The development of the Bogor taro plant genetic variation with gamma-ray radiation has resulted in three mutant clones of the hope line with high carbohydrate and low calcium oxalate advantages. The carbohydrate content may correlate with chlorophyll content which plays a role in the photosynthesis process, so it is necessary to know the chlorophyll content in the mutant clones of the Bogor taro hope line and its relationship with its growth in two locations in Kemang District and Cijeruk District, Bogor Regency. The results showed that the mutant clones of the hope line B1023 planted in Kemang District had higher chlorophyll content than the parents. Meanwhile, the mutant clones of the hope line B1511 planted in the Cijeruk District had higher chlorophyll content than the parents. The chlorophyll content of the hope mutant clones planted in Kemang District was higher than the hope mutant clones planted in Cijeruk District. Schott)Pengembangan variasi genetik tanaman talas Bogor dengan radiasi sinar gamma telah menghasilkan tiga klon mutan galur harapan dengan keunggulan tinggi karbohidrat dan rendah kalsium oksalat.  Kandungan karbohidrat mungkin berhubungan erat dengan kandungan klorofil yang berperan dalam proses fotosintesis, sehingga  perlu untuk diketahui kandungan klorofil pada klon mutan galur harapan tanaman talas Bogor dan hubungannya dengan pertumbuhannya di dua lokasi Kecamatan Kemang dan Kecamatan Cijeruk Kabupaten Bogor.  Hasil penelitian menunjukkan bahwa klon mutan galur harapan B1023 yang ditanam di Kecamatan Kemang memiliki kandungan klorofil lebih tinggi dari tetua. Sedangkan pada klon mutan galur harapan B1511 yang ditanam di Kecamatan Cijeruk memiliki kandungan klorofil lebih tinggi dari tetua. Kandungan klorofil klon mutan galur harapan yang ditanam di Kecamatan Kemang lebih tinggi dibandingkan dengan kandungan klorofil klon mutan harapan yang ditanam di Kecamatan Cijeruk. Kandungan klorofil tinggi menunjukkan hubungan negatif dengan bobot basah umbi dan kandungan karbohidrat, tetapi tidak memberikan pengaruh terhadap jumlah daun dan anakan. Kata kunci :  Klorofil a, Klorofil b, Fotosintesis, Klon mutan, Colocasia esculenta

ABSTRACT  The objective of this research was to examine the effect and quantity of rice straw mulch on the growth and production of kailan. This research was conducted inAspakusa Makmur Farms Association, Teras, Boyolali and Laboratory of Ecology and Plant Production, Faculty of Animal and Agricultural Sciences, Diponegoro University, Semarangfrom January 24th to March24th 2018. The research was assigned in completely randomized monofactor design with the factor was M0: control, M1: 2 ton/ha, M2: 4 ton/ha, M3: 6 ton/ha and M4: 8 ton/ha. Parameters observed were plant height, number of leaves, soil and temperature, plant and root wet weight, plant and root dry weight and total chlorophyll content.The results showed thatmulch treatment with a dose of 6 tons/ha was able to increase the yield of leaves, root dry weight and total chlorophyll content but did not increase crop yields, soil and temperature, plant and root wet weight, plant dry weight Keyword: Kailan, Mulch, Rice straw ABSTRAK  Penelitian bertujuan untuk mengkaji pengaruh dan takaran mulsa jerami padi terhadap pertumbuhan dan produksi tanaman kailan. Penelitian dilakukan pada tanggal24 Januari 2018 – 24 Maret 2018 di Lahan pertanian Asosiasi Aspakusa Makmur Kecamatan Teras Kabupaten Boyolali dan Laboratorium Ekologi dan Produksi Tanaman, Fakultas Peternakan dan Pertanian, Universitas Diponegoro, Semarang. Penelitian disusun dengan rancangan acak lengkap monofaktor dengan perlakuan M0: kontrol, M1: jerami padi 2 ton/ha, M2: jerami padi 4 ton/ha, M3: jerami padi 6 ton/ha dan M4: jerami padi 8 ton/ha. Parameter yang diamati adalah tinggi tanaman, jumlah daun, kelembaban dan suhu tanah, berat basah tanaman dan akar, berat kering tanaman dan akar dan kandungan klorofil total. Hasil penelitian menunjukkan bahwa Perlakuan mulsa dengan takaran 6 ton/ha mampu meningkatkan hasil jumlah daun, berat kering akar dan kandungan klorofil total tetapi tidak meningkatkan hasil tinggi tanaman, kelembaban dan suhu tanah, berat basah tanaman dan akar, berat kering tanaman Kata kunci: kailan, mulsa, jerami padi

