This experiment examined whether the transfer of DNA or RNA from conditioned planaria could make the recipients more susceptible to similar conditioning. It has been shown in prior experiments that cephalization is not necessary for memory retention in planaria, so there must be another, non-neural mechanism for memory. Thus, changes in DNA or RNA may be responsible for memory retention, and introducing these changes to naive planaria could transfer the "memory" of the conditioned donor. This study included 3 groups of 11 planaria: the control group, the DNA receival group, and the RNA receival group. After the control group was conditioned, each planarian’s RNA and DNA were extracted and given to its genetically identical counterparts in the DNA and RNA receival groups. The number of trials it took to condition the planaria in each group was measured and compared to see if the transfer of RNA or DNA made the recipients more easily conditionable than the control donor group. The results show that the transfer of RNA and DNA made insignificant changes to the conditionability of recipients. However, further research may be able to identify the location of memory bearing molecules, which, if applied to experiments similar to this one, would ensure that the planaria received a high enough concentration of genetic material for any changes present to make a significant effect.

Background: Cardiovascular complications are the leading cause of morbidity and mortality at any stage of chronic kidney disease (CKD). Moreover, the high rate of cardiovascular mortality observed in these patients is associated with an accelerated atherosclerosis process that likely starts at the early stages of CKD. Thus, traditional and non-traditional or uremic-related factors represent a link between CKD and cardiovascular risk. Among non-conventional risk factors, particular focus has been placed on anaemia, mineral and bone disorders, inflammation, malnutrition and oxidative stress and, in this regard, connections have been reported between oxidative stress and cardiovascular disease in dialysis patients. Methods: We evaluated the oxidation process in different molecular lines (proteins, lipids and genetic material) in 155 non-dialysis patients at different stages of CKD and 45 healthy controls. To assess oxidative stress status, we analyzed oxidized glutathione (GSSG), reduced glutathione (GSH) and the oxidized/reduced glutathione ratio (GSSG/GSH) and other oxidation indicators, including malondialdehyde (MDA) and 8-oxo-2’-deoxyguanosine (8-oxo-dG). Results: An active grade of oxidative stress was found from the early stages of CKD onwards, which affected all of the molecular lines studied. We observed a heightened oxidative state (indicated by a higher level of oxidized molecules together with decreased levels of antioxidant molecules) as kidney function declined. Furthermore, oxidative stress-related alterations were significantly greater in CKD patients than in the control group. Conclusions: CKD patients exhibit significantly higher oxidative stress than healthy individuals, and these alterations intensify as eGFR declines, showing significant differences between CKD stages. Thus, future research is warranted to provide clearer results in this area.

High-risk HPV is clearly associated with cervical cancer. HPV integration has been confirmed to promote carcinogenesis in the previous studies. In our study, a total of 285 DNA breakpoints and 287 RNA breakpoints were collected. We analyzed the characteristic of HPV integration in the DNA and RNA samples. The results revealed that the patterns of HPV integration in RNA and DNA samples differ significantly. FHIT, KLF5, and LINC00392 were the hotspot genes integrated by HPV in the DNA samples. RAD51B, CASC8, CASC21, ERBB2, TP63, TEX41, RAP2B, and MYC were the hotspot genes integrated by HPV in RNA samples. Breakpoints of DNA samples were significantly prone to the region of INTRON (P < 0.01, Chi-squared test), whereas in the RNA samples, the breakpoints were prone to EXON. Pathway analysis had revealed that the breakpoints of RNA samples were enriched in the pathways of transcriptional misregulation in cancer, cancer pathway, and pathway of adherens junction. Breakpoints of DNA samples were enriched in the pathway of cholinergic synapse. In summary, our data helped to gain insights into the HPV integration sites in DNA and RNA samples of cervical cancer. It had provided theoretical basis for understanding the mechanism of tumorigenesis from the perspective of HPV integration in the HPV-associated cervical cancers.

