The low doses effect of experimental zearalenone (ZEN) intoxication on the presence of Ca2+ in selected ovarian cells from pre-pubertal bitches

Abstract The objective of this study was to determine the effect of 42-day ZEN intoxication on the presence of Ca2+ in selected ovarian cells from beagle bitches, using the potassium pyroantimonate (PPA) method. Samples were collected from 30 clinically healthy, pre-pubertal, genetically homogeneous animals. The bitches were divided into three groups of 10 animals each: experimental group I - 50 μg ZEN/kg BW (100% NOAEL) administered once daily per os; experimental group II - 75 μg ZEN/kg BW (150% NOAEL) administered once daily per os; control group - placebo containing no ZEN administered per os. An electron microscopic analysis revealed that cells died due to apoptosis, depending on the ZEN dose and the type of cells exposed to intoxication. Lower ZEN doses led to apoptosis-like changes in the cells. Cell death was a consequence of excess Ca2+ accumulation in the mitochondria, followed by cell dysfunction and a decrease in or the absence of mitochondrial metabolic activity in oocytes, follicle cells and interstitial cells in experimental bitches.

Research goal: identification of the risk of oncogenesis in the liver under the influence of 2 promoter factors – partial hepatectomy (PHE) performed against the background of superinvasive opisthorchiasis (SO) in Syrian hamsters. Material and methods. SO was modeled by infecting animals (n = 86) with 50 metacercariae of O. felineus. Superinvasions (50 metacercariae) were repeated 6 and 16 days after primary infection. PHE – removal of the middle lobe of the liver – 17.3-17.7% of the organ weight was performed according to the method of G. M. Higgins, K. M. Andersson 16 days after the last superinvasion. The quantitative assessment of the regenerative process after PHE was determined by the coefficients of regeneration completeness (%) by the formula Кпрп = (М1 – М2) / М3 × 100. The histological specimens were stained with hematoxylin and eosin and by the methods of Van Gieson, Slinchenko, Samsonov, Kupriyanov. IHC–reactions were carried out with antibodies to Ki–67, CD31, CD34, CD117, Oct 4, and α–fetoprotein. Electron microscopic analysis was performed on a “JEM – 100CX” microscope (JEOL, Ltd., Japan). The quantitative characteristics were processed using the statistical software Microsoft Excel (2019) and the Statistica package (version 12.6). Results. Analysis of the obtained data indicates that by the 16th day after PHE, 3 zones have formed in the liver: zone A – the stump, zone B – is adjacent to the wound zone A, zone C – is adjacent to zone B. By the 16th day after PHE, expressed proliferation processes of the CD31, CD34, Oct 4, CD117–positive cells and differentiation into endothelial cells, cholangiocytes, hepatocytes with the formation of vessels are noted. At a later date of the experiment, hepatocyte fields with the presence of α–fetoprotein in the cytoplasm, cholangiocellular tubules were formed; at a later date (23 days), ducts lined with a cylindrical epithelium of normal structure were identified. The epithelial lining of the preexisting ducts is multi–row by the end of the experiment; no tumors were found in the liver in the experimental group. An increase in liver mass was noted in the control group (SO) by 8.2%, in the experimental group – 24.7%. Conclusion. The liver with partial hepatectomy against the background of superinvasive opisthorchiasis is an organ with permanent proliferation of CCD and HCD cells, hyperregeneration and a significant increase in mass. In the presence of 2 promoters without initiators, the risk of carcinogenesis does not increase.

Abstract Zearalenone is an estrogenic mycotoxin that often contaminates plant material used in the production of feeds for companion animals. Small daily doses of ingested zearalenone - a competitive substrate modulating the activity of enzymes participating in estrogen biosynthesis at the pre-receptor level - can induce subclinical symptoms of hyperestrogenism in bitches. The objective of this study was to determine the effects of low zearalenone doses on the presence of estrogen receptors in the ovaries of pre-pubertal Beagle bitches. The bitches were divided into three groups of 10 animals each: experimental group I - 50 μg zearalenone/kg body weight administered once daily per os; experimental group II - 75 μg zearalenone/kg body weight administered once daily per os; control group - placebo containing no ZEN administered per os. The animals were ovariorectomized at the end of the experiment, at 112 days of age. Estrogen receptors were detected in ovarian specimens by immunohistochemical methods. The results revealed an absence of estrogen receptors alpha in all groups. In both experimental groups a decrease in the positive response of estrogen receptors beta in specified structures of ovaries was observed. Very low α-zearalenol levels probably attested to the slowing down (hypostimulation) of the biotransformation process. Overall, zearalenone intoxication led to hyperestrogenism during a specific developmental stage of pre-pubertal bitches. As regards hormesis, the threshold dose of zearalenone (adaptive capability) was exceeded in the ovaries of experimental group II animals. The results obtained in both experimental groups suggest that long-term exposure to low-dose zearalenone intoxication decreased the degree of estrogen receptors beta staining in particular structures of ovaries in the experimental bitches, which initiated epigenetic modification mechanisms that inhibited ovarian development.

