Metal-induced oxidative stress level in patients with fibromyalgia syndrome and its contribution to the severity of the disease: A correlational study

BACKGROUND: Fibromyalgia syndrome (FMS) is an extra-articular rheumatological disease characterised by widespread chronic musculoskeletal pain. Metal-induced oxidative stress contributes to the severity of FMS. AIMS: First, this study evaluated the association between plasma levels of toxic heavy metals and essential metals with oxidative stress (OS) markers. Second, the OS markers and metal contents were correlated with the disease severity by assessing the Fibromyalgia Impact Questioner Revised (FIQR) and tender points (TP). METHOD: A total of 105 FMS patients and 105 healthy controls of similar age and sex were recruited. OS parameter such as lipid peroxidation (LPO), protein carbonyl group (PCG), nitric oxide (NO) and essential metals such as zinc (Zn), magnesium (Mg), manganese (Mn), copper (Cu) and toxic heavy metals such as aluminium (Al), arsenic (As), lead (Pb) were estimated. RESULTS: Levels of LPO, PCG, NO (p< 0.001) and Cu, Mn, and Al (p< 0.001), were significantly higher, and Mg (p< 0.001) and Zn (p< 0.001) were significantly lower in patients compared to controls. A positive association was observed between OS parameters, FIQR and TP with Cu, Al and Mn. A significant negative association was observed between Zn and Mg with FIQR, TP and OS parameters. CONCLUSION: Heavy metals such as Al induce OS parameters and decrease the levels of essential trace elements such as Mg and Zn, which may be responsible for the severity of FMS.

Environmental contaminants such as heavy metals are transported to the Arctic regions via atmospheric and ocean currents and enter the Arctic food web. Exposure is an important risk factor for health and can lead to increased risk of a variety of diseases. This study investigated the association between pregnant women’s levels of heavy and essential metals and the birth outcomes of the newborn child. This cross-sectional study is part of the ACCEPT birth cohort (Adaption to Climate Change, Environmental Pollution, and dietary Transition) and included 509 pregnant Inuit women ≥18 years of age. Data were collected in five Greenlandic regions during 2010–2015. Population characteristics and birth outcomes were obtained from medical records and midwives, respectively, and blood samples were analyzed for 13 metals. Statistical analysis included one-way ANOVA, Spearman’s rho, and multiple linear and logistic regression analyses. The proportion of current smokers was 35.8%. The levels of cadmium, chromium, and nickel were higher compared to reported normal ranges. Significant regional differences were observed for several metals, smoking, and parity. Cadmium and copper were significantly inversely related to birth outcomes. Heavy metals in maternal blood can adversely influence fetal development and growth in a dose–response relationship. Diet and lifestyle factors are important sources of toxic heavy metals and deviant levels of essential metals. The high frequency of smokers in early pregnancy is of concern, and prenatal exposure to heavy metals and other environmental contaminants in the Greenlandic Inuit needs further research.

In a nutshellPhysiological but not excessive levels of zinc, copper and other essential metals are required for human fertility, and may also help protect against accumulation of reproductively toxic heavy metals, e.g. lead and cadmium.There is limited RCT evidence for zinc supplements in treating infertility, and very little for other essential metals. Nevertheless, clinicians treating infertility should pay attention to the patient's mineral status.

Thirty specimens (15 adults and 15 juveniles) of Chelonia mydas found in the Cananéia estuary in the state of São Paulo on the southeastern Brazilian coast between January 2005 and September 2006, were analyzed The concentrations of Cd, Cu, Pb, Mn and Ni in liver and kidney samples of adult and juvenile green turtles were determined by Flame Atomic Absorption Spectrophotometry.The average Cd concentration found in adult livers (0.57µg.g-1) was significantly higher than that in juveniles (0.279µg.g-1). Cu concentrations were significantly higher in the liver than in the kidney, and significantly higher in adults (39.9µg.g-1) than in juveniles (20.7µg.g-1) Average Mn concentrations in liver and kidney did not differ between adults (4.32 and 4.17µg.g-1) and juveniles (4.81 and 3.82µg.g-1), whereas Ni concentrations in adults (0.28 and 0.19µg.g-1, respectively) were significantly higher than in juveniles (0.13 and 0.089µg.g-1, respectively). Pb concentrations in liver were significantly higher in adults (0.37µg.g-1) than in juveniles (0.06µg.g-1). The concentrations of essential trace elements in Chelonia mydas were generally comparable to values reported in other, similar studies. With respect to non-essential metals (Cd, Pb and Ni), Chelonia mydas presented lower values than those reported for their northern Atlantic counterparts.

