ABSTRAKSemen adalah cairan yang dikeluarkan oleh organ kelamin ternak jantan yang secara normal diejakulasikan ke dalam saluran reproduksi ternak betina melalui kopulasi. Evaluasi semen merupakan alat utama untuk memprediksi kinerja  reproduksi ternak ayam  jantan. Evaluasi kualitas semen penting dilakukan untuk menyelesaikan salah satu masalah utama dalam peternakan pembibitan ayam, yaitu   rendahnya fertilitas.  Faktor terbesar dari sumber masalah tersebut terkait dengan rendahnya kualitas semen ayam. Kualitas semen ayam  lebih banyak dipengaruhi oleh faktor non genetik, seperti faktor pakan dan nutrisi. Tujuan dari penulisan makalah ini adalah untuk mengkaji hasil-hasil studi terkait pengaruh pakan terhadap kualitas semen ayam. Hasil studi menunjukkan bahwa faktor pakan sangat mempengaruhi kualitas semen ayam, terutama imbangan energi metabolis-protein dalam pakan ayam jantan sangat penting diatur untuk menjaga berat badan ayam jantan agar tidak terjadi kelebihan atau kekurangan berat badan yang sangat mempengaruhi kualitas semen ayam. Suplementasi suplemen dan imbuhan pakan menjadi upaya yang dapat dilakukan untuk meningkatkan kualitas semen ayam. Jenis suplementasi yang dapat dilakukan dengan bahan sumber antioksidan, asam amino, mikromineral, bahan mengandung bioaktif tanaman, bahan sumber asam lemak dan probiotik. Disimpulkan bahwa faktor pakan sangat menentukan terhadap kualitas semen ayam.Kata kunci: ayam jantan, kualitas semen, pakanABSTRACTRooster semen is a liquid released by the genitals of the roosters which is normally ejaculated into the female reproductive tract during copulation. Semen evaluation is the main way to predict the reproductive performance of roosters. Semen quality evaluation is important to solve one of the main problems in poultry breeding farms, namely low fertility, whereas the biggest factor of this problem source is related to the poor of roosters’ semen quality. The quality of roosters semen is more influenced by non-genetic factors, such as feed and nutrition. The purpose of this paper was to examine the results of studies so far related to the effect of feed on the roosters’ semen quality. The results of the study indicated that the feed factor greatly influences the quality of roosters semen. The metabolic energy-protein balance in the rooster's feed was very important to regulate to maintain the weight of the rooster so as not to overweight and underweight because bodyweight greatly affects the quality of roosters semen. Supplementation of feed supplements and feed additives was an effort that can be done to improve the roosters semen quality. The kind of supplementation that could be done was supplementation of antioxidant sources, amino acids, microminerals, materials containing plant bioactive, fatty acid source ingredients, and probiotics. It was concluded that the feed factor was crucial to the quality of semen quality produced by the roosters.Keywords: feed, rooster, semen quality

The viscous seminal plasma (SP) is currently a major impediment to the handling of ejaculate and the development of some biotechnologies in South American camelids. The vas deferens-collected spermatozoa of alpacas is a useful technique to avoid this problem. On the other hand, SP contains a large protein component that has been implicated in the function of spermatozoa within the female reproductive tract. In this sense, the low fertility achieved using transcervical insemination with frozen-thawed spermatozoa in alpacas could be improved by adding SP. This study aimed to evaluate the effect of the whole SP on some in vitro parameters of alpaca spermatozoa after the freezing-thawing-process and the fertility after artificial insemination. It would contribute to a better understanding of the interaction between thawed sperm cells and SP. Spermatozoa were obtained by surgically diverted vas deferens. The samples were diluted with a Tris-based extender, packaged in straws, and frozen. At thawing, each straw was divided into two post-thawing conditions: with the addition of 10% of PBS (control) or with 10% SP (treatment). The sperm cells were evaluated using dynamic parameters, sperm cell morphology, and morphometry. Fertility was assessed by an artificial insemination trial. All in vitro parameters were analyzed by ANOVA. A heterogeneity test was scheduled for the fertility trial. After the freezing-thawing process, motility and plasma membrane functionality was improved when SP was added. No differences were found for post-thaw viability between the control and treatment samples. The percentage of normal cells was higher with SP at post-thawing, and a decrease of the presence of bent tailed spermatozoa with a droplet in the SP group was observed. The length of the head spermatozoa was 3.4% higher in the samples with PBS compared to those in which SP was added. Females pregnant at day 25 post-insemination were 0/12 (with SP inside the straw) and 1/10 (without SP inside the straw). In conclusion, the presence of 10% SP at post-thawing improves sperm cells' motility, functionality, and morphology, indicating that it would be beneficial to improve the frozen-thawed alpaca's physiology spermatozoa. More fertility trials must be developed to increase this knowledge.

