Actinomycetes are found in soils with loose, humus, dry characteristics and around plant roots. Actinomycetes produce secondary metabolite compounds as antibacterial. Escherichia coli, Pseudomonas aeruginosa and Salmonella typhi are Gram negative bacteria that can cause infection in humans. This study aims to determine the ability of the Actinomycetes metabolite filtrate from Bogor Botanical Gardens as an antibacterial agent against the growth of E. coli, P.aeruginosa and S. typhi in vitro. This study used an experimental design with samples of Actinomycetes isolats originating from the Bogor Botanical Gardens soil using the disc diffusion method on Muller Hinton Agar (MHA) media by looking at the clear zone of bacterial growth around the disc paper. Of the three concentration groups, namely 50%, 60%, and 70%, the largest average inhibition zone is found at a concentration of 70% with the average for the three test bacteria E. coli, P.aeruginosa and S. typhi respectively: 4.23 mm; 3.0 mm and 8.43 mm. The results of the Kruskal - Wallis test with p value = 0.01 showed that there was an effect of the Actinomycetes metabolite filtrate on the growth of the three tested bacteria as antibacterials.

Pneumonia is a lung parenchymal infection caused by Pseudomonas aeruginosa.It is Gram negative bacteria that have developed antibiotic resistance. Actinomycetes are Gram-positive bacteria that produce secondary metabolites which have the ability as antimicrobial. Objectives: To identified the ability of Actinomycetes isolates to inhibit the growth of the bacterium Pseudomonas aeruginosa ATCC 27853. The samples in this experiment were from Kebun Raya Bogor that had been rejuvenated on Starch Casein Agar (SCA). Methods: Six dilution series 10-1; 10-2; 10-3; 10-4; 10-5; 10-6 Actinomycetes isolates were used to observe the inhibition zone of P.aeruginosa growth on Mueller Hinton Agar (MHA) media by diffusion method. Results: The effective incubation time occurred at 24 hours, and then it resulted in the average clear zone diameter of 14.70 mm, 10.57 mm, 8.53 mm, 8.47 mm, 6.97 mm, and 5.30 mm. The results of the One – Way Anova test with p-value = 0.000 (p < 0.005) showed some differences at each concentration to inhibit the growth of P.aeruginosa ATCC 27853 at 24 hours incubation period. Conclusion: The most effective concentration of Actinomycetes isolates that can potentially be antibacterial was the concentration of 10-1 with potential solid inhibitory power.Keywords: Actinomycetes, antibacterial, Pseudomonas aeruginosa

Background: Topical fluoride products such as varnishes prevent dental caries by limiting demineralization and antibacterial properties. The structural and metabolic characteristics of cariogenic microorganisms are different from each other. Also, the formulation properties, concentration, and release behavior of fluoride vary in different varnishes. Objectives: In this study, we investigated the antibacterial effects of three types of common fluoride varnishes against two cariogenic bacteria of Streptococcus mutans (S.M.) and Lactobacillus acidophilus (L.A.). Methods: In this in vitro study, antibacterial effects of MI varnish [containing calcium phosphopeptide-amorphous calcium phosphate (CPP-ACP)], Polimo (containing xylitol), and FluoroDose varnishes were evaluated using disc diffusion method via measuring the diameter of the inhibition zone. The antibiotics of erythromycin and ampicillin were used as the positive control of the test. Analysis of variance (ANOVA) and post hoc tests were applied to compare the mean of non-growth zone diameter in the studied groups. Data were analyzed by SPSS 16. The statistical significance level was determined as P-value < 0.05. Results: The antibacterial effect of each varnish was optimal against both bacteria. In comparison, the mean diameter of the inhibition zone in MI varnish was significantly (P-value=0.019) higher than Polimo and FluoroDose brands on S.M.; however, this difference was not statistically significant for L.A. Furthermore, Polimo and FluoroDose varnishes showed similar antimicrobial effects against these bacteria. Conclusions: The use of these varnishes seems to be suitable for preventing tooth decay. MI varnish is preferable because of its higher antibacterial properties.

