ABSTRACT Areca nut (Areca vestiaria Giseke), which is a kind of wild palm, is a multi-functional plant. The North Sulawesi people empirically use this plant to cure various diseases. This study aims to determine the phytochemical content and antibacterial activity of ethanol extracts of areca nut against the inhibitory growth of Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa using 5 concentrations namely 1%, 2%, 3%, 4% and 5%. Extraction was done by maceration using 96% of ethanol. Antibacterial activity testing was using paper disc diffusion method (Kirby and Bauer diffusion). The results of this study indicate that the ethanol extracts of areca nut stem contains flavonoids, saponins, triterpenoids that have the potential to be antibacterial and has antibacterial strength against Escherichia coli, Staphylococcus aureus, and Pseudomonas aeruginosa, at concentrations of 4% and 5%, which are concentrations which are included in the group that categorize as strong to inhibit the bacteria E. coli and P. aeruginosa, while the extract concentration of 2%, 3%, 4%, and 5% which are included in the group that categorize as strong to inhibit the S. aureus bacteria. Keywords: Agar diffusion method, Antibacterial, Areca nut, Phytochemical. ABSTRAK Pinang Yaki (Areca vestiaria Giseke) yang merupakan sejenis palem liar, merupakan tanaman yang multi fungsi. Masyarakat Sulawesi Utara secara empiris menggunakan tanaman ini untuk menyembuhkan berbagai macam penyakit.. Penelitian ini bertujuan untuk mengetahui kandungan fitokimia dan aktivitas antibakteri dari ekstrak etanol tangkai buah pinang yaki terhadap daya hambat pertumbuhan bakteri Escherichia coli, Staphylococcus aureus, dan Pseudomonas aeruginosa menggunakan 5 konsentrasi yakni 1%, 2%, 3%, 4%, dan 5%. Ekstraksi dilakukan dengan cara maserasi menggunakan etanol 96%. Pengujian aktivitas antibakteri menggunakan metode difusi dengan cakram kertas (difusi Kirby dan Bauer). Hasil penelitian ini menunjukkan bahwa ekstrak etanol tangkai buah Pinang Yaki memiliki kandungan flavonoid, saponin, triterpenoid yang berpotensi sebagai antibakteri dan memiliki kekuatan antibakteri terhadap Escherichia coli, Staphylococcus aureus, dan Pseudomonas aeruginosa, pada konsentrasi 4% dan 5% merupakan konsentrasi yang termasuk dalam golongan kuat untuk menghambat bakteri E. coli dan P. aeruginosa, sedangkan pada konsentrasi ekstrak 2%, 3%, 4%, dan 5% termasuk dalam golongan kuat menghambat bakteri S. aureus. Kata kunci: Pinang Yaki, Fitokimia, Antibakteri,metode difusi agar

This study tests the antibacterial activities of Moringa oleifera leaf extract against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa, obtained from Microbiology laboratory, Al-Hikmah University Ilorin. Phytochemical analysis reveals the presence of tannins in all the three extracts (Aqueous, ethanolic and N-hexane), while. flavonoids, alkaloids and Saponins were only present in the Aqueous and ethanolic extracts. The leaves extracts were screened for antibacterial activity by agar well diffusion method, employing five different extracts concentrations (100mg/ml, 80mg/ml, 60mg/ml, 40mg/ml and 20mg/ml). The aqueous extracts had a mean activity of 11.50±0.70mm, 7.50±0.70mm and 8.5±0.70mm for S. aureus, E. coli and P. aeruginosa respectively. The ethanol extracts had a mean activity of 12.00±1.41mm, 10.00±1.41mm and 8.00±1.21mm for S. aureus, E. coli and P. aeruginosa respectively, while the N-hexane extracts exert no any activity. The antibiotics discs exert more inhibitory effect compared to the extract. The MIC for the aqueous extracts are at a concentrations of 60mg/ml for S. aureus, 80mg/ml for P. aeruginosa and 80mg/ml for E. coli. While that of the ethanol extract of the leaves are at a concentration of 60mg/ml for S. aureus, 80mg/ml for E. coli and 100mg/ml for P. aeruginosa. The MBC for the aqueous Extract of the leaves are at a concentration of 80mg/ml for S. aureus, 100mg/ml for P. aeruginosa and 100mg/ml for E. coli, while that of the ethanol extract are at a concentration of 80mg/ml for S. aureus and 100mg/ml for E. coli), while no any MBC was recorded for P. aeruginosa. The antibacterial activity tests indicate that the ethanol extract was more active. Among the three isolates, S. aureus is more sensitive to the aqueous and ethanol extracts. The activity exhibited by the extracts may be related to the presence of a number of Phytoconstituents.

