This research work was aimed at investigating the effects of vitamin C, E and garlic interaction on tramadol induced toxicity in Wistar rats. Thirty five (35) rats were used for the study and rats were shared into five study groups namely; the positive control group fed with water and rats pellets, the negative control group was induced with tramdol only, the TmVC group in which 0.2mg of tramadol was induced and 0.2ml of vitamin C were administered, The TmVE group in which 0.2 mg of tramadol was induced and vitamin E was administered. The TmG group in which 0.2mg of tramadol was induced and garlic was administered. The body weight, organ weights and serum enzyme activities of the rats were monitored during the experimental period. The enzyme activity level of ALT remained constant in the positive control and negative control groups but began to rise in  the TmVC group in which vitamin C was administered and kept rising in the TmVE group and the most significant increase came in the TmG group in which garlic was administered. AST was lower in all of the groups in which tramadol was administered compared to the positive control (group one) with the highest drop in the TmG group in which garlic was administered. ALP, however, showed significant decrease in all the groups except TmG when compared to the positive control at (P<0.05). High density lipoprotein was relatively low in negative control when compared to positive control at (P<0.05). High density lipoprotein was high in antioxidants treated groups compared to negative control at (P<0.05). The low density lipoprotein was high in negative control when compared to positive control. The antioxidant treated groups showed no significant decrease in low density lipoproteins when compared to negative control group.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Fortified milk containing Lactobacillus plantarum, L. casei, and L. acidophilus isolates and their mixture were used in dosing the male albino rats at an age of 9-12 weeks at an average of 23 g with induced hypothyroidism at a concentration of 0.6 g.kg-1 of carbimazole. Total cholesterol, triglycerides, high-density lipoprotein (HDL), low-density lipoprotein (LDL), and very-low-density lipoprotein (VLDL) were estimated. The results showed a significant increase in the level of triglycerides (TG), cholesterol and triglycerides. Low-density lipoprotein (LDL) and very-low-density lipoprotein (VLDL), with a significant decrease in the level of high-density lipoprotein (HDL) in infected male mice, compared to the control sample, and upon dosing with liquid milk fortified, it returned to its normal level without significant differences from the control group.

The efficiency of oat bran and barley bran in lowering the induced hyperlipidemia and hypercholesterolemia in blood of male Albino rats (Rattus rattus) was studied. Twenty rats were divided into four groups each consisted of five rats and fed the specified test diets for eight weeks. The first group (G1) is the negative group which was fed basal diet, the second group (G2) was fed 1.0% cholesterol, was the third group (G3) fed 1.0% cholesterol and 10% oats bran, and the fourth group (G4) was fed 1.0% cholesterol and 10% barley bran. Feeding rats on 1% cholesterol significantly increased serum total cholesterol, low density lipoprotein, and very low density lipoprotein and triglyceride and decreased serum high density lipoprotein. Furthermore, enzyme activity of alanine aminotransferase, aspartate aminotransferase, and alkaline phosphatase was increased, and lipid peroxide was increased, whereas catalase and glutathione-S-transferase were decreased. Kidney functions parameters in the cholesterol supplemented group were elevated compared with the negative control. In addition, histological alteration in kidney, liver, heart, and testes was observed, compared with the negative control. Hypercholesterolemic rats supplemented with oat bran and barley bran showed significant decrease in lipid parameters, significant increase in high density lipoprotein-cholesterol, improved antioxidant enzyme, and improved histopathology of kidney, liver, heart, and testes. In conclusion, both oat bran and barley bran had protective effects against induced hyperlipidemia and improved histological alterations. Oat bran appeared more efficient than barley bran in lowering the lipid profile levels in hypercholesterolemic rats.