AbstractOur earlier report has shown that salt substitutes (Obu-Otoyo) contain some toxic heavy metals. This study, therefore, investigated the effect of the dietary inclusion of salt substitutes (Obu-Otoyo), namely, salt “A” and “B”, on biomarkers of oxidative stress and renal function in rats. Salt “A”, which has a gray color, is the product of a process in which ash is produced by burning palm kernel shaft soaked in water overnight and extracting the residue to produce the salt substitute while Salt “B”, which has a white color, is a rock salt mined from a local site at Ilobu town, Osun-State, Nigeria. Salt substitutes were fed to normal rats as dietary inclusion at 0.5% and 1.0% for 21 days. The dietary inclusion of the salt substitutes caused a significant (p<0.05) increase in plasma activities of creatinine, urea, uric acid, and blood urea nitrogen compared with the control. Meanwhile, the dietary inclusion of the salt substitutes caused a significant (p<0.05) decrease in renal superoxide dismutase, catalase, reduced glutathione level, glutathione-S-transferase, and glutathione peroxidase activities with a concomitant increase in the malondialdehyde level compared with the control. Furthermore, there was a significant (p<0.05) increase in the concentrations of heavy metals, such as Pb, Co, Cu, Fe, Zn and Cr, in kidney of rats fed with the salt substitute Obu-Otoyo. Therefore, this finding indicates that Obu-Otoyo induces nephrotoxicity in rats. The nephrotoxicity of Obu-Otoyo could be attributed to the induction of oxidative stress as a result of the presence of some heavy metals, suggesting possible health hazards in subjects who consume it.

This study was based on determining concentration of essential and toxic heavy metal in coconut water available at a local Hazaribagh area in Dhaka, Bangladesh. All essential minerals, if present in the drinking water at high concentration or very low concentration, it has negative actions. In this study, fifteen samples and eight heavy metals were analyzed by Atomic Absorption Spectroscopy (AAS) method which was followed by wet ashing digestion method. The concentration obtained in mg/l were in the range of 0.3 to 1.5, 7.77 to 21.2, 0 to 0.71, 0 to 0.9, 0 to 0.2, 0.9 to 17.3, 0.1 to 0.9, 0 to 0.9 and 0 to 0.7 for Fe, Ni, Cu, Cd, Cr, Zn, Pb and Se respectively. From this data it was concluded that any toxic heavy metals like Cd, Cr, Pb and Ni exceed their toxicity level and some essential nutrients were in low concentration in those samples.

This study was based on to determine the concentration of macro and micro nutrients as well as toxic and nontoxic heavy metals present in the chicken feed available in Dhaka city of Bangladesh. All macro nutrients, if present in the feed at high concentration have some adverse effect, at the same time if this nutrient present in the feed at low concentration this have some adverse effect too. So that this nutrient level should be maintained at a marginal level. On the other side toxic heavy metals if present in the feed at very low concentration those can contaminate the total environment of the ecosystem. In this study six brand samples (starter, grower, finisher and layer) which was collected from different renowned chicken feed formulation industry in Bangladesh. Those samples were prepared for analysis by wet ashing and then metals were determined by Atomic Absorption Spectroscopy. It was found that 27.7 to 68.4, 57.3 to 121.9, 0.21 to 4.1, 0.32 to 2.1, 0.11 to 1.58, 0.28 to 2.11 and 0.28 to 1.78 for zinc, iron, copper, mercury, cadmium, nickel and cobalt respectively. It was found that essential macro and micro nutrients were present in the feed in low concentration on the other side mercury was present in high concentration in the feed samples.