Diarrhea is still one of the world’s major health problem especially in developing country. Foods and beverages contaminated by microorganism become the risk factor of diarrhea, including Escherichia coli (E. coli). One of the curative effort that can be done is to utilise the secondary metabolite compounds contained in binahong leaf (Anredera cordifolia (Ten.) Steenis). This study used true experimental method. The sample was binahong leaf extract with concentration of 25%, 50, and 75% using diffusion method (Kirby-Bauer) on doublelayered Mueller Hinton Agar (MHA) (base layer and seed layer) in order to measure the growth inhibition zone around the cylinder plate. The amount of population on this study was 24 isolations of E. coli. The repetition of each treatment group was counted by Federer formula. The result analyzed using Kruskal-Wallis test and showed that there was difference on binahong leaf extract effectiveness (p < 0,05), as well as on post hoc analysis using MannWhitney test showed that there was significant difference at concentration of 25%,50%, and 75%. (p < 0,001). This shows that there is a significant differences in the concentration of binahong leaf extract.

Sponge samples collected around Manado waters were obtained 30 species and their crude extracted have been tested in vitro for their activity in inhibiting bacterial growth. Based on the results of antibacterial screening in 30 sponge extracts, there were 23 sponge extracts which had bioactivity in inhibiting the growth of S. aureus, E. coli, S. saprophyticus and P. aeruginosa, Sponge extract No. 43 (of 30 sponge extracts tested) was the most active in inhibiting bacterial growth and had the widest inhibition zone diameter.Keywords: screening, sponge, crude extract, antibacterial, ManadoABSTRAKSampel spons dikoleksi di sekitar Perairan Manado sebanyak 30 jenis/spesies, dimana ekstrak kasarnya telah diuji secara in vitro aktivitasnya dalam menghambat pertumbuhan bakteri. Diarrhea is defecation with a frequency more often than usual (three times or more) a day (10 mL/kg/day) with a soft or liquid consistency, even in the form of water alone. Pathogenic bacteria, such as Escherichia coli, Salmonella typhimurium, and Shigella sp., play a role in many cases, to which antibiotics are prescribed as the first-line therapy. However, since antibiotic resistance cases are often found, preventive therapies are needed, such as consuming yogurt, which is produced through a fermentation process by lactic acid bacteria (LAB). This research aimed to determine the activity of lactic acid bacteria (Liactobacillus bulgaricus and Streptococcus thermophilus) in yogurt in inhibiting the growth of the pathogenic bacteria E. coli, S. typhimurium, and Shigella sp. The research applied in vitro with the liquid dilution test method and the true experimental design research method with post-test-only and control group design. The design was used to see the inhibitory effect of yogurt LAB on the growth of E. coli, S. typhimurium, and Shigell sp. to compare the effect of several different yogurt concentrations, namely 20%, 40%, 60%, and 80%. The results of the Least Significance Different analysis showed that there was a significant difference between yogurt with a concentration of 0% and that with various concentrations in inhibiting the growth of E. coli, S. typhimurium, and Shigella sp. with a p-value of &lt;0.05. Whereas, there was no significant difference in the various concentrations of yogurt in inhibiting the growth of the three kinds of bacteria with a p-value of &gt; 0.05.<p class="Default" align="center"> </p>

Se evaluó la actividad antibacteriana in vitro de aceites esenciales de diferentes especiesdel género Citrus frente a cepas ATCC de Staphylococcus aureus, Staphylococcus epidermidis,Klebsiella pneumoniae, Pseudomonas aeruginosa y Escherichia coli, determinandola concentración mínima inhibitoria (CMI) y la concentración mínima bactericida(CMB). Las bacterias se replicaron en medios de agar y caldos específicos. Se determinóel momento de máxima densidad óptica (DO620) para emplearlo como tiempode incubación; luego se hicieron pruebas de evaluación de sensibilidad con la exposiciónde las cepas a concentraciones a 1000 g/mL del extracto en caldo. Para solubilizarse empleó dimetilsulfóxido (DMSO) al 1%. Posteriormente, se le determinó laconcentración mínima inhibitoria mediante metodologías de microdilución en caldoy la concentración mínima bactericida. Encontrándose una actividad de los aceitesesenciales del género Citrus, con valores de CMI ≥ 600 mg/mL frente a S. aureus,S. epidermidis, K. pneumoniae, P. aeruginosa y E. coli. En función a los resultados obtenidos,se concluye que las diferentes especies del género Citrus son consideradas comopromisorias para el control del componente bacteriano.

