Diabetes mellitus is a heterogeneous group of syndromes characterized by an elevation of blood glucose caused by a relative or absolute deficiency of insulin. The incidence of diabetes is growing rapidly worldwide. Drug treatment for diabetes mellitus is expensive and carries risks for many adverse effects. Bangladesh is a rich emporium of medicinal plants useful in the treatment of diabetes. The study was performed to investigate the blood glucose lowering effect of Swietenia mahagoni seeds in experimentally induced diabetic rats. Twenty four healthy Long Evans Norwegian strain of rats were included in the study and divided into four groups (A, B, C and D) comprising 6 rats each. Diabetes was induced by alloxan (120 mg/kg body weight). Administration of ethanolic extract of Swietenia mahagoni seeds (group-B) produced no significant change in the blood glucose level as compared to control (group-A). Administration of ethanolic extract of Swietenia mahagoni seeds in group-D produced a significant reduction in blood glucose level as compared to diabetic control (group-C). Histological examination of pancreas showed destruction of beta cells in Islets of pancreas in group-C whereas retaining of islets and few degranulations of beta cells of pancreas found in group-D. These observations and results provide information that ethanolic extract of Swietenia mahagoni seeds has hypoglycaemic effect in experimentally induced diabetic rats which requires further investigation. The study was performed to compare the blood glucose lowering effect of Swietenia mahagoni seeds with an oral antidiabetic drug, rosiglitazone in experimentally induced diabetic rats. Twentyfour healthy Long Evans Norwegian strain of rats were included in the study and divided into four groups (A, B, C and D) comprising 6 rats each. Group A (control group) received standard rat food for 14 days. Diabetes was induced by a single intraperitoneal administration of alloxan 120mg/kg body weight in Group B, C and D. Group B was given standard food for 10 days and considered as diabetic control. Group C and D were treated with ethanolic extract of Swietenia mahagoni seeds 1000mg/kg and rosiglitazone 10mg/kg orally respectively. Administration of ethanolic extract of Swietenia mahagoni seeds in group C and rosiglitazone in group D produced a significant reduction in blood glucose level as compared to diabetic control (group B). Histological examination of pancreas showed destruction of beta cells in Islets of pancreas in group B whereas retaining of islets and few degranulations of beta cells of pancreas found in group C and group D.  Objective: Lawsonia inermis (Linn.) leaves are one of the alternative medicines to treat diabetes mellitus in Indonesia. We investigated the blood glucose level (BGL) of the L. inermis (Linn.) leaves ethanolic extract (LLEE) leaves and evaluated the histopathological alterations in diabetic rats.Methods: This study was an experimental study with posttest - only control group design. Alloxan (120 mg/kg, intraperitoneally)-induced diabetic rats. 35 of Wistar rats (Rattus norvegicus) were divided randomly into five groups, i.e. K: Normal control, P1: Diabetic control, and P2, P3, and P4 (200 mg/kg body weight [BW], 400 mg/kg BW, and 600 mg/kg BW of LLEE, orally) for 28 days. At the end of the treatment, the rats were sacrificed to obtain the kidney for histopathological evaluation using hematoxylin and eosin technique. BGLs were conducted using a glucose meter (GlucoDR).Results: One-way ANOVA test showed that dose 400 mg/kg BW of the LLEE was related to BGL of alloxan-induced diabetic rats (p=0.000). The histopathological of kidney showed glomerular inflammation (GI), epithelial membrane lining degeneration, vascular congestion, and interstitial tubule hemorrhage at diabetic control (P1). Meanwhile, treated with 600 mg/kg BW of LLEE (P4) showed increase cellular regeneration as normal architecture of the kidney.Conclusion: The LLEE at dose 400 mg/kg BW effective decreased BGL and was able to restore the kidney destruction of alloxan-induced diabetic rats at dose 600 mg/kg BW.