Objectives: In this paper, the effects of tobacco on aerobic exercise ability and physical fitness recovery of college students were studied. Methods: University group sports intervention form: traditional characteristic project (basketball) + Taiji soft ball (R&D intervention project). Exercise time: 3 times/week; Activity duration: 30min; Activity intensity: the heart rate is controlled at 120-140 beats/min. All the college students in the experimental group are students who have never smoked, and the college students in the control group are students who have smoked for more than two years. The other conditions are the same. Results: The exercise time and endurance of experimental groups I and II were significantly lower than those of the control group, and the cardiopulmonary function was significantly lower than that of the control group. The indexes of experimental group II changed significantly compared with experimental group I, and the difference was statistically significant. Conclusion: Cigarette smoke can significantly reduce the aerobic exercise ability and anti fatigue ability. The longer the smoking time, the more serious the adverse effects. Therefore, tobacco smoke and nicotine will damage college students’ aerobic exercise ability and have a negative impact on the recovery of physical fitness after exercise.

The objective of the present study was to find out the role of emotion in effective teaching of junior college students. The sample for the present research selected from the grantable junior college in Ahmednagar. For the research 11th class 90 students of Commerce faculty was selected. Among them 30 student selected for the positive emotion teaching group (Experimental group I). 30 students selected for the negative emotion (Experimental group II). And 30 students were selected for the neutral group (Control group III). The age range of the students were 17 to 18 years (M=35.00 SD=7.38). In the research Simple random method was used. The first hypothesis of the research was positive emotional teaching method high in English retention than negative and neutral teaching methods. Tolls 11th class English syllabus selected for the study. Result first result was showed that positive emotional teaching had significantly high English retention than the negative and neutral emotional teaching methods.

Bone decalcification is a time-consuming process. It takes weeks and preservation of the tissue structure depends on the quality and velocity of the demineralization process. In the present study, a decalcification methodology was adapted using microwaving to accelerate the decalcification of rat bone for electron microscopic analysis. The ultrastructure of the bone decalcified by microwave energy was observed. Wistar rats were perfused with paraformaldehyde and maxillary segments were removed and fixed in glutaraldehyde. Half of specimens were decalcified by conventional treatment with immersion in Warshawsky solution at 4ºC during 45 days, and the other half of specimens were placed into the beaker with 20 mL of the Warshawsky solution in ice bath and thereafter submitted to irradiation in a domestic microwave oven (700 maximum power) during 20 s/350 W/±37ºC. In the first day, the specimens were irradiated 9 times and stored at 40ºC overnight. In the second day, the specimens were irradiated 20 times changing the solution and the ice after each bath. After decalcification, some specimens were postfixed in osmium tetroxide and others in osmium tetroxide and potassium pyroantimonate. The specimens were observed under transmission electron microscopy. The results showed an increase in the decalcification rate in the specimens activated by microwaving and a reduction of total experiment time from 45 days in the conventional method to 48 hours in the microwave-aided method.

The effect of fiber rich product on child hood obesity studied with 60 sample which were collected randomly among 10-12 years and categorized into 2 groups i.e, experimental group and control group. Experimental group further categorized into three groups i.e, supplementation with exercise (n=15), only supplementation group (n=15), only exercise group (n=15) and control group consists of (n=15).Anthropometric, and diet survey conducted as parameters. A fiber rich product was prepared and supplemented for 2 months to the selected subjects and it consists of whole Bengal gram, whole green gram, green peas, barley and jaggery. Positive results were obtained in three experimental groups. Significant decrease observed inweight, and BMI, Total cholesterol, Triglycerides, LDL, VLDL significant increase observed in HDL in group I II and III. The results were (51.60-48.26kg, 24.7-23.1, 195.2-152.3 mg/dl, 168.2-145.0 mg/dl, 52.2-54.13 mg/dl, 109.4- 69.4mg/dl, 33.6-28.7mg/dl) in group I, In group II the results are (50.3-49.86kg, 23.7-23.4, 168.6-161.0mg/dl, 145.4-129.6mg/dl, 44.2-45.2 mg/dl, 95.3-90.0mg/dl, 29.1-28.3mg/dl).In group III the tesults aere (50.7-49.6kg, 24.5-23.9, 143.2-139.3mg/dl, 139-134.5mg/dl, 38.2-38.7mg/dl 76.8-74.1 mg/dl, 25.6-28.1mg/dl) .From the results it was clear that when compared to group II and III group I has shown better results.

Emotion is the ability of a person to express the state of mind which he is, but when a person is mentally, physically, emotionally, and spiritually unstable results in a psychological change and helpless on situations. The purpose of the study was to nd out Effect of SKY yoga and pranayama on emotion among college girls .To achieve the purpose of the study 45 College girls were selected randomly from Coimbatore as the subjects. The subject's age ranged from 18 to 21 years. . The selected subjects were divided into 3 groups. Experimental Group I under went on sky yoga only & Experimental Group II went on pranayama only for 50 minutes per day, six days a week for 12 weeks. The control group was not given any practice. The pre-test and post-test were conducted before and after the training for all three groups. The data collected from the groups before and after the training period were statistically analysed by using Analysis of Co-Variance (ANACOVA) to determine the signicant difference and tested at 0.05 level of signicance. The result of the study showed that there was signicant improvement in the emotion of the Experimental Groups I, II subjects than the Controlled group. Through the SKY yoga and Pranayama Emotion got controlled. The conclusion was that SKY yoga controlled emotions and got reduced the psychological effects depression and stress etc. among college girls.