Limited data are available on susceptibilities of these organisms to some of the recently made accessible antimicrobial agents. The in vitro activities of newer antibiotics, such as, ceftolozane/tazobactam (C/T) and ceftazidime/avibactam (CZA) along with some “older” antibiotics, for example fosfomycin (FOS) and colistin (CL) were determined against selected strains (resistant to ≥ 3 antimicrobial agents) of Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa. Minimum inhibitory concentrations (MIC) were determined by Clinical and Laboratory Standards Institute microbroth dilution. 133 isolates: 46 E. coli, 39 K. pneumoniae, and 48 P. aeruginosa were tested. Results showed that E. coli isolates with MIC50/90, 0.5/1 μ g / mL for CL; 4/32 μ g / mL for FOS; 0.25/32 μ g / mL for C/T; 0.25/8 μ g / mL for CZA, exhibited susceptibility rates of 95.7%, 97.8%, 76.1%, and 89.1%, respectively. On the other hand, K. pneumoniae strains with MIC50/90, 0.5/1 μ g / mL for CL; 256/512 μ g / mL for FOS; 2/128 μ g / mL for C/T; 0.5/128 μ g / mL for CZA showed susceptibility rates of 92.3%, 7.7%, 51.3%, and 64.1%, respectively. P. aeruginosa isolates with MIC50/90, 1/1 μ g / mL for CL; 128/128 μ g / mL for C/T; 32/64 μ g / mL for CZA presented susceptibility rates of 97.9%, 33.3%, and 39.6%, respectively. Higher MICs were demonstrated against most of the antibiotics. However, CL retained efficacy at low MICs against most of the isolates tested.

Urinary tract infections (UTIs) are among the most common infections requiring medical attention worldwide. The production of biofilms is an important step in UTIs, not only from a mechanistic point of view, but this may also confer additional resistance, distinct from other aspects of multidrug resistance (MDR). A total of two hundred and fifty (n = 250) Escherichia coli isolates, originating from clean-catch urine samples, were included in this study. The isolates were classified into five groups: wild-type, ciprofloxacin-resistant, fosfomycin-resistant, trimethoprim-sulfamethoxazole-resistant and extended spectrum β-lactamase (ESBL)-producing strains. The bacterial specimens were cultured using eosine methylene blue agar and the colony morphology of isolates were recorded. Antimicrobial susceptibility testing was performed using the Kirby–Bauer disk diffusion method and E-tests. Biofilm-formation of the isolates was carried out with the crystal violet tube-adherence method. n = 76 isolates (30.4%) produced large colonies (>3 mm), mucoid variant colonies were produced in n = 135 cases (54.0%), and n = 119 (47.6%) were positive for biofilm formation. The agreement (i.e., predictive value) of mucoid variant colonies in regard to biofilm production in the tube-adherence assay was 0.881 overall. Significant variation was seen in the case of the group of ESBL-producers in the ratio of biofilm-producing isolates. The relationship between biofilm-production and other resistance determinants has been extensively studied. However, no definite conclusion can be reached from the currently available data.