A study has been conducted to compare some varieties of planting media on red lettuce (Lactuca sativa var crispa L), the variables observed in this study are wet weight and chlorophyll content. This research was conducted by using complete randomized design (RAL) consisting of 5 variants of planting medium (Rockwool (as control), cocopeat, fuel husk, cotton and sponge) with 4 replications. The results showed that rockwool planting media had significant effect (P <0,05) on wet weight of red lettuce plant (Lactuca sativa var. Crispa L). Then, on the results of chlorophyll content test, it was found that the variation of burning husk medium media on red lettuce plant had significant effect (P <0,05) to chlorophyll a and total chlorophyll, while in chlorophyll b the result obtained was not significantly different (P> 0, 05). The results of this study indicate that the treatment of rockwool planting media in red lettuce plant growth is effective to increase wet weight and the treatment of burning husk medium media can increase the chlorophyll content of red lettuce plants namely chlorophyll a and total chlorophyll red lettuce plants (Lactuca sativa var. Alfalfa in dry stress conditions cannot grow optimally. Drought inhibits growth and reduces alfalfa production in many cultivation systems. Plants breeding with gamma-ray irradiation and in vitro selection using PEG found somaclonal variants adapted to grow well in drought conditions. This study aims to determine the type of mutation with the best morphology in dry stress conditions. Alfalfa mutants resulting from gamma irradiation were sub-cultured in vitro. Alfalfa mutants cultured in PEG medium, according to treatments. This experiment used completely randomized factorial design with 2 factors, namely the gamma-ray irradiation doses (0 Gy, 100 Gy, 200 Gy, 300 Gy, 400 and 500 Gy), and PEG concentrations (5%, 10%, 15%, and  20% ). The variables observed were viability, plant height, number of leaves, wet weight, medium shrinkage, leaf wilting, and leaf color. The results showed that the gamma-irradiated mutant had a better (p<0.05) morphology than the control (0 Gy) in the face of dry stress conditions. Combination of 500 Gy and 20% PEG concentration resulted the best type of mutant. Key words:        alfalfa, in vitro, gamma rays, PEG

SummaryEnhancement of the resistance level in plants by rhizobacteria has been proven in several pathosystems. This study investigated the ability of four rhizobacteria strains (Pseudomonas putida BTP1 and Bacillus subtilis Bs2500, Bs2504 and Bs2508) to promote the growth in three barley genotypes and protect them against Cochliobolus sativus. Our results demonstrated that all tested rhizobacteria strains had a protective effect on barley genotypes Arabi Abiad, Banteng and WI2291. However, P. putida BTP1 and B. subtilis Bs2508 strains were the most effective as they reduced disease incidence by 53 and 38% (mean effect), respectively. On the other hand, there were significant differences among the rhizobacteria-treated genotypes on plant growth parameters, such as wet weight, dry weight, plant height and number of leaves. Pseudomonas putida BTP1 strain was the most effective as it significantly increased plant growth by 15-32%. In addition, the susceptible genotypes Arabi Abiad and WI2291 were the most responsive to rhizobacteria. This means that these genotypes have a high potential for increase of their resistance against the pathogen and enhancement of plant growth after the application of rhizobacteria. Consequently, barley seed treatment with the tested rhizobacteria could be considered as an effective biocontrol method against C. sativus.

Indian mustard plants are the main vegetable crops consumed by urban communities. The need for this vegetable increases along with the increase in culinary tourism in each region and the government's appeal on sustainable food home. One effort made to meet the needs of vegetable mustard in urban areas can be cultivated using polybags. The main problem of vegetable cultivation on polybags is the availability of soil media. To overcome the limitations of soil media it is attempted to substitute it with organic litter enriched with MOL (local microorganisms). The purpose of research is to find the right combination of planting media for mustard plants. The results showed that the composition of K7 = soil: compost: litter of hay: litter of maize (2: 1: 1: 1) best on plant length, number of leaves, length of root leaves and number of roots while the wet weight of mustard plant (g) K7 = soil: compost: litter of maize: litter of corn (2: 1: 1: 1) and K8 = soil: compost: litter of maize: litter of maize: cocopeat (2: 1: 1: 1: 1) or soil: compost + urea. Increased weights of wet mustard by 63% compared to soil media and 20% compared to soil composition: compost + urea. Organic straw in the form of straw: litter of corn stalk: cocopeat enriched with MOL are able to become ready-made planting media on the cultivation of indian mustard plants in polybags.

Chlorophyll is an important factor which also affects the yield in maize. In this study, genetic analysis of chlorophyll content was conducted by joint segregation analysis of four generations P1, P2, F1 and F2:4 from the cross Shen3336?Shen3265 using the mixed major genes and polygenes inheritance models. Genetics of chlorophyll revealed that chlorophyll ?a? was controlled by two main gene having additive-dominanceepistasis effects. The heritability of these genes were 56.3%. Chlorophyll ?b? was controlled by two pairs of codominant major gene plus additive-dominance polygene. The heritability of these major genes and polygenes were 1.12% and 93.26% respectively. Chlorophyll ?a+b? was controlled by two pairs of additive-dominance- epistatic major genes plus additive-dominance polygene having heritability of 56.2% and 5.2% respectively.

