ANTI-ATHEROSCLEROTIC EFFECTS OF SONNERATIA ALBA FRUIT EXTRACT IN ATHEROSCLEROTIC-INDUCED RATS

Objective: The aim of this research was to investigate the potential of Sonneratia alba fruit extract to prevent atherosclerosis formation. Methods: Eighteen male Wistar rats were divided into standard or negative control (S), positive control (P) and treatment (T) groups. Atherosclerosis was induced in groups P and T by orally administering a single dose of Vitamin D3 and a high-fat diet for three days. Sonneratia alba fruit extract was given to the T-group for three days. Lipids were enzymatically measured and foam cells were counted in 10 fields of a microscopic view of the abdominal aorta. Results: This study showed higher cholesterol, LDL, HDL and triglyceride levels in the T-group, compared to the other groups. The average number of foam cells in the S-, P-and T-groups were 11.8±3.3 cells, 21.2±2.2 cells and 11.7±2.9 cells, respectively. Statistical analysis with One-Way Anova showed a significant difference in the average number of foam cells (p = 0.042). Further analysis showed a significant difference in the average number of foam cells in groups S with P (p = 0.041) and P with T (p = 0.024). Conclusion: Sonneratia alba fruit extract showed a potential effect to inhibit atherosclerosis process but could not suppress lipid levels in the blood.

BACKGROUND <br />A high-fat diet can lead to hyperlipidemia which will end up as liver damage (steatohepatitis). Ethanolic Feronia elephantum Correa fruit extract (EFEC) has an antioxidant activity which is expected to overcome hyperlipidemia in the liver. The objective of this study was to determine the effects of EFEC on liver function and morphological changes in rats.<br /><br />METHODS<br />This was an experimental study with a post-test only group design. A total of 20 male Wistar rats aged 2-4 months were randomized into 5 groups, A= negative control, B= positive control (high fat diet + 10 mg/kgBW simvastatin), C = high fat diet + 500 mg/kgBW EFEC fruit extract, D = high fat diet + 600 mg/kgBW EFEC, and E = high fat diet + 700 mg/kgBW EFEC). High-fat diet was given for 4 weeks (quail egg yolks, 10ml/200gBW). EFEC was administered for 4 weeks after induction of hypercholesterolemia. Examination of liver serum glutamic oxaloacetic transaminase (SGOT) and serum glutamic pyruvic transaminase (SGPT) was performed on rat blood serum and histopathological examination was carried out using steatohepatitis grading. One way ANOVA test and Post-Hoc Tamhane’s test were used to analyze the data. <br /><br />RESULTS<br />Administration of EFEC at 700 mg/kgBW improved the liver enzymes (p=0.00 for SGPT and SGOT) and steatohepatitis grading in high-fat diet induced rats (mild condition, E = 75% vs A = 100% mild)<br /><br />CONCLUSION<br />Ethanolic Feronia elephantum Correa fruit extract at 700 mg/kgBW was able to improve steatohepatitis in high-fat diet induced rats.

Background: The process of wound healing is influenced by various factors such as age, hormones, and wound care. Wound care is done to accelerate wound healing which can be done by various methods, one of them is traditional care. Traditional wound care can use medicinal plants. Rhodomyrtus tomentosa is a medicinal plant that has an antioxidant, anti-inflammatory, antitumor and antibacterial content. Thus this study aims to evaluate the effectiveness of the antiseptic solution of the Rodhomyrtus tomentosa leaf extract on wound healing in male Wistar rats. Method: this research is pure experimental research with post test only control group design. Thirty male white rats were divided into five groups, namely negative control, positive control, Rhodomyrtus tomentosa leaf extract 15%, 30%, and 60%. Rhodomyrtus tomentosa leaf extraction was carried out by maceration method with 70% ethano solvent. The extraction results are divided into 3 concentrations (15%, 30% and 60%). The wound healing process was evaluated by measuring the length of the wound manually from 0 to 10 days in each group. Meanwhile, the number of fibroblast cells was calculated through hematoxylin eosin (HE) staining and observed using an Olympus CX41 microscope with a 10x magnification and objective lens magnification in 3 fields. Result: There was a significant difference in the reduction in wound length (p =< 0,000) between the five experimental groups (Rhodomyrtus tomentosa leaf extract solution 15%, 30% and 60%, negative control and positive control. Solution of rhodomyrtus tomentosa leaf extract accelerated the increase in the number of fibroblasts compared to the negative control group (p = 0.003), but did not make a difference (p = 0.403) with the positive control group. Rhodomyrtus tomentosa leaf extraction solution had the same microscopic effect on the number of fibroblasts with a positive control group given 0.9% NaCl solution. Conclusion: There was a significant difference in the number of fibroblasts between all groups, but no difference in wound healing length.

