Aeromonas hydrophila bacteria was isolated from the suspected EUS-affected shing fish, Heteropneustes fossilis (Bloch). The disease investigations were primarily based on clinical signs and subsequently confirmed by the isolation of bacterial pathogen Aeromonas hydrophila from lesion of liver and kidney. The A. hydrophila isolates were identified by a series of morphological, physiological and biochemical tests. The total bacterial load in liver, intestine and kidney were 1.67 × 104 to 6.46 × 108 CFU/g, 1.71 × 103 to 1.18 × 109 CFU/g and 1.47 × 104 to 3.70 × 108 CFU/g respectively.DOI: http://dx.doi.org/10.3329/pa.v19i1.17362 Progress. Heteropneustes fossilis (shing) of 35g body weight were experimentally infected with Aeromonas hydrophila by two different methods: viz. intraperitonial and intramuscular injection. A standard dose of infection (6.4 × 107 CFU/ml) was selected based on predetermined LD50. Each method gave rise to the mortality of shing up to 85%. Clinical signs of fish included injury, hemorrhage and large ulcerative lesions on the body at the injected area. In kidney, liver and intestine pathologically, massive atrophy and focal necrosis were found. Hemorrhage, Vacuolation and atrophy of hepatic sinusoids represented by necrosis of the sinusoidal lining cells, degeneration of hepatic tissue and distribution of bacterial cell all over the tissue were found in liver. Atrophy, hemorrhage, villi missing and missing of epithelium were found in intestine of the experimentally infected shing. Bacterial cells were distributed in the whole hematopoietic tissue including the renal tubules. Tissue abscess characterized by focal necrosis, hemorrhage and Vacuolation were also found in the kidney of the infected shing. An experiment was carried out to investigate the health status of Hypophthalmichthys molitrix among three different fish farms at Mymensingh district of Bangladesh. Fish sampling and water quality parameters were collected on a monthly basis. Samples of skin, muscle, gills, liver and kidney were examined through histological technique. Results from water quality parameters suggested that water temperature had a distinct effect on fish health of H. molitrix, which was mostly evident during the colder months. Infectious symptoms were usually evident in July to October and the severity of pathology was increased in November and December. These were evident from their clinical signs that include scale loss, haemorrhagic lesion, and reddish spots or from their histopathological symptoms such as necrosis, pyknosis, inflammation, haemorrhage, hypertrophy, missing of gill lamellae, vacuums, tubular and pillar cells degeneration, inflammation, clubbing, and suspected bacterial colonies. Results suggest that the health condition of the fishes from BAU fish farm was the best over the other two farms, whereas Government fish farm followed by NGO fish farm were severely affected with EUS, protozoan and bacterial infection. Moreover, internal organs like liver and kidney were more affected than skin, muscle and gills.

The pathogenic bacteria, Aeromonas hydrophila from naturally infected silver carp, Hypophthalmichthys molitrix were isolated and identified from a fish farm in Mymensingh. Fish showed motile Aeromonas septicemia (MAS) like reddish head and anal region, pale body colour and external haemorrhages. Intestine, liver, kidney and environmental samples such as sediment, feed and water were inoculated onto the Aeromonas isolation medium (AIM) and TSA plates. The AIM colonies were undergone specific characterization for Aeromonas hydrophila. Quantitative studies of bacterial flora of fish on TSA plates showed that the total bacterial load in intestine, liver and kidney of the sampled fish, sediment, feed and water of the environment were 1.00 × 105 to 1.50 × 105 CFU/g, 2.7 × 102 to 4.46 × 104 CFU/g, 1.00 × 103 to 2.17 × 103 CFU/g, 1.90 × 104 CFU/g, 4.10 × 103 CFU/g and 2.50 × 103 CFU/ml, respectively. After isolation, A. hydrophila was finally identified by their specific morphological, physiological and biochemical characteristics. They were gram negative, rod shaped, motile bacteria that showed positive reaction for oxidase and catalase, fermented glucose and were resistant to vibriostatic agent 0/129. The study  aimed to show the effect of experimental infection with E.coli O157:H7 on some liver and kidney function in adult rabbits. Twenty five domestic rabbits (males and females), aged 6-8 weeks and weighing 1500-2000g, had a negative fecal bacteriological culture of E.coli O157:H7 were used. The rabbits were randomly divided into two groups: infected group (15 rabbits), each animal was drenched  orally with 1ml of Phosphate buffer saline containing (2×109CFU) of E.coli O157:H7, while the control group (10 rabbits) were drenched orally (1ml) of Phosphate buffer saline, the clinical signs were daily observed for 30 days, recording of body weight and blood samples were collected at (0, 3, 15 and 30) days for serum   to evaluate  biochemical tests including Alanine transaminase, Aspartate aminotransaminase and alkaline phosphatase, creatinine, urea  and total serum protein. The results showed different clinical signs after inoculation, including diarrhea, lethargy, weight loss, inability to eat or drink, urine was cloudy or milky, while control group remains in normal condition. The serum biochemical tests in infected group showed a significant increase of three enzymes at 15, and 30 days,   the highest values of Alanine transaminase; Aspartate aminotransaminase and alkaline phosphatase showed at 30 days post infection as compared with the control group. Serum creatinine and urea recorded a significant increase (P< 0.05) at 30 day post infection as compared with control group. However, total protein revealed a significant decrease in the infected group, and a significant decrease was noticed at 15 day post infection, while the control group showed an increase in total protein co-ordinated with increase of body weight and progressing age. This study concluded that the infection with E. coli O157:H7may cause changes in liver and kidney functions which were more sever at 15,30 days post infection.

