Objective:To know the anti-infective effectiveness of ascorbic acid, after a linear incision in the mucosa of the mandibular antero-inferior segment of the oral cavity of the guinea pigs, housed in the vivarium of the Biology Center of the Central University of Ecuador. Materials and methods:This study is experimental in-vivo and longitudinal, where an incision was made in the mucosa of the mandibular anteroinferior segment of the oral cavity in guinea pigs, three groups of 10 guinea pigs were formed, Group C, received mega doses ( 1.42 ml / 214.2 mg) which contained high amounts of ascorbic acid, Group B received normal doses (0.095 ml / 14.25 mg) and group Aor control group did not receive ascorbic acid, this dose was applied one day before, during, and one day after the incision. The clinical evaluation was performed at 24 and 72 hours post incision, a sample was taken by swab from the wounded area at 72 hours for the planting of bacterial cultivation. Results: The clinical examination shows that at 24 hours in group C, 0% bleeding, edema, and erythema, and 100% epithelial junction, while in group B, 30% bleeding, 50% edema, 50% erythema and 100% epithelial junction and in group A50% bleeding, 40% edema, 40% erythema and 20% epithelial junction; at 72 hours the results in group C and B showed better healing percentages in relation to group A, in this group some guinea pigs still showed bleeding edema and erythema in the wounded area. Conclusion: It is determined that the guinea pigs who received mega doses of ascorbic acid presented a better result in the process of healing after clinical examination at 24 and 72 hours and the swab exam shows a lower amount of bacterial colonization, thus demonstrating the effectiveness of ascorbic acid in mega doses of infectious processes.

The study evaluates effects of varied doses of ascorbic acid and piroxicam on behavioural signs of pain in orchidectomised Savannah Brown goats during their postsurgical pain management. The goats were divided into 6 groups of 3 goats each. Orchidectomy was performed on all animals under sedation with xylazine and linear infiltration with lignocaine. After surgery, varied doses of piroxicam (IM) and ascorbic acid (IV) were administered to the goats: Group A = piroxicam, 5 mg/kg + ascorbic acid, 100 mg/kg; Group B = piroxicam, 5 mg/kg + ascorbic acid, 200 mg/kg; Group C = piroxicam, 10 mg/kg + ascorbic acid, 100 mg/kg; Group D = piroxicam, 5 mg/kg; Group E: ascorbic acid, 100 mg/kg together with antibiotics, procaine penicillin, 20,000 IU/kg + streptomycin, 10 mg/kg (IM); and Group F (control) received only the antibiotics. After surgery, pain intensity was determined in each goat by numerical rating scale. Vocalisation, teeth grinding, rapid and shallow breathing, tail wagging, occasionally bleating, neck extension, and dorsal lip curling were recorded at varying degrees across the experimental groups. The goats in the control group showed the highest degree of behavioural signs of pain. It is concluded that treatment with a combination of piroxicam and ascorbic acid ameliorated pain more than either of the agents in orchidectomised Savannah Brown goats.

Campylobacteriosis is the most commonly reported gastrointestinal disease in humans. Campybacter jejuni is the main cause of the infection, and bacterial colonization in broiler chickens is widespread and difficult to prevent, leading to high risk of occurrence in broiler meat. Phage therapy represents an alternative strategy to control Campylobacter in poultry. The aim of this work was to assess the efficacy of two field-isolated bacteriophages against experimental infections with an anti-microbial resistant (AMR) Campylobacter jejuni strain. A two-step phage application was tested according to a specific combination between chickens’ rearing time and specific multiplicities of infections (MOIs), in order to reduce the Campylobacter load in the animals at slaughtering and to limit the development of phage-resistant mutants. In particular, 75 broilers were divided into three groups (A, B and C), and phages were administered to animals of groups B and C at day 38 (Φ 16-izsam) and 39 (Φ 7-izsam) at MOI 0.1 (group B) and 1 (group C). All broilers were euthanized at day 40, and Campylobacter jejuni was enumerated in cecal contents. Reductions in Campylobacter counts were statistically significant in both group B (1 log10 colony forming units (cfu)/gram (gr)) and group C (2 log10 cfu/gr), compared to the control group. Our findings provide evidence about the ability of phage therapy to reduce the Campylobacter load in poultry before slaughtering, also associated with anti-microbial resistance pattern.