Coronary heart disease (CHD) is the cause of 7,3 million death globally in 2008. The main pathophysiology process that leads to CHD is atherosclerosis which is caused by high LDL cholesterol level. The aim of this study was to explore bay leaf and celery leaf infusion effect on reducing LDL cholesterol level in dyslipidemic rat model. The type of this study was true experimental research using wistar rat (Rattus novergicus) as the sample. The rats were induced by high fat diet and PTU for two weeks then each of the group were given either bay leaf infusion, celery leaf infusion, simvastatin (positive control), or aquadest (negative control) for a week. The measurement of LDL level used Friedewald formulation. Data was analyzed using paired sample t test and one way ANOVA. As the result there was a significance reduction on LDL level after treatment with bay leaf (p=0,013) and celery leaf infusion (p=0,035) but there was no significance difference between groups (p=0,293). It was concluded that bay leaf and celery leaf infusion could reduce the LDL level in dyslipidemic wistar rat but there was no difference on the capability of the two different leafs on reducing LDL level. Dermatophytosis is a fungal infection on skin that one of them caused by Trichophyton rubrum. Dermatophytosis treatment by using chemical drugs has many shortcomings, such as a high cost and a drug resistance. Roselle (Hibiscus sabdariffa L.) calyx content was flavonoid, which have an antifungal effect. Flavonoids on roselle calyx include anthocyanin, gossypeptin (hexahydroxyflavone) 3-glucoside, flavonol glucoside hibiscritin, flavonoid gossypeptin, delphinidine 3-monoglucoside, cyanidin 3-monoglucoside. The objective of this study was to determine the effect of roselle calyx on Trichophyton rubrum growth in vitro. The study was performed as an experimental laboratory. The object of study was T. rubrum. The sample of T. rubrum colonies in this study was taken by a random sampling. The study used T. rubrum colonies on seven Sabouraud Dextrose Agar plates. Each plate had four holes. Each hole was filled by aquadest as a negative control, fluconazole 25 µg/mL as a positive control, and various roselle calyx extract concentrations (10%, 20%, 30%, 40% and 50%). The plates were incubated in an incubator with a temperature of 25oC for 7 days and measured for the diameter of roselle calyx extract inhibition effect. The data were collected and analyzed by One-way ANOVA and Least Significance Difference (LSD) tests on SPSS 16.0 for Windows. The result of One-way ANOVA test showed that there was a difference of inhibition diameter mean among all of the various roselle calyx extract concentration groups (p<0.05). The diameter of roselle calyx extract inhibition effect increased for each concentration up to 50%. The inhibition diameter of positive control compared to 20% roselle calyx extract concentration had no a significant difference. The study was concluded that roselle calyx extract has an antifungal effect to T. rubrum growth in vitro.

