Characteristics of the etiological structure and susceptibility to antibiotics, antiseptics of infectious pathogens of respiratory organs in children in critical states

The study of the etiological structure, the properties of pathogens of the respiratory infectious diseases in children and their resistance to antibacterial agents is particularly relevant in modern conditions, expands the search for new approaches to combating pathogens, improves the results of treatment and reduces the mortality of this pathology. The aim — study of etiological structure, sensitivity to antibiotics and antiseptics of pathogens of infectious and inflammatory diseases of respiratory organs in children. In the study there were enrolled 247 patients who were treated in Vinnytsia Regional Children’s Clinical Hospital (VRCCH) in 2016. The sensitivity of microorganisms to 23 antibacterial agents was determined by the disc-diffusion method according to the generally accepted method. The analysis of the antimicrobial activity of antiseptic drugs (decamethoxine, miramistin, chlorhexidine digluconate) was performed by a double serial dilution technique with the determination of the minimum inhibitory bacteriostatic (MIC) and bactericidal (MBcC) concentrations, by the method of successive serial dilutions of the drug in a liquid nutrient medium. In patients who were in inpatient treatment at the VRCCH in 2016 because of pneumonia there were found opportunistic microorganisms which were of etiological significance in the development of the infection. Among them there were Streptococci (47,3 %), Staphylococci (15,3 %), Candida (13,3 %), Enterococci (10,9 %), including a high proportion of owned non-fermenting gram negative bacilli (9,8%) and species of Enterobacteria (2,0 %). Isolated strains of microorganisms had moderate resistance to most modern antibiotic drugs. The sensitivity of isolated strains of microorganisms to reserved antibiotics as carbapenems, often being used in the treatment of critical states of patients in the intensive care units, was found to above 18,2%. The sensitivity to this antibiotic in Enterococcus spp. (7,1 %), Staphylococcus spp. (5,9 %) was also low. Carbapenems, fluoroquinolones (the 1st and 2nd generations), antibiotics and aminoglycosides were found to be effective against gram positive microorganisms in more then 45% of cases. According to this they were considered to be as drugs of choice in the treatment of infectious and purulent-inflammatory pathology of respiratory organs, caused metitcilin- and vancomycin-resistant strains of microorganisms. Resistance to these drugs among investigated strains did not exceed 9,0 %. The high bactericidal properties of antiseptics as decamethoxine was determined against S.pyogenes, Staphylococcus spp. Its MBcC against these bacteria (1,65±0,20 mkg/ml and 4,32±0,50 mkg/ml, respectively) proved the advantage of decamethoxine’s effectiveness in comparison with chlorhexidine digluconate 3,14 times, 2,44 times miramistin. Clinical strains of C.albicans showed the highest susceptibility to decamethoxine, which fungicidal activity was determined in the presence (16,17±2,33 mkg/ml), in comparison with chlorxedine (MFtsK 27,59±3,59 mg/ml) and miramistin activity (27,59±3,595 mkg/ml). In children with inflammatory diseases of the respiratory organs gram-positive cocci are among the predominant pathogens (73,5 %) of cases, in the association allocated – 8,0 % of pathogens. Allocated strains of microorganisms were moderately resistant to all antibiotics studied. For antimicrobial activity antiseptic drugs, especially decamethoxine, have advantages over antibiotics confirming the possibility of their use in combination with systemic antibacterials.

Pycnarrhena cauliflora (Miers.) Diels., local name sengkubak, is one of indigenous plants from West Kalimantan that has been used as natural flavor. Pycnorrhena cauliflora is one of species of Menispermaceae family which is rich in bisbenzylisoquinoline alkaloids. This alkaloids are known to have various biological activities including antiprotozoal, antiplasmodial, antifungal and antibacterial activities. This study aimed to investigate antimicrobial activity of  the P. cauliflora (Miers.) Diels. methanolic extracts against gram-positive and gram-negative bacteria. The methanolic extract of P. cauliflora (Miers.) Diels., root, leaf and stem were prepared by maceration. The disk-diffusion method was then used to determine the antimicrobial activity of the extracts against Streptococcus pyogenes, S. mutants, Staphylococcus aureus, S. epidermidis, Salmonella typhi, Shigella flexneri, Pseudomonas aeruginosa and Escherichia coli after 18-24 h incubation at 37 oC. Amoxicillin was used as positive control for gram-positive bacteria and ciprofloxacin was used as gram-negative bacteria. The inhibition zones were then measured in mm. Analysis were conducted in duplicates. The results showed in general the methanolic extracts of P. cauliflora (Miers.) Diels. root (inhibition zone diameter= 10-23 mm) were more active than that leaf (0-15 mm) and stem (0-17 mm) extracts against gram-positive bacteria. The zone inhibition diameter of amoxicillin as positive control was 8-42 mm. In addition, the methanolic extracts of P. cauliflora (Miers.) Diels. root (12-17 mm) were also more active than that leaf (0-12 mm) and stem (0-12 mm) extracts against gram-negative bacteria. The zone inhibition diameter of ciprofloxacin as positive control was 33-36 mm. In conclusion, the methanolic extract of P. caulifloria (Miers.) Diels. root is the most extract active against both gram-positive and gram-negative bacteria. Further study will be focused to isolate active compounds in the methanolic extract of the root.