Background : Toxicological consequences arising from exposure to mixtures of heavy metals especially at low, chronic and environmentally relevant doses are poorly recognised. In the present study, we evaluated effects of chronic exposure to combinations of three metals arsenic (As), cadmium (Cd) and lead (Pb) present frequently in drinking water on reproductive function and oxidative damage caused to reproductive organs of female rats. Method : Female rats were exposed to mixture of metals (As, Cdand Pb) for 90 consecutive days. The gain in body weight and weight of reproductive organs were recorded following autopsy on 91 stday. The oestrus cycle were monitored during entire treatment period. Numbers of corpora lutea, implantation sites, live fetus and survival of the fetus were evaluated in rats mated successfully with untreated male after completion of their respective treatment. Ovarian cholesterol, protein, ascorbic acid and enzyme Δ 5 -3β HSD levels were estimated. Serum levels of steroid hormones oestrogen and progesterone were estimated. Histopathological picture of both ovary and uterus were assessed. Levels of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidise (GPX) activity, amount of reduced glutathione (GSH) and malondyaldehyde (MDA) in blood, ovary and uterus were measured as biomarkers of oxidative stress. Results : The treated rats showed reduced body weight gain and reduction in the weight of ovary and uterus. Oestrus cycle was disrupted with continuous diestrous in treated animals. Number of corpora lutea, implantation sites and live fetus and the survival of fetus evaluate were reduced significantly in treated groups. The levels of ovarian cholesterol and ascorbic acid increased in treated rats with decrease Δ5 -3β HSD level. There was reduction in serum level of both the ovarian steroid hormones oestrogen and progesterone. The protein levels did not differ between the groups. There was a significant increase in levels of MDA and decrease in levels of all the antioxidant enzymes in treated group. Conclusion : The results revealed there was disruption to reproductive functions with decrease in stereoidogenic activity and associated oxidative stress in female rats treated with combination of mixture of metals (Cd, As and Pb) at low dose for 90 consecutive days.

The paper evaluates the presence and content of traces of heavy metals Hg, Pb, Ni, Cd (total forms) from coal and solid combustion products, the degree of transfer and accessibility in the area of influence of a lignite power plant. The content of toxic heavy metals in residues are characterized by RE Meiji [ 1 (Pb and Hg) and REMeij �1 (Ni and Cd) for the filter ash. Pb and Ni content in the soil exceeds normal values, and Pb exceeds and alert value for sensitive soils around the residue deposit (70.20 mg.Kg-1). The degree of accessibility of the metals in plants (TF), reported at the Khan reference value (0.5), indicates a significant bioaccumulation level for the metals: Cd (1.9) and Hg (0.6) inside the deposit; Cd (0.39) at the base of the deposit, Hg (0.8) in the area of the thermal power plant. The trace levels of heavy metals analyzed by GFAAS and CVAAS (Hg), indicates a moderate risk potential for food safety and quality of life in the studied area.

Abstract The aim of the this study was to evaluate the effects of fresh, dried and freeze-dried Centaurea depressa M. Bieb. (Asteraceae) on the oxidant and antioxidant status of the model organism D. melanogaster Meigen (Diptera: Drosophilidae) experimentally. The study was carried out from 2016 to 2019, and plant leaf extracts (0-50 mg/l) were added to insect standard artificial diets. The total protein, protein carbonyl content and glutathione-S-transferase, superoxide dismutase and catalase activities were quantified at the insect’s third larval stage. Our data showed that protein carbonyl content varied from 2.70 nmol/mg protein in the control group to 59.11 nmol/mg protein in the group fed with fresh leaf extract signifying induction of oxidative stress. All extracts increased the levels of all antioxidant enzymes and decreased the amounts of total protein. Meanwhile, the group fed with the freeze-dried extract showed no significant difference in the levels of total protein and protein carbonyl content except at the 50 mg/l concentration of the extract. Moreover, this group had superoxide dismutase and catalase activities 4 to 5 times higher than in the control group. In conclusion, induction of oxidative stress indicates that the fresh form of C. depressa leaves may have potential as a natural pesticide, whereas induction of endogenous antioxidant enzymes by the freeze-dried extract suggest its potential as an antioxidant.