Selfish genetic elements (SGEs) are ubiquitous in eukaryotes and bacteria, and make up a large part of the genome. They frequently target sperm to increase their transmission success, but these manipulations are often associated with reduced male fertility. Low fertility of SGE-carrying males is suggested to promote polyandry as a female strategy to bias paternity against male carriers. Support for this hypothesis is found in several taxa, where SGE-carrying males have reduced sperm competitive ability. In contrast, when SGEs give rise to reproductive incompatibilities between SGE-carrying males and females, polyandry is not necessarily favoured, irrespective of the detrimental impact on male fertility. This is due to the frequency-dependent nature of these incompatibilities, because they will decrease in the population as the frequency of SGEs increases. However, reduced fertility of SGE-carrying males can prevent the successful population invasion of SGEs. In addition, SGEs can directly influence male and female mating behaviour, mating rates and reproductive traits (e.g. female reproductive tract length and male sperm). This reveals a potent and dynamic interaction between SGEs and polyandry highlighting the potential to generate sexual selection and conflict, but also indicates that polyandry can promote harmony within the genome by undermining the spread of SGEs.

The uterus is responsible for the nourishment and mechanical protection of the developing embryo and fetus and is an essential part in mammalian reproduction. The Mayer-Rokitansky-Kuester-Hauser (MRKH) syndrome is characterized by agenesis of the uterus and upper part of the vagina in females with normal ovarian function. Although heavily studied, the cause of the disease is still enigmatic. Current research in the field of MRKH mainly focusses on DNA-sequencing efforts and, so far, failed to decipher the nature and heterogeneity of the disease, thereby holding back scientific and clinical progress. Here, we developed long-term expandable organoid cultures from endometrium found in uterine rudiment horns of MRKH patients. Phenotypically, they share great similarity with healthy control organoids and are surprisingly fully hormone responsive. Transcriptome analyses, however, identified an array of dysregulated genes that point at potentially disease-causing pathways altered during the development of the female reproductive tract. We consider the endometrial organoid cultures to be a powerful research tool that promise to enable an array of studies into the pathogenic origins of MRKH syndrome and possible treatment opportunities to improve patient quality of life.

AbstractThe establishment of repositories of frozen semen, for the conservation of agricultural genetic resources, is not a simple matter of collecting and freezing semen in the hope that one day it will be suitable for use in an artificial insemination procedure. Important genetic issues need to be considered; for example, how many samples should be stored and from how many individuals? Aside from these, many biological and logistic issues must be considered. Cryopreservation technology does not work equally well in all species, often because of anatomical differences in the female reproductive tract leading to significant variability in the number of spermatozoa needed in order to achieve an acceptable conception rate. Moreover, spermatozoa from different species are not equally susceptible to cryoinjury. However, it is also emerging that semen samples from individuals within a species are also of different quality; several studies have revealed that these differences reflect the quality of DNA within the spermatozoon itself and also the efficacy of biochemical functions, including metabolic and signalling systems, within individual cells. As new possibilities to select spermatozoa for insemination arise, especially the use of flowsorting for gender selection, these issues may become more significant. In this article we interpret the way in which some of this new information may impact upon the practical implementation of genetic resource conservation.