An exploratory study based on laboratory experiment was carried out to determine the antibacterial effect of Dimethyl sulfoxide (DMSO) extract of Aloe vera leaf gel (DAE) against standard strains of Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli and Klebsiella pneumoniae in the Department of Pharmacology & Therapeutics in collaboration with the Department of Microbiology, Mymensingh Medical College, Mymensingh, Bangladesh. DMSO extract was used in five different concentrations (100, 200, 300, 400 and 500 μg/ml). Dose dependent inhibitory effect was seen against the test organisms using disc diffusion method. Zone of inhibition (ZOI) were 8 mm, 13 mm, 15 mm, 16 mm and 21 mm against S. aureus; 0 mm, 8 mm, 13 mm, 15 mm and 18 mm against P. aeruginosa; 8 mm, 11 mm, 13 mm, 16 mm and 20 mm against E. coli; 0 mm, 9 mm, 12 mm, 14 mm and 18 mm against K. pneumoniae at 100, 200, 300, 400 and 500 μg/ml respectively. The minimum inhibitory concentration (MIC) was assessed by broth dilution technique. The MICs of DAE for S. aureus, P. aeruginosa, E. coli and K. pneumoniae were 300 μg/ml, 400 μg/ml, 400 μg/ml and 450 μg/ml respectively. From the study it was observed that DMSO extract of Aloe vera leaf gel possesses antibacterial effect against the test pathogens. The findings highlight the need for further extensive study to detect and isolate the biologically active ingredients present in the Aloe vera leaves which are responsible for antibacterial effect. This study aimed to determine the effect of concentration ethanol extract from tembelakan leaf (Lantana camara Linn)  on bacteria growth of Staphylococcus aureus and Escherichia coli. Extraction was done by maceration using ethanol 96% and then separated using rotary evaporator. Antibacterial activity test of the ethanol extract by Well agar diffusion method. Variation in crude extract saponin used in this study was 5%, 10%, 15%, 20%, 25% and positive controls were used for comparison with Amoxicilin and Chloramphenicole concentration of 25 µg/mL and DMSO as a negative control. The results of antibacterial activity test is indicated by the formation of growth inhibitory region S. aureus and E. coli. The result of growth inhibitory regions was analyzed by One way ANOVA. One way ANOVA test results indicate that there are effects of ethanol extract concentration of tembelekan leaf (L. camara Linn) against S. aureus and E. coli. Effective concentration of ethanol extract tembelekan leaf (L. camara Linn) when compared with positive control to inhibit the growth of S. aureus and E. coli is at 25% with a relatively strong antibacterial activity. Test with phytochemicals screening method which is showed that tembelekan leaf contains the flavanoid, saponins, and tannins compounds as antibacterial

Background: Eucheuma spinosum seaweed contain flavonoid, triterpenoid, alkaloids and polyphenol which has been widely used in antibacterial activity. Purpose(s):The aim of the research are to determine antibacterial activities of metanol extracts of E. spinosum against Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922; to formulate hand wash of seaweed of methanol extract E. spinosum that has antibacterial activity and has physical and chemical stability. Methods: Seaweed E.spinosum methanol extract was derived by maceration method. Antibacterial activities of the extract were tested by liquid dilution and solid dilution method. Hand wash was formulated by mechanical dissolved methods. Antibacterial activity of hand wash were tested by liquid dilution and solid dilution method. Physical and chemical stabilities were conducted by cycling test. Results:. These were showed through minimum inhibitory concentration (MIC) of ethanol extracts of E. spinosum against S. aureus ATCC 25923 at concentrations of 6% and E. coli ATCC 25922 at concentrations of 6%. The minimum bactericidal concentration (MBC) of ethanol extracts of