Screening of Antidiabetic Activity of Extract and Fraction of Biophytum sensitivum in Rats Using STZ-Nicotinamide Induced Diabetes Model

The study was aimed at investigate the effect of methanolic extract of Phaseolus semierectus(L.) (Leguminosae) on general mating behaviour, libido, and adverse effects on sexually normal male albino rats. Methanolic extract was administered orally at the dose of 100, 200, and 400 mg / kg, to different groups of male rats (n = 8) once a day for 14 days. All the doses resulted in significant increase in mount frequency, intromission frequency and anogenital sniffing when compared to normal. The methanolic extract of Phaseolus semierectus (L.) seeds at higher concentration (400 mg/kg body weight) showed significant aphrodisiac activity on male Wister albino rats as evidenced by an increase in number of mounts and mating performance. Thus, in experimental rats, the results of the present study suggest that the methanolic extracts of Phaseolus semierectus (L.) exert significant aphrodisiac activity.

Background: Diabetes prevalence is estimated to increase annually. Numerous people use traditional medicine, such as India also considered as the diabetic capital in the world. Diabetes is a metabolic disorder characterized by disturbances in lipid, carbohydrate and protein metabolism. The present study to evaluate the antidiabetic potential of coriandrum sativum. linn fruits methanolic extract in streptozocin induced diabetic wistar albino rats model.Methods: Diabetes induction in wistar albino rats by administration of streptozocin (50mg/kg, i.p.) in citrate buffer. 30 wistar albino rats were divided into 5 groups (A, B, C, D, E). Group A: served as normal control, whereas Group B: diabetic control, Group C, D methanolic coriandrum sativum Linn. fruits extract (CSFME) at a dose of 100, 200mg/kg orally, Group E was given standard drug Glibenclamide (0.5mg/kg) orally. All groups are administered for the period of 14 consecutive days and blood sugar levels was measured at regular intervals up to end of the study.Results: This present research study confirms that the test drug compound CSFME has sustained oral hypoglycaemic activity and statistically significant (p ≤0.05) and which is comparable with standard drug Glibenclamide.Conclusions: This research study confirms that the CSFME has antidiabetic activity against streptozocin induced wistar diabetic albino rats. It could be a novel antidiabetic agent and also a dietary adjunct in the type 2 diabetes management and its complication. Further studies are necessary required to confirm the antidiabetic activity of individual phytochemical compounds of Coriandrum sativum.

This study is aimed at comparing the antidiabetic and antioxidant potential of fenugreek and buckthorn which are commonly used in modulating diabetes in the Middle East. In this study, the antioxidant and antidiabetic activity of the aqueous extracts of the leaf and seed of fenugreek and buckthorn was tested in streptozotocin-induced diabetic male rats fed with a fat-rich diet for 8 weeks. Thirty-six male albino rats were divided into 6 groups ( n = 6 ); the 1st group was the negative control. Diabetes was induced in the other 30 rats using streptozotocin, which were then divided into 5 groups; the 2nd was the untreated positive diabetic group, the 3rd was treated with fenugreek leaf aqueous extract, the 4th was treated with the fenugreek seed aqueous extract, the 5th was treated with buckthorn leaf aqueous extract, and the 6th was treated with buckthorn seed aqueous extract. The positive control group showed an increase in blood sugar, glycated hemoglobin, liver function enzymes, lactate dehydrogenase, kidney indices, total cholesterol, triglycerides, low- and very-low-density lipoprotein, immunoglobulins, and lipid peroxidation and a decrease in high-density lipoprotein, albumin, and antioxidant activity. The histology of the liver and testes showed severe histopathological alterations. Rats of groups 4-6 that were treated with the aqueous extract of the leaf and seed extract of fenugreek and buckthorn showed improvement of all biochemical and histopathological parameters. The seed extract of fenugreek and buckthorn showed more antioxidant activity than their leaves.

Background: Raphia vinifera (Arecaceae) is a medicinal plant commonly used as a sexual enhancer. Objective: To investigate the aphrodisiac potential of aqueous extract (AE) and methanolic extract (ME) of R. vinifera in sexually experienced male rats. Materials and Methods: Thirty male Wistar rats were randomly distributed into six groups (5 rats per group) and administered for 14 days with distilled water (10 ml/kg), sildenafil citrate (1.44 mg/kg), and AE or ME of R. vinifera (100 or 500 mg/kg). The copulatory activity was tested on days 0, 7, and 14 using receptive females. Further, on day 14, rats were sacrificed and biochemical analyses (testosterone, total protein, and acid phosphatase) were performed. Results: Sildenafil citrate significantly decreased the intromission latency (day 14, p = 0.04) and frequency (days 7 and 14, p = 0.03) but increased the mount frequency (day 14, p = 0.04), compared with control. Remarkably, R. vinifera enhanced the sexual activity by significantly decreasing the intromission latency (AE and ME, 500 mg/kg, day 14, p = 0.04) and increasing the mount frequency (AE and ME, 100 mg/kg, day 7, p = 0.02) compared with control. Moreover, R. vinifera improved plasmatic (AE, 100 mg/kg, p = 0.03; AE, 500 mg/kg, p = 0.001; ME, 100 mg/kg, p = 0.01) and testicular (AE, 100 mg/kg, p = 0.001; AE, 500 mg/kg, p = 0.01; ME, 100 mg/kg, p = 0.001; ME, 500 mg/kg, p = 0.01) testosterone levels as well as plasmatic total proteins concentration (ME, 500 mg/kg, p = 0.04). Conclusion: These findings showed that R. vinifera possesses an aphrodisiac property which could further justify its folkloric use in traditional medicine as a sexual enhancer.