The genera Cistus present groups of about twenty species of shrubs belon- ging to the Cistaceae family are very common throughout the Mediterranean region. Cistus salviifolius, known as Melliya is a species of this genus that occurs in the forest region of Mount Ouarsenis (northwest Algeria), exactly in Thniet El Had Na onal Park in the Wilaya of Tissemsilt. This study was un- dertaken to evaluate the an bacterial effects of C. salviifolius and its chemical composition in addition the effect of shade drying me on essen al oil yield in order to op mize this parameter which is most o en not controlled for producers. The op mal yield recorded is about 0.07± 0.02%. Its characte- riza on by gas chromatography coupled with mass spectrometry shows a dominance of hydrocarbon compounds. The minimum inhibitory concentration (MIC) is established using 6 strains of Gram-positive and negative bacteria. The antibiotic susceptibility test is performed using the giliose- environment diffusion method, a bacterial suspension is prepared and subsequently well homogenized. Opacity must be adjusted to 0.5 McF (McFarland), from a young exponen al culture (18 to 24 hours). The an mi- crobial ac vity of C. salviifolius EO was tested against three Gram-posi ve and three Gram-nega ve bacteria. In fact, a strong inhibitory and bactericidal effect has been demonstrated against all Gram-positive and Gram- negative bacteria tested. It appears from this study that the EO of C. salviifo- lius has a very high antimicrobial activity, it shows a wide spectrum covering Gram positive and Gram negative bacteria. Taking into account the results of this study, new an bacterial agents could be developed and the use of C.salviifolius should be encouraged in the pharmaceutical and perfumery industries.

Biofouling and biofilms exist as ubiquitous, undesirable accumulation of flora and fauna upon a given substrate when being immersed into an aquatic medium. Therefore, a novel antifouling based materials with the incorporation of nanotechnology has been developed for the prevention of biofouling in its initial stage through photocatalytic treatment. This study investigated the antimicrobial properties of photoactive Cerium (Ce) doped ZnO powder and explores its potential properties for future antifouling application. ZnO nanoparticles was doped with 0.4 mol% Ce was synthesized through the combination of modified citrate gelation technique and solid state sintering. The successful preparation of Ce doped ZnO was confirmed by XRD and SEM. The antimicrobial activity of Ce doped ZnO against E. coli and S. aureus was determined through antibacterial susceptibility test by agar well diffusion method whilst its photocatalytic inactivation efficiency against selected bacteria was analysed through photodegradation testing under UV light irradiation. The findings demonstrated that the synthesized Ce doped ZnO powder exhibited antibacterial effect against Gram-positive bacteria (S. aureus) and excellent photocatalytic efficiency to inactivate both Gram-negative (E. coli) and Gram-positive (S. aureus). 2 g/L of Ce doped ZnO catalyzed the 100% disinfection of both bacteria in 180 min of UV light exposure. Thus, this proved that Ce doped ZnO powder has the potential as efficient antifouling agent.