ABSTRACT The characterization of microbial assemblages within solid gas hydrate, especially those that may be physiologically active under in situ hydrate conditions, is essential to gain a better understanding of the effects and contributions of microbial activities in Gulf of Mexico (GoM) hydrate ecosystems. In this study, the composition of the Bacteria and Archaea communities was determined by 16S rRNA phylogenetic analyses of clone libraries derived from RNA and DNA extracted from sediment-entrained hydrate (SEH) and interior hydrate (IH). The hydrate was recovered from an exposed mound located in the northern GoM continental slope with a hydrate chipper designed for use on the manned-submersible Johnson Sea Link (water depth, 550 m). Previous geochemical analyses indicated that there was increased metabolic activity in the SEH compared to the IH layer (B. N. Orcutt, A. Boetius, S. K. Lugo, I. R. Macdonald, V. A. Samarkin, and S. Joye, Chem. Geol. 205:239-251). Phylogenetic analysis of RNA- and DNA-derived clones indicated that there was greater diversity in the SEH libraries than in the IH libraries. A majority of the clones obtained from the metabolically active fraction of the microbial community were most closely related to putative sulfate-reducing bacteria and anaerobic methane-oxidizing archaea. Several novel bacterial and archaeal phylotypes for which there were no previously identified closely related cultured isolates were detected in the RNA- and DNA-derived clone libraries. This study was the first phylogenetic analysis of the metabolically active fraction of the microbial community extant in the distinct SEH and IH layers of GoM gas hydrate.

The emergence of antibiotic-resistant bacteria is considered a serious problem. The resistance of bacteria against antimicrobial substances becomes important in the repair systems for damage to DNA and RNA molecules. The role of the antioxidant system in the development of bacterial resistance against antibiotics is not yet practically studied. The article studied the expression regulation of the genes of antioxidant enzymes and enzymes involved in the genetic information in E. coli cells with the antibiotic resistance against apramycin and cefatoxime. The study was conducted on bacterial cells resistant against these two antibiotics. The genes blaOXA-1, blaSHV, blaTEM, mdtK, aadA1, aadA2, sat, strA, blaCTX, blaPER-2, tnpA, tnpR, intC1 and intC1c were identified in bacterial cell case. This indicates the presence of plasmids in bacteria with these genes, which provide bacterial resistance to apramycin and cefatoxime. It was established that during the formation of cefotaxime resistance, there was a sharp increase in the expression of the Cu, Zn superoxide dismutase gene: in comparison with the control group, the representation of its transcripts increased 141.04 times for cefotoxime and 155.42 times for apramycin. It has been established that during the formation of resistance to the studied antibiotics in E. coli, an increase in the expression of the end4 and end3 genes is observed. There is tendency toward an increase in the number of transcripts of the pol3E gene observed in the formation of resistance against cefotaxime and apromycin.

Background: The role of the learning methods is changing from teacher centered learning to student centered learning (SCL). One of the SCL methods is collaborative and cooperative learning. The past twenty years have seen an increase in collaborative and cooperative learning based pedagogies in colleges, but not in community based health education. The aim of this study is to identify the efectivity of collaborative and cooperative learning in community based health educationMethod: This research used quasi experimental non-randomized control group post test design. Respondent of the research are 111 geriatric cadres of Kecamatan Kepanjen and 72 geriatric cadres of Kecamatan Singosari.Results: Data shows that cadre’s knowledge in first training, 30 and 90 days after first training increased in both groups (p<0,001). There is no relationship between individual characteristic (age, education, occupation, time to work as a cadre, training) and increase of knowledge (p>0,05).Conclusion: Collaborative and cooperative method can increase cadre’s knowledge of stroke effectively and maintain memory retention longer than conventional method.

