Background: Ultraviolet B exposure is one of the causes of extrinsic skin aging. Ultraviolet exposure may lead to skin collagen damage. This study about mesenchymal stem cells from umbilical cord (UCMSC) compared with platelet rich plasma (PRP) with the aim was to prove that the intradermal injection of UCMSC was more effective than PRP in increasing amount of collagen and fibroblasts in Wistar rats' back skin exposed to Ultraviolet B rays. Methods: This study used post-test only control group design. Subjects were 36 Wistar rats aged 2.5 months. A control group consisting of 18 Wistar rats treated with intradermal injection of PRP and the treatment group with UCMSC. Both groups were exposed to UVB with a total radiation dose of 840 mJ/cm2. The parameter of collagen was determined by picro sirius red staining while fibroblasts by Haematoxylin-Eosin staining. Result: The mean amount of collagen in the control group was lower (57,82±6.52%) than the treatment group (65.69±4.51%) with p<0.001. The result showed there were differences in the number of collagen in both groups (p<0.05). The mean number of fibroblasts in the control group was lower (22.63±5.98 cell/field of view) than the treatment group (28.20±8.90 cell/field of view) with p=0.034. The result showed there were differences in mean of fibroblasts in both groups (p<0.05). Conclusion: It can be concluded that the intradermal injection of UCMSC was more effective in increasing the amount of collagen and fibroblasts than intradermal injection of PRP in Wistar rats' back skin exposed to UVB rays.

Background : The 1st and 2nd degree hemorrhoids is managed non-operatively with the anti-inflammatory and plebothropic drugs. Graptophyllum pictum extract (GPE) has already been used widely in Indonesia to treat hemorrhoid with good result, however, the mechanism is not supported by the molecular research.Objective : This study is intended to study the phlebothropic effects of GPE by measuring the degree of edema and extravassal leucocytes of experimental wistar hemorrhoid.Methods : Experimental study with Randomized Controlled Trial Post-test only design in male wistar rats, weight around 200 gr, induced for the development of a disease-like condition of hemorrhoids by 6% croton oil on the anus for 3 days. Random allocation was performed to divide the 14 wistar rats in 2 groups. Group I as control got normal saline solution, while group II was treated with GPE 100mg/kgbw, started on day 4th for 5 consecutive days. On 9th day blood was extracted from retroorbital fossa and anus was resected up to 2 cm from anal verge and weighted. The degree of anal edema was measured by rectoanal coefficient, that is rasio between anal weight (miligrams) and body weight (grams).Results : Until the end of study, all wistar rat were still alive. The mean (±SD) of body weight   of control group was 173.84 ­(±13.37) and the treatment group was 171.70  (±13.10), and there was no significant differences (p = 0.833). The mean (±SD) of rectoanal coefficient in the treatment group was 2.46 (±0.41) and it was significantly lower than control group  (3.13 ± 0.85) (p = 0.029). The mean (±SD) of extravassal leukocytes in the treatment group was 900.14(±48.09) and it was significantly lower than the control (1003.28 ± 99.30) (p = 0.042).Conclusions : Graptophyllum pictum extract at doses of 100 mg/Kgbw have phlebotonic effect  in decreased edema and extravassal leukocytes.

This study aims to determine the morphology of Leydig cells in hyperlipidemic wistar rats after having administered with sembung (Blumea balsamifera) extract orally. This study utilised a randomized post-test only control group design. The sample in this study were 16 adult male wistar rats (Rattus novergicus) aged 3-4 months with a body weight of 150-200 grams, which were equally and randomly divided into two groups, namely control group (hyperlipidaemia and sterile aquadest) and treatment group (hyperlipidaemia and sembung leaf extract). Hyperlipidaemia was induced with lard administration for 50 days. Data were analysed statistically using the Independent t-test. The results showed that the number of Leydig cells in the treatment group was higher than the control group with 68.13 ± 1.89 and 55.63 ± 1.92 cells respectively (P <0.05). In addition, the mean Leydig cell core diameter of the control group, 5.00 ± 0.34 µm, was smaller compared to the treatment group which was 5.80 ± 0.20 µm (P <0.05). It can be inferred that sembung leaf extract provides a protective effect against damage to Leydig cells due to hyperlipidaemia.