E. spinosum against S. aureus ATCC 25923 at concentrations of 8% and E. coli ATCC 25922 at concentrations of 8%. Formulation of hand wash seaweed E. spinosum methanol extract at concentrations of 8% and 10%. The test of stabilities results of hand wash changes were organoleptic, viscosity, pH, and foaming ability were accordance to qualified standard. The antibacterial activity of hand wash contains seaweed E. spinosum methanol extract at concentration of  8% and 10% have bactericidal activity againts S. aureus ATCC 25923 and E. coli ATCC 25922. Conclusion: Overall, these results suggested that formula of hand wash contains E. spinosum metanol extracts have antibacterial properties against S. aureus ATCC 25923 and E. coli ATCC 25922.Keywords: antibacterial, Eucheuma spinosum, hand wash, physical and chemical stability Latar Belakang: Rumput laut Eucheuma spinosum mengandung flavonoid, triterpenoid, alkaloid, dan polifenol yang memiliki aktivitas antibakteri. Tujuan: Penelitian ini bertujuan mengetahui aktivitas antibakteri ekstrak metanol E. spinosum terhadap Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922; membuat sediaan sabun cuci tangan dari ekstrak metanol E. spinosum yang memiliki aktivitas antibakteri dan stabil secara fisika dan kimia. Metode: Ekstrak metanol E. spinosum diperoleh dengan metode maserasi. Uji aktivitas antibakteri ekstrak dilakukan dengan metode dilusi cair dan dilusi padat. Sabun cuci tangan diformulasi dengan metode pencampuran mekanik. Uji aktivitas antibakteri sabun cuci tangan dilakukan dengan metode dilusi cair dan dilusi padat. Uji stabilitas fisika kimia dilakukan dengan metodecycling test. Hasil: Konsentrsi Hambat Minimum (KHM) ekstrak metanol E. spinosum terhadap S. aureus ATCC 25923 adalah 6% dan terhadap E. coli ATCC 25922 adalah 8%. Ekstrak metanol E. spinosum dapat diformulasi menjadi sabun cuci tangan dengan konsentrasi 8% dan 10%. Uji stabilitas menunjukkan bahwa perubahan organoleptik, viskositas, pH, dan kemampuan membentuk busa masih berada dalam nilai yang dipersyaratkan. Uji aktivitas antibakteri sabun cuci tangan yang mengandung ekstrak metanol E. spinosum dengan konsentrasi 8% dan 10% memiliki aktivitas antibakteri terhadap ATCC 25923 and E. coli ATCC 25922. ABSTRACT  Akway (Drimys piperita) is a woody, evergreen and aromatic plant that belongs to family winteraceae. This plant is used by Sougb tribe lived in Sururey village, District of Anggi, to enhance the vitality of body. The objectives of the research were to determine antimicrobial stability of akway bark extracts influenced by heating time of 100OC, levels of acidity (pH) and salt contents.. Antimicrobial assays were done by using agar well diffusion method against four species of bacteria, i.e.  Escherichia coli ATCC25922, Bacillus cereus ATCC10876, Pseudomonas aeruginosa ATCC27853 and Staphylococcus aureus ATCC25923. The results showed that ethanolic extracts of akway bark only inhibited growth of B. cereus and S. aureus with minimum inhibitory concentration 0,99% and 0,89% . The levels of concentration and acidity of ethanol extracts  influenced the antimicrobial capacity of extracts.. Whereas heating time on 100OC during 25 minutes and salt contents  up to 5% of extract solution did not influence the antimicrobial stability of  akway bark extracts. Key words : akway, extracts, antimicrobe, pH, Heating, salt ABSTRAK Akway (Drimys piperita) adalah tumbuhan berkayu, aromatik dan hijau sepanjang tahun dan tergolong dalam suku winteraceae. Tumbuhan ini digunakan oleh Suku Sougb yang bermukim di desa Sururey Distrik Anggi, untuk mengobati malaria dan meningkatkan vitalitas tubuh. Tujuan dari penelitian ini adalah untuk menentukan stabilitas antimikroba ekstrak kulit kayu akway pada waktu pemanasan ekstrak pada 100OC, tingkat keasaman (pH) dan kandungan garam. Pengujian antimikroba dilakukan dengan menggunakan metode difusi agar terhadap empat spesies