In every year, Subclinical mastitis (SCM) frequently occurs and results huge economic losses in livestock industry of Bangladesh. This study was redacted to estimate the present status of SCM in cow in selective area of Barisal district. For determining sub-clinical mastitis, a total of 152 milk samples of clinically suspected cows were subjected to White Side Test (WST) and Surf Field Mastitis Test (SFMT). Specifically, the milk samples of each individual cows which were positive to both WST and SFMT considered as SCM indicating 35.52% prevalence. Other than the local breed cows, the cross bred cows showed significantly higher prevalence, is 47.06%. In consideration to age variation, highest prevalence was observed at 5 to 7 years of age in both types where cross breed was 54.71% and 36.67% for local breed cows. Though highest prevalence was recorded as 34.78% in 2nd parity of local breed cows but the cross breed cows showed highest prevalence 54.55% in their 3rd parity. Afterward, non-pregnant cows showed the insignificantly higher prevalence (41.06%) than pregnant cows were 26.32%. Additionally, this study reported that the cross breed cows yielding more than 10 liters of milk (prevalence was 61.54%) were more prone to SCM than the others and also >5 to 10 liters milk producing local breed cows (prevalence was 31.03%) were more susceptible to SCM than others. In a short, this study revealed that high milk yielding cross breed cows are more likely to SCM after their 3rd calving.

Subclinical mastitis (SM) is a major concern in the dairy industry because it causes economic losses and can lead to clinical mastitis. The mechanisms of the onset and progression of SM are not fully understood, and a new procedure for the detection and appropriate prediction of SM leading to clinical mastitis is necessary for dairy cow management. Inflammatory cytokines such as interleukin (IL)-6 are known to be present in inflamed mammary glands at the onset of mastitis, and IL-6 concentrations correlate with the levels of inflammation. In this study, the detection of IL-6 was examined for the evaluation for the future prediction of SM in 77 quarter milk samples from 20 cows. IL-6 concentrations in quarter milk were measured by sandwich ELISA, and the data were compared with milk somatic cell count (SCC) levels to diagnose SM. Average IL-6 concentration was significantly higher in SM quarter milk (207·0±441·6 pg/ml) than in healthy control quarter milk (12·6±33·4 pg/ml, P<0·01). Results of the cross-tabulation table show that SM prediction accuracy based on IL-6 concentration was almost equal or superior to SM prediction accuracy based on SCC. The detection of IL-6 in milk indicated SM earlier than did the detection of elevated SCC. Thus, the detection of IL-6 in milk could be a future prediction marker for SM.

Mastitis is the most prevalent disease of dairy animals, imparting huge economic losses to the dairy industry. There is always a dire need to monitor the prevalence of mastitis, its bacteriology, and evaluation of antimicrobial susceptibilities for mastitis control and prevention. Therefore, the objectives of this study were to investigate: (i) the prevalence of mastitis in cattle and buffaloes; (ii) identification of bacteria associated with mastitis; (iii) antimicrobial susceptibility of bacterial isolates. Milk samples (n = 1,566) from cattle (n = 1,096) and buffaloes (n = 470) were processed for detection of mastitis using the California mastitis test in the year 2018–19. A total of 633 mastitic milk samples were further processed for bacteriology and antimicrobial susceptibility testing by the disc diffusion method. Overall, the prevalence of clinical and subclinical mastitis was 17 and 57% in both species. Clinical mastitis was higher in cattle (20%) compared to buffaloes (11%), whereas subclinical was higher in buffaloes (66%) than cattle (53%). Besides, month-wise prevalence was higher in hot and humid months in both species. Staphylococci spp. (34%) were the most predominant bacterial isolates from mastitic milk, followed by Escherichia coli (19.4%), Streptococci spp. (9%), and Klebsiella spp. (8%). Most of the bacteria were susceptible to gentamicin (92%) and enrofloxacin (88%), when a panel of 16 different antimicrobials was tested. Nevertheless, most of the isolates were resistant to sulphamethoxazole (99%), lincomycin (98%), oxytetracycline (89%), ampicillin (86%), and doxycycline (85%). This study concludes a high prevalence of mastitis caused by Staphylococcal spp. in cattle and buffaloes belonging to the northwest of Pakistan, and gentamicin and enrofloxacin might be appropriate antimicrobial agents in the treatment of bovine mastitis.

