ABSTRACTStaphylococcus aureus is a normal bacterial flora found on the skin and mucous membranes of humans. Staphylococcus aureus bacteria can also overgrow and invade under certain conditions, act as pathogenic bacteria that cause various diseases or disorders in the body, one of which is a respiratory tract infection. The cayenne pepper plant is widely used by people in the cayenne pepper fruit section as a spice ingredient in various traditional dishes, however, the cayenne pepper leaves are still not widely used. Cayenne pepper leaves contain saponin and phenol group compounds, namely flavonoids, which have inhibitory activity against Staphylococcus aureus bacteria. The purpose of this study was to determine the most active fraction inhibiting the growth of staphylococcus aureus bacteria in vitro. It carried testing the antibacterial activity of the test plants out by the solid diffusion method. The results showed that the ethyl acetate extract fraction from cayenne pepper leaves (Capsicum frutescent L.) Was the most active antibacterial fraction against staphylococcus aureus bacteria with a minimum inhibitory level (MIC) of 10% w / v with a very strong category.

ABSTRACTBackground Staphylococcus aureus is a pathogenic bacteria that can cause infection. Biduri leaf has antibacterial compound such as tannin, flavonoid, saponin, and polyphenol.Objective This study aimed to know the difference of growth inhibition zone of Staphylococcus aureus at the various concentration of ethanol extract of biduri leaf and to determine an effective concentration.Methods This study was the true experiment with posttest only control design, used Kirby-Bauer disk diffusion method with five concentrations (20%, 40%, 60%, 80%,100%), positive control (chloramphenicol 30 µg) and negative control (ethanol 96%).Result The result of this study showed the average of inhibition zone diameter of each concentration consecutively 26,2 mm, 28,3 mm, 29,7 mm, 31 mm and 31,5 mm. One Way Anova statistic analysis showed that the value of p is 0.000 so there is the difference of growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of a biduri leaf.Conclusion The conclusion of this study is there are differences in growth inhibition zone of Staphylococcus aureus at various concentrations of ethanol extract of biduri leaf and an effective concentration that can inhibit of Staphylococcus aureus is concentration 20% with mean inhibition zone is 26,2 mm. Keywords: ethanol extract of biduri leaf; Staphylococcus aureus; inhibition zone

Crude methanol extracts of fruits, seeds, and bark of Zanthoxylum armatum were investigated in vitro for antimicrobial activities against 9 different bacterial strains: Bacillus subtilis, Enterococcus faecalis, Proteus vulgaris, Pseudomonas aeruginosa, Salmonella typhi, Shigella dysenteriae, Staphylococcus aureus, methicillin-resistant Staphylococcus aureus (MRSA), and Staphylococcus epidermidis using agar well diffusion method, and the MBC values were determined. Only 5 bacteria, i.e., Bacillus subtilis, Enterococcus faecalis, MRSA, Staphylococcus aureus, and Staphylococcus epidermidis exhibited antibacterial properties against the different extracts. The fruit and seed extracts showed activities against 5 bacteria, while the bark extract was active against 2 bacteria only (MRSA and Staphylococcus aureus). Staphylococcus aureus was found to be more susceptible for all the extracts compared to other strains. The maximum ZOI of 20.72 mm was produced by fruits (wild) and 18.10 mm (cultivated) against Staphylococcus aureus. Likewise, the least ZOI of 11.73 mm was produced by seeds (wild) and 11.29 mm (cultivated) against Escherichia faecalis. Similarly, the lowest MBC value of 0.78 mg/mL was obtained for fruit extracts against MRSA, 1.56 mg/mL for fruits, seeds, and bark extracts against Bacillus subtilis, MRSA, and Staphylococcus aureus, and highest value of 50 mg/mL for fruits and seeds extracts against S. epidermidis. The fruits, seeds, and bark extracts of Z. armatum exhibited remarkable antibacterial properties against different pathogenic bacteria causing several diseases, which suggests the potential use of this plant for treating different bacterial diseases such as skin infection, urinary tract infection, dental problems, diarrhea, and dysentery.

