The present study was designed to investigate antioxidant, antimicrobial and cytotoxic effects of petroleum ether, chloroform and hydromethanol fraction of methanol extract of Citrus aurantifolia peel. Preliminary phytochemical screening of the fractions was done following the standard procedure. Antioxidant activity was measured using DPPH free radical scavenging assay besides measuring total phenolic and flavonoid content using Folin-Ciocalteu reagent and aluminum trichloride method, respectively. The antimicrobial activity was conducted by disc diffusion method and cytotoxic activity was determined by brine shrimp lethality bioassay. The results of phytochemical screening were indicative of the presence of steroids, alkaloids, saponins, glycosides, flavonoids in the fractions. A dose dependent scavenging activity was observed in DPPH radical scavenging assay where chloroform fraction demonstrated the highest activity with IC50 value of 153.68 ± 3.60 μg/ml. The highest phenolic content was observed in chloroform fraction (308.0 ± 6.55 mg/g gallic acid equivalent) and highest flavonoid content was found in hydromethanol fraction (132.66 ± 2.36 mg/g quercetin equivalent). The chloroform fraction showed excellent antibacterial activity against all the tested bacteria where highest zone of inhibition (19 mm) was produced against Bacillus cereus. In brine shrimp lethality bioassay, LC50 values for petroleum ether, chloroform and hydromethanol fractions were 367.39 μg/ml, 228.14 μg/ml and 296.52 μg/ml, respectively. The present findings suggested that C. aurantifolia peel could be a potent source of medicinally important secondary metabolites and further investigations can be done to identify those active compounds responsible for such bioactivity. The current study was designed to investigate the antimicrobial and cytotoxic activities of methanol extract (MEGM), petroleum ether fraction (PEFGM), carbontetrachloride fraction (CTFGM), chloroform fraction (CFFGM) and aquous fraction (AQFGM) of stem bark of Glochidion multiloculare (Euphorbiaceae). Antimicrobial activity was evaluated by disc diffusion method and cytotoxic activity by brine shrimp lethality bioassay. In case of antimicrobial screening the CFFGM showed moderate inhibitory activity against B. subtilis having the zone size 12 mm and against Escherichia coli and Escherichia coli were 11 mm, while in the brine shrimp lethality bioassay, the petroleum ether soluble fraction revealed the highest cytotoxicity having LC50 of 3.11 g/ml. The work described in this paper details the biological investigation on Bombax ceiba, species of Malvaceae. The methanol crude extract of Bombax ceiba was fractionated with kupchan method and n-hexane, carbon tetrachloride, chloroform fraction were made for  screening the antimicrobial and antitumor potentials using disc diffusion method and brine shrimp lethality bioassay respectively. An established antibiotic (kanamycin, 30µg/disc) and cytotoxic agent (vincristine sulphate) were used to compare the results. From the graphs the concentration of methanolic crude extract give LC50 (50% mortality) value of 3.90mg/ml. LC90 was also determined from the graph to establish the therapeutic index and the value was found 150.0mg/ml. The four fractions were assayed for antimicrobial screening and the carbon tetrachloride fraction showed most prominent zone of inhibition against a number of bacterial and fungal strains.Keywords: Bombax ceiba; Brine shrimp lethality bioassay; Disc diffusion method.© 2011 JSR Publications. Wulfenia amherstiana belongs to the Scrophulariaceae family and various plants of this family are known for their biological activities. The present study was focused on the isolation of bioactive compounds including a novel flavone 6,7,4′-trimethyl flavone (TMF) along with three known flavonoids such as quercetin, rutin, and a steroid β-sitosterol which were isolated from the ethanolic extract of W. amherstiana (Himalayan Wulfenia) through column chromatography and purified by using HPLC. Their structures were identified and elucidated through electron ionization mass spectroscopy (EIMS), 1DNMR (1H-NMR and 13C-NMR), and 2DNMR (COSY, HMQC, and HMBC) spectroscopy. The antimicrobial activities of this novel compound were evaluated through agar well diffusion method, while antioxidant and cytotoxic activities were assessed through 2,2-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging assay and brine shrimp lethality assay, respectively. The NMR data revealed that TMF is a novel compound. TMF showed potential antibacterial and antifungal activities against Staphylococcus aureus (MIC = 128 μg/ml) and Candida albicans (MIC = 128 μg/ml). The cytotoxic potential of TMF was determined from brine shrimp lethality assay with LD50 of 127.01 μg/ml. The free-radical scavenging potential of TMF at various concentrations implicated its strong antioxidant activity in vitro. The results revealed that TMF demonstrated substantial antimicrobial activity against S. aureus and C. albicans, strong antioxidant activity, and moderately cytotoxic activity.