Background: Immune responses to eliminate Salmonella infection are by activating macrophage and by producing NO. Chlorophyllin is a chlorophyll derivate that has immunomodulator properties. Objective: The aim of this study was to prove effect of chlorophyllin in macrophage phagocytosis index and NO level. Methods: A post test only controlled group design was conducted in 5 groups Balb/c mice (negative control, positive control, dosage 100 µg/200 g BW, dosage 200 µg/200 g BW and dosage 380 µg/200 g BW). Macrophage phagocytosis index was measured by counting cells that phagocyte latexs particles. NO level was measured by Griess method. Macrophage phagocytosis index difference was analyzed by one way anova and NO level deference was analyzed by Kruskall-Wallis test (α 0,05).Results: Means of macrophage phagocytosis index were 0,7(±0,80), 1,8(±0,80), 2(±0,22), 2,5(±0,43) and 3,2(±0,68) respectively in negative control, positive control, chlorophyllin dosage 100 µg/g BW/day, 200 µg/g BW/day and 380 µg/g BW/day. There was a significant difference of macrophage phagocytosis index between group (p 0,000). Mean of NO level were 0,4 µM(±0,10), 0,6 µM(±0,60), 0,8 µM(±0,64), 0,6 µM(±0,67) and 0,4 µM(±0,26) respectively in negative control, positive control, chlorophyllin dosage 100 µg/g BW/day, 200 µg/g BW/day and 380 µg/g BW/day. There was no difference of  NO level between group (p 0,813).Conclusion: There was a significant difference of macrophage phagocytosis index between chlorophyllin administered group and control. The higher chlorophyllin dosage, the higher macrophage phagocytosis index. Therewas no difference of  NO level between chlorophyllin administered group and control.

ABSTRAK Kurma merupakan salah satu buah dengan kandungan gizi terlengkap. Selain tinggi energi terutama karbohidrat (glukosa, fruktosa), kurma juga mengandung mineral besi yang berperan dalam metabolisme energi. Tujuan penelitian ini untuk mengetahui pengaruh kurma terhadap berat badan tikus (Rattus novergicus). Jenis penelitian eksperimental laboratorik dengan pre-post test with control grup. Sampel adalah 24 ekor tikus putih jantan (Rattus novergicus) strain Wistar, dibagi menjadi 4 kelompok, yaitu kelompok K1 (kontrol negatif), dan K2 (kontrol positif (diberikan suplemen FeSO )), P1 (perlakuan ekstrak air kurma mentah), dan P2 (perlakuan sari kurma). Analisis data dengan uji paired t-test untuk mengetahui perbedaan berat badan pre-post test, ANOVA untuk mengetahui perbedaan berat badan post test antar kelompok, yang dilanjutkan dengan post hoc test LSD, dengan α=0,01. Hasil penelitian menunjukan terjadi peningkatan berat badan pada semua kelompok sampel (K1, K2, P1, dan P2). Peningkatan berat badan tertinggi pada kelompok kontrol positif (K1) 11,12%, diikuti oleh kelompok perlakuan SK (P2) 11,00%, dan perlakuan EAKM (P1) 10,90%, sedangkan yang terendah pada kelompok kontrol negatif (K1) hanya 5,32%. Hasil analisis paired t-Test pada kelompok kontrol positif (K2), perlakuan EAKM (P1) dan SK (P2) menunjukan terdapat perbedaan berat badan pre- post test yang signifikan (p<0,01). Hasil analisis One Way Anova, diperoleh nilai p=0,000 (p<0,01) yang menandakan terdapat perbedaan berat badan post test yang signifikan antar 4 kelompok, di mana post hoc test LSD menunjukan perbedaan terletak antara kontrol negatif (K1) dengan kelompok yang diberikan perlakuan (K2, P1, dan P2), sedangkan diantara kelompok K2, P1, dan P2 tidak ada perbedaan yang signifikan. Kesimpulan: Kurma dapat meningkatkan berat badan tikus yang mengalami anemia defisiensi besi. Pengaruh kurma terhadap berat badan tikus sama dengan pengaruh dari pemberian suplemen FeSO4. Peningkatan berat badan tikus tanpa perlakuan sangat rendah jika dibandingkan dengan peningkatan berat badan tikus yang diberikan kurma dan suplemen FeSO .Kata Kunci: kurma (Pheonix dactylifera L.); ekstrak air kurma mentah (EAKM), sari kurma (SK), suplemen FeSO4, berat badan tikus. ABSTRACT Dates are one of the most nutritious fruits. In addition to high energy, especially carbohydrates, dates also contain iron minerals that play a role in energy metabolism. The purpose of this study was to determine the effect of dates on rat body weight (Rattus novergicus). Types of laboratory experimental studies with pre-post test with control group. The sample was 24 male white rats (Rattus novergicus) Wistar strain, divided into 4 groups, ie group K1 (negative control), and K2 (positive control (given FeSO4 supplement)), P1 (crude dates juice treatment), and P2 (dates juice treatment). Analysis of data with paired t-test to determine the difference of pre-post test weight, One Way Anova to know difference of post test between group weight, followed by post hoc test of LSD, with α = 0,01. The results showed that there was an increase in weight across all sample groups (K1, K2, P1, and P2). The highest weight gain in the positive control group (K1) was 11.12%, followed by the group of dates juice treatment (P2) 11.00%, and crude dates juice treatment (P1) 10.90%, while the lowest in the negative control group (K1) only 5 , 32%. The result of paired t-test analysis in positive control group (K2), crude dates juice treatment (P1) and dates juice treatment (P2) showed significant difference of pre-post test weight (p <0,01). The results of One Way Anova analysis showed that p = 0,000 (p <0,01) indicated that there was significant difference of post test weight between 4 groups, where post hoc test of LSD showed the difference between negative control (K1) and group that given treatment (K2, P1, and P2), whereas between groups K2, P1, and P2 there was no significant difference. Conclusion: Dates can increase the weight of rats with iron deficiency anemia. The effect of dates on body weight of rats is similar to that of FeSO4 supplementation. Increased rats weight without treatment is very low when compared with the increased weight of rats given dates and supplements of FeSO4.   Keywords: dates (Pheonix dactylifera L.); raw dates crude extract, dates juice, FeSO4 supplement, weight of rats.

