The current study was designed to investigate the antimicrobial and cytotoxic activities of methanol extract (MEGM), petroleum ether fraction (PEFGM), carbontetrachloride fraction (CTFGM), chloroform fraction (CFFGM) and aquous fraction (AQFGM) of stem bark of Glochidion multiloculare (Euphorbiaceae). Antimicrobial activity was evaluated by disc diffusion method and cytotoxic activity by brine shrimp lethality bioassay. In case of antimicrobial screening the CFFGM showed moderate inhibitory activity against B. subtilis having the zone size 12 mm and against Escherichia coli and Escherichia coli were 11 mm, while in the brine shrimp lethality bioassay, the petroleum ether soluble fraction revealed the highest cytotoxicity having LC50 of 3.11 g/ml. The present study was designed to investigate antioxidant, antimicrobial and cytotoxic effects of petroleum ether, chloroform and hydromethanol fraction of methanol extract of Citrus aurantifolia peel. Preliminary phytochemical screening of the fractions was done following the standard procedure. Antioxidant activity was measured using DPPH free radical scavenging assay besides measuring total phenolic and flavonoid content using Folin-Ciocalteu reagent and aluminum trichloride method, respectively. The antimicrobial activity was conducted by disc diffusion method and cytotoxic activity was determined by brine shrimp lethality bioassay. The results of phytochemical screening were indicative of the presence of steroids, alkaloids, saponins, glycosides, flavonoids in the fractions. A dose dependent scavenging activity was observed in DPPH radical scavenging assay where chloroform fraction demonstrated the highest activity with IC50 value of 153.68 ± 3.60 μg/ml. The highest phenolic content was observed in chloroform fraction (308.0 ± 6.55 mg/g gallic acid equivalent) and highest flavonoid content was found in hydromethanol fraction (132.66 ± 2.36 mg/g quercetin equivalent). The chloroform fraction showed excellent antibacterial activity against all the tested bacteria where highest zone of inhibition (19 mm) was produced against Bacillus cereus. In brine shrimp lethality bioassay, LC50 values for petroleum ether, chloroform and hydromethanol fractions were 367.39 μg/ml, 228.14 μg/ml and 296.52 μg/ml, respectively. The present findings suggested that C. aurantifolia peel could be a potent source of medicinally important secondary metabolites and further investigations can be done to identify those active compounds responsible for such bioactivity. The petroleum ether, ethyl acetate and methanol extracts of the stem bark of Callistemon citrinus were subjected to screenings for antimicrobial and antioxidant activities and brine shrimp lethality. The ethyl acetate  extract and its column fractions XVIII and XIV exhibited moderate antimicrobial activity, while the methanol extract  revealed significant antioxidant activity having IC50 of 3.84 ?g/ml. This study was carried out to evaluate the antioxidant, brine shrimp lethality, bioassay and antitumor effects of nhexane, petroleum ether, methanol and chloroform extracts of Annona reticulata leaves. All extracts demonstrated good cupric reducing activity with increasing concentrations keeping methanol extract at the top and n-hexane extract at the second position. The methanol extract was found to have the highest antioxidant capacity 28±26.88 followed by n-hexane 16.95±11.39 and chloroform 12.85±10.32 in the form of mg of ascorbic acid equivalent per gram of crude extract. The methanol extract also showed the highest values of 2.41±0.963 mg of gallic acid equivalent phenolic content and 6.54±4.08 mg of quercetin equivalent flavonoid content per g of crude extract. In the brine shrimp lethality bioassay, LC50 values of methanol, chloroform, petroleum ether, and n-hexane extracts were found to be 1.084?g/ml, 1.096?g/ml, 1.995?g/ml and 3.126 ?g/ml, respectively. These findings suggest that A. reticulata leaves can be a good source of antioxidants and possess significant cytotoxic effect. In a narrow spectrum investigation on antitumor activity of the extracts, we found that the methanol extract inhibited the tumor growing process (control group, chloroform group and petroleum ether group showed significant rise in body weight; i.e., 8% - 14% of initial weight compared to that of methanol group showing a rise of 0.45% - 0.94% of initial weight) and improved ILS (Percentage Increase in Life Span) with increasing doses (Doses of 50mg/kg, 100mg/kg and 200mg/kg showed ILS values of 13.70%, 19.20% and 21.92%, respectively) to some extent when compared with control group and groups receiving other extracts.Bangladesh J Microbiol, Volume 29, Number 2, Dec 2012, pp 70-74