Introduction: Diabetes mellitus is a complex and progressive disease which often lead to several debilitating complications that partly caused by by free radicals which can be overcome with antioxidants. Bay leaves contain essential oils, tannins, flavonoids and terpenoids which have considerable antioxidant properties. Therefore, this study aims to determine the effect combination treatment of vildagliptin and bay leaf (Syzygium polyanthum) extract toward pancreatic beta cells density and glycated albumin levels in diabetic male Wistar rats (Rattus norvegicus). Methods: An experimental posttest only control group study was conducted using 36 albino male rats, aged 2-3 months, weighing 180-200 grams. The rats were divided into 2 groups (n= 18) with control group treated with 1.8mg/200g body weight vildagliptin and 2cc placebo (aquabidest) while the treatment group received 1.8mg/200g BW vildagliptin and 250 mg/200g BW bay leaves extract. All treatment lasted for 21 days. Results: The results showed that pancreatic beta cell counts in the treatment group  was significantly higher than the control group  (109.07 ± 20.47 cells/field of view vs 90.87 ± 13.91 cells/field of view; p<0.01). However, the levels of Glycated albumin between two groups were not significantly different (treatment vs control: 17.33 ± 4.51 vs 20.18 ± 4.57; p=0.068). Conclusion: This study suggested that combination treatment of vildagliptin and bay leaf extract increased pancreatic beta cells but did not reduce glycated albumin levels in diabetic male Wistar rats.

Abstract: Excessive physical activity increases the consumption of oxygen which can result in increased levels of reactive oxygen species (ROS) causing oxidative stress characterized by the elevation of F2-isoprostane as a byproduct of lipid peroxidation. Willow (Justicia gendarussa Burm. f.) has an antioxidant activity that is able to inhibit and break the chain reaction of free radicals and prevent lipid peroxidation. This study was aimed to prove that willow leaf extract could inhibit the elevation of F2-isoprostane levels in male Wistar rats (Rattus norvegicus) urin induced by excessive physical training. This was a true experimental study using 16 male Wistar rats (Rattus norvegicus), aged 2-3 months, weighing 180-200 g as subjects. The control group (P0) was treated with a placebo and excessive physical training, and the treatment group (P1) was treated with the willow leaf extract of 200mg/rat/day and excessive physical training. Before and after 14 days of treatment, the urine samples were collected and their levels of F2-isoprostane 8-isoPGF2α were examined by using enzyme immunoassay kit (EIA). The results showed that the mean level of F2-isoprostane in the control group (P0) before treatment (pretest) was 5.20±0.483 ng / mL while in the treatment group (P1) was 5.46±0.655 ng / mL (P > 0.05). On the other hand, after treatment for 7 days (post-test), the mean level of F2-isoprostane in the control group (P0) was 5.34±0.476 ng/mL and in the treatment group (P1) was 3.61±0.389 ng/mL (P < 0.01). Conclusion: The extract of willow (Justicia gendarussa Burm. f.) leaf could inhibit the elevation of F2-isoprostane levels in male Wistar rats (Rattus norvegicus) urin induced by excessive physical training.Keywords: willow leaf, F2-isoprostane, excessive physical trainingAbstrak: Aktivitas fisik berlebih akan meningkatkan komsumsi oksigen yang dapat mengakibatkan peningkatan kadar reactive oxygen species (ROS) sehingga menyebabkan stres oksidatif yang ditandai dengan meningkatnya F2-isoprostan sebagai salah satu produk peroksidasi lipid. Penelitian ini bertujuan untuk membuktikan bahwa pemberian ekstrak daun gandarusa dapat menurunkan kadar F2-isoprostan urin tikus Wistar (Rattus norvegicus) jantan yang diinduksi latihan fisik berlebih. Jenis penelitian ialah eksperimental dengan subjek 16 ekor tikus (Rattus norvegicus), galur Wistar, sehat, berumur 6 bulan, berat badan 180-200 gr, dengan kadar F2-isoprostan >2ng/mL. Kelompok kontrol (P0) diberikan pelatihan fisik berlebih dan plasebo aquadest 2ml sedangkan kelompok perlakuan (P1) diberikan pelatihan fisik berlebih dan ekstrak daun gandarusa dosis 200mg/ekor/hari. Sebelum dan setelah 14 hari perlakuan, sampel urin ditampung dan diperiksa kadar F2-isoprostan menggunakan 8-isoPGF2α enzyme immunoassay kit (EIA) dari assay design untuk data pretest dan post-test. Hasil penelitian menunjukkan rerata kadar F2-isoprostan pada kelompok P0 sebelum perlakuan (pretest) ialah 5,20±0,483 ng/mL, sedangkan pada kelompokP1 ialah 5,46±0,655 ng/mL (P > 0,05). Setelah perlakuan selama 14 hari (post-test), rerata kadar F2-isoprostan pada kelompok P0 ialah 5,34±0,476 ng/mL dan pada kelompok P1 ialah 3,61±0,389 ng/mL (P < 0,01). Simpulan: Ekstrak daun gandarusa (Justicia gendarussa burm. f.) dapat menurunkan kadar F2-isoprostan urin tikus Wistar (Rattus norvegicus) jantan yang diinduksi latihan fisik berlebih.Kata kunci: daun gandarusa, F2-isoprostan, pelatihan fisik berlebih

