The purpose of this research is to clarify aspects of the pathogenesis of Salmonella Enteritidis in experimentally inoculated day-old turkeys. Three treatments were conducted among a total of 120 turkeys; one control group and two treatment groups in which 6 x 102 CFU mL-1 and 7 x 105 CFU mL-1, respectively, of Salmonella Enteritidis was inoculated in the crops. Two birds from each treatment were sacrificed and necropsied at 1, 3, 4, 12, 18, and 24 hours, and 3, 4, 38, and 49 days post-inoculation. We re-isolated Salmonella, measured lymphocytes, and conducted immunohistochemical tests. Six hours post-inoculation, Salmonella was found in the investigated organs (yolk sac, cecum, fragments of spleen, and bursa of Fabricius) with conventional bacteriology and immunohistochemistry, and was continuously detected in almost all analyzed organs until turkeys were four-days old. Further, Salmonella was detected after 38 days in cecum, when the concentration 7 x 105 CFU mL-1 was given. At both inoculation concentrations, the number of lymphocytes was similar; larger quantities were found in the first hour post-inoculation, followed by a gradual reduction, reaching the lowest levels at 24 hours after inoculation. Afterwards, lymphocytes increased discreetly, remaining at the same level until 49 days after inoculation. In conclusion, inoculation concentration influences mitigation, dissemination, elimination, and persistence of this pathogen in turkeys. Lower concentrations promote less invasion as well as lower cell stain and lower lymphocyte count.

Mastery of keyboarding being part urgent to communicate through the computer, and very important, especially students who study in the field of business. On the other side, individual performance also influenced by the work environment, and music is one form the environment that could effect keyboarding performance. A model that can describe this is the stimulus-responsible model of mehrabian-russell which explains that music can affect the performance of individual writing. Almost all of Indonesia has traditional music. Therefore, research is needed to identify the effects of traditional music to enrich the area of innovation in strategies and techniques of learning keyboarding skills. In this study conducted an experiment on two groups, the treatment groups(traditional music) and control group (without traditional) and questionnaire to strengthen the experimental results. In preliminary tests of typing, both groups had a mean AWPM identical or similar. Furthermore, tests carried one, where the treatment group listened to five kinds of music Java, Sunda, and Bali. The test result and the result of the questionnaire showed that (typing skills shown by the average value of AWPM) will increase significantly used of traditional music as the background. So, the genre of traditional music can be stimulant to improve average performance of AWPM. There are three traditional kinds of music that can improve student typing speed and accuracy significantly, a regional music cublek-cublek suweng, kebiar bali, and oleg tambuliningan, but cublek-cublek suweng is a genre of traditional music that has the best effect. Therefore, the process of learning courses keyboarding skills needs to use instrumental traditional music, especially instrumental music that has a fast and cheerfully tempo.

AbstractIntroductionSunset Yellow FCF (SY), used frequently in ready-made foods, cosmetics, and the pharmaceutical industry, may cause many health problems. This study is intended to evaluate the morphological and cellular effects of SY on the embryonic chicken immune system throughout incubation.Material and MethodsBabcock white leghorn eggs were randomly divided into four groups. Besides a control group, there were three treatment groups which received a single injection of 200, 1,000, or 2,000 ng of SY into the air sac just before incubation. The eggs were opened on the 10th, 13th, 16th, and 21st days of incubation. Samples of the bursa of Fabricius, thymus, and spleen were taken from embryos. Serial sections of 5 μm thickness were stained with histological methods and routine histological procedures were performed.ResultsAn increase in the spleen volume was determined as the hatching time of the chicks approached. The highest eosinophil ratio was found in the SY1,000 and SY2,000 groups (P < 0.05), where the most significant change was developmental retardation in the thymus. In the bursa of Fabricius, there was less lymphocyte accumulation and eosinophilic cell infiltration with increasing doses.ConclusionIt was concluded that in ovo administered SY has undesired effects on embryonic development of the bursa of Fabricius, spleen, and thymus, and on spleen volume.