The aim of this research is to know the ability of meniran ethanol extract (Phyllanthus niruri L.) in inhibiting the growth of Candida albicans. The research design is an experimental study. The extract used was herbal meniran extract made by maceration using 70% ethanol solvent, the content of ethanol extract of meniran tested against Candida albicans using disc diffusion method were 5%, 10%, 20%, 30% and 40%. Activity test conducted on Sabaraound Dextrose agar media that has been smeared with Candida albicans culture which has been standardized with Mc Farland standard. The pre-tested disc paper is first dipped into the extract with various concentrations. Incubated at 30 ° C for 24 hours and the inhibit zone formed was measured. The result showed that herbal ethanol extract of meniran resulted in drag zone diameter at Candida albicans ie 8,5 mm; 10.3 mm; 12.6 mm; 14.1 mm and 14.3 mm, for negative control 0 mm and positive control 20.1 mm. Data obtained from the results of the study were analyzed using one way anova method showed p - value <0,05, it was concluded that there was significant difference between negative control, 5%, 10%, 20%, 30%, 40% and positive control.

Apridamayanti P, Fajriaty I, Etni Hatita E. 2018. Antioxidant activity and analgesic assessment of Lansium domesticum stem bark infusion. Nusantara Bioscience 10: 71-75. Langsat (Lansium domesticum Correa) is empirically used as a medicine for fever and pain. This study intended to determine the effectiveness of stem bark infusion of langsat as an antioxidant and analgesic. Antioxidant activity test was performed by DPPH method (2,2 Diphenyl-1-picrilhydrazil), with infusion at concentration of 1000, 2000, 3000, 4000 and 5000 ppm. Absorbance of the sample was measured using a UV-Vis spectrophotometer. The analgesic test was performed on male Swiss mice by Writhing test using acetic acid 0.6%, CMC-Na induced negative control, Paracetamol 65 mg/kg BW induced positive control, dose I (65 mg/kg BW), dose II (130 mg/kg BW) and dose III (195 mg/kg BW). Data was analyzed using One Way ANOVA in SPSS and the percentage of writhing protection at each dose was calculated. The results of phytochemical metabolites screening of stem bark infusion showed the presence of flavonoids, terpenoids/steroids, tannins, phenols and saponins. Antioxidant activity measured by spectrophotometric measurements showed that the antioxidant activity of stem bark infusion was IC50 2820 μg/ml. The percentage of writhing protection on dose I, II, and III was 57.52%, 42.48% and 24.51% respectively, showing a significant difference with negative control at minute 5, 10 and 15 (P <0, 05). There were no significant differences between positive control and dose I, at minute 30 to minute 60 (P> 0,05). The effective dose of stem bark infusion was obtained as an analgesic at dose of 65 mg/kg BW from 30 to 60 minutes.