Endometriosis is a chronic disease of the female reproductive system, which is characterised by the presence of endometrial tissue outside the uterus especially in the pelvic cavity that includes ovaries, fallopian tubes and it may occur extra genitally in some rare cases. 176 million women all over the world including 26 million in India suffer from endometriosis and high frequency was observed between the age group of 26-30 years, resulting in increased primary infertility. Various studies surveying the female reproductive tract have confirmed the existence of microbiota starting in the vagina, cervix, uterus, fallopian tubes and ovaries. These microbes play a crucial role at different stages ofthe reproduction, in forming a physical barrier against pathogen and the stimulation of host defence mechanisms. Direct and indirect mechanisms are observed including the production of biochemically active compounds that directly kill or inhibit pathogens. Women suffering from endometriosis lead an impaired quality of life and continue to deal with endometriosis-associated symptoms even after diagnosis and treatment of the disease. In endometriosis condition, adhesions are observed in the fallopian tubes and ovaries that block tubal motility and damage the oocyte-pickup. Progesterone resistance is familiar in endometriosis which changes the implantation window period causing the loss of implantation markers and finally leading to infertility. Malignant tumours of ovaries have also been identified and known to arise from endometriosis. Microbiome can be used as a novel diagnostic tool for endometriosis as there is a variation observed in the composition and distribution along the female reproductive tract of healthy women and endometriosis patients. A non-invasive diagnosis is achieved through this, aiming at early diagnosis and alternative treatment for endometriosis.

The aim: This study aims to develop and implement a complex of pregravid preparation based on endogenous melatonin level, development of oxidative stress and violation of apoptosis regulation in women with tubal-peritoneal infertility. Materials and methods: We have examined 100 women of reproductive age, including 65 women with tubal-peritoneal infertility treated by ART. All studied women were defined levels of endogenous melatonin, oxidative status and level of apoptosis in the endometrium and in follicular fluid (infertile women only). As a powerful free radical neutralizer, we used melatonin to improve the microenvironment of maturing oocytes and create favourable conditions for implantation in the endometrium. Results: Treatment by exogenous melatonin showed better results than standard therapy without antioxidants: the quality of oocytes and embryos, the level of ongoing pregnancies and childbirth was significantly higher. Conclusions: We achieved such results due to restoring the balance in pro / antioxidant system and apoptosis in female reproductive tract.

Candida albicans, along with other closely related Candida species, are the primary causative agents of vulvovaginal candidiasis (VVC)—a multifactorial infectious disease of the lower female reproductive tract resulting in pathologic inflammation. Unlike other forms of candidiasis, VVC is a disease of immunocompetent and otherwise healthy women, most predominant during their child-bearing years. While VVC is non-lethal, its high global incidence and profound negative impact on quality-of-life necessitates further understanding of the host and fungal factors that drive disease pathogenesis. In this review, we cover the current state of our understanding of the epidemiology, host response, fungal pathogenicity mechanisms, impact of the microbiome, and novel approaches to treatment of this most prevalent human candidal infection. We also offer insight into the latest advancements in the VVC field and identify important questions that still remain.

The article deals with the possibility of using feed additives in the feeding of broiler chickens and the peculiarities of their effect on the poultry organism. Contemporary data on the biological role of nutrients and energy in the feeding of broiler chickens are presented. The characteristics of the main feeds for poultry and the ways of reducing their prime cost in the conditions of broiler farms are given. The basic principles of effective feeding of poultry with forages of different content of metabolic energy and crude protein are considered, taking into account their assimilation in the body. Provided compound feed recipes and practical tips. It is established that high growth rate of broiler chickens in the start and groove periods (5–39 days) is provided by high-protein compound feeds – 23% (1–2 weeks), 21% (3–5 weeks) and exchange energy of 298 kcal and 305 kcal, respectively. With further cultivation in the finish forage the protein content in the forage decreases and the level of metabolic energy increases (up to 309 kcal). However, for each cross applies its own technology of cultivation and different rates of nutrients in raw materials. It is clear that effective breeding of broiler chickens is not possible without the proper rationing and use of protein and feed energy. Of course, provided that the supply of biologically active substances also corresponds to the normalized amount. Thus, the quality of poultry products is directly dependent on the quality of feed and the efficiency of transformation of feed energy into products.