This study was designed to evaluate the effects of Moringa oleifera (L) aqueous seed extract on aphrodisiac, gonadal and epididymal sperm reserves of Wistar rats. Twenty-five male and fifteen female Wistar rats aged two months weighing 150 – 200 g were purchased and housed in cages at the Faculty of Pharmaceutical Science, Ahmadu Bello University Zaria. The Wistar rats were provided with a 12 hours light and dark cycle, fed with pellets of broiler starter and drinking water were provided ad libitum. The rats were acclimatized for 14 days and they were randomly divided into 5 groups A, B, C, D and E. Group B, C and D as treatment groups, whereas, group A and E were negative and positive controls, respectively, with 5 rats in each group and each was kept singly in separate cage. Groups A and E received 1 ml of distilled water and 5 mg of sildenafil citrate orally respectively. Groups B, C and D received suspension of Moringa oleifera aqueous seed extract orally at the dose rate 100, 200 and 300 mg/kg respectively, between 9:00 - 10:00 am daily for 21 days. Female rats were paired with males at a ratio of 1:1, and mating behaviour recorded. Group C and E male rats showed a significant (p < 0.05) increase in mounting frequency (MF), respectively. Intromission frequency (IF) was significantly (p < 0.05) increased in group C and E, respectively. Gonadal and epididymal sperm reserves were significantlydifferent (p < 0.05)between the M. oleifera treated and control groups.

Background: This study was designed to evaluate the effect of sesbania grandiflora linn leaf extracts on diuretic activity in wistar rats.Methods: Healthy male wistar albino rats divided into 9 groups and weighing 150-200gm.diuretic activity was expressed by Natriuretic activity, saluretic activity, carbonic anhydrase inhibition, Diuretic index or Diuretic action, saluretic index, Natriuretic index. Animalgroups are divided into: group1 treated as control, 2and 3 treated as standards, group 4, 5 and 6 treated with aqueous leaf extract of Sesbania grandiflora Linn. in different doses of 150,300,450mg/kg. Group 7,8and 9 treated with methanolic leaf extract of Sesbania grandiflora Linn. Doses of 150,300,450mg/kg.Results: No change in body weight group treated with AE-150 (p>0.05) with AE-150 (0.1%), increase in urine volume with increasing doses both with aqueous and methanolic extracts, sodium excretion with methanolic extract was high compared with the aqueous extract, significant increase in potassium excretion in both aqueous and methanolic extract treated groups.All the statistical tests were done by using SPSS version 6. The level of significance was 5% (p≤0.05).Conclusions: All the electrolytes concentration was increased in the urine, hence excluding osmotic diuretic action. Diuretic index, saluretic activity, saluretic index and natriuretic index of ME-450 were more than the value of hydrochlorothiazide hence excludes thiazide diuretic action. But these values were less than furosemide.

Background: Diabetes is associated with chronic hyperglycaemia, long-term damage, dysfunction, and organ failure. This study aims to evaluate the antidiabetic activity of the twigs of Andrographis paniculata and its toxicological markers on Streptozotocin (STZ)-induced diabetic Albino rats. Methods: A total of thirty rats were randomly divided into five groups of six animals each. Non-diabetic animals were treated with distilled water as non-diabetic sham control group 1, while diabetic animals (group 2, 3, 4 and 5) were treated with 60 mg/kg bw STZ intravenous (iv) and 100 mg/kg body weight (bwt) of metformin orally for group 2, distilled water for group 3, and 250 and 500 mg/kg bwt of Andrograhis paniculata (A. paniculata) for groups 4 and 5, respectively. The animals were dosed for 28 days, after which they were sacrificed. Liver and kidney function tests as well as livid profile tests were used as the biomarkers of toxicological assessment. Fasting blood glucose was carried out weekly. Oral Glucose Tolerance Test (OGTT) was conducted on the 28th day of the antidiabetic assessment. Results: A. paniculata groups 4 and 5 were significant at different doses (p < 0.05) in reducing the blood glucose level in comparison with metformin. There were significant changes in total and direct bilirubin, total protein, potassium, triglyceride and inorganic phosphorus in 500 mg/kg bwt of the treated group in comparison with the metformin and diabetic group groups. A. paniculata at 500 mg/kg bwt is most effective for its antidiabetic and organ protecting effects.

This study was conducted to compare the ameliorative effect ofNigella sativaand propolis methanol extract onstreptozotocin-induced diabetic male rats and treating diabetic nephropathy. Forty male Albino rats were divided into four groups; the first group was the negative control fed standard diet. The other 30 rats were injected withstreptozotocin to inducediabetes by a single intravenous injection and then divided equally into three groups; the second group was the positive diabetic control; the third and the fourth groups were treated orally with 20% w/wNigella sativaseeds methanol extract and propolis methanol extract (20% w/w), respectively. The rats of the second group showed increased glucose levels and lipid peroxide accompanied with reduction in superoxide dismutase, catalase, and glutathione-S-transferase enzyme activities compared with the negative control. Carboxymethyl lysine, interleukin-6, and immunoglobulins were also increased as a result of diabetes. Kidney function parameters were also elevated, while potassium and sodium levels were decreased. Moreover, tissues of kidney and pancreas showed severe histopathological changes. Treating the diabetic rats withNigella sativaand propolis methanol extract in the third and fourth groups, respectively, ameliorated all altered biochemical and pathological examinations approaching the negative control. Propolis was more effective thanNigella sativa.

Experiment on the mating behaviour of male albino rats paired with receptive feamles established that in comparison with control animals, males under the influence of ginseng (a) began ejaculation earlier and repeated the action more often in a 45-minute observation period and (b) deposited more copulation plugs in 10 days. The data suggest that ginseng facilitates the mating behavoir of male rats.
