AbstrakPenelitian eksperimental ini bertujuan mengetahui pengaruh asap rokok terhadap sistem imun seluler. Ada tiga kelompok tikus yang dipajan selama 7, 14, dan 21 siklus estrus dibandingkan dengan kontrol yang tidak terpajan asap rokok. Parameter yang dinilai adalah persentase blastranformasi limfosit T yang dikultur selama 72 jam. Hasilnya pada pemajanan selama 7 dan 14 siklus estrus tidak menunjukkan perbedaan yang bermakna dibandingkan kontrol, sedangkan pemajanan pada 21 siklus terdapat perbedaan yang bermakna. Hal ini dapat diartikan baru pada pemajanan 21 siklus terjadi gangguan imunitas seluler. Pada pemajanan 7 siklus dibandingkan 21 siklus terdapat perbedaan blastransformasi limfosit T secara signifikan (p=0.004), sedangkan pada pemajanan 14 dibandingkan 21 siklus terlihat perbedaan blastransformasi limfosit T tidak bermakna (p=0.986). Dari hasil tersebut dapat disimpulkan imunitas seluler tikus akan terganggu setelah pemajanan asap rokok 21 siklus estrus. Berdasarkan hasil penelitian ini kemungkinan imunitas seseorang yang terpajan asap rokok cukup lama dapat terganggu. Perlu penelitian lebih lanjut dengan pemajanan lebih dari 21 siklus untuk mengetahui timbulnya penyakit kanker dan kardiovaskuler. AbstractExperimental research was done to understand the effect of cigarette smoke to cellular immunity. In this research, 3 groups of rats were exposed by cigarette smoke for 7, 14, and 21 estrous cycles. Each group had a control group that was not exposed by cigarette smoke. Percentage of T lymphocytes blastransformation that cultured in 72 hours was used as the parameter. T Lymphocytes blastransformation was measured with the blood culture preparation. The result showed the groups exposed in 7 and 14 cycles had no significant influence in cellular immunity. But the groups exposed in 21 cycles showed a decline of the cellular immunity function. This research has provided that the cellular immunity of the rats declines by the cigarette smoke in 21 cycle’s exposure. Based on this research, people who are exposed by cigarette smoke for long time can decrease of their immunity, especially cellular immunity.

The aim of the study – to evaluate changes in cellular immunity in rhesus-sensitized women in response to IVIG administration and the prognostic effectiveness of a method for the prevention of isoimmunization in the next pregnancy. Materials and Methods. The study was performed on the basis of City Maternity Hospital No. 7" (Odesa) in 2014–2019. 37 rhesus-sensitized women were randomly splited in two clinical groups: main clinical group (n=19) where patients received human immunoglobulin for intravenous administration, and control group (n=18) where patients did not receive IVIG. Results and Discussion. The state of cellular immunity in rhesus-sensitized women is characterized by a moderate decrease in the absolute and relative indices of T-lymphocytes while increasing the number of B-lymphocytes. The NK cell population did not differ from the control group. When analyzing subpopulations of T-lymphocytes, it can be concluded that the number of T-helper cells is increased and the number of T-suppressors is proportionally reduced. These changes explain the increase in the number of B-lymphocytes as a result of increasing antigenic load on cell receptors. In the group of women who received IVIG therapy, the ratio of chances of normalization of cellular immunity was 18.41 (95 % CI 2.62–166.74), T-helper – 14.93 (95 % CI) 2.45–107.8), T-suppressors – 14.57 (95 % CI 2.13 –127.57) and B-lymphocytes – 31.87 (95 % CI 4.1–333.41). According to the ROC analysis, the quality of the statistical model of IVIG application corresponds to AUC = 0.843 (95 % CI 0.689–0.941) According to the ROC analysis, the level of β-lymphocytes in the compared AUC groups = 0.58 (95 % CI 0.405–0.742).