Background: View to its interesting role in the peptidoglycan biosynthesis pathway the enzyme UDP-N- acetylglucosamine enolpyruvyl transferase is an attractive target to develop new antibacterial agents, it catalyzes the first key step of this pathway and its inhibition leads to the bacterial cell death. Fosfomycin is known as the natural inhibitor of MurA. Objective: Call new inhibitors of MurA by virtual screening of different chemical compounds libraries, and test the best scored “virtual hits” against three pathogenic bacteria: Escherichia coli, Bacillus subtilis, and Staphylococcus aureus. Methods: A Virtual screening of the structural analogues of fosfomycin downloaded from PubChem database was performed on one side and of the French National Chemical Library as well as using ZINC database to identify new structures different from fosfomycin on the other, FlexX was the software used for this study. The antibacterial testing was divided into methods: disk diffusion and broth dilution. Results: A set of virtual hits was found with better energy score than that of fosfomycin, seven between them were tested in vitro. Therefore, disk diffusion method explored four compounds exhibited antibacterial activity: CID-21680357 (fosfomycin analogue), AB-00005001, ZINC04658565, and ZINC901335. The testing was continued by broth dilution method for both compounds CID-21680357 and ZINC901335 to determine their minimum inhibitory concentrations, and ZINC901335 had the best value with 457µg/ml against Staphylococcus aureus. Conclusion: Four compounds were found and proven in silico and in vitro to have antibacterial activity: CID-21680357, AB-00005001, ZINC04658565, and ZINC901335.

Overuse of antibiotics has become the major factor for the emergence and dissemination of multi-drug resistant strains of several groups of microorganisms and this lead to search for agents that may have antibacterial effects. Vitamin E emerged as an essential, fat-soluble nutrient in the human body and it is essential, because the body cannot manufacture its own vitamin E, so foods and supplements must provide it. The aim of the present study was to evaluate the effect of vitamin E against pathogenic bacteria. Gram positive and gram negative bacteria were selected as the test microorganisms based on their importance in infections. In this study vitamin E used in four concentrations (50,100,200,400) IU/ml. The agar diffusion method was used to determine antibacterial activity. Results showed that gram negative bacteria were shown to be more resistant than gram positive bacteria. The resistance of gram negative bacteria towards antibacterial substances may be related to lipopolysaccharides in their outer membrane.

Abstract Pseudomonas azotoformans known for the benefit on agriculture area as soil fertility enhancer. Pseudomonas azotoformans UICC B-91 was successfully isolated from tissues of N. altissima which traditionally known for treating infectious diseases. Therefore, endophytic P. azotoformans might contain antimicrobial compound. This research aims on exploring the antimicrobial activities of several extracts P. azotoformans UICC B-91 against pathogenic microbes. Isolates were extracted with dichloromethane, chloroform, and ethyl acetate. Each crude extract was observed for its potential antimicrobial activity with disc diffusion method against several pathogen microbes. Bioactive compound was assayed by thin layer chromatography (TLC) and screened by bioautography. Results indicated that all crude extract have inhibitory effect on Escherichia coli ATCC 8739, Bacillus cereus ATCC 10876, Staphylococcus aureus ATCC 6583, Staphylococcus aureus ATCC 25923, Salmonella typhimurium ATCC 25241, Pseudomonas aeruginosa ATCC 15442, Bacillus subtilis ATCC 19659 and Candida albicans ATCC 10231. Highest clear zone was on dichloromethane extract. TLC fraction of dichloromethane extract was able to inhibit growth on pathogenic bacteria (except for B. subtilis ATCC 19659 and S. aureus ATCC 6583) and fungi at minimum concentration in the range 125-1,000 μg/mL and 125 μg/mL, respectively. Bioactive compound from endophytic P. azotoformans extracted with dichloromethane have antibacterial and anticandidal effect.