The work described in this paper details the biological investigation on Bombax ceiba, species of Malvaceae. The methanol crude extract of Bombax ceiba was fractionated with kupchan method and n-hexane, carbon tetrachloride, chloroform fraction were made for  screening the antimicrobial and antitumor potentials using disc diffusion method and brine shrimp lethality bioassay respectively. An established antibiotic (kanamycin, 30µg/disc) and cytotoxic agent (vincristine sulphate) were used to compare the results. From the graphs the concentration of methanolic crude extract give LC50 (50% mortality) value of 3.90mg/ml. LC90 was also determined from the graph to establish the therapeutic index and the value was found 150.0mg/ml. The four fractions were assayed for antimicrobial screening and the carbon tetrachloride fraction showed most prominent zone of inhibition against a number of bacterial and fungal strains.Keywords: Bombax ceiba; Brine shrimp lethality bioassay; Disc diffusion method.© 2011 JSR Publications. Scopoletin (1) was isolated from the chloroform soluble fraction of a methanol extract of the stem bark of Phyllanthus reticulates (Family: Euphorbiaceae). The petroleum ether, carbon tetrachloride and choloroform soluble fractions of this methanol extract were subjected to antimicrobial screening and brine shrimp lethality bioassay. All of the partitionates showed moderate to strong inhibitory activity to microbial growth while the chloroform soluble fraction showed strongest cytotoxicity having LC50 1.99 ?g/ml. The methanol extract of the stem bark of Anthocephalus chinensis as well as its n-hexane, carbon tetrachloride, chloroform and aqueous soluble fractions were subjected to brine shrimp lethality bioassay and microbiological investigation. In the brine shrimp lethality bioassay, the aqueous soluble partitionate of the methanolic extract revealed the highest cytotoxicity having LC50 of 1.19 mg/ml while in case of antimicrobial screening, the chloroform soluble materials demonstrated moderate inhibition of growth of test organisms. The antibacterial activity of methanol extract from the root bark of Akanda (Calotropis gigantea L.) and its petroleum ether, chloroform and ethyl acetate fractions were investigated. Both of methanol extract and its chloroform fraction showed activity against Sarcina lutea, Bacillus megaterium and Pseudomonas aeruginosa. Petroleum ether fraction showed activity against Bacillus subtilis and Shigella sonnei whereas ethyl acetate fraction showed activity against Pseudomonas aeruginosa and Escherichia coli at 20ìg/disc, 30ìg/disc and 40ìg/disc doses. Among the tested materials, methanol extract and its chloroform fraction showed comparatively better results. Minimum inhibitory concentration (MIC) for methanol extract and each fraction were also determined by serial dilution technique. Antibacterial activity of Moringa oleifera Lam. stem bark has been tested against Staphylococcus aureus and Escherichia coli. The extraction of Moringa oleifera Lam. stem bark was used maceration method with methanol solvent and has obtained extract yield of 6.1%. The antibacterial activity test of Moringa oleifera stem bark extracts used a well diffusion method. The concentration of Moringa oleifera stem bark extract was varied to four concentrations of 1% 2% 3% 4% (w/v). The inhibition zone of methanol extract of Moringa oleifera stem bark against Staphylococcus aureus at extract concentrations of 1%, 2%, 3%, and 4% was 10.08 mm, 11.8 mm, 15.00 mm, and 17.02 mm, respectively. The methanol extract of Moringa oleifera stem bark at concentrations of 1%, 2%, 3%, and 4% could also inhibition of Escherichia coli growth with inhibition zone of 14.01 mm, 16.50 mm, 17.09 mm, and 17.10 mm, respectively. The fractionated crude extracts and three isolated pure compounds XM-1, XM-2 and XM-3 from stem bark of Xylocarpus mollucensis were screened for their antibacterial and antifungal activities and cytotoxicity against brine shrimp nauplii. Petroleum ether, ethyl acetate (EtOAc) and methanol (MeOH) extracts and the compounds isolated from EtOAc fractions were studied for their antimicrobial activities. Cytotoxic activities were conducted only with EtOAc extract and its selected fractions. The EtOAc extract showed promising antimicrobial activities against all the gram positive and gram negative bacteria whereas petroleum ether extract showed moderate activities and the MeOH extract did not show any antimicrobial activities. The isolated pure compounds XM-1, XM-2 and XM-3, whose structures were not elucidated, exhibited activities against most of the bacterial strains. The cytotoxicity towards brine shrimp nauplii of the crude EtOAC extract and its selected fractions were studied. The LC50 values of the EtoAc extract was 12.6 ?g/ml and for the fractions 2, 5, 8 and 13 were 17.78, 13.34, 14.13 and 15.85 ?g/ml, respectively. In this study, three compounds, stigmasterol (1), lupeol (2), 16-hydroxylupeol (3) were isolated from the petroleum ether soluble fraction of a methanol extract of whole plant of Justica gendarussa (Burm. f). The crude methanol extract as well as its petroleum ether, carbon tetrachloride and chloroform soluble Kupchan fractions were studied for antioxidant, antimicrobial and cytotoxic activities. Among the different fractions tested for antioxidant activity, the chloroform soluble partitionate was the most potent with IC50 value of 18.80 μg/ml as compared to tertbutyl- 1-hydroxytoluene (IC50=17.69 μg/ml). Antimicrobial screening of the different extractives was conducted by the disc diffusion method and all the fractions except chloroform soluble partitionate exhibited mild antimicrobial activity with zone of inhibition ranging from 7-10 mm. In brine shrimp lethality bioassay, the petroleum ether soluble materials demonstrated the highest toxicity with LC50 of 1.27μg /ml. This is the first report of isolation of compounds from J. gendarussa, and its antioxidant, antimicrobial and cytotoxic properties.
