Test of Burn Wounds Healing Effects of Collagen From Snakehead Fish (Channa striata) Bone in The Preparation of Cream on Male White Rats (Rattus norvegicus)

Abstract. The use of snakehead fish (Channa striata) collagen in cream is an alternative toreduce fish wastes. It has been proved that snakehead fish (Channa striata) contain collagen.This research aims to study the burn wound healing effectivity of bone’s collagen fromsnakehead fish (Channa striata) that is formulated into a cream. This study used theexperimental method with the bone of snakehead fish (Channa striata) as a sample. This studyconsisted of 4 (four) such as isolation of collagen from snakehead fish's (Channa striata) bone,characterization of collagen by analyzing moisture, ash, protein, fat and functional groupanalysis with FTIR, formulation, and evaluations of cream such as organoleptic test,homogeneity test, pH measurement, stability test, and irritation test, and burn wound healingtest in male white rats (Rattus norvegicus) and the result is analyzed using SPSS 22.0 Free trialand One Way ANOVA and Post-Hoc Tukey HSD. The results showed that fish collagen couldbe isolated from snakehead fish (Channa striata) and the yield obtained is 33.3%. The results ofcollagen characterization and evaluation test met the collagen standard requirements. Results ofthe burn wound healing test on male white rats (Rattus norvegicus) showed that K1 burn woundrecovered on day 21, K2 on day 12, K3 on day 15, K4 on day 18, and K5 on day 18.Measurement of burn wound diameter on day 21 showed K1 = 1.20 cm, K2 = 0.15 cm, K3 =0.10 cm, K4 = 0.45 cm, K5 = 0.40 cm. The results of the statistical analysis of burn wounddiameter showed a significant difference p = 0,000 (p <0.05) between each group. K3 showedthe reduction in the burn wound diameter is faster and the smallest, it can be concluded that theoptimal dose of snakehead fish bone’s collagen cream is a concentration of 3%.

Burns are one of the injuries that require the best care to achieve optimal cure. Several previous studies have suggested that leaf extract of Centella asiatica and chinese petai leaf potential as burning healing agent for mild and moderate degree besides medical treatment. The purpose of this study was to explain the difference of effectiveness of leaf extract of pegagan leaves 25% by giving 30% petai chinese extract to heal burns. The samples were white rats (Rattus norvegicus) with total of 28 tails selected according to inclusion criteria then divided into 4 groups randomly is group of leaf extract of pegagan leaf 25% (K1), 30% petai petroleum extract (K2) Gel base (K3) and control (K4). The design of this study used the randomized posttest only control group design and analyzed using Anova and post Hoc Duncan test. The measured variable is the duration of wound healing. Anova test results and Post Hoc Duncan test duration of burn wound healing show that 30% Chinese petai extract group has significant difference with other group with p-value <0,05. Treatment of burns by using chinese petroleum extract 30% more effective than 25% of pegagan leaf extract in healing burns in white rats (Rattus norvegicus) as evidenced by the fastest healing time of burns.

Background: Gelatin has been used as a hemostatic agent to stop bleeding and it can be absorbed by human body within 6 hours. The patin fish (Pangasius djambal) gelatin has a high amount of amino acid, especially glutamine which has an important role in increasing the proliferation of fibroblast in wounds. The purpose of this study was to determine the effect of patin fish gelatin to the number of fibroblast in wound after tooth extraction.Method: Rat’s tooth extraction is done on the lower left incisive tooth. Thirty white rats were divided into two groups randomly with 3 time series. Control group was the rats without given of patin fish gelatin and treatment group was the rats given 1cc of patin fish gelatin with pipette. Socket taken for histologically processed by Hematoxylin Eosin staining.Result: It showed there were significant differences in the number of fibroblasts between groups. The Post Hoc Tukey test result showed there was a significant difference between the control group and the treatment group.Conclusion: Patin fish gelatin has effect increasing the number of fibroblast inwound after white rat’s (Rattus norvegicus) tooth extraction

AbstractThe purpose of this study was to evaluate whether cryopreserved (frozen) adipose-derived regenerative cells (ADRCs) have a therapeutic effect on burn wound healing as well as freshly isolated (fresh) ADRCs.Full thickness burns were created on dorsum of nude mice and burn wound was excised. The wound was covered by artificial dermis with; (i) fresh ADRCs, (ii) frozen ADRCs, and (iii) PBS (control). The assessment for wound healing was performed by morphological, histopathological and immunohistochemical analyses.In vivo analyses exhibited the significant therapeutic effect of frozen ADRCs on burn wound healing up to the similar or higher level of fresh ADRCs. There were significant differences of wound closure, epithelized tissue thickness, and neovascularization between the treatment groups and control group. Although there was no significant difference of therapeutic efficacy between fresh ADRC group and frozen ADRC group, frozen ADRCs improved burn wound healing process in dermal regeneration with increased great type I collagen synthesis compared with fresh ADRCs.These findings indicate that frozen ADRCs allow us to apply not only quickly but also for multiple times, and the cryopreserved ADRCs could therefore be useful for the treatment of burn wounds in clinical settings.