Objective: To determine the anti-hyperglycemic effects of interleukin-1 inhibitor (diacerein) in alloxan induced diabetic albino wistar rats. This experimental study was performed at the Department of Animal Husbandry and Veterinary Sciences, Sindh Agriculture University, Tando Jam within 6 months from April 2016 to September 2016. Total of 160 adult Albino Wistar Rats having an average of 200 to 300 grams body weights were selected. Animals were categorized into 4 groups as; Group A (n=15): Control rats – receive 0.9% normal saline as placebo Experimental Groups Group B (n=15): Experimental Control (Diabetic rats) - Alloxan50 mg/kg body weight intraperitoneal. Group C (n=15): Diabetic rats + Diacerein (30 mg/kg/day) orally daily. Group D (n=15): Diabetic rats + Diacerein (50 mg/kg/day) orally daily. Animals were kept and treated as per the NIH Guideline for Use and Care of Laboratory Animals. Diabetes mellitus was induced via a single intraperitoneal injection of 50 milligram/kg alloxan monohydrated dissolved in aseptic 0.9% saline. After 72 hours, blood specimens were taken from the caudal vein of the rats and glucose level>200 mg/dL was taken as diabetes. Experimental rats were given diacerein approximately 30 and 50 mg orally for 6 weeks. At the completion of experiment the body weight was measured of each animal by electronic measuring balance and blood sample was taken from each animal of all groups to assess the blood glucose level and HbA1c level. Data were recorded via self-made proforma and analysis was done by using SPSS version 20. Results: Average body weight of Diabetic control (Group B) was 193.33±22.50 grams, which was lower in contrast to Diacerein treated group C 202.47±25.70 grams and significantly lower as compared to Diacerein treated group D as  212.6±23.43 grams. A significant increase in blood glucose levels 182.07±10.63 mg/dl was noted in the Diabetic control (Group B) compared to Diacerein treated group C (110.13± 8.54 mg/dl) and group D (85.87±8.41 mg/dl) (P=0.001). HbA1c was markedly raised in the Group B- diabetic controls, while diacerein treated diabetic rats (groups C and D) showed a significant decrease in HbA1c (P=0.001). Conclusion: It was concluded that Diacerein achieves the Euglycemic state by reducing the levels of blood glucose and glycated hemoglobin (HbA1c) in Alloxan-Induced diabetes mellitus in Wistar Albino Rats.

Alhagi maurorum(camel thorn plant) is a promising medicinal plant due to the presence of flavonoids and phenolic compounds as major contents of its constituents. No previous study has been conducted before onA. maurorum extractsas an antioxidative stress and/or antidiabetic herb in STZ-induced DM in rats. Therefore, four groups of rats were allocated as control (C), STZ-induced DM (D), and STZ-induced DM supplemented with 300 mg/kg BW of either aqueous extract (WE) or ethanolic extract (EE) ofA. maurorum. The plasma levels of glucose, TG, TC, LDL-C and VLDL-C, MDA, and bilirubin and the activities of transaminases and GR were significantly increased in the diabetic group. Also, diabetic rats showed severe glucose intolerance and histopathological changes in their livers. In addition, levels of insulin, total proteins, GSH, and HDL-C and the activities of SOD, GPx, and GST were significantly decreased in the diabetic rats compared to those of the control group. The ingestion ofA. maurorumextracts lowered the blood glucose levels during the OGTT compared to the diabetic rats and restored all tested parameters to their normal levels with the exception of insulin level that could not be restored. It is concluded thatA. maurorumextracts decreased elevated blood glucose levels and hyperlipidemia and suppressed oxidative stress caused by diabetes mellitus in rats.

The purpose of this project is to study the anti-diabetic effect of on a diabetic rat model. A total of Twenty male Sprague rats were used and it randomly distributed into four groups which are Group I: , Group II: negative control, Group III: and Group IV: and . In diabetic model were induced with via injection at the dosage of 65mg/kg. and FBG (Fasting Blood Glucose) level of diabetic rats were assessed every three days. Blood was collected via cardiac puncture at day 21 after the induction of treatment. Insulin level of the rats was assessed with the Mercodia Rat Insulin ELISA kit. FBG level of group I (12.16 ±3.96, p<0.05) and group IV (11.34 ±3.67, p<0.05) were significantly decreased. Meanwhile, the for all rats did not show any significant increase. However, the insulin level was escalated in group IV (0.74+0.25, p<0.05) significantly. The present study shows that the and the combination of and lowered blood glucose level and enhanced insulin secretion.

Background: There is a growing interest in the correlation between antioxidants and periodontal disease. In this study, we aimed to investigate the effect of oxidative stress and the impact of two antioxidants, curcumin and rutin, respectively, in the etiopathology of experimentally induced periodontitis in diabetic rats. Methods: Fifty Wistar albino rats were randomly divided into five groups and were induced with diabetes mellitus and periodontitis: (1) (CONTROL)—control group, (2) (DPP)—experimentally induced diabetes mellitus and periodontitis, (3) (DPC)—experimentally induced diabetes mellitus and periodontitis treated with curcumin (C), (4) (DPR)—experimentally induced diabetes mellitus and periodontitis treated with rutin (R) and (5) (DPCR)—experimentally induced diabetes mellitus and periodontitis treated with C and R. We evaluated malondialdehyde (MDA) as a biomarker of oxidative stress and reduced glutathione (GSH), oxidized glutathione (GSSG), GSH/GSSG and catalase (CAT) as biomarkers of the antioxidant capacity in blood harvested from the animals we tested. The MDA levels and CAT activities were also evaluated in the gingival tissue. Results: The control group effect was statistically significantly different from any other groups, regardless of whether or not the treatment was applied. There was also a significant difference between the untreated group and the three treatment groups for variables MDA, GSH, GSSG, GSH/GSSG and CAT. There was no significant difference in the mean effect for the MDA, GSH, GSSG, GSH/GSSG and CAT variables in the treated groups of rats with curcumin, rutin and the combination of curcumin and rutin. Conclusions: The oral administration of curcumin and rutin, single or combined, could reduce the oxidative stress and enhance the antioxidant status in hyperglycemic periodontitis rats.