Genetic markers associated with inflammatory responses could help in selecting the animals susceptible/tolerant to mastitis. The selective breeding assisted by these markers could help to reduce the huge economic losses that are posed by various forms of mastitis. Onepossible marker is CXCR1, a chemokine receptor that is required for neutrophil migration to infection site. Therefore the present study was planned to identify genetic polymorphism (if any)in CXCR1 gene and associate it with subclinical mastitis in Riverine buffalo of Northern India. For this,two hundred healthy lactating water buffalo were randomly chosenfrom the herds maintained by various farmers in Kapurthala District of Punjab, India.Blood and milk samples of selected buffaloes werecollected. Screening of the animals for sub clinical mastitis was done by SCC and CMT assays of milk samples.Genomic DNA was isolated from blood samples by phenol chloroform method. The DNA of good quality was used for further analysis. PCR-SSCP was used to explore the polymorphism in 311 bp fragment of partial exon2 of CXCR1 gene. The 311 bp fragment of CXCR1 gene was found to be monomorphic in all the DNA samples screened of Indian water buffalo.Int J Appl Sci Biotechnol, Vol 4(2): 228-232

Mycoplasmas are an important and economically significant cause of mastitis in dairy cows in various parts of the world. The organisms are highly contagious, with the main reservoir of infection originating from cows with subclinical mastitis. In 1998 the 1st cases of bovine mastitis due to Mycoplasma bovis were diagnosed in Ardabil State, Iran. An investigation was carried out with the aim of establishing the extent of mycoplasma infections in dairy cows in Ardabil State. Milk samples obtained from 80 cows with clinical mastitis were cultured in the laboratory for the presence of mycoplasmas. Similarly, 48 bulk-tank milk samples were examined for the presence of mycoplasmas. A modified Hayflick broth was used to isolate the mycoplasmas and an immunoperoxidase test used for the species identification of the isolates. Mycoplasma bovis was isolated from 39 (48.75 %) of the clinical mastitis samples and from 48 of the bulk-tank milk samples tested. This indicated that mycoplasma udder infections were more prevalent in dairy cows in Ardabil State than previously thought.

Subclinical mastitis in goats causes economic losses and risks to public health. Given the need for research that shows the most isolated staphylococci species and sensibility tests comparing the resistance between coagulase-negative (CNS) and positive Staphylococcus (CPS) goats with subclinical mastitis, the aim of this study was to identify the microorganisms isolated from milk samples of goats with subclinical mastitis, as well as define the staphylococci species and determine the sensitivity profile of Staphylococcus spp. to antimicrobials. To collect samples, tests were performed for mug of black background and California mastitis test (CMT), collecting milk from CMT positive animals. A total of 226 samples from seven herds of dairy goats was collected and forwarded to the laboratory, where they were seeded for the isolation of the microorganism and implementing the antibiotic sensibility test. Of these, 122 samples had bacterial growth and the most isolated staphylococci species were: S. epidermidis (24.55%), S. lugdunensis (15.40%) and S. intermedius (13.64%). Samples showed increased resistance to antimicrobials: penicillin (81.8%), oxacillin (60.0%) and ampicillin (55.5%). Greater sensitivity to: enrofloxacin (99.1%), erythromycin (98.2%), gentamicin (98.2%) and vancomycin (98.2%) were observed. The S. epidermidis showed higher antimicrobial resistance to amoxicillin and penicillin than S. lugdunensis and S. intermedius. Similar resistance in vitro between CNS and CPS was observed to most antimicrobials. It is important to control the overuse of antibiotics to prevent the emergence of resistant strains.

