The present study was undertaken to test the chemopreventive effects of one herbal medicinal plant, Indigofera aspalathoides, on chemically induced carcinogenesis in rats. A well-known polyaromatic hydrocarbon, namely, 20-methylcholanthrene, which is a known carcinogenic substance, was used to induce fibrosarcoma in Wistar strain of male albino rats. Fibrosarcoma rats were treated with aqueous extracts of Indigofera aspalathoides. The rats were divided into four groups, each consisting of six animals. Group I served as normal control, Group II served as fibrosarcoma-induced animals, Group III were fibrosarcoma-bearing animals treated with aqueous extracts of Indigofera aspalathoides, and Group IV animals, which were normal healthy animals treated with Indigofera aspalathoides aqueous extract, served as drug control set. Group III and Group IV animals were treated with aqueous extract of Indigofera aspalathoides intraperitoneally at a dose of 250 mg/kg. b.w. for 30 days. The fibrosarcoma was proved by pathological examinations. The activity levels of nucleic acids such as total DNA and RNA and hexose, hexosamine, and sialic acid in liver and kidney of treated rats were used to monitor the chemopreventive role of the plant extract. The observed increase in the levels of DNA, RNA, hexose, hexosamine, and sialic acid in liver and kidney tissues of fibrosarcoma-bearing animals reached near normal state after the treatment with aqueous extracts of Indigofera aspalathoides, suggesting that Indigofera aspalathoides does have a chemotherapeutic role.

Pyrethroid cyhalothrin (PC) is an insecticide that is used worldwide for pest control in agriculture and household use. Samoa extract (SE) is a potent antioxidant protecting cells from oxidative stress. The present study investigates the protective and therapeutic effect of SE on PC-induced changes in sperm quality in male rats. Fifty adult male albino rats were divided into five groups: group I: served as control; group II: received PC i.p. only (6.2 mg/kg b.wt.); group III: received SE only (100 mg/kg b.wt., p.o.) for eight weeks; group IV: received SE as a protective agent daily for eight weeks, then followed by the administration of PC (i.p.) three times a week for two weeks; group V: exposed to PC (i.p.) three times a week for two weeks, then treated with the SE daily for 8 weeks. Results showed that PC caused markedly impaired sperm quality (a count, viability, motility, and abnormality). Compared to PC-treated animals, SE in the protective group markedly restored the alteration of sperm indices. However, SE in the curative group was found to be less effective in restoring PC-induced alterations. In conclusion, the data of this study revealed that the SE as a protective agent is more effective than as a therapeutic agent. Keywords: Samoa; Pyrethroid; Sperm quality; Rat

The present study aimed to investigate the ameliorative effects of fenugreek seeds and curcumin on hematotoxicity induced by nicotine in male albino rats. 30 male F-344/NHsd Fischer rats, weighing from 180 to 200g were used in the present study. The animals were divided into five groups (6 rats for each); Group I (control group), Group II (nicotine treated group), Group III (nicotine/fenugreek seeds co-administered), Group IV (nicotine/curcumin co-administered), and Group V (nicotine/curcumin& fenugreek seeds co-administered). At the end of the experimentation and 24 hours after the last dose, all animals were anaesthetized with ether and blood samples were collected by heart puncture. The samples were collected in clean dry tubes containing the anticoagulant substance EDTA and used for the hematological studies. The results showed that the animals treated with nicotine for 4 weeks showed a significant decrease in RBCs count, hemoglobin concentration, hematocrit value, MCH, MCHC, and platelets count, and increased MCV and WBCs count as compared to the control group. Co-administration of nicotine with fenugreek and/or curcumin caused improvement in all hematological parameters when compared with nicotine group. It can be concluded that nicotine had a strong effect on the hematological parameters. The ingestion of fenugreek and/or curcumin prevent the hematoxicity induced by nicotine. The current study suggests that fenugreek and curcumin may be useful in combating free radical-induced hematotoxicity induced by nicotine.

Background: Anxiety has become a very important area of research interest in psychopharmacology as it affects one-eighth of the population worldwide. Benzodiazepines are still the most commonly used drugs for anxiety despite a number of side effects.There is a need for newer antianxiety drugs with increased safety and efficacy, hence this study was undertaken to evaluate the anxiolytic activity of Nerium oleander flowers.Methods: Aqueous extract of Nerium oleander flowers (NA) was prepared using soxhalet apparatus. A total of 24 Albino rats aged 8-10 weeks of either sex weighing about 100-150g were obtained and divided into 4 groups of 6 rats each. Group I: Normal saline 10mL/kg. Group II: Diazepam 2mg/kg Group III: NA at a dose of 200 mg/kg Group IV: NA at a dose of 400mg/kg. The anxiolytic activity of Aqueous extract of NA was tested by elevated plus maze and digital actophotometer models. Data was analysed using one way ANOVA followed by Posthoc Tukey’s test.Results: In EPM model, the NA at 200, 400mg/kg doses showed that the number of entries and time spent in the open arms were increased significantly as compared to the control animals. (p<0.001). In Actophoptometer model, two different doses of NA (200 and 400mg/kg) showed a dose-dependent decrease in the locomotor activity, when compared to the control animals (p<0.001).Conclusions: Both the doses of aqueous extract of Nerium oleander flowers (200mg and 400mg/kg) possess anxiolytic activity and could be used as a new approach to treat anxiety.