This study aimed to determine the effect of snakehead fish extracts (Channa striata) and turmeric extract (Curcuma domestica) towards incision wound healing in rats (Rattus norvegicus). Twenty-four male rats, aged 8-week-old, 200-300 grams of body weight divided into 4 groups: control (P0), treatment with povidone iodine as positive control (P1), treatment with snakehead fish extract (P2) and treatment with turmeric extract (P3). The rats were given premedication consisting of acepromazine 0.5 mg kg / BW and atropine 10 mg / kg BW. The rats were then anesthetized using ketamine 0.1 mg / kg BW. An incision was made on each rats’ back area along 2 cm with 0.05 mm depth and observed for 14 days. Observations were carried out every day with data collection at <7 days, 7-14 days, and 14 days. The One-Way ANOVA statistical analysis resulted that the wound fluid parameters and the duration of wound healing showed no significant difference between P0, P1, P2 and P3. This study concluded that the administration of povidone iodine, snakehead fish extract, and turmeric extract influenced the healing of incision wounds in rats.

Burn wound healing process consists of three phases of inflammation, proliferation and remodelling. The number of fibroblasts has increased during the proliferation phase. Oxidative stress in burn may disrupt proliferation through TGF-β inhibition, result in decreased number of fibroblasts and poor burn wound healing. Genistein in edamame seeds has anti-inflammatory, antibacterial and antioxidant properties. Edamame contains vitamins A, C, and E which is also have antioxidant properties. Edamame has been suggested to protect burn wound healing from oxidative stress. This study aimed to understand the effectiveness of edamame seed (Glycine max L. Merril) ethanolic extract (ED) to fibroblast count in second degree burn wound healing. This is true experimental research with post test only control group design. The sample of this research was 24 rat models of second degree burn which was made by attaching hot metal (60 ° C) for 5 seconds on the shaved back of the rats. The sample was divided into 6 groups with topical treatment, among others: positive control group (silver sulfadiazine), negative control group (Na CMC 0.5%), group with ED 20%, ED 40%, ED 60%, and ED 80%. Treatment was given for 15 days and skin sample was taken on the 16th day. Observation of the number of fibroblasts was done by taking pictures of histhologic preparations in Hematoxylin and Eosin staining under 400x magnification of light microscope. In one way ANOVA test, significance value of 0.011 (p≤0,05) indicated that there was at least a significant difference between 2 groups. LSD’s post hoc analysis showed that there was a significant difference in the group ED 20, 40, and 60% compared with negative control. Background. For centuries, medicinal plants have been extensively used in wound healing of burn injuries. The aim of this study is to analyze comparatively the effects of curcumin and Hypericum perforatum (HP) on second-degree burn wounds in rats. Materials and Methods. This experimental study was conducted on 24 male Sprague-Dawley rats with second-degree burns. The animals were randomly divided into three groups. The burns were treated with curcumin (Group B) and Hypericum perforatum (Group C) with daily application. Group A was considered as the control group and received no medication. Histological parameters, reepithelization, granulation tissue formation, inflammation, and angiogenesis were assessed after the scar biopsy at the end of research. Results. All histological parameters of the control group showed statistically significant difference than the other groups (p<0.05). There was no statistically significant difference between Groups B and C in terms of reepithelization and inflammation (p=0.351, p=0.067). In the curcumin group compared with the HP group, a significant difference was observed for two parameters of fibrosis and angiogenesis (p<0.05). Conclusion. Both curcumin and Hypericum perforatum oil are effective in burn wound healing. Our findings showed a better quality of healing in curcumin-treated rats.

