The current study evaluated the effects of a methanol extract from Chaetomorpha aerea (a green alga) on non-specific immune responses and resistance against Edwardsiella tarda infection in Labeo rohita. Different doses of the extract (5, 50 and 500 mg/kg of body weight) were injected into the fish intraperitoneally while a control group was injected with 0.2 mL of sterile physiological salt solution. Variations in several immunostimulatory parameters (i.e., neutrophil, serum lysozyme, myeloperoxidase, serum antiprotease, and ceruloplasmin activity), reactive oxygen species (ROS) and reactive nitrogen species (RNS) were assessed after 7, 14, 21, and 28 days of post stimulation. E. tarda culture was injected into the fish after 28 days of post stimulation to induce infection to monitor fish mortality within 14 days. Interestingly, all doses of methanolic extract enhanced neutrophil, lysozyme, and myeloperoxidase activity, ROS and RNS, while a dose of 50 mg/kg was the most effective. Fish injected with this optimal dose were also protected against infection with virulent strain of E. tarda. The results of the study suggest that C. aerea extract is a potential prophylactic agent against bacterial infections in finfish.

The current study focused on assessing the outcome of methanol extract of Chaetomorpha antennina (MECA) on the growth performance and immune modulation in both specific and non-specific immune responses through the assessment of neutrophil, serum lysozyme, serum myeloperoxidase, antiprotease, ceruloplasmin, reactive oxygen species (ROS), and reactive nitrogen species (RNS) activity in Labeo rohita (rohu) at 28 days post treatment along with assessment of the disease resistance capacity against Edwardsiella tarda at 30days post immunization. Fishes (n = 144; average weight 50.0 ± 0.23 g) were evenly divided into four treatments, with 12 fishes per tank in triplicates. The MECA was injected intraperitoneally in the fishes at different doses as 0, 25, 75, and 150 mg/kg of the body weight. The results demonstrated that fish treated with MECA have an increased body weight, specific growth rate, and feed conversion ratio (p < 0.05) with respect to the control group. Results suggested that the MECA inclusion can significantly enhance (p < 0.05) the levels of serum lysozyme, neutrophil function, serum antiprotease activity, cellular RNS, and ROS production. Exposure to MECA of 75 mg/kg showed a significantly higher survival percentage against E. tarda disease infection. These results indicate MECA as a stimulant of immunity in L. rohita against E. tarda. The results suggested that MECA is a potent immunostimulant in finfish aquaculture and can offer higher economic welfare.

AbstractDue to increased anthelmintic resistance, complementary methods to drugs are necessary to control gastrointestinal nematodes (GIN). Vaccines are an environmentally-friendly and promising option. In a previous study, a Teladorsagia circumcincta recombinant sub-unit vaccine was administered to two sheep breeds with different levels of resistance against GIN. In the susceptible Canaria Sheep (CS) breed, vaccinates harboured smaller worms with fewer eggs in utero than the control group. Here, we extend this work, by investigating the cellular and humoral immune responses of these two sheep breeds following vaccination and experimental infection with T. circumcincta. In the vaccinated CS group, negative associations between antigen-specific IgA, IgG2 and Globule Leukocytes (GLs) with several parasitological parameters were established as well as a higher CD4+/CD8+ ratio than in control CS animals, suggesting a key role in the protection induced by the vaccine. In the more resistant Canaria Hair Breed (CHB) sheep the vaccine did not significantly impact on the parasitological parameters studied and none of these humoral associations were observed in vaccinated CHB lambs, although CHB had higher proportions of CD4+ and CD8+ T cells within the abomasal lymph nodes, suggesting higher mucosal T cell activation. Each of the component proteins in the vaccine induced an increase in immunoglobulin levels in vaccinated groups of each breed. However, levels of immunoglobulins to only three of the antigens (Tci-MEP-1, Tci-SAA-1, Tci-ASP-1) were negatively correlated with parasitological parameters in the CS breed and they may be, at least partially, responsible for the protective effect of the vaccine in this breed. These data could be useful for improving the current vaccine prototype.