The plant of castor is widely spread in the Iraqi land, and characterized with containing ricin toxin, which has a very serious effects, and because the seeds of this plant scattered in the agricultural soil and rivers water, which increases the exposure of humans and animals to these beans. Objective: This experiment was designed to study the effect of high concentration of castor bean powder in some physiological and biochemical parameters and changes in some tissues of the body, as well as trying to use doxycycline to reduce the effects of ingestion of these seeds. Materials and Methods: In the experiment, 24 local rabbits were raised and fed in the Animal House of the Faculty of Medicine / Al-Muthanna University, then divided into four groups and treated for three weeks (21 days), Control group: treated with normal saline solution (0.9) orally throughout the experiment, G1: was treated orally with a concentration of 25 mg / kg of castor bean powder daily during the experiment, G2 : orally treated 25 mg / kg of castor bean and 25 mg / kg of doxycycline, G3: orally treated 25 mg / kg of castor powder with 50 mg / kg of doxycycline daily throughout the trial period. Results: The results of the experiment showed significant changes (P less than 0.05) in all physiological and biochemical blood tests when compared with control group. There was a significant decrease in PCV, Hb, RBC, T.protein and body weights, while demonstrated a significant increase in WBC, Urea, Creatinine, ALT, AST and ALP, with distortions in liver and kidney of animals that treated with Castor beans. In contrast, the treatment with doxycycline and caster beans showed significant improvement reflected by a normal proportion in physiological tests and biochemical tests with improvement in the tissues when compared to control group. Conclusions: It can be concluded from this study that castor bean has high toxic and pathogenic effects that may be dangerous to the life of the organism. Therefore, it is advisable to be cautious of these pills and avoid exposure to them, also recommended to take high concentrations of doxycycline treatment when infected with castor bean poisoning.

Purpose: This research was aimed to screen and identify multifunctional phosphorus-dissolving bacteria of a Chinese fir (Cunninghamia lanceolata) plantation and study its phosphorus-dissolving characteristics in order to provide strain resources and a theoretical basis for developing the appropriate bacterial fertilizer of a Chinese fir plantation. Methods: First, phosphorus-dissolving bacteria were isolated from the woodland soil of a Chinese fir plantation by Pikovskava inorganic phosphorus medium (PVK). Then, some growth-promoting indicators of primary screening strains were determined, including the capacity of phosphorus-solubilized, nitrogenase activity, 1-aminocyclopropane-1-carboxylic acid (ACC) deaminase activity, production of indole-3-acetic acid (IAA), secretion of iron carrier and so on. Finally, the screening multifunctional phosphorus-dissolving bacteria were identified, which were combined with colony characteristics, physiological and biochemical tests and molecular biotechnology. Results: (1) Thirteen phosphorus-dissolving bacteria were isolated and screened in total, and P5 (195.61 mg·L−1) had the strongest capacity of phosphorus-solubilized. Five phosphorus-dissolving bacteria were provided with nitrogenase activity, and the highest activity of nitrogenase was P10 and P5 (71.90 C2H4 nmol·mL−1·h−1 and 71.00 C2H4 nmol·mL−1·h−1, respectively). Four strains were provided with ACC deaminase activity, and the highest activity of ACC deaminase was P5 and P9, (0.74 μmol·mg−1·h−1 and 0.54 μmol·mg−1·h−1, respectively). Most strains could secrete IAA, and three strains of bacteria had a strong secretory ability, which could secrete IAA with a concentration greater than 15 mg·mL−1, and P5 was 18.00, P2 was 17.30, P6 was 15.59 (mg·mL−1). P5 produced carriers of iron better than others, and the ratio of the diameter of the iron production carrier ring to the diameter of the colony was 1.80, respectively, which was significantly higher than other strains. Combining all kinds of factors, P5 multifunctional phosphorus-dissolving bacteria were screened for eventual further study. (2) Strain P5 was identified as Burkholderia ubonensis, based on the colony characteristics, physiological and biochemical tests, 16SrDNA sequence analysis and phylogenetic tree construction. Conclusion: P5 has a variety of high-efficiency growth-promoting capabilities, and the ability to produce IAA, ACC deaminase activity and siderophore performance are significantly higher than other strains, which had great potential in the development of microbial fertilizer.