Mastitis is a major health problem of the udder in dairy sheep breeds. For diagnosis of subclinical mastitis, somatic cell count (SCC) is commonly used. The presence of pathogens in the udder causes the increase of leukocytes and thus SCC in milk. Therefore, the aim of this study was to evaluate the presence of pathogens in the milk of ewes and the possible relationship with SCC. The changes of leukocytes subpopulation in milk samples with high SCC were evaluated as well. The experiment was carried out on a dairy farm with the Lacaune breed. This study was conducted on 45 ewes (98 milk samples) without signs of clinical mastitis. Based on somatic cell count, samples were divided to five SCC groups: SCC1 &lt; 200 000 cells/ml (45 milk samples); 200 000 ≤ SCC2 &lt; 400 000 cells/ml (10 milk samples); 400 000 ≤ SCC3 &lt; 600 000 cells/ml (six milk samples); 600 000 ≤ SCC4 &lt; 1 000 000 cells/ml (six milk samples); SCC5 ≥ 1 000 000 cells/ml (31 milk samples). No pathogens were observed in the majority of milk samples (60.20%). Coagulase-negative staphylococci (CNS) were the most commonly isolated pathogens from the milk of ewes (86.11%). Staphylococcus epidermidis had the highest incidence from CNS (35.48%). In the SCC5 group, up to 79.31% of bacteriological samples were positive. The percentage of leukocytes significantly increased (P &lt; 0.001) in the samples with higher SCC (≥ 200 × 10<sup>3</sup> cells/ml) in comparison to the group SCC1. Also, the percentage of polymorphonuclear cells (PMNs) was significantly higher with increasing SCC (P &lt; 0.001). In conclusion, the presented results showed that the high SCC was caused by the presence of the pathogen in milk. Thus SCC &lt; 200 000 cells/ml and leukocyte subpopulation, especially PMNs, could be considered as important tools in udder health programs applied in dairy ewes.

This study was carried out to determine the prevalence of coagulase-negative staphylococci in clinical and subclinical mastitis in commercial and small-scale farms in Zimbabwe. Thirty five quarter milk samples from clinical mastitis cases and 371 quarter milk samples from cows with subclinical mastitis were cultured for bacterial pathogens. The most frequent pathogens isolated in clinical mastitis were the enteric bacteria (31.4 %), followed by coagulase negative staphylococci (22.9 %) and then Staphylococcus aureus (17.1 %), whereas in subclinical mastitis S. aureus (34.2 %) and coagulase-negative staphylococci were (33.2 %) the most common. Bacillus species were only isolated in milk samples from subclinical mastitis. Coagulase-negative staphylococci were observed in mixed infections with other bacteria in only 2.2 % of the 406 milk samples from clinical and subclinical mastitis where they were isolated together with Bacillus species in 6 of the 9 mixed infection cases. About 95 % of the milk samples from which 131 coagulase-negative staphylococci were isolated had correspondingly high somatic cell counts. The coagulase-negative staphylococci isolated most frequently were S. chromogenes (7.9 %), S. epidermidis (7.4 %) and S. hominis (5.9 %). They were all associated with high somatic cell counts. All the coagulase-negative staphylococci isolates were susceptible to cloxacillin and erythromycin, and more than 90 %of the isolates were susceptible to neomycin, penicillin and streptomycin. The highest resistance was to tetracycline (17.6 %), followed by lincomycin (13.7 %). About 8 % of the isolates were resistant to both penicillin and streptomycin.

