The aim of this study was to detect genes for enterotoxins (hbla, entFM and bceT) and for emetic toxin (cer), to determine antibiotic resistance, and to estimate intraspecies diversity in B. cereus isolates by RAPD analysis. B. cereus was identified in 12 out of 117 indigenous Bacillus spp. using the classical microbiological methods and PCR. All isolates were resistant to penicillin and ampicillin, two to tetracyclin and four to trimethoprim-sulphamethoxazole. Also, all isolates produced inducible penicillinases and ?-lactamase. Toxin genes were detected with PCR. EntFM and cer genes were present in all isolates, hbla in all, but two, and bceT in none. RAPD analysis was performed with four different primers, two of them designed for this study. The intraspecies diversity revealed 10 different patterns at the 90% similarity level. Two separate clusters were formed regardless of a soil type or utilization. The detection of genes encoding toxins in all B. cereus isolates indicated these bacteria as potentially pathogenic and seriously for human health. Regardless of a soil type or utilization, the RAPD analysis showed high intraspecies heterogeneity in B. cereus isolates. To the best of our knowledge, this is the first study to analyse the presence of entero- and emetic toxin genes and genetic heterogeneity in B. cereus isolates from different soil types and different soil utilization in Serbia.

AbstractMany studies report organic carbon stabilization by clay minerals, but the effects of land use and soil type on the properties of humic acids (HAs) are missing. The aim of the paper is to determine the effects of land use and soil types on the characteristics of HAs, which have a considerable influence on organic matter quality. It was hypothesised that the effect of the land use on HAs properties depends on the particular size distribution. The research was performed in three ecosystems: agricultural, forest, and meadow, located in Slovakia. From each of them, the samples of 4 soil types were taken: Chernozem, Luvisol, Planosol, and Cambisol. The soil samples were assayed for the content of total organic carbon (TOC) and the particle size distribution. HAs were extracted with the Schnitzer method and analysed for the elemental composition, spectrometric parameters in the UV-VIS range, and hydrophilic and hydrophobic properties, and the infrared spectra were produced. The research results have shown that the properties of HAs can be modified by the land use and the scope and that the direction of changes depends on the soil type. The HAs of Chernozem and Luvisol in the agri-ecosystem were identified with a higher “degree of maturity”, as reflected by atomic ratios (H/C, O/C, O/H), absorbance coefficients, and the FT-IR spectra, as compared with the HAs of the meadow and forest ecosystem. However, as for the HAs of Cambisol, a higher “degree of maturity” was demonstrated for the meadow ecosystem, as compared with the HAs of the agri- and forest ecosystem. The present research has clearly identified that the content of clay is the factor determining the HAs properties. Soils with a higher content of the clay fraction contain HAs with a higher “degree of maturity”.

Abstract Ceftriaxone resistance in the Enterobacterales is typically the result of production of ESBLs or AmpC β-lactamases. The genes encoding these enzymes are often co-located with other antibiotic resistance genes leading to resistance to aminoglycosides, quinolones and trimethoprim/sulfamethoxazole. Carbapenems are stable to ESBLs and AmpC giving them reliable in vitro activity against producers of these β-lactamases. In contrast, piperacillin/tazobactam and amoxicillin/clavulanate are compromised by co-production of OXA-1, which is not inhibited by tazobactam or clavulanate. These in vitro findings provide an explanation for the MERINO trial outcomes, where 3.7% (7/191) randomized to meropenem died compared with 12.3% (23/187) randomized to piperacillin/tazobactam as definitive treatment of bloodstream infection due to ceftriaxone-resistant organisms. No randomized trials have yet put cefepime and carbapenems head to head, but some observational studies have shown worse outcomes with cefepime. We argue that carbapenems are the antibiotics of choice for ceftriaxone-resistant Enterobacterales.

Bacterial burn is one of the major diseases affecting pear trees worldwide, with serious impacts on producers and economy. In Morocco, several pear trees (Pyrus communis) have shown leaf burns since 2015. To characterize the causal agent of this disease, we isolated fourteen bacterial strains from different parts of symptomatic pear trees (leaves, shoots, fruits and flowers) that were tested in planta for their pathogenicity on Louise bonne and Williams cultivars. The results showed necrotic lesions with a significant severity range from 47.63 to 57.77% on leaves of the Louise bonne cultivar inoculated with isolate B10, while the other bacterial isolates did not induce any disease symptom. 16S rRNA gene sequencing did not allow robust taxonomic discrimination of the incriminated isolate. Thus, we conducted whole-genome sequencing (WGS) and phylogenetic analyzes based on gyrA, gyrB and cdaA gene sequences, indicating that this isolate belongs to the Bacillus altitudinis species. This taxonomic classification was further confirmed by the Average Nucleotide Identity (ANI) and the in silico DNA-DNA hybridization (isDDH) analyzes compared to sixty-five Bacillus spp. type strains. The genome was mined for genes encoding carbohydrate-active enzymes (CAZymes) known to play a role in the vegetal tissue degradation. 177 candidates with functions that may support the in planta phytopathogenicity results were identified. To the best of our knowledge, this is the first data reporting B. altitudinis as agent of leaf burn in P. communis in Morocco. Our dataset will improve our knowledge on spread and pathogenicity of B. altitudinis genotypes that appears as emergent phytopathogenic agent, unveiling virulence factors and their genomic location (i.e., within genomic islands or the accessory genome) to induce trees disease.