bakteri yaitu Escherichia coli ATCC25922, Bacillus cereus ATCC10876, Pseudomonas aeruginosa ATCC27853 and Staphylococcus aureus ATCC25923. Hasil menunjukkan bahwa ekstrak etanol kulit kayu akway hanya dapat menghambat bakteri Bacillus cereus dan Staphylococcus aureus dengan konsentrasi hambat minimum masing-masing adalah 0,99% dan 0,89%. Tingkat konsentrasi dan keasaman (pH) mempengaruhi kapasitas antimikroba ekstrak etanol kulit kayu akway. Sedangkan perlakuan pemanasan pada suhu 100OC dengan lama pemanasan sampai dengan 25 menit dan penambahan garam NaCl sampai konsentrasi 5%  tidak berpengaruh pada stabilitas antimikroba ekstrak etanol kulit kayu akway.

Objective: This research aims to analyze the ability of robusta coffee leaves fraction extract to inhibit the growth of Staphylococcus aureus and Escherichia coli and also determine the minimum inhibitory concentration (MIC). Methods: Antibacterial activity evaluated by the disc diffusion method observed in four types of fraction of extract robusta coffee leaves (n-hexane, ethyl acetate, ethanol, and water). Each extract divided into three various concentrations, 5%, 10%, and 15%. Determination of antimicrobial activity in vitro by the disk diffusion method. Results: Ethyl acetate fraction of coffee leaves extract produced the largest diameter zone of inhibition of bacterial growth compared to other extraction fractions of 17.28 mm in E. coli and 18.58 mm in S. aureus. The MIC of coffee leaves extract fraction water, ethyl acetate, and n-hexane on E. coli and S. aureus is 5%, while the fraction ethanol MIC is 10%. Conclusion: The antibacterial effect of ethyl acetate fraction of coffee leaves extract showed an antibacterial effect that was better than the fraction of n-hexane, ethanol, and water.

Porites merupakan genus karang yang memiliki persebaran luas di Indonesia, termasuk perairan Gunungkidul, Yogyakarta. Penelitian menunjukkan bahwa bakteri simbion karang Porites memiliki potensi antibakteri dalam menanggulangi bakteri patogen. Penelitian ini betujuan untuk mengetahui jenis bakteri simbion karang Porites dari Perairan Gunungkidul Yogyakarta yang memiliki aktivitas antibakteri patogen S. aureus dan E. coli. Bakteri simbion diisolasi dari fragmen jaringan karang dengan pengenceran bertingkat, kemudian uji aktivitas antibakteri dilakukan dengan menggunakan metode overlay dan difusi paperdisk. Delapan dari 64 isolat aktif menghambat kedua bakteri patogen Staphylococcus aureus dan Escherichia coli. Dua diantaranya merupakan isolat unggul yang menunjukkan aktivitas antibakteri paling baik. Kedua isolat selanjutnya diidentifikasi karakteristik molekular DNA dengan sekuen gen 16S rRNA. Hasil identifikasi 16S rRNA menunjukkan isolat GKP1.4.3 memiliki kesamaan 99% dengan Bacillus pumilus strain NBRC 12092, dan isolat GKP3.2.2 memiliki kesamaan 99% dengan Vibrio natriegens strain NBRC 15636.Porites is a coral which has distributed widely in Indonesia, including Gunungkidul Waters, Yogyakarta. Research has shown that Porites coral symbiont bacteria have antibacterial potency against pathogenic bacteria.This study aims to determine the type of Porites coral symbiont bacteria collected from the waters of Gunungkidul Yogyakarta which has antibacterial activity of S. aureus and E. coli. Bacteria symbionts were isolated from coral tissue fragments by serial dillution method, while antibacterial activity was performed by using overlay and paperdisk diffusion method. Eight of the 64 active isolates inhibited both pathogenic bacteria S. aureus and E. coli. Two of 8 isolates showed stronger antibacterial activity. The two isolates subsequently identified the molecular characteristics of DNA with the 16S rRNA gene sequence. The identification of 16S rRNA showed that GKP1.4.3 isolate had 99% similarity with Bacillus pumilus strain NBRC 12092, and GKP3.2.2 isolate had 99% similarity with Vibrio natriegens strain NBRC 15636.

