<p>Although growth hormone (<em>GH</em>) gene mutations are described in several species, the studies concerning their variabilities and associations with economic traits in rabbits are scarce, particularly associations with semen traits. A total of 149 rabbit bucks from five populations (V-line=36, Moshtohor line=28, APRI line=42, cross ½A½M=23, and Gabali=20) were used in the present study to identify polymorphism of c.-78 C&gt;T single nucleotide polymorphism (SNP) of <em>GH</em> gene among these populations and to investigate the association of <em>GH</em> gene polymorphism with body weight (BW), daily weight gain (DG) and semen traits. DNA was extracted from blood samples for genotyping of c.-78 C&gt;T SNP of <em>GH</em> gene based on polymerase chain reaction with the restriction fragment length polymorphism (PCR-RFLP) technique. The genetic diversity of SNP C&gt;T of <em>GH</em> gene was assessed in terms of genotypic and allelic frequencies, effective number of alleles (Ne), observed (Ho) and expected (He) heterozygosity, Hardy-Weinberg equilibrium (HWE), reduction in heterozygosity due to inbreeding (F<sub>IS</sub>) and polymorphism information content (PIC). Three genotypes of TT, CC and TC of PCR product of 231 bp of <em>GH</em> gene were detected and all the populations were in HWE in terms of <em>GH</em> gene. The highest Ne was obtained for the Moshtohor line (1.978), while the lowest allelic numbers were obtained for V-line (1.715) and Gabali breed (1.800). The highest genotype frequency of <em>GH</em> gene was 0.48 in TT genotype of V-line, 0.21 in CC genotype of Moshtohor line, 0.67 and 0.56 in TC genotype of ½A½M and Gabali rabbits (<em>P</em>&lt;0.05). The highest frequency for C allele was recorded by Moshtohor line (0.45) and the lowest frequency by Gabali (0.32). The genetic diversity scores for <em>GH</em> gene were intermediate (Ho=0.551, He=0.471, PIC=0.358). The values of Ho ranged from 0.444 in V-line to 0.667 in ½A½M cross, while the values of He were 0.425 in V-line and 0.508 in Moshtohor line. The values of PIC were moderate and ranged from 0.332 in V-line to 0.375 in M-line. The highest F<sub>IS</sub> was observed in Moshtohor line (0.042) and the lowest value was observed in ½A½M cross (–0.413). The CT genotype of <em>GH</em> gene showed the highest and significant values for body weights at 4, 8, 10 and 12 wk (542, 1131, 1465 and 1861 g) and daily gains at intervals of 4-6 and 8-10 wk (23.1 and 26.5 g). Additionally, the CT genotype recorded the highest and significant values for volume of ejaculate (1.1 mL), sperm motility (57.6%), live sperm (85.6%), normal sperm (93.1%) and sperm concentration in semen (611×106/mL), along with the lowest and significant values for dead sperms (14.4%) and abnormal sperms (6.9%).</p>

In commercial bovine in vitro fertilization (IVF) companies, there is a continuous need to improve results. Efforts to maximize in vitro embryo production have included modifications in the use of sperm separation gradients. The development of commercially available sperm centrifugation gradients represents a new possibility of increasing the number of viable sperm that can be obtained from low concentration (fresh or frozen, sexed or unsexed) semen samples in order to improve the efficiency of the IVF system to make embryo production as efficient as possible. The objective of this study was to compare two different separation gradients, as follows: Group 1: Percoll (Sigma, St. Louis, MO, USA), in 45% and 90% gradients; Group 2: EquiPure (Nidacon, Gathenburg, Sweden), in top and bottom layers. Before and after separation, sperm were evaluated at 200× magnification for total motility, and then stained to assess viability at 400× with fast-green/eosin stain (Sigma). Sperm separation was performed using frozen/thawed semen from one bull. Semen was separated by centrifugation at 200g for 30 min in both density gradients. Results obtained from Groups 1 and 2 were compared by chi-square test. Sperm separation with Percoll yielded lower numbers of sperm (average sperm concentration after separation of 92 × 106, vs. 159 × 106 sperm/mL for EquiPure; P < 0.05) but resulted in higher motility (60% vs. 39%, respectively; P < 0.05) of separated sperm. Rates of live sperm cells were not significantly different between groups (69.5% vs. 70%, respectively; P > 0.1). These results indicate that the commercial separation medium EquiPure may be associated with higher sperm concentration levels but with lowered sperm motility when compared to Percoll for bovine sperm separation. However, Equipure provided similar percentages of live sperm when compared to Percoll, which is currently used in our laboratory.

The genetic Variation of Reed Grass <em>Calamagrostis arundinacea</em> (L.) Roth was investigated in 25 populations in various geographic regions of Poland. A total of 907 individuals were sampled for electrophoretic analysis of peroxidase loci (11 allozymes). Populations were characterised by genetic parameters e.g. heterozygosity level, Wright's fixation index (F) and polymorphism coefficient (Pg). Mean values of interpopulation variability level (GST=0.0310), total genetic diversity (HT=0.4102) and gene flow between populations (Nm=7.805) were also examined. All the populations were polymorphic and they remain in Hardy Weinberg equilibrium.