Abstract: High blood cholesterol is a major risk factor for atherosclerosis. Gedi plants contain polyphenolic compounds that can lowering blood cholesterol levels. This study aimed to find out the effect of gedi (Abelmoschus manihot L) leaves extract on histopatologic features of aorta of those wistar rats, which had been administered an atherogenic diet. This was an experimental study on animal using 20 Wistar rats as samples. The rats were divided into 4 different groups, 5 rats for each group. Negative control group was fed only pelet, positive control group was fed pork fat as much as 2 ml/day, group C was fed on pork fat and gedi leaf extract, and group D was fed on pork fat then continued with gedi leaf extract. The results indicated that administration of 30 mg gedi leaf extract showed a histopathologic feature which is decreasing of foam cells. Conclusion: Microscopically, aorta of those rats fed on pork lard diet showed a number of foam cells in tunica intima and media whereas the rats that were fed on pork lard along with gedi leaf extract, generally there were no foam cells, but there aresome parts which contain few foam cells. On the group of Wistar rats which were fed on pork lard, continued with gedi leaf extract showed less foam cells compared to the positive control group.Keywords: Gedi leaf extract, pork lard, foam cell.Abstrak: Kadar kolesterol darah yang tinggi merupakan faktor risiko utama terjadinya aterosklerosis. Tumbuhan gedi mengandung senyawa polifenol yang dapat menurunkan kadar kolesterol darah. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun gedi terhadap gambaran histopatologi aorta tikus wistar dengan diet aterogenik. Hewan uji yang digunakan berupa 20 ekor tikus Wistar yang dibagi ke dalam 4 kelompok, masing-masing kelompok terdiri dari 5 ekor tikus. Kelompok kontrol negatif hanya diberikan pelet biasa, kelompok kontrol positif diberikan diet lemak babi 2ml/hari, kelompok C diberikan diet lemak babi bersamaan dengan ekstrak daun gedi, dan kelompok D diberikan lemak babi kemudian dilanjutkan dengan pemberian ekstrak daun gedi. Hasil penelitian memperlihatkan bahwa pemberian ekstrak daun gedi 30 mg mampu memberikan gambaran histopatologi aorta berupa berkurangnya jumlah sel busa. Simpulan: Secara mikroskopik, aorta tikus wistar yang diberi lemak babi menunjukkan adanya penumpukan sel-sel busa pada tunika intima dan media, sedangkan pada aorta tikus wistar yang diberi lemak babi bersamaan dengan ekstrak daun gedi maupun yang dilanjutkan dengan ekstrak daun gedi ditemukan berkurangnya jumlah sel-sel busa.Kata Kunci : Ekstrak daun gedi, Lemak babi, Sel busa.

Trigona honey (TH) is well known for its therapeutic characteristics. To date, the study of Trigona honey as a prophylactic or immune booster prior to the bacterial infection of the invivo model is not well covered. This study aims to investigate anti-inflammatory and immune activities in Wistar rats infected with respiratory infection following with Trigona honey. 25 Wistar rats were assigned to possitive groups, negative control group, positive control group was fed TH (5 g / kg body weight) orally, the untreated group was infected with Staphylococcus aureus to induce respiratory infection, the treated group has been infected with S. aureus followed by treatment with TH at a dose of 1.5 ×108 CFU / mL and the preventive group ingested TH one week before S. aureus infection. Blood was obtained for biochemical analysis. Lung tissues have been collected for molecular examination. The results showed a significant decrease in serum levels of ALT, AST, urea and creatinine in the preventive and treated groups, serum IgG increased significantly (P<0.05) in the preventive and treated groups, IFN-y increased in the preventive group while decreased in the treated group, and IL-8 increased in the treated group while decreased in the preventive group. The mRNA expression of AGP is up-regulated in the positive control, preventive and treated groups. The α2-MG, TNF-α , and mRNA expressions showed lower regulation after administration of TH in preventive and treated groups. The results show the ability of TH to counteract immune and inflammatory changes in serum levels and gene expressions.

  ABSTRAK Hiperkolesterolemia merupakan suatu kondisi dimana kolesterol dalam darah meningkat melebihi ambang normal yang ditandai dengan meningkatnya kadar kolesterol total terutama Low Density Lipoprotein (LDL) dan diikuti dengan penurunan kadar High Density Lipoprotein (HDL) darah, Tujuan penelitian adalah menganalisis pengaruh pemberian asupan jus pepaya terhadap kadar LDL dan kolesterol total pada ibu rumah tangga usia 40-70 tahun yang menderita hiperkolesterolemia. Metode: Jenis penelitian ini merupakan penelitian true eksperimental dengan desain penelitian pre and post test with control group. Total subjek sebanyak 20 orang yang dibagi kedalam dua kelompok, yaitu Intervensi Jus pepaya dan kelompok kontrol. Hasil penelitian menunjukkan bahwa kadar kolesterol total pada kelompok jus pepaya mengalami penurunan yang bermakna (p<0,05). Sedangkan pada kelompok kontrol tidak bermakna (p>0,05). Kadar LDL pada kelompok jus pepaya mengalami penurunan yang bermakna (p<0,05). Sedangkan pada kelompok kontrol tidak bermakna (p>0,05). Kesimpulan jus pepaya (carica papaya l) dapat menurunkan kadar LDL (Low Density Lipoprotein) dan kolesterol total.   Kata kunci: jus pepaya, kolesterol total, LDL.       ABSTRACT Hypercholesterolemia is a conditions where cholesterol in the bloods increased beyond the normal standard, its showed with cholesterol total level’s was increased especially Low Density Lipoprotein (LDL) and followed by the decreasing of High Density Lipoprotein (HDL) in blood, The purpose of this research is analyzing the influence’s of papaya juice intake for LDL’s levels and cholestrol’s total at the age of 40 – 70 years old for housewives with hypercholesterolemia. The method this research is true experimental researchs with pre and post test control group designs. The subject in this research are 20 people and separated into two groups, the first group is papaya juice’s intervention group, and the second is control group. The result of this research show that papaya juice’s has significant result for decreasing cholesterol’s total with p value’s < 0,05 , while the control group has no significant result because its p value’s >0,05. The result for LDL’s levels show that papaya juice’s has significant result too for decreasing LDL’s levels with p value’s < 0,05, while the control group has no significant result because its p value’s >0,05. The conclusion in this research is papaya juice (carica papaya l) can decreased ldl’s levels and cholesterol’s total.   Keywords: cholesterol’s total, LDL, papaya juice.