A series of new tetracyclic oxathiine-fused quinone-thioglycoside conjugates based on biologically active 1,4-naphthoquinones and 1-mercapto derivatives of per-O-acetyl d-glucose, d-galactose, d-xylose, and l-arabinose have been synthesized, characterized, and evaluated for their cytotoxic and antimicrobial activities. Six tetracyclic conjugates bearing a hydroxyl group in naphthoquinone core showed high cytotoxic activity with EC50 values in the range of 0.3 to 0.9 μM for various types of cancer and normal cells and no hemolytic activity up to 25 μM. The antimicrobial activity of conjugates was screened against Gram-positive bacteria (Staphylococcus aureus, Bacillus cereus), Gram-negative bacteria (Pseudomonas aeruginosa and Escherichia coli), and fungus Candida albicans by the agar diffusion method. The most effective juglone conjugates with d-xylose or l-arabinose moiety and hydroxyl group at C-7 position of naphthoquinone core at concentration 10 µg/well showed antimicrobial activity comparable with antibiotics vancomicin and gentamicin against Gram-positive bacteria strains. In liquid media, juglone-arabinosidic tetracycles showed highest activity with MIC 6.25 µM. Thus, a positive effect of heterocyclization with mercaptosugars on cytotoxic and antimicrobial activity for group of 1,4-naphthoquinones was shown.

The structure of the microflora of the urogenital tract of a woman is variable and diverse, changing its qualitative and quantitative composition can affect various physiological processes in the body of a woman, including the course of pregnancy. In this study, the results of cultures of 1415 samples of urine and cervical canal discharge of pregnant women were analyzed. Species identification was carried out by MALDI-ToF mass spectrometry using Microflex LT (Bruker) mass spectrometer. Gram-positive bacteria (69.5%) dominated the structure of the cervical canal microflora, among which Staphylococcus spp prevailed., Enterococcus spp. and Lactobacillus spp. Among gram-negative bacteria most often encountered microorganisms of the order Enterobacteriales, the predominant species among which was E. coli. Also, yeast-like fungi were isolated from the material of the cervical canal, their number was 11% of the total number of crops. Qualitative microbiological composition of urine was represented by gram-positive flora (68.7%), gram-negative flora (30.1%) and Candida fungi (1.2%). There is a significant predominance of coagulase-negative staphylococci (97.3%) over coagulase-positive (2.7%) in the structure of gram-positive microorganisms. The composition of gram-negative flora is mainly represented by bacteria of the order Enterobacteriales (71.4%). The study identified microorganisms that can cause postpartum complications and the development of inflammatory diseases of the newborn, which suggests the need for regular microbiological examination for pregnant women.

Antimicrobial activity of methanol extracts of four Parmeliaceae lichens (Hypogymnia physodes (L.) Nyl., Evernia prunastri (L.) Ach., Flavoparmelia caperata (L.) Hale and Parmelia sulcata Taylor) against a panel of microbial strains (11 Gram-positive (Enterococus sp., Bacillus subtilllis, Sarcina lutea, Micrococus luteus, Staphylococcus aureus, Clostridium sporogenes) and Gram-negative bacteria (Escherichia coli, Proteus vulgaris, Salmonela enteritidis, Pseudomonas aeruginosa, Klebsiella pneumoniae), the filamentous fungus A. niger and the yeast C. albicans) was assayed using a disk diffusion method (1 mg of the extract per disc; extracts were dissolved in methanol, 25 mg/mL). All tested extracts showed moderate antimicrobial activity. Multivariate statistical treatment (agglomerative hierarchical clustering analysis, AHC) of the obtained results allowed grouping of the samples according to their antimicrobial potential against different strains: antimicrobial profile of H. physodes and E. prunastri extracts were comparable; the similar is true for F. caperata and P. sulcata samples. In addition, based on the similarities/ dissimilarities in their susceptibility toward the tested extracts, two groups of microorganisms could be distinguished: Group I - P. vulgaris, K. pneumoniae (Gram-negative bacteria), A. niger and C. albicans; Group II - E. coli, S. enteritidis, P. aeruginosa (Gram-negative bacteria) and all of the assayed Gram-positive strains.

Introduction. Antiseptic drugs currently occupy an important place in the treatment and prevention of ocular infectious and inflammatory diseases. Often microorganisms are characterized not only by resistance to a single antibiotic, but also by the presence of multiple resistances, which limits the choice of an effective drug. This problem requires a detailed study and monitoring of the sensitivity of the main pathogens of ocular infections, not only to antibiotics but also to antiseptics. The aim was to study the species composition of conjunctival microflora in patients with ocular surface infection and to evaluate the antimicrobial activity of antiseptic eye drops. Materials and methods. Investigation was carried out in 20122018 in 4237 bacterial conjunctivitis patients. The sensitivity to antiseptic preparations of pathogens isolated from patients with conjunctivitis was detected. Results. 1068 strains of microorganisms isolated from the conjunctival cavity of patients were tested. Gram-positive cocci dominated among clinically significant pathogens 47.4%. Antimicrobial activity of Vitabact eye drops against gram-positive cocci was higher than that of antiseptic Okomistin. Summary. Antimicrobial activity of the studied antiseptics against gram-positive and gram-negative pathogens was different. It is necessary to conduct further research on the antimicrobial activity of antiseptic eye drops.