An exploratory study based on laboratory experiment was carried out to determine the antibacterial effect of Dimethyl sulfoxide (DMSO) extract of Aloe vera leaf gel (DAE) against standard strains of Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumoniae in the Department of Pharmacology & Therapeutics in collaboration with the Department of Microbiology, Mymensingh Medical College, Mymensingh, Bangladesh. DMSO extract was used in five different concentrations (100, 200, 300, 400 and 500 μg/ml). Dose dependent inhibitory effect was seen against the test organisms using disc diffusion method. Zone of inhibition (ZOI) were 8 mm, 13 mm, 15 mm, 16 mm and 21 mm against S. aureus; 0 mm, 8 mm, 13 mm, 15 mm and 18 mm against P. aeruginosa; 8 mm, 11 mm, 13 mm, 16 mm and 20 mm against E. coli; 0 mm, 9 mm, 12 mm, 14 mm and 18 mm against K. pneumoniae at 100, 200, 300, 400 and 500 μg/ml respectively. The minimum inhibitory concentration (MIC) was assessed by broth dilution technique. The MICs of DAE for S. aureus, P. aeruginosa, E. coli and K. pneumoniae were 300 μg/ml, 400 μg/ml, 400 μg/ml and 450 μg/ml respectively. From the study it was observed that DMSO extract of Aloe vera leaf gel possesses antibacterial effect against the test pathogens. The findings highlight the need for further extensive study to detect and isolate the biologically active ingredients present in the Aloe vera leaves which are responsible for antibacterial effect. Мета роботи. Визначення антимікробної активності бедринцю ломикаменевого підземних органів екстракту густого. Матеріали і методи.  Oб’єктoм для дoслідження обрано бедринцю ломикаменевого підземних органів екстракт густий (БЕГ-1, одержаний екстрагуванням 75 % етанолом і БЕГ-2, екстрагент 85 % етанол). В якості тест-культур використовували 5 музейних штамів: грампозитивні мікроорганізми Staphylococcus aureus АТСС 6538, спорову культуру Bacillus subtilis АТСС 6633, грамнегативну культуру Escherichia coli АТСС 25922 та Pseudomonas aeruginosa АТСС 9027. Антифунгальну дію з’ясовували відносно Candida albicans АТСС 885-653. Бактеріостатичні властивoсті дoсліджуваних oб’єктів встанoвлювали за результатами росту еталонних штамів мікроорганізмів у нативному водному розчині бедринцю та в розведені 1:2 та 1:4 в м'ясо-пептонному бульйоні; бактерицидні – за відсутністю росту вмісту пробірок із розведенням екстракту на щільних пoживних середoвищах (м’ясо-пептонний агар – МПА) для грампозитивних мікроорганізмів S. aureus, B. subtilis, для грамнегативної культури E. coli, P. аeruginosa. Агар Сабуро використовували для дріжджеподібних грибів роду Candida (C. аlbicans). Результати й обговорення. Бедринцю ломикаменевого підземних органів екстракт густий має широкий спектр антибактеріальної активності. Нативний екстракт БЕГ-1 по відношенню до тест-штамів S. aureus, B. subtilis, C. аlbicans проявляв бактерицидну дію. Бактеріостатичну дію він проявляв до P. aeruginosa, а по відношенню до E. coli антимікробного ефекту не виявлено. Екстракти в розведені 1:2 та 1:4 проявили бактерицидну дію відносно B. subtilis, бактеріостатичну – до S. aureus, C. albicans і не виявлено протимікробної дії до E. coli та P. aeruginosa. Нативний зразок БЕГ-2 по відношенню до усіх досліджуваних 5 тест–штамів проявив бактерицидну активність; у розведені 1:2 та 1:4 проявляв бактерицидну та бактеріостатичну дію відносно S. aureus, B. subtilis, C. albicans. Антимікробна дія по відношенню до E. coli та P. aeruginosa не виявлена. Антимікрoбну активність нативного бедринцю ломикаменевого екстракту густого також вивчали у дoслідах in vitro методом дифузії в агар –  метод “колодязів”. Штами S. aureus, C. albicans, B. subtilis, P. aeruginosa, E. coli проявили чутливість до нативного бедринцю ломикаменевого підземних органів екстракту густого. Грампозитивні бактеріальні штами S. aureus,а також гриби C. albicans, B. subtilis є найбільш чутливими до  БЕГ-2, а БЕГ-1 проявляв порівняно меншу чутливість. Грамнегативні культури P. aeruginosa та E. coli також проявили помірну чутливість до  БЕГ-1, але значно більшу чутливість до БЕГ-2. Висновки. 1. Експериментально встановлено, що обидва досліджувані зразки бедринцю ломикаменевого підземних органів екстракту густого проявляють антибактеріальну активність. 2. Бедринцю ломикаменевого підземних органів екстракт густий проявляв більш виражену антимікробну активність по відношенню до грампозитивної мікрофлори, тому є перспективним для створення лікарського засобу з антимікробними властивостями.