Bovine mastitis continues to be a leading cause of heavy economic losses in the dairy industry and a public health hazard globally. This cross-sectional study investigated the prevalence, etiologies of clinical and subclinical mastitis, and associated predisposing factors in Embu and Kajiado counties in Kenya. A semistructured questionnaire was administered to 154 smallholder dairy farmers to collect data on management practices, animal factors, and disease history. A total of 395 dairy cows were initially screened for subclinical mastitis using the California mastitis test (CMT), and milk samples were aseptically collected. Both CMT positive and CMT negative samples were analyzed using conventional bacteriological isolation and identification procedures. In the present study, the overall prevalence of mastitis based on CMT and clinical examination was 80% (316/395), out of which 6.8% (27/395) was clinical mastitis, while 73.1% (289/395) was subclinical mastitis. Based on culture, the overall prevalence of clinical and subclinical mastitis was 51.6% (815/1580), 74.4% (294/395), and 76.6% (118/154) at the quarter, cow, and farm level, respectively. From the 1574 milk samples analyzed by cultured, 1016 bacteria were yielded. The predominant bacteria were coagulase-negative Staphylococcus (CNS), 42.8% (435/1016), and in decreasing order, Streptococcus species, 22.2% (226/1016), Staphylococcus aureus, 15.7% (160/1016), and Pseudomonas aeruginosa, 5.1% (52/1016), and the least was Enterobacter species, 0.7% (7/1016), while 23.7% of the sample yielded no bacterial growth. Risk factor analysis revealed that milking mastitic cows last (p=0.002), using a clean udder drying towel for each cow (p=0.033) and previous history of mastitis (p=0.046) were significantly associated with presence of mastitis. The current study has shown a relatively high prevalence of subclinical mastitis with CNS as predominant bacteria. Therefore, control measures are urgently warranted. Management factors such as milking mastitic cows last, using a clean towel for udder drying for each cow, and culling mastitic cows should be considered and included in the Kenyan mastitis control programs.

Six hundred and thirty clinically-normal milk samples from dairy goat flocks comprising a mixed population of German Alpine, Toggenburg, Saanen and Galla crosses were exam-ined over a 3-month period to determine the prevalence of bacterial organisms. Bacteria were isolated in 28.7 % of the milk samples (181/630) either singly (92.8 %) or in combination (7.2 %). The most prevalent bacterial organisms were Staphylococcus spp. (60.3 %), followed by Micrococcus spp. (17.7 %), Acinetobacter spp. (5 %), Actinomyces spp. (5%) and Streptococcus spp. (1.1 %). The Staphylococcus spp. were mainly coagulase negative (64.3 %). Coagulase-- negative staphylococci and coagulase-positive staphylococci accounted for 37.5 % and 22.7 % respectively of the total bacteria isolated. The isolation of bacteria, some of which are important in clinical and subclinical mastitis, in apparently normal caprine milk, indicates that particular attention should be given to the management of these dairy goat flocks in order to avoid the development of cases of clinical mastitis.

The aim of this study was to isolate yeasts from subclinical mastitic cows and to investigate the slime production by the isolated yeasts. The material used in this study included 339 milk samples from 152 dairy cattle with subclinical mastitis.Milk wasplated onto blood agar, MacConkey agar and Sabouraud dextrose agar. Forty-onesamples (12.1% of total milk samples) were found positive for the yeast by API 20 C AUX identification system. The isolated yeasts were classified into four genera ofCandida, Trichosporon, CryptococcusandSaccharomyces.TheCandidaspecies were following:C. krusei,C. kefyr,C. guilliermondii,C. famata,C. rugosaandC. utulis. Other yeasts were identified asTrichosporon mucoides,T. asahii,Cryptococcus laurentii,C.  neoformansandSaccharomyces cerevisiae. Slime production was tested on Congo red brain heart infusion agar and evaluated according to Congo red phenomenon. Fifteen (36.6%) strains were slime factor positive: seven wereC. krusei, fourC. kefyr, oneC. guilliermondii, oneC. famata, oneT. asahii, and oneC. laurentii. The results of the present study indicate that yeast mastitis is significant for causing economic losses and slime production is mostly found in non-albicans Candidaspecies. Therefore, non-albicans Candidaspecies should be examined for slime production.