Chee is 1 of 4 important native chicken breed in Thailand. Genetic selection can be used to improve growth and carcass performance. The objective of this study was to compare growth and carcass performance of native chickens (Chee) with a population selected for economic traits (Chee KKU12) and natural selection (Chee N). Two hundred Chee KKU12 and Chee N chickens were divided into 5 replicates, 20 chickens per replication. Record growth and carcass quality for data analysis. The results showed that at 12 weeks of age, Chee KKU12 chickens (1,279.484 g) had a higher body weight than did Chee N chickens (1,180.212 g). The averages daily weight gain at 4-6, 6-8, and 0-12 weeks of age of Chee KKU12 chicken (17.861,19.230, and 14.843 grams) was higher than Chee N chickens (16.284, 17.497, and 13.668 g) (P<0.05). The carcass quality with mixed gender showed that Chee KKU12 chickens had higher breast (20.859%) and abdominal fat (0.659%) than Chee N chicken (19.585% and 0.217%, respectively) (P<0.05), but Chee KKU12 chicken thigh (17.007%) was significantly lower than Chee N chickens (18.627%). Regression analysis revealed that the selection of Chee KKU12 chickens for gain in weight will result in better carcass composition including wing (0.074 g), breast (0.089 g), and drumstick (0.134 g), while Chee N chicken had better thigh (0.189 g) when selected for high growth performance (P<0.05). It was concluded that chicken population selected for economic traits has a better growth performance in open housing conditions than naturally selected chickens.

ABSTRAK. Tujuan dari penelitian ini adalah untuk mengidentifikasi pengaruh polimorfisme gen heat shock protein 70 (HSP70) terhadap sifat toleransi panas. Genotiping dilakukan pada 110 itik lokal dari 4 populasi itik Sumatera Barat (pitalah, bayang, kamang, dan payakumbuh). Enzim restriksi yang digunakan adalah SacII. Cekaman panas dilakukan selama 1 jam dengan sampel itik lokal sebanyak 24 ekor untuk menganalisa status hematologi. PCR-RFLP dan Sanger sequencing assay digunakan untuk mengidentifikasi polimorfisme. Analisis polimorfisme dilakukan dengan menggunakan Software MEGA 7 dan pengaruhnya dianalisis dengan ANOVA menggunakan software SAS 9.4. Produk amplifikasi yang dihasilkan yaitu 466 pasang basa. Lokus HSP70/SacII mengenali satu situs SNP (Single nucleotide Polymorphism) g.1702TC, menghasilkan dua alel (T dan C) dan tiga genotype (TT, TC, CC). Gen HSP70/SacII bersifat polimorfik pada semua populasi. Hasil penelitian menunjukkan bahwa alel T memiliki frekuensi yang lebih tinggi daripada alel C pada semua populasi. Analisis chi-kuadrat (χ2) menunjukkan bahwa semua populasi itik lokal memenuhi kaidah kesetimbangan Hardy-Weinberg. Pengaruh genotipe gen HSP70 terhadap status hematologi menunjukkan perbedaan (P0,05) terhadap nilai heterofil dan nilai H/L dengan genotipe CT menunjukkan toleransi panas yang lebih baik dibanding genotipe lainnya. Gen HSP70/SacII dapat dijadikan marka molekuler sifat toleransi panas pada itik lokal berdasarkan keragaman dan pengaruhnya terhadap status hematologi.  (The effect of heat shock protein 70 (HSP70) SacII gene polymorphism on heat tolerance of West Sumatera local duck) ABSTRACT. The objective of this study was to identify the effect of HSP70/SacII gene polymorphism associated with the thermotolerance trait. Genotyping was performed on 110 local duck from the West Sumatera ducks population (pitalah, bayang, kamang, and payakumbuh). The restriction enzyme used was SacII. Heat stress was done for 1 hour with samples used as much as 24 birds to analyze hematological status. PCR-RFLP and Sanger sequencing assays were used to identify polymorphism. Analysis of polymorphism was conduct by MEGA 7 software and its effect is analyzed with ANOVA using Statistical SAS 9.4 software. The product of amplification was 466 bp. HSP70/SacII recognized one SNP g.1702TC, producing two alleles (T and C) and three genotypes (TT,TC,CC). The HSP70/SacII locus were polymorphic in all population. T allele had a higher frequency than C allele in all populations. The analysis of chi-square (χ2) showed that all local ducks population were fit with Hardy-Weinberg equilibrium. The effect of HSP70/SacII gene on hematological status showed differences (P0,05) of heterophil and H/L ratio with CT genotype show better heat tolerance than other genotypes. HSP70/SacII gene can be used as a marker of heat tolerance in local ducks based on polymorphism and its effect on hematological status.