A total of four compounds namely betulin (1), erythrodiol (2), lupeol (3), and stigmasterol were isolated from the leaf extract of Ixora arborea for the first time. The structures of the isolated compounds were established by extensive spectroscopic studies. In our preliminary screening, the petroleum ether, carbon tetrachloride, dichloromethane and aqueous soluble fractions of the methanol extract were subjected to antioxidant, antimicrobial and brine shrimp lethality bioassays. All of the fractions showed significant antioxidant activity along with the total phenolic content, of which the methanolic extract of leaves (MEL) showed the highest free radical scavenging activity with IC50 value 33.0 μg/ml. In the brine shrimp lethality bioassay, among all the extractives the petroleum ether soluble fraction (PESF) demonstrated significant cytotoxicity with LC50 value of 0.622 μg/ml. However, in case of antimicrobial screening, only the methanolic crude extract showed mild growth inhibitory activity against the tested microorganisms. <p><strong>Objective: </strong>To evaluate the antimicrobial activity and cytotoxicity of hexane, ethyl acetate and methanol extracts of <em>Alysicarpus vaginalis </em>var<em>. nummularifolius </em>(DC) along with phytochemical analysis.</p><p><strong>Methods: </strong>The crude extract of hexane (AVH), ethyl acetate (AVE) and methanol (AVM) of <em>Alysicarpus vaginalis </em>var<em>. nummularifolius </em>(DC) were prepared and analysed for phytochemical constituents using standard methods. The cytotoxicity activity of the plant extracts was predicted using brine shrimp lethality assay (BSLA). The antimicrobial activity and the minimal inhibitory concentration (MIC) of the plant extracts were examined against 5 bacterial and 2 fungal strains using agar well diffusion method, and two fold serial dilution method, respectively.</p><p><strong>Results: </strong>The phytochemical screening studies showed a higher concentration of saponins, alkaloids, flavonoids, phenols, quinones and terpenoids in AVM than the other two extracts. The LC<sub>50</sub> value of AVH and AVE were found to be 900.05 µg/ml and 754.35 µg/ml respectively using BSLA while that of AVM was&gt;1000 ug/ml. All the extracts of the plant showed antimicrobial activity against most of the test organisms. The MIC values of AVM were lower than AVE for all the microbial strains except for <em>Pseudomonas aeroginosa</em> where AVE (107.87µg/ml) exhibited higher value than AVM (51µg/ml).</p><p><strong>Conclusion: </strong>The present study concluded AVM with a high presence of phytochemicals. The AVE and AVM were found to possess promising antimicrobial activity when compared with the standards. The AVM exhibited lesser toxicity when compared with AVH and AVE.<strong></strong></p><p> </p>

The objective of this study was to evaluate the phytochemical and pharmacological screening of fruit extracts of Aegle marmelos using various in vitro methods. The antioxidant activity of the extracts was evaluated by using DPPH free radical scavenging and reducing power method, while well diffusion method was used for antimicrobial activity. Phytochemical screening indicated the presence of flavonoids, and phenolics were abundantly present in methanolic and aqueous extracts of A. marmelos. Methanolic extract showed significant antioxidant potential, which was however inferior to that of ascorbic acid. The methanolic extract showed maximum zone of inhibition against the S. aureus (18 mm) and it was higher than that of the standard tetracycline. The HPLC data predict that the presence of scopoletin, marmelosin and umbelliferone, which may contribute to their pharmacological action.

Excessive of ultraviolet light causes abnormality of melanin production. Antioxidants and antityrosinase agents are able to reduce hyperpigmentation by interrupting the process of melanin production. The purpose of this study is to examine the antioxidant and antityrosinase activities as well as toxicity of both 80% ethanol and aqueous extracts of Alpinia nigra by DPPH free radical scavenging assay, mushroom tyrosinase assay and brine shrimp lethality bioassay. Alpinia nigra extracts showed positive result on antioxidant and antityrosinase activities. We found that extract of A. nigra’s leaf has the most effective activity of antioxidant and antityrosinase among other parts of this plant. The ethanol and aqueous extracts from the leaf of A. nigra at the concentration of 125 μg/mL showed % inhibition for free radical scavenging as 94.97% and 93.35%, respectively. The IC50 values of antioxidant were 39.83±16.21 and 46.33±15.22 μg/mL, respectively. In addition, ethanol extract of the leaf from A. nigra at the concentration of 1,000 μg/mL produced 92.61% inhibition of mushroom tyrosinase activity, whereas aqueous extract of A. nigra’s leaf at the same concentration produced 74.47% inhibition. The IC50 of antityrosinase activities were 142.81±13.32 and 406.88±66.43 μg/mL for ethanol and aqueous extracts, respectively. Moreover, the brine shrimp lethality bioassay showed that all extracts were non-toxic (LC50 >1,000 μg/mL). In conclusion, the ethanol extract of A. nigra’s leaf may be beneficial and provide the novel and safe source for antioxidant and whitening agent.