Objectives: Investigating the Renoprotective effects of Ascorbic acid ingentamicin induced renal injury (GIRI) in male Wistar rats. Study design: Experimental study.Place and Duration: Animal house of Al-Tibri from September 2016 to December 2016.Methodology: 60 male Wistar rats were selected by non-probability purposive sampling,and were divided into three groups; Group A- control, Group B- Gentamicin induced renalinjury (GIRI) (70 mg/kg/bwt i.m) daily , and Group C- GIRI + AA (Ascorbic acid - 0.2 mg/kg/bwt) daily. Cardiac puncture was performed by a Disposable Syringe for blood sampling.Sera were used for biochemical testing. Renal tissue was stained with H & E for histologicalexamination. Statistix 8.1 software (USA) was used for data analysis at P≤ 0.05. Results:Blood urea and serum creatinine were elevated in the group B (GIRI) compared to GIRI+AAand the controls (P=0.0001). Serum superoxide dismutase, glutathione peroxidase, andcatalase were low in GIRI compared to GIRI+AA and control groups. Renal tissue in ascorbicacid treated rats showed improved tissue architecture. Conclusion: It is concluded that theAscorbic acid exerts Renoprotective effects in gentamicin induced renal injury in rat model.

Background: The benefits of chocolate have attracted significant attention from clinicians, especially the active compound of caffeine on bone metabolism. The bone density significantly affected the rate of tooth movement. Purpose: This study aims to analyse the effect of the dose and the duration of caffeine consumption in chocolate on alveolar bone mineral density in orthodontic tooth movement. Methods: Forty-eight male guinea pigs (Cavia cobaya) aged between 3-4 months and weighing 300-350 grams were divided into four groups (group A control, group B caffeine dose of 2.3 mg, group C caffeine dose of 3.45 mg, and group D caffeine dose of 4.6 mg). An open coil spring was applied to the mandibular inter-incisor with an orthodontic force of 35 grams. Guinea pigs were sacrificed using lethal doses of anaesthetics on days 0, 1, 7, and 14 after an orthodontic appliance installation. Mandibular alveolar bone mineral density in compression sites was analysed with an atomic absorption spectrophotometer (AAS). Experiment data results were analysed using two-way ANOVA with a 95% degree of confidence. Results: Caffeine consumption with a dose of 4.6 mg on day 7 had the lowest alveolar bone mineral density and the highest was at a dose of 2.3 mg on day 14, but there were no differences between the dose groups, the duration groups and interactions between both of them (p>0.05). Conclusion: The consumption of caffeine in chocolate did not decrease the bone mineral density in the compression site of orthodontic tooth movement.

The present study was undertaken to elucidate the effect of ascorbic acid on alcohol-induced reproductive toxicity and also to compare it with that of abstention. A total of thirty-six male guinea pigs were divided into two groups and were maintained for 90 d as control and ethanol-treated groups (4 g/kg body weight (b.wt.)). After 90 d, ethanol administration was stopped and animals in the control group were divided into two groups and then maintained for 30 d as the control and control+ascorbic acid groups and those in the ethanol-treated group as ethanol abstention and ethanol+ascorbic acid (25 mg/100 g b. wt.) groups. Animals treated with ethanol showed a significant decline in sperm quality (P< 0·001), decreased activity of steroidogenic enzymes (P< 0·05) and reduced serum testosterone (P< 0·05), luteinising hormone and follicle-stimulating hormone levels, decrease in the activity of testicular succinate dehydrogenase, adenosine triphosphatase, sorbitol dehydrogenase and reduction in fructose content (P< 0·05). It also caused an increase in testicular malondialdehyde levels (P< 0·05) and decrease in the levels of glutathione content (P< 0·001) of testes. Ascorbic acid levels in testes and plasma were also reduced (P< 0·001) in ethanol-fed animals. Ascorbic acid supplementation altered all these parameters and produced a better and faster recovery from alcohol-induced reproductive toxicity than abstention. The mechanism of action of ascorbic acid may be by reducing the oxidative stress and improving antioxidant status, which eventually changed the microenvironment of testes and enhanced the energy needed for motility of sperms, improved the sperm morphology and elevated the testosterone and gonadotropin levels.

Large doses of ascorbic acid may mobilise Fe from Fe-binding proteins in vivo which in turn could catalyse lipid peroxidation, a process associated with degenerative diseases. This hypothesis was tested in vitro in the serum of Fe-loaded animals. Eighteen male guinea pigs weighing about 500 g on arrival were allocated to two groups of nine. Fe loading was induced in one group by two intraperitoneal injections of 200 mg iron dextran given on days 1 and 5. Blood (6 ml) was drawn from all animals on day 12 by cardiac puncture. Serum and LDL were separated. Serum was tested for loosely-bound Fe (bleomycin assay) and lipid peroxidation (thiobarbituric acid reactive substances (TBARS) assay) and LDL for susceptibility to in vitro oxidation (TBARS and conjugated diene assays). On day 12, another intraperitoneal injection of 200 mg iron dextran was given to the animals in the Fe-loaded group. On day 19, all animals were given 75 mg ascorbic acid by intraperitoneal injection. Blood (6 ml) was drawn 4 h later by cardiac puncture. Serum and LDL assays were repeated. Ascorbic acid increased loosely-bound Fe and in vitro oxidation in the serum from animals of the Fe-loaded group but not in the serum from animals of the control group. Susceptibility of LDL to in vitro oxidation increased after the ascorbic acid injection in the control group but there was no further increase in the Fe-loaded group. These data suggest that large doses of ascorbic acid promote Fe mobilisation and in vitro oxidation in the serum of Fe-loaded animals.