AbstractPhospholipase D3 (PLD3) and PLD4 polymorphisms have been associated with several important inflammatory diseases. Here, we show that PLD3 and PLD4 digest ssRNA in addition to ssDNA as reported previously. Moreover, Pld3−/−Pld4−/− mice accumulate small ssRNAs and develop spontaneous fatal hemophagocytic lymphohistiocytosis (HLH) characterized by inflammatory liver damage and overproduction of Interferon (IFN)-γ. Pathology is rescued in Unc93b13d/3dPld3−/−Pld4−/− mice, which lack all endosomal TLR signaling; genetic codeficiency or antibody blockade of TLR9 or TLR7 ameliorates disease less effectively, suggesting that both RNA and DNA sensing by TLRs contributes to inflammation. IFN-γ made a minor contribution to pathology. Elevated type I IFN and some other remaining perturbations in Unc93b13d/3dPld3−/−Pld4−/− mice requires STING (Tmem173). Our results show that PLD3 and PLD4 regulate both endosomal TLR and cytoplasmic/STING nucleic acid sensing pathways and have implications for the treatment of nucleic acid-driven inflammatory disease.

Luego se realizó la respectiva coloración en Orceína acética al 2% de acuerdo a la técnica de Tjio y Levan, observándose con el microscopio compuesto un total de 1500 células meristemáticas en cada uno de los tratamientos. Los resultados aplicando tratamiento después de 8 horas de exposición a la concentración de 2 g/L mostraron una disminución del índice mitótico de 11.26 %, mientras que se incrementó el índice profásico en 95.57 %. Se produjeron también alteraciones como rupturas cromosómicas, puentes anafásicos y células binucleadas. Las pruebas y análisis de varianza, mostraron diferencias significativas entre los diferentes tratamientos, confirmado mediante la prueba de comparación múltiple de promedios de Duncan. Se evidencia un probable efecto genotóxico en el material genético por el Sorbato de Potasio y una alteración en el ciclo celular de los meristemos de A. cepa L.; así mismo, se comprueba la función de biosensor de A. cepa L. en el estudio de sustancias que puedan afectar el ciclo celular. ABSTRACT In order to determine the effect of Potassium Sorbate on the cell cycle in root meristems of Allium cepa L. “onion” set three experimental groups were exposed to solutions of Potassium Sorbate at concentrations of 0.5, 1 and 2g /l, for 6, 7 and 8 hours each and a control group. Later the respective color in 2% Acetic orcein according to the technique of Tjio and Levan, observed with the compound microscope a total of 1500 meristematic cells in each of the treatments. The appropriate treatment results after 8 hours of exposure to the concentration of 2 g / L showed a decrease in mitotic index of 11.26%, while it increased the rate at 95.57% prophasic index. There were also alterations as chromosome breaks, anaphasic bridges and binucleated cells. The tests and analysis of variance showed significant differences between the different treatments, as confirmed by multiple comparison test of Duncan averages. It shows a probable genotoxic effect in the genetic material by Potassium Sorbate and an alteration in the cell cycle of meristems of A. cepa L., likewise, is found biosensor function of A. cepa L. in the study of substances that may affect the cell cycle. Bacterial infections and infestations of man can be caused by both microbes and non-microbes. Microbes include bacteria, viruses, fungi, and protozoa. Non-microbes include worms, insects, and arachnids. This chapter concentrates on the basic biology of bacteria. A pathogen is an organism that is able to cause disease in its host and the pathogenicity of any organism is its ability to produce disease. Microbes express their pathogenicity by means of their virulence. The virulence of any pathogen is determined by any of its structural, biochemical, or genetic features that enable it to cause disease in the host. The relationship between a host and a potential pathogen is non- static; the likelihood of any pathogen causing disease in its host depends both on the virulence of the pathogen and the degree of resistance or susceptibility of the host, due mainly to the effectiveness of the host’s defence mechanisms. Two of the main factors influencing a bacteria’s pathogenicity are its ability to invade and it ability to produce toxins—either exotoxins or endotoxins. Bacteria are unicellular prokaryotic micro-organisms, unlike human cells, which are eukaryotic. Fungi, protozoa, helminths, and arthropods are also eukaryotic. Prokaryotic organisms contain both DNA and RNA, but their genetic material exists unbound in the cytoplasm of the cell as, unlike eukaryotic cells, they have no nuclear membrane. Sometimes bacteria contain additional smaller circular DNA molecules, called plasmids. The main features of a bacterium are the cell wall, cytoplasm, and cell membrane. However, some bacteria have additional features such as spores, capsules, fimbriae (pili), and flagellae. The construction of the cell wall is different in different bacteria, but all cell walls contain peptidoglycan. The structure of the cell wall determines the staining characteristics when stained using the Gram stain. Although its first use was over a hundred and fifty years ago, is still the standard method for primary classification of bacteria. Occasionally, bacteria do not have a cell wall. Gram staining of a fixed smear of bacteria is used to separate bacteria into Gram positive or Gram negative, and also to demonstrate their shape. Bacteria with a thick peptidoglycan layer but with no outer membrane stain purple and are called Gram positive.