This study was initiated on 24 prepubertal Holstein x Kankrej crossbred heifers of nearly identical age (7-9 months) and body weight (130-140 kg) at University farm to evaluate the effect of high plane of nutrition on blood biochemical and minerals profile and the age at puberty. Twelve heifers were managed under routine farm feeding (control) and the rest 12 under ideal optimum feeding regime (treatment) that included extra 1 kg concentrate, 30 g min mix and ad-lib dry fodder. The body weight and ovarian ultrasonography together with blood sampling was carried out at monthly interval from 10 to 18 months of age to study the ovarian dynamics and blood biochemical changes. High plane of nutrition to growing heifers was beneficial in reducing the age of onset of puberty (by 2-3 months) compared to routine farm fed group. The mean plasma total protein and cholesterol concentrations showed a rising trend with significant variations from 10 to 16 months of age, where it got mostly stabilized indicating adult profile. The activity of enzymes GOT and GPT also rose gradually and significantly from 10 months till 14-15 months of age, and thereafter it remained more or less static till 18 months of age. The levels of both these enzymes were higher, with lower protein and cholesterol, in control than the treatment group from 15-16 months of age onwards. The mean plasma levels of both calcium and phosphorus increased gradually and significantly with advancing age till 16-17 months of age, with little higher values in supplemented than a control group. The plasma levels of zinc, iron, copper, and cobalt also showed rising trend with significant differences between 10th and 12th-14th months of age, and from 15th to 18th months of age the levels were statistically the same in all the groups with slightly higher values in the treatment group.

Abstract Background This study assessed the safety and efficacy of intrathecal injection of umbilical cord tissue mesenchymal stem cells (UCT-MSC) in individuals with cerebral palsy (CP). The diffusion tensor imaging (DTI) was performed to evaluate the alterations in white-matter integrity. Methods Participants (4–14 years old) with spastic CP were assigned in 1:1 ratio to receive either UCT-MSC or sham procedure. Single-dose (2 × 107) cells were administered in the experimental group. Small needle pricks to the lower back were performed in the sham-control arm. All individuals were sedated to prevent awareness. The primary endpoints were the mean changes in gross motor function measure (GMFM)-66 from baseline to 12 months after procedures. The mean changes in the modified Ashworth scale (MAS), pediatric evaluation of disability inventory (PEDI), and CP quality of life (CP-QoL) were also assessed. Secondary endpoints were the mean changes in fractional anisotropy (FA) and mean diffusivity (MD) of corticospinal tract (CST) and posterior thalamic radiation (PTR). Results There were 36 participants in each group. The mean GMFM-66 scores after 12 months of intervention were significantly higher in the UCT-MSC group compared to baseline (10.65; 95%CI 5.39, 15.91) and control (β 8.07; 95%CI 1.62, 14.52; Cohen’s d 0.92). The increase was also seen in total PEDI scores (vs baseline 8.53; 95%CI 4.98, 12.08; vs control: β 6.87; 95%CI 1.52, 12.21; Cohen’s d 0.70). The mean change in MAS scores after 12 months of cell injection reduced compared to baseline (−1.0; 95%CI −1.31, −0.69) and control (β −0.72; 95%CI −1.18, −0.26; Cohen’s d 0.76). Regarding CP-QoL, mean changes in domains including friends and family, participation in activities, and communication were higher than the control group with a large effect size. The DTI analysis in the experimental group showed that mean FA increased (CST 0.032; 95%CI 0.02, 0.03. PTR 0.024; 95%CI 0.020, 0.028) and MD decreased (CST −0.035 × 10-3; 95%CI −0.04 × 10-3, −0.02 × 10-3. PTR −0.045 × 10-3; 95%CI −0.05 × 10-3, −0.03 × 10-3); compared to baseline. The mean changes were significantly higher than the control group. Conclusions The UCT-MSC transplantation was safe and may improve the clinical and imaging outcomes. Trial registration The study was registered with ClinicalTrials.gov (NCT03795974).