Abstract: The aim of this study is to investigate the antipyretic effect of leaf-flower extract (Phyllanthus niruri L.) in Wistar rats induced by DPT-HB vaccine. Samples were 15 Wistar rats as test animals and were divided into five groups of three in each. The negative control group was given orally distilled water, positive control group was given paracetamol and 3 experimental groups were given ethanolic leaf-flower extracts (100 mg, 200 mg and 300 mg /200 g body weight of rats). Pyrexia was induced in rats by intramuscular injection of 0.2 ml DPT-HB vaccine. Antipyretic activity was measured at 0, 30, 60, 90, 120, 150, 180 minutes after administration of leaf-flower extract, paracetamol and distilled water. The results showed that leaf-flower extract at a dose of 300 mg /200 g BW decreased the rectal temperature greater than with doses of 100 and 200 mg /200 g BW for 180 minutes of measurement. Conclusion: Leaf-flower extract has antipyretic effect in Wistar rats.Keywords: antipyretic, phyllanthus niruri, DPT-HB, wistar ratAbstrak: Penelitian ini bertujuan untuk menguji efek antipiretik ekstrak meniran (Phyllanthus niruri L.) pada tikus Wistar yang diinduksi vaksin DPT-HB. Metode: Hewan uji yang digunakan adalah 15 ekor tikus Wistar yang dibagi ke dalam 5 kelompok, masing-masing kelompok terdiri dari 3 ekor tikus. Kelompok kontrol negatif diberikan aquades, kelompok kontrol positif diberikan parasetamol, dan 3 kelompok eksperimental diberikan ekstrak tanaman meniran masing-masing dengan dosis 100mg, 200mg dan 300 mg/200 grBB tikus. Induksi demam pada hewan uji menggunakan vaksin DPT-HB 0,2ml secara intramuskular. Pengukuran suhu rektal dilakukan sebelum dan sesudah pemberian vaksin dan setelah pemberian bahan uji yaitu pada menit ke-30, 60, 90, 120, 150 dan 180. Hasil penelitian memperlihatkan pemberian ekstrak meniran dengan dosis 300 mg/200 grBB menunjukkan penurunan suhu rektal lebih besar dibanding dengan dosis 100 dan 200 mg/200 grBB selama 180 menit pengukuran. Simpulan: Ekstrak meniran memiliki efek antipiretik pada tikus Wistar.Kata kunci: antipiretik, phyllanthus niruri, DPT-HB, tikus wistar

This study investigated the hepatoprotective effect of methanol seed extract of Citrus tangerina on liver damage induced by paracetamol in laboratory rats. Wistar rats were used in this study and categorized into five groups. Groups 1 and 2 received 10 ml/kg normal saline orally, groups 3 and 4 were administered 200 mg/kg and 400 mg/kg respectively of Citrus tangerina seed extract orally, while silymarin 100 mg/kg served as standard drug treatment for group 5. Following six (6) days of pretreatment with the extract, hepatotoxicity was induced with paracetamol 3 g/kg (orally) in all the groups except the positive control group. At the end of the experiment (24 hours after induction), blood samples were collected under diethyl ether anaesthesia for biochemical markers of liver enzymes and antioxidative stress and the liver was harvested for histopathological studies. Both doses (200 mg/kg and 400 mg/kg) of Citrus tangerina seed extract significantly (p < 0.05) reduced the liver enzymes level, but significantly (p < 0.05) increased antioxidant enzymes when compared with the negative control group. Liver histology showed that the Citrus tangerina seed extract prevented hepatic injury induced by paracetamol. The methanol seed extract of Citrus tangerina possesses antioxidative and hepatoprotective effects.