Myopathy is one of the side effects of colchicine, manifested as pain, dysfunction, and weakness of muscles. Ginseng extract antioxidant  and anti-inflammatory activities have been reported in muscular tissue. This study was conducted to investigate whether administration of ginseng ameliorates colchicine-induced skeletal muscles damage in adult male albino rats. Forty adult male albino rats were randomly  divided into four groups (ten/group) – Group I (control group): rats received normal diet and orally given normal saline, Group II: ginseng extract was administered via oral gavage daily for one month at the doses of 300 mg/kg, Group III (Colchicine treated group): rats were  given colchicine (50 μg/kg/day) via oral gavage for one month, and Group IV: concomitant administration of Ginseng and colchicine for one month. Serum creatine kinase (CK) levels were measured, and skeletal muscles specimens were processed for light and electron microscopic examination. Administration of colchicine (group III) showed elevated serum CK, and histologically myofibrillar disarray foci together with cellular infiltration and edema. Group IV showed reduction in most of those manifestations. Concomitant administration of ginseng with colchicine ameliorated most of the symptoms related to colchicine induced-myopathy.

Background: Nicotine is the active alkaloid in cigarettes. It was reported that tobacco smoking has many hazards; one of these hazards is the effect on the cognitive function of the prefrontal cortex. The aim of our study is to investigate the antioxidant effects of ginger, cinnamon oils, and their combination on morphological changes in the prefrontal cortex that were induced by nicotine. Materials and methods: Fifty adult male albino rats were divided into five groups: group I (control group), group II (nicotine), group III (nicotine + cinnamon), group IV (nicotine + ginger), and group V (nicotine + cinnamon + ginger). The coronal sections from the anterior part of the rat brain at the site of prefrontal cortex were examined by light microscope for (H&E and immunohistochemical staining with TNF-α and GFAP), while the ultrastructure morphology was examined by transmission electron microscopy. Levels of the oxidative stress markers (MDA, GSH) in the rats’ brain tissue homogenate were biochemically assessed. Results: Compared to the control group, the rats that were treated with nicotine (group II) showed a significant oxidative stress in the form of marked elevation of MDA and decrease in GSH, apoptotic changes especially in the pyramidal cells in the form of neuronal cell degeneration and pyknosis, and an elevation in the inflammatory marker TNF-α and GFAP expressions. These changes were observed to a lesser degree in rat group (III) and group (IV), while there was a marked improvement achieved by the combined usage of cinnamon and ginger oils, together compared to the nicotine group. Conclusions: Ginger and cinnamon are powerful antioxidants which ameliorate the degenerative and oxidative effects produced by nicotine on a rat’s prefrontal cortex.

AbstractForty male albino rats were paired and tested for five trials, one trial daily, in a dominance test situation. At the end of the five trials the dominant-submissive relationships of the pairs of animals were determined and the pairs assigned randomly to one of four groups. The criterion for dominance was total time in control of a food source during the trials. In the second five trials of the study, animals assigned to Group I, which served as a control group, were given injections of saline solution one hour before a dominance test. In Group II, submissive animals were administered .5 mg/kg. of chlorpromazine one hour prior to testing while dominant animals received saline solution. In Group III, dominant animals were given the drug while submissive animals received saline and in Group IV all animals received chlorpromazine. At the conclusion of the five drug trials, all pairs of animals were tested on five more trials during a port-drug stage. Results showed that: 1. When either a dominant or submissive animal of a pair was administered chlorpromazine, the drugged animals tended to control the food source for longer periods of time than did the non-drugged animals. Administration of chlorpromazine to both dominant and submissive animals of a pair caused both animals to spend more time in control of the food although the dominant animals tended to control the food source for longer periods of time than the submissive animals. 2. Fighting behavior was significantly reduced in all drug groups during the drug stage of the study.

AbstractObjectivesRheum ribes L. is a perennial plant that belongs to the family of Polygonaceae, which is often used in traditional therapy because it possesses many bioactivities, such as antioxidant and antibacterial ones. Here we examined the effect of different R. ribes L. extracts on oxidative stress in experimental diabetic rats.MethodsThirty-six rats were divided into six groups as follows: group I, control group; group II, diabetic rats; group III, diabetic rats treated with the aqueous extract of R. ribes L. by gavage at 50 mg/kg for 15 days; group IV, diabetic rats treated by gavage with the ethanolic extract of R. ribes L. at 50 mg/kg for 15 days; group V, nondiabetic rats treated by gavage with the aqueous extract of R. ribes L. at 50 mg/kg for 15 days; group VI, nondiabetic rats treated by gavage with the ethanol extract of R. ribes L. at 50 mg/kg for 15 days. After 15 days, the animals were sacrificed and the liver and kidney tissues of each animal were isolated. Superoxide dismutase (SOD), catalase (CAT), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) activities in the tissue samples were measured, and histopathologic examination was carried out.ResultsR. ribes L. was effective in reducing the oxidative stress and increasing the levels of the antioxidant enzymes. Increased levels of MDA and decreased levels of SOD, CAT and GSH-Px were observed in both the liver and kidney tissues in group II. Decreased levels of MDA and increased levels of SOD, CAT and GSH-Px were observed in group III compared with group II. In group IV, decreased levels of MDA and increased levels of SOD, CAT and GSH-Px were observed in comparison with group II.ConclusionsDiabetes increases oxidative stress and causes a decrease in antioxidant enzyme levels. Both aqueous and ethanolic extracts of R. ribes L. decrease oxidative stress activity and increase the levels of antioxidant enzymes. The ethanol extract of R. ribes L. has a higher antioxidant effect than the aqueous extract.