The present investigation aims to examine the diabetic potential of the plant Ficus racemosa in normal and alloxan induced diabetic rats. The bark extract with water, petroleum ether and with alcohol were screened for blood glucose lowering activity and the alcoholic extract having better therapeutic potential was prepared through Soxhlet extraction for further study. Alcoholic and aqueous extract of bark of Ficus racemosa at a dose of 400 mg/Kg was given to normal and alloxan induced diabetic rats and the blood samples taken from the retero-orbital plexus vein were analyzed for blood glucose level as per standard protocol with available kits through Auto-analyzer. The comparison of blood sugar level as per model schedule showed that in normal group the ethanolic extract, at a dose of 400 mg/Kg intra-peritoneal, the blood glucose lowering 28.66 % while in aqueous extract given group it was 25.90 %. In alloxan induced diabetic rats decrease in blood glucose level in aqueous and ethanolic extract group was found to be 27.01 % and 45.03 % respectively. In conclusion, the ethanolic extract of Ficus racemosa reflected anti-diabetic potential through its glucose lowering activity in experimental animals. It supported the folklore claim of anti-diabetic activity of the plant.

Metformin is now being recognized as the standard therapy in T2D patients who are overweight. Metformin has many drug-disease interactions that can increase the risk of metformin-associated lactic acidosis. Therefore this study was conducted to evaluate any possible pharmacodynamic interactions between metformin and drugs used to treat chronic diseases e.g. Hypertension. The rats were fasted overnight before inducing diabetes with streptozotocin. The rats were given an intraperitoneal injection of streptozotocin (50 mg kg−1) freshly prepared in 0.1M sodium citrate buffer. The diabetic state was confirmed 72 h after streptozotocin injection. Diabetic rats were grouped into seven groups each group of five rats and distributed among the normal control group diabetic control group and the treatment groups. The treatment continued for 10 days. Blood samples were taken before treatment and after 10 days and analyzed for serum glucose, cholesterol, HDL, LDL, and triglycerides. In the diabetic control group which was given STZ alone the blood glucose level decreased significantly (p < 0.05) after 10 days but still above the hyperglycemic level (200mg/dl). The same was observed in the group treated with metformin. The group treated with nifedipine and aspirin showed significant reduction (p < 0.01) in the glucose level below the hyperglycemic level (200mg/dl). While the groups treated with (Metformin + Nifedipine) and (Metformin +Aspirin) showed highly significant reduction (P<0.001) in blood glucose level. These results conclude that the combination of (metformin +Nifedipine) and the combination of (Metformin + Aspirin) have highly significant hypoglycemic effect. It also showed that Nifedipine has promising role in reducing blood glucose level, lipid profile especially LDL-cholesterol, and body weight.

Background: Effective control of blood glucose and activities of antioxidant are key factors that prevent diabetes mellitus (DM) and its complications. There are lots of herbal plants that have those both effects. Ceplikan leaves (Ruellia tuberosa L.) is a traditional medicine which is empirically used to lower blood glucose level. Instead of antioxidant compound, there is assumed other compound in ceplikan leaves that has side effect to pancreatic beta cells.Objective: To identify the effect of ceplikan leaves extract to blood glucose level and pancreas histology description in white diabetic rats (Rattus norvegicus).Method: Thirty subjects of Wistar strain male white rats of 2-3 months old and of 150-200 grams weight were made diabetic with aloxan and randomly divided into 5 groups. Group I consisted of diabetic rats with aquadest, group II with glibenclamide, and Group III-V were given extract of ceplikan leaves in different concentrations that were 1.6 mg, 3.2 mg, and 6.4mg, respectively. Treatment was given orally per day within 30 days. Level of blood glucose was measured in the day of 0, 3, 4, and 30. Statistical analysis used repeated measures and t-test.Result: The supply of ceplikan leaves extract could reduce level of blood glucose of diabetic rats, although the decrease was insignificant. Average diameter of wider Langerhans island occurred to the group of diabetic rats that were given extract of ceplikan leaves dosage 6.4 mg. There was no significant difference (p > 0.05) in changes of blood glucose level before and after experiment in diabetic rats. Pancreas histological description of rats showed that there was improvement as indicated by greater quantity of Langerhans Island and wider diameter of Langerhans Island.Conclusion: Ceplikan leaves was safe and efficacious, so that self-medication of DM using ceplikan leaves could be sustained through formal approach.