Buah naga super merah (Hylocereus costaricencis)saat ini dibudidayakan oleh Kebun Benih Holtikultura Baturaden sebagai produk unggulan yang berpotensi meningkatkan kesejahteraan masyarakat Banyumas. Buah naga super merah diyakini memiliki efek anti bakteri oleh karena kandungan polifenol yang terdapat pada daging buah maupun kulit buah naga. Penelitian ini bertujuan untuk melihat daya hambat ekstrak buah naga super merah terhadap pertumbuhan bakteri Streptococcus mutans yang merupakan salah satu pencetus terjadinya penyakit periodontal. Sebagai langkah awal, dilakukan uji determinasi buah naga super merah untuk memastikan bahwa buah naga yang digunakan adalah buah naga super merah. Selanjutnya dilakukan pembuatan ekstrak dengan metode maserasi. Hasil penelitian antar perlakuan menunjukan bahwa tidak terdapat perbedaan daya hambat bakteri yang signifikan (p>0,05) antara ekstrak buah naga super merah, kontrol positif dan kontrol negatif. Penelitian ini menyimpulkan bahwa ekstrak buah naga super merah dapat digunakan sebagai bahan antibakteri dalam terapi periodontal. Hasil penelitian ini diharapkan akan menjadi acuan untuk langkah berikutnya yaitu pengujian efek anti bakteri larutan buah naga super merah terhadap akumulasi plak pada rongga mulut.   Super red dragon fruit (Hylocereus costaricencis) is currently cultivated by the Baturraden Horticultural Seed Garden as a superior product that has the potential to improve the welfare of the Banyumas community. Dragon fruit is believed to have anti-bacterial effects because of the polyphenol content found in fruit flesh and dragon fruit skin. This study aims to see the inhibitory power of super red dragon fruit extract on the growth of Streptococcus mutans bacteria which is one of the triggers of periodontal disease. As a first step, a determination of dragon fruit is determined to ensure that the dragon fruit used is super red dragon fruit. Furthermore, the extract was made by maceration method. The results of the study between treatments showed that there was no significant difference in bacterial inhibition (p> 0.05) between super red dragon fruit extract, positive control and negative control. This study concluded that super red dragon fruit extract can be used as anti bacterial agent for periodontal therapy. The results of this study are expected to be a reference for the next research, analyzing the antibacterial effect of super red dragon fruit solution to the accumulation of plaque on the oral cavity

The incidence of diabetes mellitus in Indonesia continues to increase. There are some complications in the diabetic condition, one of which is diabetic ulcer. Diabetic ulcers in diabetes patient can increase the risk of amputation and expensive treatment costs, so the alternative treatmeant such as Merremia mammosa wich has antiinflamatory and antidiabetic is needed. This study aimed to determine the comparison of the number of macrophage in the incisional wound in hyperglycemic male wistar rats between treatment with Merremia mammosa extract and NaCl. The in vivo test was done by creating wound incision on the mice backs and treated with gentamycin ointment 5% in a positive control group, NaCl in a negative control group and Merremia mammosa extract in a dose of 100m), 200mg, 400mg in treatment group. The result obtained by counting the number of macrophage in histopatholgy examination. The result showed the number of macrophage were 0,36 cells/field of view in a negative control group, 0,52 cells/field of view in a positive control group, 0,48 cells/field of view in all Merremia mammosa treatment groups. The data analysis showed no significant difference with p-value of 0,729. In conclusion, there was no significant difference between the used of Merremia mammosa extract and NaCl on full thickness incisional wounds of hyperglycemic male wistar rats.
