The plant phyllosphere is colonized by a complex ecosystem of microorganisms. Leaves of raw eaten vegetables and herbs are habitats for bacteria important not only to the host plant, but also to human health when ingested via meals. The aim of the current study was to determine the presence of putative probiotic bacteria in the phyllosphere of raw eaten produce. Quantification of bifidobacteria showed that leaves of Lepidium sativum L., Cichorium endivia L., and Thymus vulgaris L. harbor between 103 and 106 DNA copies per gram fresh weight. Total cultivable bacteria in the phyllosphere of those three plant species ranged from 105 to 108 CFU per gram fresh weight. Specific enrichment of probiotic lactic acid bacteria from C. endivia, T. vulgaris, Trigonella foenum-graecum L., Coriandrum sativum L., and Petroselinum crispum L. led to the isolation of 155 bacterial strains, which were identified as Pediococcus pentosaceus, Enterococcus faecium, and Bacillus species, based on their intact protein pattern. A comprehensive community analysis of the L. sativum leaves by PhyloChip hybridization revealed the presence of genera Bifidobacterium, Lactobacillus, and Streptococcus. Our results demonstrate that the phyllosphere of raw eaten produce has to be considered as a substantial source of probiotic bacteria and point to the development of vegetables and herbs with added probiotic value.

Occurrence of mineral forming and other bacteria in mats is well demonstrated. However, their high diversity shown by ribotyping was not explained, although it could explain the diversity of formed minerals. Common biomarkers as well as phylogenic relationships are useful tools to clustering the isolates and predict their potential role in the natural niche. In this study, combination of MALDI-TOF MS with PCA was shown a powerful tool to categorize 35 mineral forming bacterial strains isolated from Dohat Fshaikh sabkha, at northwest of Qatar (23 from decaying mats and 12 from living ones). 23 strains from decaying mats belong to Virgibacillus genus as identified by ribotyping and are shown highly involved in formation of protodolomite and a diversity of minerals. They were used as internal references in categorization of sabkha bacteria. Combination of isolation of bacteria on selective mineral forming media, their MALDI TOF MS protein profiling and PCA analysis established their relationship in a phyloproteomic based on protein biomarkers including m/z 4905, 3265, 5240, 6430, 7765, and 9815. PCA analysis clustered the studied strains into 3 major clusters, showing strong correspondence to the 3 phyloproteiomic groups that were established by the dendrogram. Both clustering analysis means have evidently demonstrated a relationship between known Virgibacillus strains and other related bacteria based on profiling of their synthesized proteins. Thus, larger populations of bacteria in mats can be easily screened for their potential to exhibit certain activities, which is of ecological, environmental and biotechnological significance.

Manufacturing of novel foodstuffs supplemented with live probiotic bacteria has recently been intensively investigated. The supplementation of confectionery with probiotics is troublesome since some unit technological processes are conducted at high temperatures and the products are usually stored at ambient temperature. Our group has developed a method of the production of milk chocolate, sweetened with either sucrose or isomalt and aspartame, containing 32, 36, or 40 g/100 g fat, and supplemented with live cells of probiotic bacterial strains: Lactobacillus casei and paracasei. This new milk chocolate displayed the same sensory properties as the reference, probiotic-free chocolate. The number of live bacterial cells was maintained at the functional level of 10<sup>6</sup> &divide; 10<sup>8</sup> cfu/g after keeping for 12 months irrespective of the temperature. The highest number of live probiotic bacteria survived in the chocolate kept at 4&deg;C. Thus the product can be regarded as functional food.

At present, plant diseases caused by soil borne plant pathogens have major constraints on crop production. Which include genera Fusarium spp, Phytophtora spp, Sclerotinia and Altenaria. Due to this reason, chemical fungicides are routinely used to control plant disease, which is also true in Mongolian case. However, use of these chemicals has caused various problems including environmental pollution with consequence of toxicity to human health also resistance of some pathogens to these fungicides are present. Fortunately, an alternative method to reduce the effect of these plant pathogens is the use of antagonist microorganisms. Therefore, some species of the genus Bacillus are recognized as one of the most effective biological control agent.Our research was focused to isolate Bacillus licheniformis, with antifungal potential, from indigenous sources. In the current study, 28 bacterial cultures were isolated from soil and fermented mare’s milk also named as koumiss. Isolated bacterial cultures were identified according to simplified key for the tentative identification of typical strain of Bacillus species. As a result 8 strains were positive and further screened for antifungal activity against Fusarium spp and Alternaria solani. Out of these 8 strains 5 strains are selected based on their high effectiveness against fungal pathogens and for further confirmation Polymerase Chain reaction run for effective bacterial strains using specific primers B.Lich-f and B.Lich-r.