  Objective: Salmonella enteritidis ghosts (SEGs) is a non-living empty bacterial cell envelopes which were generated using a different concentration of sodium hydroxide (NaOH) 6.4 mg/mL and evaluated as a vaccine candidate in specific pathogen-free (SPF) chicken. SEGs have been produced by chemical-mediated lysis and evaluated the potential efficacy of chemically induced SEG vaccine and its ability to induce protective immune responses against virulent S. enteritidis challenge in SPF chickens.Methods: SPF chickens were divided into three groups: Group A (non-vaccinated control), Group B (vaccinated with prepared vaccine), and Group C (vaccinated with commercial vaccine).Results: Vaccination of SPF chicken with SEGs induced higher immune responses before and after virulent challenge. SPF chicken vaccinated with SEGs showed increasing in serum enzyme-linked immunosorbent assay (ELISA) antibodies. During the vaccination period, Groups B and C showed higher serum antibody titer compared to Group A. The minimal inhibitory concentration (MIC) of NaOH was capable of inducing non-living SEGs, and it has successfully generated non-living SEGs by MIC of NaOH.Conclusion: It is a one-step process which means easy manufacturing and low production cost compared to protein E-mediated lysis method. Chemically induced SEG vaccine is a highly effective method for inducing protective immunity. This study strongly suggests that SEGs will be a permissive vaccine, as the method of inhibition of S. enteritidis was safe and cheaper than other methods, and it gave a good protection.

SUMMARYSalmonella Enteritidis is the main cause of foodborne salmonellosis worldwide. The limited effectiveness of current interventions against this pathogen has been the main incentive to develop new methods for the efficient control of this infection. To investigate the use of DNA vaccines against S. Enteritidis in humans, immune responses stimulated by two plasmids containing the genes designated SEN1002, located in the pathogenicity island SPI-19 and encoding a Hcp protein involved in transport mechanisms, and SEN1395, located in the genomic island ΦSE14 and encoding a protein of a new superfamily of lysozymes, were evaluated. Humoral and cellular responses following intranasal immunization of two groups of BALB/c mice with the plasmids pV1002 and pV1395 plus adjuvant were evaluated and it was observed that the IgG2a/IgG1 ratios were sixfold higher than control groups. Both plasmids stimulated specific secretory IgA production. Increased proliferation of lymphocytes and IFN-γ production were detected in both experimental groups. DNA-vaccinated mice developed protective immunity against a virulent strain of S. Enteritidis, with nearly 2 logs of protection level compared to the negative control values in the spleen. Therefore, DNA vaccines are efficient at stimulating cellular and humoral immune responses at systemic and mucosal levels.

This study was conducted to know some Immune responses in women with vaginal candidias is by Candidia species. Hemoglubin (Hb) was decreased significantly in patient women comparing with control group. The differential count of Blood cells revealed that neutrophils and monocytcs were increased significantly in patient women comparing with control group. The percentage of the Phagocytosis index was decreased significantly in patient women (66.45 + 15.05) comparing with control group (73.72 + 3.77) and the T- lymphocytes were decreased also in patient women (57.75 + 18.787) comparing with control group (74.25 + 7.759).

Abstract BACKGROUND: The effects of inorganic trace minerals (ITM) replaced by low-dose glycine-complexed trace minerals (GCM) on growth, serum parameters, immunity, intestinal morphology, and mineral excretion in piglets were investigated. One hundred and twenty-eight weaned piglets (14.18 ± 0.33kg body weight (BW)) were randomly assigned to 4 treatments with 4 replicates, 8 piglets per replicate. Treatments consist of: (T1) basal diet + 100% inorganic trace mineral (ITM) as the control group (20 ppm Cu, 150 ppm Fe, 150 ppm Zn, and 30 ppm Mn from sulfates); (T2) basal diet + 50% ITM (Cu, Fe, Zn, and Mn from sulfates, 50% of control) + 50% organic trace minerals (OTM, Cu, Fe, Zn, and Mn from glycine complexed trace minerals (GCM), 50% of control); (T3) basis diet + 50% OTM from GCM; (T4) basal diet + 70% OTM from GCM. The feeding period lasted 28 d and was divided into 2 stages (0 to 14 d and 15 to 28 d). After feeding trial , 6 pigs per treatment were randomly selected to slaughter for sampling. RESULTS: Average daily gain, feed intake, and G:F were not affected by dietary treatments during the overall period. During the second, and the overall feeding phases, the digestibility of Zn and Fe in T3 and T4 was higher than that of T1 ( P < 0.05). The concentration of serum ferritin in T2 was significantly higher than T3 and T4. Serum immunoglobulin A concentration in the ileal mucosa of T2 was higher than that of T1 ( P < 0.05), and the higher duodenum villus height was observed in T4 compared with the rest treatments ( P < 0.05). The lowest trace mineral excretion was overserved in T3 ( P < 0.01); in addition, the urinary concentrations of Zn and Fe in T2 were lower than that in T1 ( P < 0.05). CONCLUSION: These results indicate that GCM have higher bioavailability than ITM, and that supplementation of low-dose GCM to replace full dose ITM could reduce mineral excretion without affecting performance, blood profiles, immune responses, and intestinal morphology in piglets.