This study was carried out to evaluate the antioxidant, brine shrimp lethality, bioassay and antitumor effects of nhexane, petroleum ether, methanol and chloroform extracts of Annona reticulata leaves. All extracts demonstrated good cupric reducing activity with increasing concentrations keeping methanol extract at the top and n-hexane extract at the second position. The methanol extract was found to have the highest antioxidant capacity 28±26.88 followed by n-hexane 16.95±11.39 and chloroform 12.85±10.32 in the form of mg of ascorbic acid equivalent per gram of crude extract. The methanol extract also showed the highest values of 2.41±0.963 mg of gallic acid equivalent phenolic content and 6.54±4.08 mg of quercetin equivalent flavonoid content per g of crude extract. In the brine shrimp lethality bioassay, LC50 values of methanol, chloroform, petroleum ether, and n-hexane extracts were found to be 1.084?g/ml, 1.096?g/ml, 1.995?g/ml and 3.126 ?g/ml, respectively. These findings suggest that A. reticulata leaves can be a good source of antioxidants and possess significant cytotoxic effect. In a narrow spectrum investigation on antitumor activity of the extracts, we found that the methanol extract inhibited the tumor growing process (control group, chloroform group and petroleum ether group showed significant rise in body weight; i.e., 8% - 14% of initial weight compared to that of methanol group showing a rise of 0.45% - 0.94% of initial weight) and improved ILS (Percentage Increase in Life Span) with increasing doses (Doses of 50mg/kg, 100mg/kg and 200mg/kg showed ILS values of 13.70%, 19.20% and 21.92%, respectively) to some extent when compared with control group and groups receiving other extracts.Bangladesh J Microbiol, Volume 29, Number 2, Dec 2012, pp 70-74

The antioxidant, antibacterial and cytotoxic properties of methanol extract of Urtica crenulata (syn: Laportea crenulata Gaud) stem has been investigated in the present study. The antioxidant and cytotoxic activity of the Urtica crenulata methanol extract were assessed by DPPH free radical scavenging method and brine shrimp lethality bioassay method, respectively. The antibacterial activity of the plant extract (500 μg/disc) was also carried out by disc diffusion technique. Stem extract showed DPPH (2,2-diphenyl-1-picrylhydrazyl) free radical scavenging effect compared with ascorbic acid. IC50 value of ascorbic acid and stem extract was found 14.72 μg/ml and 1468.9 μg/ml, respectively. In antibacterial experiment, Urtica crenulata stem extract showed 8, 14 and 10 mm of diameter of zone inhibition against Salmonella typhi, Shigella flexneri and Shigella sonnei, respectively and 9 and 8 mm of diameter of zone inhibition against Bacillus subtilis and Bacillus cereus but no activity was observed against Staphylococcus aureus. In brine shrimp lethality assay, the LC50 value of the extract was found 104.0 μg/ml, which indicates that the extract has high cytoxic effect. The present study demonstrates that methanol extract of Urtica crenulata stem has significant cytotoxic effect. Ruta angustifolia was used in this study in order to evaluate the antimicrobial activity and antioxidant properties and its correlation with the polyphenolic content. Two Gram-positive bacteria (Staphylococcus aureus ATCC 25923, Bacillus cereus ATCC 11778) and two Gram–negative bacteria (Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 8739) were used to determine the antibacterial activity. Aqueous maceration extract was used for antioxidant activities and methanolic maceration extract was used for antibacterial activity. The antioxidant properties and activities were evaluated by using total phenolic content (TPC), total flavonoid content (TFC), DPPH free radical scavenging activity and beta-carotene bleaching method. Whereas, the antibacterial activity was examined using disc diffusion method against selected microorganism at concentration 1.0 mg/disc. The results showed the phenolic content of R. angustifolia extract was 18.89 g GAE/100 g extract while the flavonoid content was 14.170 g QE/100 g extract. R. angustifolia exhibited good radical scavenging with IC50 value of 2.04 mg/ml. The result for disc diffusion method showed no inhibition zone against all the strains of bacteria at 1.0 mg/disc concentration of the extract. Based on the results, it can be concluded that the R. angustifolia aqueous extract has the antioxidant properties and there is correlation between polyphenolic content of the extract with its antioxidant activity. However, R. angustifolia methanolic extract did not show any antibacterial activity.