To investigate the sensitivity of guinea pig tissues to ascorbic acid depletion, as distinct from inanition, young male guinea pigs were maintained with either ascorbic acid restriction or total diet restriction for 8 weeks. One group (A) received no ascorbic acid for 3 weeks, then 0·5 mg/d for 5 weeks; one group (B) was weight-matched by restricted food intake to the first group; a third (marginally deficient) group (C) received 1 mg ascorbic acid/d throughout; a fourth was weight-matched to this group (D); and a fifth group received the control dietad lib.(E). Both of the groups with restricted ascorbic acid intakes (A and C) developed very low tissue ascorbic acid contents, but only the first group (severely deficient group A) also exhibited a severely reduced growth rate. This group also exhibited reduced femur calcium and hydroxyproline contents and reduced skin hydroxyproline content. These changes were not seen in the corresponding weight-matched group (B). Neither plasma alkaline phosphatase (EC3.1.3.1) activity, nor a variety of indices of vitamin D status exhibited changes which could be attributed specifically to reduced ascorbic acid intake and hence to lowered tissue ascorbic acid content. It is concluded that low tissue ascorbic acid levels in guinea pigs clearly alter the connective tissue composition of growing femur and skin, but do not necessarily produce a major, specific effect on vitamin D status. Moreover, the control of inanition is crucial to permit interpretation of the changes in metabolism that are caused by ascorbic acid deficiency.

Introduction The oral cavity and pharynx have a rich sensory system composed of specialized receptors. The integrity of oropharyngeal sensation is thought to be fundamental for safe and efficient swallowing. Chronic obstructive pulmonary disease (COPD) patients are at risk for oropharyngeal sensory impairment due to frequent use of inhaled medications and comorbidities including gastroesophageal reflux disease. Objective This study aimed to describe and compare oral and oropharyngeal sensory function measured using noninstrumental clinical methods in adults with COPD and healthy controls. Method Participants included 27 adults (18 men, nine women) with a diagnosis of COPD and a mean age of 66.56 years ( SD = 8.68). The control group comprised 11 healthy adults (five men, six women) with a mean age of 60.09 years ( SD = 11.57). Spirometry measures confirmed reduced functional expiratory volumes (% predicted) in the COPD patients compared to the control participants. All participants completed a case history interview and underwent clinical evaluation of oral and oropharyngeal sensation by a speech-language pathologist. The sensory evaluation explored the detection of tactile and temperature stimuli delivered by cotton swab to six locations in the oral cavity and two in the oropharynx as well as identification of the taste of stimuli administered in 5-ml boluses to the mouth. Analyses explored the frequencies of accurate responses regarding stimulus location, temperature and taste between groups, and between age groups (“≤ 65 years” and “> 65 years”) within the COPD cohort. Results We found significantly higher frequencies of reported use of inhaled medications ( p < .001) and xerostomia ( p = .003) in the COPD cohort. Oral cavity thermal sensation ( p = .009) was reduced in the COPD participants, and a significant age-related decline in gustatory sensation was found in the COPD group ( p = .018). Conclusion This study found that most of the measures of oral and oropharyngeal sensation remained intact in the COPD group. Oral thermal sensation was impaired in individuals with COPD, and reduced gustatory sensation was observed in the older COPD participants. Possible links between these results and the use of inhaled medication by individuals with COPD are discussed.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

Aim: To evaluate the surface detail reproduction of dental stone this is immersed in different disinfectant solution and studied under stereomicroscope. Methodology: Total number of 30 specimens of dental stone (Type III) were made with measurements of 1.5cm diameter and 1cm height .This samples are divided in to 3 groups group A,B,C. were A is immersed in Distilled water which was taken as control group ;B is immersed in 2% Glutaraldehyde and C is immersed in 5%sodium hypochlorite. Each specimen were immersed in the disinfectant solution for 15 minutes and dried under room temperature for 24 hrs. After 24 hrs each specimens are studied under stereomicroscope for surface details. Result: The results showed no significant difference in the surface irregularities and porosities for a group 1 and group 2 except group 3 which showed significant increase in the porosities, surface irregularities and erosions after disinfection with 5% NaHOCl by immersion method. Conclusion: The surface detail reproduction capacity of die stone was adversely affected when 5% Sodium hypochlorite was used as disinfectant solution when compare d to control group and 2% Glutaraldehyde