The Effect of Treatment Using Stingless Bee Propolis Extract on Burn Wound Healing in Rattus norvegicus Burn would occurs as a result of direct or indirect fire burns, exposure to sunlight and chemicals. This study aims to analyze the effect of stingless bee propolis extract (Tetragonula fuscobalteata) on the burn wound healing of the back skin of Rattus norvegicus. An experimental research was carried out using the control and experimental groups. Twenty five R. norvegicus were divided into 5 groups: the negative control group was given 70% ethanol, the positive control group was given Bioplacenton® and 3 groups were treated with propolis extract with a concentration of 50%, 70% and 100%. Measurement of burn diameter was carried out every 2 days for 21 days. Data were analyzed statistically using the one way Anova method. The results of phytochemical screening showed that the stingless bee propolis extract contained flavonoids, phenolics, tannins and saponins. The test of the effectiveness of propolis extract against burn wound obtained a sig value of 0.00 (< 0.05). stingless bee propolis extract could heal burn wound in R. norvegicus. Luka bakar dapat terjadi akibat terbakar api secara langsung atau tidak langsung, paparan sinar matahari dan bahan kimia. Penelitian ini bertujuan untuk menganalisis pengaruh ekstrak propolis lebah tanpa sengat (Tetragonula fuscobalteata) terhadap penyembuhan luka bakar pada kulit punggung Rattus norvegicus. Metode yang digunakan merupakan penelitian true experimental menggunakan kelompok kontrol dan eksperimen. Dua puluh lima ekor R. norvegicus dibagi menjadi 5 kelompok: kelompok kontrol negatif diberi etanol 70%, kontrol positif diberi Bioplacenton®, dan 3 kelompok perlakuan ekstrak propolis dengan konsentrasi 50%, 70%, dan 100%. Pengukuran diameter luka bakar dilakukan setiap 2 hari selama 21 hari. Data dianalisa secara statistik menggunakan metode Anova satu arah. Hasil skrining fitokimia ekstrak propolis lebah tanpa sengat positif mengandung senyawa flavonoid, fenolik, tanin, dan saponin. Uji efektivitas ekstrak propolis terhadap luka bakar didapatkan nilai sig 0,00 (< 0,05). Ekstrak propolis lebah tanpa sengat dapat menyembuhkan luka bakar pada R. norvegicus.

Eupatorium odoratum L. is traditionally used to treat open wounds. Aim of this study was toinvestigate the healing activity of ointment containing extract of E. odoratum in burns induced in malealbino mice. Mice were divided into 5 groups, all were induced for burn wound using a heat stamp intemperature 80°C for 20 minutes. Group I was treated with ointment base (control), group II, III and IVwere treated with extract ointment in concentration of 5%, 10%, and 20% w/w respectively, and the lastgroup V was treated with ointment reference. Observations were made during 21 days exactly on the 7th,14th and 21st day, which included parameters: percentage of healed area, epithelialization time andcollagen scores. The result showed that on 14th day, mean of healed area in group III (75.89%±7.76%)and IV (76.29%±6.981%) were significantly higher than other groups (P<0.05). On the 14th and 21th day,collagen scores in groups III (2.33±0.577 and 2.67±0.577) and IV (2.67±0.577 and 2.33±0.577) are equalwith reference group, these score are significantly higher than in groups I and II (P<0.05). Meanwhile,though epithelialization time in group III and IV (both 14.33 days±0.577days) are more quickly thanothers, statistical analysis showed that there is no significant difference on time between all groups(P>0.05).

<p>Haruan is Kalimantan’s indigenous fish which has the potency to accelerate wound healing. Haruan extract has substantial properties suchas albumin, Zn,Cu, andFeto accelerate wound healing. The purposeof this study is to assess the effect of haruan extract on wistar rats buccal mucosa wound healing on day 1, 3, 5, and 7 histopathologically. This study was purely experimental with pretest-posttest with control group design. Samples were divided into 4 treatment groups, haruan extract 25%, 50%, 100% treatment groups and aquadest treatment group as negative control, each treatment wasperformed for 7 days. Neocapillariescount reached its peak on day 5. Mean neocapillaries scoring of each group was 12, 16.5, 18.5, 6 respectively. Two way Anova and post hoc LSD tests presented a significant difference between negative control and haruan extract groups. In conclusion, haruan extract treatment significantly increased neocapillaries count in wound healing process.</p><p>Haruan adalah ikankhas Kalimantan yangberpotensi mempercepat penyembuhanluka. Ekstrak ikan haruan mengandung zat penting seperti albumin, Zn , Cu, dan Fe untuk mempercepat penyembuhan luka. Penelitian ini dimaksudkan untuk mengetahui pengaruh ekstrak ikan haruan pada proses penyembuhan luka pada mukosa bukal tikus wistar hari ke-1, 3, 5, dan 7 secara histopatologi. Penelitian eksperimental murni dengan pretest-posttest with control group ini, meliputi 4 kelompok,yaitu kelompok hewan yang diberi ekstrak haruan selama7 hari dengan konsentrasi berturut-turut 25%, 50%, 100%, dan kelompok kontrol negatif diberi akuades. Hasilnya menunjukkan bahwa jumlah neokapiler mencapai puncak pada hari ke-5. Rerata jumlah neokapiler pada tiap kelompok berturut adalah 12; 16,5; 18,5; dan 6. Uji Anova 2-jalur dan uji post hoc LSDmenunjukkan adanya perbedaan bermakna antara kontrol negatif dan ekstrak ikan haruan. Berdasarkan hasil penelitian, disimpulkan bahwa pemberian ekstrak ikan haruan secara bermakna meningkatkan jumlah neokapiler pada proses penyembuhan luka.</p>