Background: Foodborne diseases are caused by acquired pathogenic bacteria such as Salmonella enteritidis. It causes an intestinal imbalance and the microbial toxins found in the gastrointestinal tract induce symptoms such as diarrhea. Coffee contains active ingredients such as antioxidants and is used as an anti-inflammatory agent by reducing pro-inflammatory cytokine levels in the body. Aim: The purpose of this study was to determine the interaction between Lampung’s robusta coffee and tissue damage in chickens infected by S. enteritidis. Methods: This study used first-day-old Isa brown layer chickens (n = 60), which were divided into five treatment groups. The negative control group consisted of healthy and normal chickens, whereas the positive control group consisted of chickens infected with S. enteritidis bacteria at a concentration of 108 CFU/ml. Groups T1, T2, and T3 were given coffee extract with doses of 500 mg/kg BW (low dose), 1,000 mg/kg BW (moderate dose), and 1,500 mg/kg BW (high dose), respectively, and then infected with S. enteritidis bacteria at a concentration of 108 CFU/ml. The coffee extract and bacteria were given orally via a feeding tube at a volume of 0.5 ml per chick. The extract was given for 14 days (from day 3 to day 16), and the bacteria were given on days 16 and 17. On day 18, the chickens were necropsied. The malondialdehyde (MDA) level was analyzed using one-way analysis of variance test with the GLM procedure (<0.05), while the tissue histopath was analyzed using a descriptive qualitative study to examine the ileal damage Results: The results showed that the MDA levels (nmol/l) decreased in treatment groups T1, T2, and T3 compared to the positive control. On the contrary, we found improvements in the ileum histopathology of group T1 and T2 in the form of normal and regular intestinal epithelium arrangement of the ileum, long intestinal villi, and decreased total leukocytes. Conclusion: Green coffee robusta has the potential to increase antioxidants and reduce inflammation in the small intestine of chickens infected with S. enteritidis.

Sleep deprivation is now recognized as an increasingly common condition inherent to modern society, and one that in many ways, is detrimental to certain physiological systems, namely, immune function. Although sleep is now viewed by a significant body of researchers as being essential for the proper working of a host of defense systems, the consequences of a lack of sleep on immune function remains to be fully comprehended. The aim of the current study was to investigate how paradoxical sleep deprivation (PSD) for 24 and 96 h and sleep restriction (SR) for 21 days by the modified multiple-platform method, and their respective 24-h recovery periods, affect immune activation in rats. To this end, we assessed circulating white blood cell counts, lymphocyte count within immune organs, as well as Ig and complement production. The data revealed that PSD for 96 h increased complement C3 and corticosterone concentration in relation to the control group. In contrast, the spleen weight, total leukocytes, and lymphocytes decreased during SR for 21 days when compared with the control group, although production of a certain class of immunoglobulin, the IgM, did increase. After recovery sleep, lymphocyte count in axillary lymph nodes grew when rats had rebound sleep after PSD for 24 h, neutrophils increased after PSD 96 h and lymphocytes numbers were higher after SR 21 days. Such alterations during sleep deprivation suggest only minor alterations of nonspecific immune parameters during acute PSD, and a significant impairment in cellular response during chronic SR.

Avian infectious bronchitis virus (IBV) is prevalent in all countries with intensive poultry flocks. This disease is characterised primarily by respiratory signs, but some IBV strains may also infect other organs such as the intestinal and urogenital tracts. The aim of this study was to characterise the histopathological lesions and tissue tropism of Iranian isolate IR/773/2001(793/B) of avian infectious bronchitis virus in different organs of experimentally infected SPF chickens. Forty-two one-day-old, specific pathogen-free (SPF) chicks were divided randomly into two groups (21 chicks to each group). At the age of 12 days, one group was inoculated intra-ocularly with 103 EID50 of the 793/B isolate, and the other was kept as the control group. Tissue samples were collected at 2, 4, 6, 8, 10 and 12 days post-inoculation (PI). The IBV virus was detected in the caecal tonsils and cloaca from the 2nd to the 12th day PI. The virus was also detected in the kidneys from days 4–10 PI and in the bursa of Fabricius from days 4–12 PI. The virus was detected in the trachea, lungs and thymus. The most obvious histopathological lesions were found in the trachea, kidney, lungs and bursa of Fabricius. Amongst the lymphoid tissues, histopathological changes were found most frequently in the bursa of Fabricius. The results of this study indicated that the 793/B serotype of IBV is unlikely to cause mortality, severe clinical signs or gross lesions in infected chickens, but its replication in some tissues including the bursa of Fabricius could render birds susceptible to other micro-organisms.