Abstrak Pemberian rosella (Hibiscus sabdariffa Linn) diharapkan melindung hepar tikus dari kerusakkan akibat stres oksidatif pada keracunan karbon tetraklorida (CCl 4). Senyawa yang sering dijadikan petunjuk adanya kerusakan tersebut adalah malondialdehid (MDA). Rosella mengandung vitamin C, flavonoid, polifenol dan beta karoten. Tujuanpenelitian ini adalah menentukan pengaruh pemberian ekstrak rosella (Hibiscus sabdariffa Linn) terhadap MDA dan aktivitas katalase tikus yang terpapar CCl 4. Ini adalah penelitian eksperimental dengan desain Post test Only Control Group Design. Sampel 24 ekor tikus Strain Wistar berumur 2-3 bulan, berat 150-200 gr. Sampel diambil secara acakdan dibagi 4 kelompok terdiri dari kelompok kontrol negatif, kontrol positif (CCl 4), perlakuan 1 (CCl 4 dan ekstrak rosella 250 mg/kg bb) dan perlakuan 2 (CCl4  dan ekstrak rosella 500 mg/kg bb). Pemberian CCl 4secara oral dosis tunggal, setelah 24 jam kemudian diberi ekstrak rosella secara oral selama 14 hari. Data dianalisis dengan uji Anova, tingkatkepercayaan 95%.Pemberian ekstrak rosella secara statistik didapatkan perbedaan yang signifikan rerata kadar MDA dan katalase antar kelompok (p < 0,05). Disimpulkan bahwa ekstrak rosella dapat menurunkan kadar MDA dan meningkatkan aktivitas katalase tikus yang terpapar CCl 4. Rosella contains vitamin C, flavonoids, polyphenol and beta carotene. Compounds which was often used as marker of the damage caused by free radicals wa MDA. The objective of this study was to determine the effect of extracts of roselle (Hibiscus sabdariffa Linn) on MDA and catalase activity of rats exposed to CCl4. Experimental research design with Post test Only Control Group Design. Samples of 24 male Wistar Strain rats were 2-3 months old. weighing 150-200 gr. Samples were taken at random and divided into 4 groups consisting of a negative control group, positive control (CCl4), treatment 1 (CCl4 and roselle extract 250 mg / kg bw) and treatment 2 (CCl4 and roselle extract 500 mg / kg bw). CCl4 was given a single dose orally, after 24 hours, the subjects were given rosella extract orally for 14 days. Data were analyzed by ANOVA with a confidence level of 95%. Rosella extract obtained statistically significant differences of MDA and catalase levels among groups (p <0.05). It can be concluded that the rosella extract can reduce levels of MDA and increase the activity of catalase mice exposed to CCl4. From the research it can be concluded that the rosella extract can reduce levels of MDA and increas the activity of catalase mice exposed to CCl4. It is needed further research on the toxicity of extracts of rosella and organ damage caused.Keywords: carbon tetrachloride, MDA, catalase, rosella

Abstract Objectives Diethylnitrosamine (DEN) is found in workplaces, processed meats, tobacco smoke, whiskey, etc. It is capable of forming DNA-adducts. Fluted pumpkin (Telfairia occidentalis [To]) is a medicinal plant, and its herbal preparations have been employed variously in ethnomedicine. Furthermore, it has been reported to possess anti-oxidant, anti-cancer, anti-inflammatory properties. We investigated the possible mitigating effect of the leaf paste of To on DEN-induced deleterious effects in male Wistar rats. Methods Forty-five rats weighing between 100 and 150 g were equally divided into nine groups and treated thus: Group 1 (negative control), Group 2 (0.05 mg/kg carboxymethyl cellulose [CMC] daily), Group 3 (positive control, 25 mg/kg bw DEN administered intraperitoneally thrice per week), Group 4 (25 mg/kg bw quercetin [QUE] daily alone), Groups 5 and 6 (100 and 200 mg/kg bw To daily, respectively), Group 7 (25 mg/kg bw DEN and QUE), Groups 8 and 9 (25 mg/kg bw DEN with 100 and 200 mg/kg bw To, respectively). Blood glucose levels, liver damage biomarkers (aspartate aminotransferase [AST], alanine aminotransferase [ALT] and gamma-glutamyltransferase [γ-GT]), frequency of micronucleated polychromatic erythrocyte (mPCEs), and liver histology were assessed. Results DEN significantly (p<0.05) increased blood glucose levels, activities of ALT, AST and γ-GT, and frequency of mPCEs. Histologically, DEN caused a severe architectural anarchy. However, the intervention groups demonstrated the remarkable protective properties of To by ameliorating the adverse effects caused by DEN. Conclusions Taken together, the leaf paste of To is capable of mitigating DEN-induced hepatotoxicity and clastogenicity in male Wistar rats.