ABSTRACT Cattle are a known reservoir of Shiga toxin-producing Escherichia coli. The prevalence and stability of Shiga toxin and/or Shiga toxin genes among native wild ungulates in Idaho were investigated. The frequency of both Shiga genes and toxin was similar to that reported for Idaho cattle (∼19%).

During the past 20 years, great landscape changes took place in the northwest of China. Landscape change resulted in soil type transformations. This paper discusses the changes and fractal of soil types in oasis. In order to do it, the soil type maps of Manasi River Basin in 1987 and 2006 were used. 13 types of soil and 2 types of land-use were classified and analyzed in the study area. Results indicated many variations in characteristics. Firstly, all soil types underwent remarkable changes from 1987 to 2006 in the study area: the identified changed area was about 30% or 6506.33 km2. Secondly, in comparison with 1987, in 2006 2/3 of the area's soil types increased, while 1/3 decreased. Rapid expansion of Aquicambids (415.28 km2), and rapid decrease of Petrocambids (797.05 km2) and Aquisalids (415.93 km2) were the noticeable findings. Furthermore, Haplocambids obtained largest gains from other soil types, while Petrocambids lost largest area to other types. Additionally, the fractal relationship objectively existed between the perimeter and area of soil patches. The fractal dimension of Aquisalids, Petrocalcids and Ustifluvents became higher and their shapes became more complex during this period. The stability index was higher in 2006 which indicated that the spatial structure of soil type was more stable than in 1987. These chaotic and occasional changes were largely caused by human activities and natural conditions. Consequently, environmental managers should pay more attention to soil changes in the arid and semiarid region. Santrauka Per pastaruosius 20 metų šiaurės vakarų Kinijoje įvyko didelių kraštovaizdžio pokyčių, lėmusių ir dirvožemio tipų pakitimus. Remiantis 1987–2006 m. Manasi upės baseino dirvožemio žemėlapiais, aptariami dirvožemio tipų pokyčiai ir fraktalai oazėse. Pasirinktoje teritorijoje išskirta ir analizuota 13 dirvožemio tipų ir dvejopa žemėnauda. Nustatyta daug kintamųjų parametrų. Pirma, tirtõs teritorijos visų tipų dirvožemiai nuo 1987 iki 2006 m. žymiai pakito. Nustatytoji pokyčių zona apima apie 30 % teritorijos, arba 6 506,32 km2. Antra, palyginti su 1987 m., 2006 m. 10 dirvožemio tipų teritorija padidėjo, o 5 tipų sumažėjo. Sparčiai padidėjo Aquicambids (415,28 km2), sparčiai sumažėjo Petrocambids (797,05 km2) ir Aquisalids (415,93 km2), pokyčiai buvo žymūs. Iš visų kitų pakitusių dirvožemių tipų Haplocambids plotai padidėjo daugiausia, o labiausiai, palyginti su kitais, sumažėjo Petrocambids plotai. Be to, pastebėta, kad tarp dirvožemio teritorijos plotų ir perimetrų objektyviai egzistuoja fraktalinės sąsajos. Fraktalinės dimensijos Aquisalids, Petrocalcids ir Ustifluvents per minėtą laikotarpį padidėjo, o jų formos tapo sudėtingesnės. Stabilumo indeksas 2006 m. buvo didesnis. Tai rodė, kad erdvinė dirvožemio struktūra mažai pakito, tapo stabilesnė, palyginti nei buvo 1987 m. Šiuos atsitiktinius pokyčius iš esmės lėmė žmogaus veikla ir gamtinės sąlygos. Prieita prie išvados, kad sausojo ar pusiau sauso klimato regionuose kraštotvarkos vykdytojai dirvožemio pokyčiams turėtų skirti daugiau dėmesio.