In order to authenticate and ascertain the various claim by the rural dwellers that depend on chewing sticks for their oral hygiene and protection against innumerable diseases, the antibacterial activity of the roots of Fagara zanthoxyloides and Distemonanthus benthamianus were evaluated. Preliminary phytochemical screening was carried out using standard methods. In-vitro antibacterial activity of the methanol extracts of both plants was also carried out using the agar well diffusion method against standard strains of Staphylococcus aureus ATCC 25923, Pseudomonas aeruginosa ATCC 27853, Escherichia coli ATCC 25922, Salmonella typhii ATCC 14028, Citrobacter freundii ATCC 8090 and also some clinical isolates of Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, Klebsiella pneumoniae and Proteus mirabilis at different concentrations and ratios. Preliminary phytochemical screening showed that the extracts contained alkaloids, saponin, tannins, steroids, flavonoids, anthraquinones, cardiac glycosides and phenol. All the test microorganisms were susceptible to the inhibitory effect of the extracts at concentrations of 200 mg/ml and 500 mg/ml. F. zanthoxyloides had better antimicrobial activity with zones of inhibition ranging from 21.0 to 26.0 mm at 200 mg/ml and 24.3 to 29.3 mm at 500 mg/ml while D. benthamianus had zones of inhibition ranging from 17.7 to 26.7 mm at 200 mg/mL and 19.0 to 26.7 mm at 500 mg/mL. Some of the test microorganisms were resistant to the standard antibiotics (cefuroxime, ciprofloxacin, ofloxacin and gentamicin). Combinations of the extracts in ratios 50:50 and 75:25 yielded no increase in activity. The MIC and MBC for D. benthamianus ranged from 6.25 mg/ml to 100 mg/mL and 25 to 200 mg/ml, respectively while that of F. zanthoxyloides ranged from 1.56 mg/ml to 12.5 mg/mL and 50 to 200 mg/ml, respectively. The antimicrobial activity demonstrated by F. zanthoxyloides and D. benthamianus indicates that they would be valuable in the management of urinary, respiratory and gastrointestinal tracts infections while at the same time helping to mitigate the problem of antimicrobial resistance. The emergence of resistant bacteria strain has become a global health concern. It encourages the exploration of potential antibacterial agents, particularly from natural sources. The aim of this study was to investigate the antibacterial activity of ethanol extract of root, stems, leaves, and flowers of Polygonum pulchrum Blume against Staphylococcus aureus and Escherichia coli, through disc diffusion method using cup-plate method. Inhibition zone against S. aureus from roots, stems, leaves, and flowers ethanol extract were 3.5 mm, 2.5 mm, 2.25 mm, and 2.62 mm, respectively, while the inhibition zone against E. coli were 2.25 mm, 2.12 mm, 1.62 mm, and 1.75 mm, respectively. In conclusion, ethanol extract of root, stem, leaves, and flower of P. pulchrum Bl possessed weak antibacterial activity against S. aureus and E. coli.Keywords: P. pulchrum Bl, antibacterial, E. coli, S. aureus, cup-plate technique