This study was conducted to investigate the safety of the most commonly used Newcastle disease vaccines in Al-Sulaimaniyah province (LaSota and Clone30). A total of 225 one-day-old broiler chicks of Ross 308 breed were investigated for their maternal-derived antibody (MDA) titers by ELISA test. Subsequently, these chicks were divided randomly into 3 equal groups; ( 2 treatment groups, a T1 group which was vaccinated by LaSota vaccine and T2 group which was vaccinated by the Clone30 vaccine). and control non-vaccinated group. ELISA test was used to investigate the antibody titers against NDV in all groups on day 10 post second vaccination at 34 days of chicks age and tissue biopsies were obtained for histopathological examination from the trachea, spleen, Bursa of Fabricius and thymus to explore the tissue changes that may be induced by the vaccine. Significant variation was observed in the means of antibody titers against NDV between the control and treatment groups, whereas, no significant variation was observed between the treatment groups themselves. The histopathological examination results showed that a reactive lymphocytic response was observed in both treatment groups compared to the control group. In addition, focal epithelial sloughing and mucopurulent exudates were observed in the trachea of T1 group chicks only. The result of this study showed that the NDV vaccine of clone30 is approximate of the same efficiency and more secure than LaSota vaccine.

This study aimed to evaluate the liver, kidney damage caused by S. typhimurium and to estimate the oxidative damage in association with this bacteria. A highly virulent isolates of S. typhimurium were obtained from the department of internal and preventive medicine/ College of Veterinary Medicine/ University of Baghdad. A twenty five local rabbits of both genders with age range (2-4 months) weeks old were used for this study, the rabbits were divided randomly into five groups each group contains 5 rabbits :- group 1: drenched orally with 5 ml of normal saline and consider as control group, group 2: were drenched orally with (5 ml) suspension which contain (5��109 CFU) of Salmonella typhimurium and regarded as infected group, group 3 were drenched orally with (5 ml) suspension which have (5��109 CFU) of Salmonella typhimurium then treated with a single dose of gentamicin alone at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation), group 4 were drenched (5 ml) suspension having (5��109 CFU) of Salmonella typhimurium then treated with a single dose of Ca-EDTA alone at 40mg/kg orally after presence of signs (after 24hrs. post inoculation) and group 5 were drenched (5 ml) suspension that contain (5��109 CFU) of Salmonella typhimurium then treated with a single dose of combined gentamicin at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation) and Ca-EDTA 40mg/kg after presence of signs (after 24hrs. post inoculation).The results of biochemical profile showed a significant increase (p less than 0.05) in ALT, creatinine and urea levels in infected group as compared with control group, while, the treated groups especially group 5 showed a significant improvement in ALT, Urea and creatinine levels which returned to relative normal levels as compared with infected group after 96hrs. post treatment. Also, the results of oxidative stress showed a significant increase in the levels of MDA in G2, G3, G4 and G5 after 48 hrs. post treatment, while the level of GSH showed a significant decrease in the level at 48hrs., both were returned to relative normal levels after 96hrs.post treatment especially in group 5.In conclusion, S. typhimurium can causing liver and kidney damage which is manifested by increase ALT, Urea and Creatinine. Also, MDA and GSH is increased due to salmonellosis.

