4th groups of rats with Streptozotocin-induced diabetes supplemented with Ogiri protein isolates (200, 600 mg/kg in diet). And the 5th group of rats with Streptozotocin-induced diabetes administered glibenclamide in a dose 500 ug/kg in diet [17]. The OPI was administered for 6 weeks. The administration of OPI reduced the blood glucose concentration of the STZ-induced diabetic rats. Sera and hepatic superoxide dismutase, activities of the STZ-induced diabetic rats were significantly (P< 0.05) increased in comparison with the diabetic control rats. Lipid peroxidation of the supplemented OPI diabetic rats was significantly (P< 0.05) decreased in comparison with the diabetic control rats as the administration of OPI to the STZ-induced diabetic rats significantly increased the enzymes’ activities. The concentration of low-density lipoproteins in the OPI supplemented rats was significantly elevated. These data demonstrate that OPI supplements might be beneficial for correcting hyperglycemia but the consumption of OPI can modulate some tissue lipids in a direction not beneficial for CVD risk in patients with diabetes.

 Objective: Lawsonia inermis (Linn.) leaves are one of the alternative medicines to treat diabetes mellitus in Indonesia. We investigated the blood glucose level (BGL) of the L. inermis (Linn.) leaves ethanolic extract (LLEE) leaves and evaluated the histopathological alterations in diabetic rats.Methods: This study was an experimental study with posttest - only control group design. Alloxan (120 mg/kg, intraperitoneally)-induced diabetic rats. 35 of Wistar rats (Rattus norvegicus) were divided randomly into five groups, i.e. K: Normal control, P1: Diabetic control, and P2, P3, and P4 (200 mg/kg body weight [BW], 400 mg/kg BW, and 600 mg/kg BW of LLEE, orally) for 28 days. At the end of the treatment, the rats were sacrificed to obtain the kidney for histopathological evaluation using hematoxylin and eosin technique. BGLs were conducted using a glucose meter (GlucoDR).Results: One-way ANOVA test showed that dose 400 mg/kg BW of the LLEE was related to BGL of alloxan-induced diabetic rats (p=0.000). The histopathological of kidney showed glomerular inflammation (GI), epithelial membrane lining degeneration, vascular congestion, and interstitial tubule hemorrhage at diabetic control (P1). Meanwhile, treated with 600 mg/kg BW of LLEE (P4) showed increase cellular regeneration as normal architecture of the kidney.Conclusion: The LLEE at dose 400 mg/kg BW effective decreased BGL and was able to restore the kidney destruction of alloxan-induced diabetic rats at dose 600 mg/kg BW.