BACKGROUND: The increased strength of zirconia has resulted in its widespread application in clinical dentistry. Nevertheless, the fracture of veneering porcelains remains one of the key reasons of failure. OBJECTIVE: The objective of this study was to compare and analyze the influence of surface conditioning methods on the core-veneer bond strength of zirconia restorations. METHODS: Thirty specimens of zirconia core with sizes 10 × 5 × 5 mm were layered with porcelain of sizes 5 × 3 × 3 mm. On the basis of different surface conditioning methods, four groups were made: Group I: abrasion with airborne alumina particles of 110 μm size, Group II: sandblasting with silica coated alumina particles of 50 μm in size, Group III (modified group): alteration with a coating of zirconia powder prior to sintering, and Group IV (control group): metal core specimens. The shear force of all specimens was tested using a universal testing machine with a 0.5 mm/min crosshead speed. One-way analysis of variance (ANOVA) and Tukey’s post hoc pair wise comparison (p= 0.05) were performed to analyze the shear bond strength. A scanning electron microscope was used to assess the fractured specimens. RESULTS: A statistically significant difference was noted between the groups. The mean value of shear bond strength was 40.25 MPa for Group I, 41.93 MPa for Group II, 48.08 MPa for Group III and 47.01 MPa for Group IV. CONCLUSIONS: The modified zirconia group and control group demonstrated a significantly higher mean bond strength than that of Group I, where airborne particle abrasion was used. The scanning electron microscope showed that cohesive fracture in the porcelain veneers was the main problem of failure in altered zirconia. The modified zirconia specimens in Group III demonstrated significantly improved values of shear bond strength.

The toxicity of lead acetate (L. A.) concerned to public health disruptor due to its persistence in the environment and it has the adverse influence on the human and animal health as well. It causes physiological,biochemical, and neurological dysfunctions in humans. Histologically it has a negative effect on the liver which is considered one of the major target organs where acts as detoxification machine by elimination the toxic substance from the blood in rich with it.  As well as it affects kidneys that are the two of the most filtering organs. Therefore the present study was aimed to investigate the histopathological effect of L.A. on liver and kidney tissues in male rats. Twenty male rats involved in the study were equally and randomly divided into two groups each of them involved 10 animals. Group I (castrated rats) and Group II (control) each group received 80mg/L of lead acetate dissolved in one liter distilled water by drinking for 15 days. Histological sections showed some alterations including abnormal architecture, cell degeneration, nuclear degeneration, hyperchromatic hepatocytes, immune cells, degeneration in tubules, dilation in sinusoids, dilation in central vein of liver increased bowman's space glomerular atrophy degeneration of tubular cells in liver and kidney tissues of rats in castrated rats from control group. But the size of degenerated tissue was more severe in castrated male rats. It was concluded that the castration process could produce a hypogonadism and decreased testosterone which owns many receptors in kidney and liver may produce adverse influence with L.A. administration.

To evaluate the effect of Aqueous extract of Terminalia belliricafruit pulp (AETB) on learning by Hebb William maze model in mice with acute alcohol consumption.Swiss albino mice (n=48) of either sex weighing 20-30g will be divided into eight groups of six mice each. Drugs were given orally after 12 hours of fasting. Group I mice received 10ml/kg of Normal Saline, Group II mice received Piracetam 200mg/kg, Group III received AETB 36mg/kg, Group IV received ethanol 1.5g/kg orally, Group V received ethanol(1.5g/kg )+ piracetam (200mg/kg), Group VI mice received ethanol(1.5g/kg) +AETB(9mg/kg), Group VII mice received ethanol(1.5g/kg) +AETB (18mg/kg), Group VIII mice received ethanol(1.5g/kg) +AETB(36mg/kg). Time taken by the animal to reach the reward chamber from the start chamber (TRC) in Hebb-William maze was used as a parameterto evaluate the learning.Acute alcohol administration showed increase in TRC. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp showed a decrease in TRC when compared to the control group. The TRC values for the groups that were administered AETB along with acute alcohol administration showed decrease in TRC values compared to the negative control.Current study showed acute alcohol administration caused impairment of thelearning ability in mice. Whereas, acute administration of Aqueous extracts of Terminalia belliricafruit pulp (AETB)caused enhancement of learning. Pre-treatment with AETB before acute alcohol administration indicated protective action of AETB on alcohol affected learning in mice.