Цель исследования - изучение перепрограммирования мононуклеарных лейкоцитов на модели системного ювенильного идиопатического артрита (сЮИА), воспроизводимой у крыс Wistar с использованием полного адъюванта Фрейнда и липополисахарида. Методика. сЮИА воспроизведен у 6-месячных крыс-самцов Wistar. На 40-е сут. эксперимента животные были разделены на 3 группы: 1-я группа - контроль; 2-я - группа доксициклина; 3-я - группа дексаметазона. Взятие проб крови у животных проводили на нулевые, 41-е и 55-е сут. Мононуклеарные клетки периферической крови выделяли гравиметрически, после чего окрашивали их на маркеры и внутриклеточные цитокины. Дифференцировали моноциты (CD3-CD4+) и Т-хелперы (CD3+CD4+). Анализировали динамику внутриклеточной экспрессии интерлейкина IL-4 (рассматривали как маркер про-М2 фенотипа, так как в случае выделения из клетки ИЛ-4 служит стимулятором М2 поляризации макрофагов) и IFN-g (как маркер про-М1 фенотипа) по данным проточной цитофлуориметрии. Применяли непараметрический статистический тест Mann-Whitney-Wilcoxon в программе R для статистической обработки данных. Результаты и заключение. При моделировании сЮИА выявлено закономерное изменение фенотипа моноцитов. Применение же доксициклина и дексаметазона приводило к более ранней поляризации их по про-М2-пути в отношении моноцитов (на 41-е сут.) в сравнении с контролем. Про-М1 эффект (на 55-е сут., в сравнении с контролем) выявлен также в группах доксициклина и дексаметазона. У животных разных групп обнаружены характерные динамические изменения внутриклеточной экспрессии цитокинов. Важно, что различная направленность поляризации фенотипа при сЮИА и применении препаратов наблюдается не только у моноцитов, но и у Т-хелперов. The study objective was to evaluate targeted reprogramming of peripheral blood mononuclear cells in systemic juvenile idiopathic arthritis (sJIA) modeled in 6-month-old male Wistar rats by co-administration of complete Freund’s adjuvant and lipopolysaccharide. Methods. On day 40 of the experiment, rats were divided into three groups: control, doxycycline, and dexamethasone groups. Blood samples were collected on days 0, 41, and 55. Peripheral blood mononuclear cells were isolated gravimetrically and stained for markers and cytokines. Monocytes (CD3-CD4+) and T-helpers (CD3+CD4+) were differentiated as target cells. IL-4 was considered a marker for the pro-M2 phenotype since IL-4 can activate M2 macrophage polarization; IFN-g was considered a marker for the pro-M1 phenotype. Time-related changes in the intracellular expression of IL-4 and IFN-g were studied using flow cytometry. Data were analyzed using nonparametric statistical tests (Mann-Whitney-Wilcoxon) in the R environment for statistical computing. Results and conclusions. Monocytes (like macrophages) underwent reprogramming during the development of modeled sJIA disease. In monocytes of doxycycline and dexamethasone treatment groups, pro-M2 effects were observed earlier (day 41) than in the control group. Pro-M1 effects were observed in monocytes of doxycycline and dexamethasone groups on day 55, as compared with the control group. Characteristic time-related changes of intracellular cytokine expression were described for different groups. Importantly, the differently directed phenotype polarization was observed in sJIA and treatment groups for both monocytes and T-helpers.

Background: The activation of Nrf2/HO-1 pathway has been shown to protect against cisplatin- induced nephrotoxicity by reducing oxidative stress. Berberine (Ber), an isoquinoline alkaloid, has demonstrated antioxidant, anti-inflammatory and anti-apoptotic activities in various experimental models. Aim: To check the effect of Ber on cisplatin-induced nephrotoxicity and to explore the involved mechanism. Methods: Adult male Wistar rats were divided into 6 groups: Normal, cisplatin-control, treatment groups and per se group. Normal saline and Ber (20, 40 and 80 mg/kg; p.o.) was administered to rats for 10 days. A single intraperitoneal injection of cisplatin (8 mg/kg) was injected on 7th day to induced nephrotoxicity. On 10th day, rats were sacrificed, the kidney was removed and stored for the estimation of various parameters. Results: As compared to cisplatin-control group, Ber pretreatment improved renal function system and preserved renal architecture. It also diminished oxidative stress by upregulating the expression of Nrf2/HO-1 proteins. In addition, Ber attenuated the cisplatin mediated inflammation and apoptosis. Furthermore, it also reduced the phosphorylation of p38/JNK and PARP/Beclin-1 expression in the kidney. Conclusion: Ber attenuated renal injury by activating Nrf2/HO-1 and inhibiting JNK/p38MAPKs/ PARP/Beclin-1 expression which prevented oxidative stress, inflammation, apoptosis and autophagy in renal tissue.