Tanaman kelapa memiliki nilai ekonomi yang tinggi karena hampir semua bagian tanaman kelapa memiliki manfaat ekonomis. Penelitian bertujuan untuk  mengembangkan metode analisis klorofil pada daun dan menentukan bagian anak daun yang memiliki kandungan klorofil tertinggi. Analisis kandungan klorofil dilakukan dengan metode ektraksi pelarut yang absorbansinya dibaca pada spektrofotometer UV-Vis. Kandungan klorofil a dalam daun bervariasi pada setiap bagian anak daun tanaman kelapa. Kandungan klorofil a pada posisi tengah anak daun kelapa (763.19 µmol/m2) dan kandungan klorofil b (196.22 µmol/m2). Kandungan klorofil a dan b pada setiap bagian anak daun dan posisi kanan dan kiri anak daun memiliki nilai yang relatif sama.Kata kunci: Kelapa, klorofil a, klorofil b Abstract Many coconut plants are cultivated as agricultural crops. Coconut plants have high economic value because almost all parts of coconut plants have economic benefits. The study aims to develop a method of chlorophyll analysis on leaves and determine the part of leaflets which have the highest chlorophyll content. Analysis of chlorophyll content was carried out by a solvent extraction method in which the absorbance was read in a UV-Vis spectrophotometer. The content of chlorophyll a in leaves varies with each part of the leaves of the coconut plant. Chlorophyll a content in the middle position of coconut leaf child (763.19 µmol / m2) and chlorophyll b content (196.22 µmol / m2). The content of chlorophyll a and b in each part of the leaf child and the right and left positions of the leaves have relatively the same value.Keywords: Coconut, Chlorophyll a, Chlorophyll b

Level of anthocyanin content in flush is different between fine flavor cocoa and bulk cocoa. This study was aimed to determine the potential of anthocyanin and chlorophyll content in flush as one of the selection indicator for fine flavor cocoa. Flush sampling was carried out at Cocoa Germplasm Collections in Kaliwining Experimental Station, Indonesian Coffee and Cocoa Research Institute, while anthocyanin and chlorophyll analysis was conducted at Laboratory of Ma Chung Research Center for Photosynthetic Pigments, Ma Chung University, Malang.The research utilized a completely randomized design consisted of four genotypes of fine flavor cocoa, namely DR 2, ICCRI 02, DRC 16, PNT 16 and five genotypes of bulk cocoa, namely Sulawesi 1, MCC 01, MCC 02, KW 617 and PA 191.Each treatment was repeated three times. Anthocyanin and chlorophyll content in flush was measured. Anthocyanin analysis was done by destructive methods, while flush color was observed based on Munsell Color Chart for Plant Tissues Data analysis used orthogonal contrast, multivariate and simple correlation. The results showed that there were differences in flush color on fine flavor cocoa and that of bulk cocoa. The flush of the fine flavor cocoa had a tendency towards yellow and that of bulk cocoa had a tendency toward red with different levels of variation among genotypes. The analysis results of chlorophyll content in flush showed that chlorophyll content of fine flavor cocoa was higher than bulk cocoa with a range of 2376-3692 µmol/L for fine flavor cocoa and 1567-2628 µmol/L for bulk cocoa. On the contrary, anthocyanin content in fine flavor cocoa was lower than bulk cocoa with a range of 3.82-5.72 µmol/L for fine flavor cocoa and 6.01-11.88 µmol/L for bulk cocoa. The analysis results of orthogonal contrasts showed that chlorophyll content value was more stable than anthocyanin. There was negative relationship between anthocyanin and chlorophyll in flush with the regression Y = -221,1X + 3935 with a value of correlation r = -0.50. The anthocyanin and chlorophyll content in flush had a potential to be used as selection markers for fine flavor cocoa hybrid

This study aims to know the effect of Benzyl Amino Purine (BAP) and Coconut Water on shoot bud growth and Broccoli Sulforaphane content (Brassica oleracea L. var italic Plenck). The study was conducted in the laboratory of Biotechnology Department of Aquaculture, Faculty of Agriculture of Sam Ratulangi University, Manado, that conducted from August-December 2017. This study used a Complete Randomized Design (RAL), consisting of 8 treatments and each repeated as many 4 times, so we get 32 unit experiment. The variables observed were number of buds, number of leaves, plant height, wet weight, root number and Sulforaphane content analysis. The result of research shows that analysis of variance showed that in the use of Benzyl Amino Purine (BAP) concentration 3 ppm tends to increase the number of leaves aged 4 Weeks After Culture (MSK) and increase the number of shoots age 2 and 6 Weeks After Culture (MSK). Benzyl Amino Purine (BAP) 3 ppm can increase the wet weight of age 6W eeks After Culture ((MSK). Coconut water 20% tends to increase the number of leaves at age 6 Weeks After Culture (MSK) and increase the number of shoots aged 6 Weeks After Culture (MSK), while for combination of 3 ppm Benzyl Amino Purine (BAP) and coconut water 20% tends to increase the number of leaves aged 2 Weeks After Culture (MSK) and the number of shoots aged 2 Weeks After Culture (MSK). Combination of coconut water and Benzyl Amino Purine (BAP) is not detected by the content of Sulforaphane.