Acetone extracts of selected plant species were evaluated for theirin vitrocytotoxicity against a noncancerous African green monkey kidney (Vero) cell line and an adenocarcinoma cervical cancer (HeLa) cell line. The plants studied wereOriganum vulgareL. (Oregano),Rosmarinus officinalisL. (Upright and ground cove rosemary),Lavandula spicaL. (Lavender),Laurus nobilisL. (Bay leaf),Thymus vulgarisL. (Thyme),Lavandula x intermediaL. (Margaret Roberts Lavender),Petroselinum crispumMill. (Curly leaved parsley),Foeniculum vulgareMill. (Fennel), andCapsicum annuumL. (Paprika). Antioxidant activity was determined using a quantitative DPPH (1,1-diphenyl-2-picryl hydrazyl) assay. The rosemary species exhibited effective radical scavenging capacity with 50% inhibitory concentration (IC50) of3.48±0.218 μg/mL and10.84±0.125 μg/mL and vitamin C equivalents of 0.351 g and 1.09 g for McConnell’s Blue and Tuscan Blue, respectively. Cytotoxicity was measured using XTT (Sodium3′-[1-(phenyl amino-carbonyl)-3,4-tetrazolium]-bis-[4-methoxy-6-nitro] benzene sulfonic acid hydrate) colorimetric assay. OnlyL. nobilisandO. vulgareexhibited pronounced effects on the HeLa cell line. Dose-dependent studies revealed IC50of34.46±0.48 μg/mL and126.3±1.00 μg/mL on the HeLa cells and on the Vero cells 124.1 μg/mL ± 18.26 and 163.8 μg/mL ± 2.95 forL. nobilisandO. vulgare, respectively. Light (eosin and haematoxylin staining) and confocal microscopy (Hoechst 33342, acridine orange, and propidium iodide staining) were used to evaluate the cytotoxic mechanism of action forL. nobilisandO. vulgare.

The application of living bacteria as probiotics in food or food supplements requires a careful safety assessment. This review summarizes key issues concerning the safety aspects of bacteria added to particular products marketed for improvement of general health or treatment of (post)infectious symptoms. The bacteria used in such products should be completely safe; however, it can be challenging to provide evidence for absence of all virulence properties. In some cases, virulence factors have been detected in probiotic bacterial strains, and the implications of these traits for safety assessments are discussed. Horizontal gene transfer can result in acquisition of virulence genes or antimicrobial resistance in probiotic bacteria. Antimicrobial resistance in these bacteria can possibly aid the spread of undesired resistance in intestinal bacterial populations. The relative risk of such gene transfers is considered. The generation of complete bacterial genome sequences can both resolve and create safety issues. Current practices of safety assessment procedures in the United States and the European Union are briefly reviewed and a future outlook is provided.

AbstractDue to their small sizes and high reactivity, nanoparticles have a completely different toxicity profile than larger particles, and it is difficult to predict their potential ecological impact. There is a need for broad ecotoxicological studies of nanomaterials in order to specify their environmental impact and ensure safe application of nanotechnology products. In this work, we have assessed the toxicity of Au and Au/ZnO metal nanoparticles obtained with the use of Tanacetum parthenium (herba) extract. The obtained nanoparticles were characterized by UV–Vis spectrophotometry (UV–VIS), Transmission electron microscopy (TEM), Atomic force microscopy (AFM), and Fourier transform infrared spectroscopy (FTIR). In order to assess the toxicity of biologically synthesized nanoparticles, we used seeds of various plants: Lepidium sativum, Linum flavum, Zea mays, Salvia hispanica-chia, Lupinus angustifolius, Petroselinum crispum subsp. Crispum, Beta vulgaris, Phaseolus vulgaris. The in vitro phytotests showed that gold nanoparticles at a specific range of concentrations for all plants stimulated their growth. The highest growth activity was exhibited by the solution at the concentration of 0.300 mg/ml towards corn (Aw ≈ − 135 ± 16) and flax (Aw ≈ − 44 ± 10). Only for parsley the IC50 was determined at 0.57 mg/ml, but solutions at the concentration of 0.030 to 0.150 mg/ml also stimulated plant growth. Au/ZnO had a toxic effect at all concentrations applied in the study.

Conjugation between microsymbiont was used to create genetic variations in rhizobia for diverse rabi legumes (chickpea, pea and lentil) with better characteristics in nodulation and nitrogen fixation process. Ten antibiotics were used as selectable markers for the screening of twenty four bacterial strains to be used in mating experiments for obtaining transconjugants. All bacterial strains were sensitive to gentamycin and resistant to streptomycin, kanamycin and sulphanilamide. Total five fusants were obtained from each rhizobial cross combination with the help of electro-poration. Modified transconjugants, rhizobial strains had promiscuous infection with 50-122% more nodules showed significant increase in shoot fresh weight, dry weight and total nitrogen content in chickpea, pea and lentil plants. Electrofusantsrhizobial strains improved shoot nitrogen content up to 67% in lentil and 54% in pea and chickpea plants. The amount of nitrogen fixed in chickpea was highest (3.71gm) by transconjugants DP-C6- HLN followed by DP-C6-HP14 (3.56gm). Transconjugants DP-HP14-HLN fixed the highest amount of nitrogen (3.92gm) in pea and 4.06gm in lentil plants. Plasmids were also analyzed in order to characterize their role in the evolution of rhizobial symbionts and their involvement in symbiotic behaviour. The developed Rhizobium strains with improved symbiotic association and ability to infect across strict specificity for host legumes would be of great help for the farming community at large.