The aim of this study was to research the association between leptin gene polymorphism in Holstein cows with their dairy and meat traits of productivity. To conduct DNA testing on the LEP gene blood samples were selected from 228 Holstein heifers. Animal genotyping according to the leptin gene was performed by AC-PCR. All possible genotypes of the LEP gene were identified in the course of DNA testing. Frequency of occurrence of genotypes and alleles was ranked as fol-lows: CC – 32.5 %, TC – 49.1 %, TT – 18.4 % and C – 0.57, T – 0.43. The results of this study show that animals with genotype TT of LEP gene can be useful in breeding for the improvement of population of Holstein cattle by economic traits associated to the dairy and meat productivity. The aim of this study was to evaluate the effect of age and season on the epididymal sperm and level of testosterone in camel. A total 103 camel testes samples were collected from Al-Najaf slaughter house during a cold season (December 2012, January and February 2013) and moderatehot season (March, April and May, 2013). Testes were divided into 3 Gs according to camel age, G1 included the testes of 2-3years, G 2 (3 – 4years) and over 4 years (G 3). Blood samples were collected for determination of serum testosterone level. The sperms were obtained from the tail of epididymis from all animals groups and the results of the sperms individual motility percentage was increased at the level of (P< 0.05) significantly with age progress in both seasons. Also, sperm motility of G3 recorded a significantly higher than those of G1 and G2 in cold and moderate-hot seasons .The live percentage of G 3 animals was 90.01% in cold season with a significantly higher than those of other Gs and in both seasons. However, the live sperm percentage of G 3 during moderate-hot season was 87.82% and G 2 during moderate-hot and cold seasons were 88.58 and 88.72% respectively, showed significantly higher than those of G1 during cold and moderate-hot seasons .The concentration of sperm obtained from epididymis tail of bulls camel significantly increase with age progress in both seasons. However, the mean of the sperm concentration in cold season showed significance higher than those in moderate-hot season in animals of G 1 and 3, respectively. The abnormal morphologically sperm percentage of animals G 1 in both cold and moderate-hot seasons were significantly higher than those of G3.The testosterone levels of the young animals (under 4 years) increased gradually and reached its peak in February 2.28 ng/ml and March 2.27ng/ml. In the same trend older animal (more than 4 years) was showed 8.14 and 7.35 ng/ml, respectively. The older animals showed a significantly monthly, higher values than those of the younger animals in their testosterone level started from January up to May. In conclusions during cold months the camel over 4 years shows higher percentage of epididymal sperms parameters (live and individual motility) and sperms concentration as well as serum testosterone level.

Objective: To determine the effect of age and frequency of collection on quality parameters of Jersey bull semen at National Livestock Breeding Center (NLBC), Pokhara. Materials and Methodology: Nine Jersey bulls were selected randomly from NLBC. To obtain the effect of age, six bulls of three different age interval (3-4, 5-7 and 8-9 years) were selected randomly. Remaining three bulls were used to determine the effect of frequency of collection on semen quality. Studies were conducted for three months (Nov/Dec to Dec/Jan, 2016). Semen quality was analyzed using CASA system and SMILE software. ANOVA and paired t- tests were applied to find out level of significance. All the statistical analyses were done using SPSS 17.0 Results: Sperm concentration, post- thaw motility, live sperm, and abnormal sperm were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. Significantly higher value for semen volume, sperm concentration and post- thaw motility was obtained on second collection (P<0.05). However, volume initial motility, pre-filling motility, live sperm and abnormal sperm percentage did not differ significantly among different age intervals and between the frequencies of collection. Conclusion: Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentages were higher in 5-7 years old bulls and in second collection of semen. The present study was undertaken to determine the effect of age and frequency of collection on quality parameters of Jersey bull semen at National Livestock Breeding Center (NLBC), Pokhara, Kaski district. Remaining three bulls were used to determine the effect of frequency of collection on semen quality. Studies were conducted for three months (Nov/Dec to Dec/Jan 2017). Semen quality was analyzed using CASA system and SMILE software. ANOVA and paired t- tests were applied to find out level of significance. All the statistical analyses were done using SPSS 17.0. Sperm concentration (0.551 ±0.004 to 1.084 ± 0.014x109), post- thaw motility (42.44±0.11 to 51.72±0.03 %), live sperm (45.62 ± 0.13 to 51.23±0.15 %), and abnormal sperm (11.57±0.12 to 19.59±0.24%) were significantly affected by the age of the bulls. Semen quality was found superior in the bulls belonging to 5-7 years of age intervals. On the other hand, volume (3.98±0.09 to 6.72±0.06 ml), initial motility (72.68±0.02 to 78.64±0.15 %) and pre-filling motility (71.34±0.17 to 77.62±0.02 %) did not differ significantly among different age intervals. Significantly, higher value for semen volume (3.98±0.12 to 6.02±0.14 ml), sperm concentration (0.572±0.119 to 1.095±0.015) and post- thaw motility (40.34±0.21 to 48.54±0.06% was obtained on second collection. However, initial motility (70.58±0.06 to 75.64±0.13 %), prefilling motility (68.11±0.12 to 71.62±0.02 %), live sperm (53.27±0.54 to 56.17±0.02 %) and abnormal sperm (12.31±0.12 to 14.3±0.47 %) did not differ significantly between the frequencies of collection. Different quality parameters of semen like volume, concentration, initial motility, pre-filling motility, post thaw motility, live and normal spermatozoa percentage were higher in 5-7 years old bulls and in second collection of semen.