The search for bioactive compounds sources it continue to do because of the emergence of pathogenic bacteria that are resistant to antibiotics. Iodine plants(Jatropha multifida Linn) has flavonoids, alkaloids, tannins, saponins and jatropins. The flavanoid content in iodine plants can inhibit bacterial growth. Staphylococcus aureus is a pathogenic bacterium that can cause infections characterized by tissue damage accompanied by purulent abscesses. The purpose of this study was to determine the antibacterial activity of methanol extract of iodine plant stem against Staphylococcus aureus in vitro. Types of laboratory experimental research. The sample used in the study was iodine plant (Jatropha multifida Linn) extrac concentration used 20%, 30%, 40%, 50%, 60%, 70%, 80%, 90%, 100%. Using the Kirby bauer diffusion method. The results showed that there was antibacterial activity on iodine stems which showed a clear zone around the cactam disk at concentrations of 40% (10mm) 60% (12 mm), 80% (14mm), 100% (17 mm).

The antimicrobial action of the dietary supplement Oxidal® was tested using the classic Bauer and Kirby agar-gel diffusion method. Clinical and reference strains of Staphylococcus aureus and Escherichia coli were used in the studies. The tested dietary supplement showed a well-pronounced inhibitory effect against the microbial strains commensurable with that of the broad-spectrum chemotherapeutic agent Enrofloxacin and showed even higher activity than the broad spectrum antibiotic Thiamphenicol. The proven inhibitory effect of the tested dietary supplement against the examined pathogenic bacteria is in accordance with the established clinical effectiveness standards for antimicrobial agents.

ABSTRACT Pseudomonas aeruginosa is a gram-negative opportunistic human pathogen often infecting the lungs of individuals with the heritable disease cystic fibrosis and the peritoneum of individuals undergoing continuous ambulatory peritoneal dialysis. Often these infections are not caused by colonization with P. aeruginosa alone but instead by a consortium of pathogenic bacteria. Little is known about growth and persistence of P. aeruginosa in vivo, and less is known about the impact of coinfecting bacteria on P. aeruginosa pathogenesis and physiology. In this study, a rat dialysis membrane peritoneal model was used to evaluate the in vivo transcriptome of P. aeruginosa in monoculture and in coculture with Staphylococcus aureus. Monoculture results indicate that approximately 5% of all P. aeruginosa genes are differentially regulated during growth in vivo compared to in vitro controls. Included in this analysis are genes important for iron acquisition and growth in low-oxygen environments. The presence of S. aureus caused decreased transcription of P. aeruginosa iron-regulated genes during in vivo coculture, indicating that the presence of S. aureus increases usable iron for P. aeruginosa in this environment. We propose a model where P. aeruginosa lyses S. aureus and uses released iron for growth in low-iron environments.

Cow’s milk containing pathogenic bacteria is an important threat to the consumers. The objectives of the present study were to identify the bacterial agents of public health importance in milk samples (n=35) of different locations and to determine their sensitivity to different antibiotics. The milk samples were collected and transported aseptically and subsequently allowed for culture in bacteriological media, Gram’s staining and biochemical tests for the identification of bacterial species. The bacteria identified were Staphylococcus aureus, Escherichia coli and Salmonella typhi, and their prevalence, in case of vendor milk specimens (n=28), were 96.43%, 53.57% and 35.71% respectively, and of brand milk specimens (n=7), were 42.86 %, 28.57% and 0%, respectively. This suggests that cautionary measures should be taken for quality milk production and consumption. The antibiotic sensitivity test was done by disc diffusion method and the average inhibition zones, in case of Staphylococcus aureus, were 32 mm for oxytetracycline, 26 mm for amoxicillin, 35 mm for ciprofloxacin, 27 mm for cefotaxime, 30 mm for ceftriaxone, 30 mm for azithromycin, and 26 mm for erythromycin; in case of Escherichia coli, were 5 mm for oxytetracycline, 9 mm for amoxicillin, 22 mm for ciprofloxacin, 30 mm for cefotaxime, 31 mm for ceftriaxone, 15 mm for azithromycin, and 0 mm for erythromycin; in case of Salmonella typhi., were 25 mm for oxytetracycline, 24 mm for amoxicillin, 38 mm for ciprofloxacin, 31 mm for cefotaxime, 34 mm for ceftriaxone, 24 mm for azithromycin, and 0 mm for erythromycin. Therefore, ciprofloxacin and ceftriaxone may be the antibiotics of first choice, and cefotaxime and azithromycin may be the second choice among the test antibiotics for the treatment of illness caused by these bacteria.Asian J.