AbstractAnimal manure application to soils is considered to be one of the main cause of antibiotic and bacterial pathogen spread in the environment. Pig livestock, which is the source of one of the most used fertilizer for cultivated land, is also a hotspot for antibiotics and antibiotic-resistant bacteria. Besides harsh chemical and physical sanitization treatments for the abatement of antibiotics and bacterial load in livestock waste, more sustainable and environmentally friendly strategies need to be considered. In this context, the use of natural substances which are proved useful for pest and disease control is currently under exploration for their role in the reduction of bacterial pathogen population. Among these, plants and derived products from the Brassicaceae family, characterized by the presence of a defensive glucosinolate-myrosinase enzymatic system, have been successfully exploited for years in agriculture using the so-called biofumigation technique against crop diseases. Although the application of biofumigation to suppress a range of soil borne pests has been well documented, no studies have been examined to reduce bacterial population in animal waste. In the present study, the release and the antibacterial activity of bioactive compounds deriving from different Brassicaceae defatted seed meals against pathogens and bacterial population in pig manure is addressed. Rapistrum rugosum and Brassica nigra defatted seed meals were found to be the most active products against tested pathogens and able to significantly reduce the bacterial load in the manure.

A novel, extremely halophilic archaeon B3T was isolated from shrimp-salted seafood. Its morphology, physiology, biochemical features and 16S rRNA gene sequence were characterized. Strain B3T is non-motile, Gram-variable, requires at least 10 % (w/v) NaCl for growth and grows in the ranges of 21–50 °C and pH 6.5–9.0. The DNA G+C content of strain B3T was 63.2 mol%. Phylogenetic analysis based on the 16S rRNA gene sequences indicated that strain B3T belonged to the genus Halalkalicoccus and was phylogenetically closely related to the type strain Halalkalicoccus tibetensis (98.64 %). However, DNA–DNA hybridization experiments showed 7.0 % relatedness between strain B3T and a strain of a reference species of the genus Halalkalicoccus. Combined analysis of 16S rRNA gene sequences, DNA–DNA relatedness data, physiological and biochemical tests indicated that the genotypic and phenotypic characteristics differentiate strain B3T from other Halalkalicoccus species. On the basis of the evidence presented in this report, strain B3T represents a novel species of the genus Halalkalicoccus, for which the name Halalkalicoccus jeotgali. sp. nov. is proposed. The type strain is B3T (=KCTC 4019T=DSM 18796T=JCM 14584T=CECT 7217T).

Aeromonas hydrophila is an opportunistic microorganism. It is a secondary biological agent that contributes to the occurrence of fish diseases and its deterioration. This research was undertaken to determine the prevalence of A. hydrophila in some freshwater fishes collected from three different fish markets of Dhaka City and to test their antibiotic susceptibility. Total bacterial count and total aeromonas on different aeromonas selective media were enumerated using serial dilution technique. Bacterial isolates were characterized to identify A. hydrophila using biochemical tests and with comparison to reference strain (ATCC 7966). The lowest Aeromonas count was detected to be 2.83±0.40×102 cfu/g in Anabas testudineus and the highest was 1.03±0.153×103 cfu/g in Oreochromis mossambicus. On market basis highest aeromonas count was found in Anando Bazar (8.10±1.09×102 cfu/g) and lowest in Hatirpool Bazar (5.63±0.90×102 cfu/g) with no significant difference. Maximum susceptibility to amikacin and gentamicin was observed whereas all of the isolates were found resistant to a commonly used antibiotic amoxycillin. The obtained results point that antimicrobial susceptibility was more or less similar regardless of the origin of the samples collected. All the fishes investigated in this study contained A. hydrophila in their different organs.

In November 2018, a horse owner from the Czech Republic experienced the loss of two of his stallions in consequence of probable poisoning. The affected animals were kept in the stable only. The mares, which were grazing on pasture during the day, were not affected. All the animals were fed the same type of feed. The day before the animals became sick, between 15:00 to 19:00 h the stable was not secured against the entry of strangers. When the mares were brought back to the stable and all animals were closed and locked for the night, no signs of health issues were observed. In the following morning, the two stallions were found in lateral recumbency with seizure attacks. Despite symptomatic treatment (infusion with saline, flunixine meglumine, activated charcoal) and applications of myorelaxants, the symptoms persisted until the afternoon, and the owner agreed with euthanasia. Based on the clinical signs and pathological examination, poisoning by pesticides was suspected. The results of toxicological analyses revealed that carbofuran, metaldehyde, and strychnine were not responsible for the poisoning as they were not detected in the samples. The only positive finding was a high aluminium concentration in the stomach content, liver and kidney samples. Based on the observed symptoms and the high aluminium concentration, it was presumed that aluminium phosphide could be the source of poisoning. As there was no evidence of the possession of such product by the horse breeder and the stable stayed unsupervised for a few hours, intentional poisoning cannot be excluded.