Introduction. Pseudomonas aeruginosais the most frequently isolated organism as it acts as the opportunistic pathogen and can cause infections in immunosuppressed patients. The production of different types of beta-lactamases renders this organism resistant to many commonly used antimicrobials. Therefore, the aim of this study was to document the antibiotic resistance rate inPseudomonas aeruginosaisolated from different clinical specimens.Methods. Pseudomonas aeruginosarecovered was identified by standard microbiological methods. Antibiotic susceptibility testing was performed by modified Kirby-Bauer disc diffusion method following Clinical and Laboratory Standard Institute (CLSI) guidelines and all the suspected isolates were tested for the production of ESBLs, MBLs, and AmpC.Results.Out of total (178) isolates, 83.1% were recovered from the inpatient department (IPD). Majority of the isolates mediated resistance towards the beta-lactam antibiotics, while nearly half of the isolates were resistant to ciprofloxacin. Most of the aminoglycosides used showed resistance rate up to 75% but amikacin proved to be better option. No resistance to polymyxin was observed. ESBLs, MBLs, and AmpC mediated resistance was seen in 33.1%, 30.9%, and 15.7% isolates, respectively.Conclusions. Antibiotic resistance rate and beta-lactamase mediated resistance were high. Thus, regular surveillance of drug resistance is of utmost importance.

ABSTRACTThe genusExiguobacteriumcan adapt readily to, and survive in, diverse environments. Our study demonstrated thatExiguobacteriumsp. strain S3-2, isolated from marine sediment, is resistant to five antibiotics. The plasmid pMC1 in this strain carries seven putative resistance genes. We functionally characterized these resistance genes inEscherichia coli, and genes encoding dihydrofolate reductase and macrolide phosphotransferase were considered novel resistance genes based on their low similarities to known resistance genes. The plasmid G+C content distribution was highly heterogeneous. Only the G+C content of one block, which shared significant similarity with a plasmid fromExiguobacterium arabatum, fit well with the mean G+C content of the host. The remainder of the plasmid was composed of mobile elements with a markedly lower G+C ratio than the host. Interestingly, five mobile elements located on pMC1 showed significant similarities to sequences found in pathogens. Our data provided an example of the link between resistance genes in strains from the environment and the clinic and revealed the aggregation of antibiotic resistance genes in bacteria isolated from fish farms.

Six samples of fruits (apple, banana, mango, orange, pineapple and watermelon) and six samples of vegetables (cabbage, carrot, cucumber, lettuce, potato and tomato) were purchased from wokem market in Choba town PH, Rivers State and microbiological analysis were carried out these fruits and vegetables samples and the effects of the antimicrobial agents (Ethanol and Hydrogen peroxide) on the microbial load. The Total viable count, Total coliform count and Escherichia count were determined via standard microbiological methods. The Total viable count ranged from 4.1x10106 to 1.0x107 cfu/g Total coliform count from 4.5x107 to 1.5x106 cfu/g nd the E.coli count ranged from 5.1x102 to 1.0x102 cfu/g. The Total viable count for apple ranged between to 5.1 x107 to 2.07x109 , Cabbage 1.45x109 to 4.9x107 , banana 1.10x109 to 5.9x107 , cucumber 1.82x108 to 6.0x107. The microorganisms isolated were Escherichia coli 42%, Staphylococcus aureus 30%, Pseudomonas spp 18%, Bacillus spp,6% and Serratia spp, 2%. Hydrogen perioxide had a high pathogen reduction rate than ethanol.The isolation of E coli and other pathogenic microorganisms indicates the presences of faecal contamination and the poor hygienic production, storage and transport system. Fruits and vegetables should be properly handled from their seeds planted in the farm to the matured ones sold in the market

The destruction of the cotton crop residues (cotton stalks) is a mandatory procedure in Brazil for prophylactic issues, but is a subject unexplored by the research and there are few studies that deal with this issue. However, this is not encouraged in recent decades, studies aimed at developing and evaluating equipment for this purpose. The present study had the objective to evaluate six methods for mechanical destruction of cotton crop residues. Each method was defined based on the principle of operation of the active parts of the equipment, which were tested in medium texture soil and in a clayey one. The variables used to evaluate the efficiency of the equipment were the regrowth rate, the theoretical field capacity and energy demand. The equipment with convergent concave disks (DCC) and flat cutters discs from manufacturer A (CPS-a) showed the best results in cotton stalks destruction in both soil types. The harrow disc (GPD) was efficient only in clay soil. It was concluded that the equipment with convergent concave disks, among those tested, was the most efficient to destroy cotton stalks, regardless of soil type, and that the harrow disc was not included among the best performers.

The results obtained in two coffee fertilizer tests performed with the Puerto Rican variety of Coffea arabica on "Catalina Clay" are presented, statistically analyzed, and discussed. Nitrogen and phosphoric acid applications seem to be of greater importance as regards market-coffee production of the above variety in the soil type used, than are the applications of potash, which had no significant effects on the yields. These results are in sharp contrast with the results obtained by McClelland, who found potash applications to be essential and phosphoric acid applications to be not essential for maximum coffee production in Puerto Rico. It should be noted that McClelland's experiments were carried out on other soil types, which were probably not in condition to provide the coffee trees with their potash requirements.