The feasibility of training sheep to approach a stimulus was investigated in three experimental groups and a control group of fine wool Merino sheep (n = 11 in each group). The experimental groups (n = 11) were trained to approach either a visual (V), auditory (A), or visual + auditory (V+A) stimulus over eight training sessions and were subsequently tested in a T-maze for memory retention on six occasions over a 4-month period. Four testing occasions were spaced at greater than 30 days apart while two were less than 3 days apart. Sheep learned to approach the cues during the training period and the tests indicated that the sheep retained memory of the cues for over 130 days without reinforcement. The controls received no contingency exposure and made no choice in the T-maze test. The proportion of correct stimulus choices (±s.e.m.) in the T-maze averaged over the four longer-spaced testing occasions were V 0.61 (0.06), A 0.50 (0.11), V+A 0.77 (0.04). These differences approached significance (P = 0.058). Sheep trained to the V+A stimulus performed significantly (P < 0.05) better in the T-maze than sheep trained to the A stimulus alone. Comparisons over the shorter-spaced testing durations indicated that the sheep quickly learnt not to approach the stimulus (temporary extinction) when no food reward was available. Individual temperament of the animals was not related to their learning. This study highlights the potential for the use of V and A cues in manipulating the movement of sheep which may be useful for farm management purposes.

Cells from different origins behave differently regarding the incorporation of exogenous genetic material and the formation of transgenic cells. In this context, the objective of this study was to verify the potential of transfection of bovine mesenchymal stem cells from Wharton's jelly and adipose tissue, comparing two transfection protocols, using Lipofectamine LTX and Plus or Xfect reagents, with the integration of humanized anti-CD3. Skin fibroblasts were used as a control group. Humanized anti-CD3 is a monoclonal antibody that interacts with the CD3 molecule of the T-cell receptor, leading to the suppression of T-cells. This antibody is considered an option in the treatment of human autoimmune diseases and against the rejection of transplanted organs. Humanized anti-CD3 was used in this work for the production of bovine transgenic cells that, in the future, will be used in the development of bioreactor animals. In all steps of this study, cell types were cultured in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal calf serum (FCS) and antibiotics, in an incubator at 39°C with 5% CO2 in air with saturated humidity. All cells were plated at 5×105 into 24-well culture dishes and co-transfected with vector pBC1-anti-CD3-IRES-FEO and pEF-NEO-GFP using Lipofectamine LTX with reagent Plus or Xfect. Forty-eight hours after transfection, neomycin was added in each treatment and cells were cultured for 2 weeks. Treated cells were submitted to fluorescence microscopy, flow cytometry, and PCR evaluations. Wharton's jelly cells were sensitive to treatments and started necrosis. In the flow cytometry assay, the median fluorescence was higher in adipocytes than in fibroblasts, for both the Xfect reagent (20.057±1.620.7 and 10.601±702.86, respectively, P&lt;0.05) and for LTX (19.590±113.84 and 10.518±442.65 respectively, P&lt;0.05). These results, associated with the evaluation of epifluorescence, demonstrated that adipocytes presented a better response to transfection than did other cells, independent of the kit used. Performing PCR on co-transfected adipocytes and fibroblasts demonstrated the presence of anti-CD3, making this approach feasible in future experiments.