Determining which forms of mineral feed additives can increase farm animal productivity is a key area of research. This study assessed the mineral composition of ruminal fluid and the effectiveness of calcium and phosphorus used by animals after the introduction of finely dispersed particles (FDP) of SiO2 (group I) and FeCo (group II) with a hydrodynamic radius of 109.6 ± 16.6 and 265 ± 25 nm, respectively. The deposition and use of calcium and phosphorus in the body of the experimental animals exceeded the control values. In group I, 30.8% more calcium was deposited (p ≥ 0.05), and in group II, the value was 30.3% (p ≥ 0.01). In the experimental groups, the calcium utilization rate was 27.3% higher in group I (p ≥ 0.05), and 28.2% higher in group II (p ≥ 0.01) compared to in the control. Phosphorus deposition was 34% higher (p ≤ 0.01) in experimental group I and 6% higher in experimental group II, compared with the control. Group I had a high utilization rate of phosphorus from the feed (with a 29% difference compared to the control). Comparison of the experimental groups revealed that the introduction of SiO2 FDP promoted an increase in the concentration of silicon, phosphorus, and calcium in the ruminal fluid. The introduction of FeCo FDP was accompanied by a decrease in the concentration of iron and cobalt in the ruminal fluid. Thus, the use of feed additive in finely dispersed form in the diet of animals was accompanied by an increase in the use of calcium and phosphorus by the animal’s body, which is advisable when intensifying milk and meat productivity. In this paper present the results of research of influence of the preparation «Kvatronan-Se» and carboxylates of Se, Cu, Mn, Cr, Ge in different combinations on haematological parameters of blood of experimental animals. Study was carried out on the basis of PC Haleks-Agro. It is located in the village Gul'sk of Novograd Volynskiy district of Zhytomyr region. In the farm to bred of Simmental cows. For the experiment was formed four groups. We took into account age, live weight of 650–700 kg and the average milk yield 5000–6000 kg. Animals is located in identical conditions of feeding and maintenance. Research experiment was carried out on animals couples counterparts.  Preparations introduced animals three times at 10–12 days after insemination. The cows first experimental group were injected complex nanokarboksylativ Ge, Cu, Mn and Cr, second – drug Kvatronan–Se and animals third experimental group was administered a complex composed of Se, Cu, Mn, Cr. On 13 day after insemination in experimental animals were taken blood from tail vein and examined the contents of hematological parameters. Established that injection nanokarboksylativ enhance the fertility of cows in the first and third group, the figure rose to 20% in the second by 40% compared with the control. Research hematological parameters blood  animals shows that after entering content drugs of blood cells had minor changes but remained within the physiological norm. The results show that in  concentration monocyte the second group  was at 5.53% (P > 0.01) higher compared to the control group and 2.3% compared to the first and third. The content of lymphocytes in this group decreased by 26.3% (P > 0.05), 22% and 16% relative to the control, the first and third groups. Also, it should be noted that the hemoglobin in the experimental groups decreased in group I by 7.9% in the II – 3.9% and III by 7.3% compared with the control group. Also, these groups distinguished downward trend eosinophils 1.8%, 1.5%, 2.05%. Then, as the number of segment-nuclear neutrophils contrast, in the first group increased by 18.2% in the second and third at 16.9% comparatively control. The number of monocytes in the second group doslfidniy the suspicion was higher by 66.3% and 0.3% higher than the physiological norm – it may indicate a high immune response.

This paper shows the results of production and chemical compo­sition of cow’s milk on a farm MILKO-HF, Prilep, R. Macedonia with capacity of 100 cows of Holstein-Frisian breed divided into two groups (control and experimental). In control group I which was fed in a standard way of nutrition (ration used on the farm), average daily production of milk per cow ws 22.49 kg with the following chemical composition of milk: average daily content of milk fat is 3.75%, protein 3.57%, lactose 4.65%, nonfat dry matter 9.56% and total dry matter 13.31%. In experimental group II which were fed with the ration according to normatives, average daily production of milk per cow is 24.04 kg with the following chemical composition of milk: average daily content of milk fat is 3.99%, protein 3.57%, lactose 4.65%, non-fat dry matter 9.58% and total dry matter 13.57%. The obtained results show the increase in milk production for 1.55 kg-6.89% and better chemical composition of milk in the experimental group of cows. Thus, dry matter was increased from 13.31% to 13.57%-1.95%, milk fat content from 3.75% to 3.99%-6.4%, nonfat dry matter from 9.56% to 9.58%-0.21%, while the content of protein and lactose stayed on equal level. It was determined that the normed nutrition has influenced on production increase and better chemical composition of milk without unnecessary spent high level of nutrient i.e. nutrition of cows according to recommended normative.