ABSTRACT: In recent years, compounds with biological properties produced by plants have received attention as an alternative to control microorganisms. Essential oils extracted from green leaves of Eucalyptus sp. have been demonstrated to have antimicrobial activities, but so far there are no reports of antimicrobial activity of essential oils extracted from dried leaves of Eucalyptus staigeriana. So, the objectives of this study were to determine the chemical composition of the essential oils obtained from dried leaves of E. staigeriana (EOdlES) and to evaluate in vitro antimicrobial and antibiofilm activities of EOdlES against gram-positive and gram-negative, resistance and multiresistant Enterococcus faecalis isolated from food and clinical samples. The characterization of EOdlES was performed by gas chromatography-mass spectrometry (GC/MS). For this study, 26 bacterial strains were used, which included 11 reference strains and 15 antibiotic resistant and multiresistant E. faecalis strains. Antimicrobial activities of EOdlES against gram-positive and gram-negative were determined using the disc diffusion method. The minimum inhibitory concentration (MIC) value was evaluated by a microbroth dilution technique. The antibiofilm effects were assessed by microtiter plate method. As a result, 21 compounds were identified, being oxygenated monoterpenes (69.58%) the major chemical family. EOdlES showed only antimicrobial activity against gram-positive strains. E. faecalis resistant and multiresistant strains show the lowest MIC (3.12 to 6.25%), when compared with reference E. faecalis strain. EOdlES has the ability to inhibit the biofilm formation, but little or none ability to inhibit the preformed biofilm. This study demonstrates that EOdlES is a promising alternative to control important foodborne and clinic gram-positive resistant bacteria.

: A Three component Synthesis of novel 5-phenyl-2-(thiophen-2-yl)-4-(trifluoromethyl)-5H-indeno [1,2-b] [1,8] naphthyridin-6(11H)-one derivatives (4a-n) were prepared using 6-phenyl/(thiophen-2-yl)-4-(trifluoromethyl)pyridin-2-amine, 1H-indene-1,3(2H)-dione and aryl aldehyde using 40% aq. HF with good yield. All the synthesized compounds were screened against Gram-positive and Gram-negative bacterial strains and different Candida strains by well diffusion method. Compounds 4c, 4f and 4g showed promising activity on Bacillus subtilis strain and compounds 4c and 4g showed promising activity towards Candida albicans starains.

The traditional medicine still plays an important role in the primary health care in Yemen. The current study represents the investigation of 16 selected plants, which were collected from different localities of Yemen. The plants were dried and extracted with two different solvents (methanol and hot water) to yield 34 crude extracts. The obtained extracts were tested for their antimicrobial activity against three Gram-positive bacteria, two Gram-negative bacteria, one yeast species and three multiresistantStaphylococcusstrains using agar diffusion method, for their antioxidant activity using scavenging activity of DPPH radical method and for their cytotoxic activity using the neutral red uptake assay. In addition, a phytochemical screening of the methanolic extracts was done. Antibacterial activity was shown only against Gram-positive bacteria, among them multiresistant bacteria. The highest antimicrobial activity was exhibited by the methanolic extracts ofAcalypha fruticosa,Centaurea pseudosinaica,Dodonaea viscosa,Jatropha variegata,Lippia citriodora,Plectranthus hadiensis,Tragia pungensandVerbascum bottae.Six methanolic extracts especially those ofA. fruticosa,Actiniopteris semiflabellata,D. viscosa,P. hadiensis,T. pungensandV. bottaeshowed high free radical scavenging activity. Moreover, remarkable cytotoxic activity against FL-cells was found for the methanolic extracts ofA. fruticosa,Iris albicans,L. citriodoraandT. pungens. The phytochemical screening demonstrated the presence of different types of compounds like flavonoids, terpenoids and others, which could be responsible for the obtained activities.