Background: Excessive sun exposure results in increased activity of the tyrosinase enzyme and the amount of melanin in the skin which causes hyperpigmentation, a sign of premature aging. The purpose of this study was to prove that Ashitaba (Angelica keiskei) leaves extract cream 8% inhibited the increase of melanin and tyrosinase enzymes as effective as hydroquinone cream 4% in the ultraviolet B-exposed guinea pig (Cavia porcellus) skin. Methods: This study used a randomized post-test only control group design. The subjects were 36 male guinea pigs (Cavia porcellus). Subjects were divided into two groups, the hydroquinone cream 4% group and the Ashitaba leaves extract cream 8% group exposed to UVB light. Skin samples were collected 48 hours after the last exposure to measure the tyrosinase enzyme levels using the ELISA method and the amount of melanin was examined by Masson-Fontana staining. Results: The mean levels of tyrosinase in the hydroquinone cream 4% group was 19.51 ± 5.16 ng/L and the Ashitaba leaves extract cream 8% group was 23.76 ± 3.09 ng/L (p = 0.005). The mean amount of melanin in the  hydroquinone cream 4% group was 2.98 ± 2.27% and the Ashitaba leaves extract cream 8% group was 4.77 ± 3.33% (p = 0.069). Conclusion: The administration of Ashitaba (Angelica keiskei) leaves extract cream 8% inhibited the increase of melanin amount as effective as hydroquinone cream 4% and inhibited the increase of tyrosinase enzymes not as effective as hydroquinone cream 4% in the ultraviolet B-exposed guinea pig (Cavia porcellus) skin.

Background: A Kader refers to a Village Health Worker (VHW), a volunteer, which becomes one of the sources of community reference. Commonly, they sustain a mother's knowledge regarding adequate complementary feeding. However, there are still some VHW who have not possessed a health education background nor been able to be independent.Objectives: This study aimed to analyze the impact of collaborative models on VHW's competence as in knowledge, self-efficacy, attitudes, and counseling skill on the topic of complementary feeding.Materials and Methods: The design of this research is a quasi-experiment control group pretest-postest, with the retrieval of subjects using purposive sampling. Subjects were selected based on the location of the posyandu. The population in this research was VHWs in the Lembang district were 40 VHWs in each group. The treatment group was given training intervention for 1 month with a collaborative model, combining several methods into a series. The main topic was counseling and complementary feeding. The control group was given booklets and leaflets. VHW's competence was measured using questionnaires. This research was conducted in January-April 2020.Results: Statistical test results before the treatment of both groups showed no difference (p>0,05) in each variable. Two months after the intervention, there were significant differensces in the mean score of knowledge (p=0,001), attitude (p=0,001), and VHWs self-efficacy (p=0,000) in both groups. VHW counseling skills (p=0,149) until the first month there was not a significant difference. Unexpectedly, in the second month, the VHW counseling skills could not be observed due to the global COVID-19 pandemic. The mean score of each group increased significantly, but the score of the treatment group was higher than the control group.Conclusions: A collaborative model is effective when compared to only providing booklets and leaflets in increasing VHW's knowledge, attitudes, and self-efficacy but not effective yet for VHW counseling skills.

OBJECTIVE: The success of a dental dental implant treatment focuses on a phenomenon called osseointegration. Evaluation of Bone Area (BA) and Bone-Dental implant Contact (BIC) through histomorphometric analysis is the most widely used parameter to measure osseointegration. The aim of this study was to see post-placement Bone Dental implant Contact (BIC) value of dental implant coated and not coated with PRP.MATERIALS & METHODS: This study was an experimental laboratory conducted at Laboratory of Veterinary Faculty, Hasanuddin University. The sample was baby buck rabbit, aged 4-8 months old, weight 1500–2000 gram, divided into 2 groups each group consist of 12 rabbit, control group not coated with PRP and treatment group coated with PRP. Data analysis using SPSS version 20.0 (SPSS Inc., Chicago, IL, USA).RESULTS: There was a significant difference between the mean BIC values and the 20% increase in BIC values in LP1, LP2 and LP3 between treatment and control group on day 0, 3, 7, and 14.CONCLUSIONS: There was a difference in the average of post-placement BIC value of dental implant coated and not coated with PRP.

The soybean aphid Aphis glycines Matsumura (Hemiptera: Aphididae) is a primary pest of soybeans and poses a serious threat to soybean production. Our studies were conducted to understand the effects of different concentrations of insecticides (imidacloprid and thiamethoxam) on A. glycines and provided critical information for its effective management. Here, we found that the mean generation time and adult and total pre-nymphiposition periods of the LC50 imidacloprid- and thiamethoxam-treatment groups were significantly longer than those of the control group, although the adult pre-nymphiposition period in LC30 imidacloprid and thiamethoxam treatment groups was significantly shorter than that of the control group. Additionally, the mean fecundity per female adult, net reproductive rate, intrinsic rate of increase, and finite rate of increase of the LC30 imidacloprid-treatment group were significantly lower than those of the control group and higher than those of the LC50 imidacloprid-treatment group (P < 0.05). Moreover, both insecticides exerted stress effects on A. glycines, and specimens treated with the two insecticides at the LC50 showed a significant decrease in their growth rates relative to those treated with the insecticides at LC30. These results provide a reference for exploring the effects of imidacloprid and thiamethoxam on A. glycines population dynamics in the field and offer insight to agricultural producers on the potential of low-lethal concentrations of insecticides to stimulate insect reproduction during insecticide application.