Despite a huge number of studies, the uniqueness of antiparasitic immunity is so great that there is still insufficient knowledge of the factors contributing to the manifestation of the characteristics of immunity in mixed parasitic diseases of rabbits. Therefore, the question of the influence of the association of pathogens Treponema cuniculi and Eimeria sp. on indicators of cellular immunity of rabbits is relevant. The study was conducted on 59 male rabbits age 3–5 months of the Californian breed, selected by analogy. Animal were separated into two groups: healthy animals (control group) and sick animals (research group). Intensity of invasion was determined by the method of the Mac-Master. It has been established that the level of damage of rabbits by spirochetosis and eimeriosis was, on average, 1155.17 ± 184.87 and 6668.97 ± 284.16 pathogens in 1 g of feces. The count of T- and B-lymphocytes was determined by the method of spontaneous rosette-formation with sheep erythrocytes. Parasitizing the association of pathogens Treponema cuniculi and Eimeria sp. was revealed a high number of leukocytes (1.22 times, P < 0.001), which increased mainly due to lymphocytes, which were 1.45 times higher (P < 0.001), as well as neutrophilic metamyelocytes – 1.48 times (P < 0.05), eosinophils – 1.68 times (P < 0.001) and basophils – 1.57 times (P < 0.001) compared with similar blood parameters of healthy animals. In the blood of sick rabbits, the absolute number of T-lymphocytes (1.56 times, P < 0.001) and B-lymphocytes (3.02 times, P < 0.001) was significantly higher in comparison with a low number of O-lymphocytes (3.46 times, P < 0.001) compared with the control. This indicates the redistribution of lymphocytes to cells that carry T and B lymphocyte receptors on the plasma membrane. The absolute number of T-lymphocytes became high due to T-helpers, which in these animals were higher both in absolute (1.87 times, P < 0.001) and percentage (by 9.18%, P < 0.001) compared to control. Moreover, the percentage of T-suppressors in the blood of rabbits of the experimental group was significantly lower on 5.46% (P < 0.05) compared with the same blood count of healthy animals. Such a redistribution of the T-cell population in the peripheral blood of this group of rabbits led to an increase in the immunoregulatory index by 1.64 times (P < 0.01) than in healthy ones. High IRI and the number of T-active lymphocytes (by 28.23%, P < 0.05) in the blood of rabbits with parasitism of the association of pathogens Treponema cuniculi and Eimeria sp. indicate increased immune system tension.

Regulatory framework and methodological approaches to evaluation of immunological effects of vaccination against brucellosis are not established, and the degree of immunological post-vaccinal rearrangement is not yet developed. Due to leading role of cellular immunity in formation of immune protection against brucellosis, evaluation the cellular response in response to antigenic stimulation may be considered the most informative and objective approach to analysis of immune changes in the body during vaccination. In order to develop the most diagnostically informative methods for design of antigen-stimulation cell tests in vitro, a careful selection of a stimulating agent (antigen) is required, which should have a sufficient activating potential, thus providing specificity of reaction under in vitro conditions. The aim of the present study is to study the in vitro specific activity of a protein-polysaccharide antigenic complex from the Brucella abortus 19 BA strain (BrAg), and an opportunity of its application in order to assess the formation of post-vaccinal cellular immunity against brucellosis.The study was performed with white laboratory mice (n = 50) immunized with the Brucella abortus 19 BA strain. The control group (n = 50) consisted of laboratory mice that received a sterile saline solution in a volume of 0.5 ml. Blood samples were taken from immunized and control animals before vaccination, and 7, 14, 21, and 30 days after immunization. By means of flow cytometry, the activation molecules CD25, CD69, MHC II and CD95, expressed on T lymphocytes (CD3+CD69+, CD3+CD25+, CD3+CD95+, CD3+MHC+) were determined. To observe the development of immunity, the intensity of expression of T lymphocyte activation markers was calculated using the stimulation quotient. BrAg was used for specific in vitro stimulation of T lymphocytes. The liquid brucellosis allergen (brucellin) was used as an antigen for comparison, when studying opportunity of BrAg usage for assessing the postvaccinal immunity development.The following results were obtained: BrAg has pronounced specific activity, it did not cause non-specific in vitro reactions (activation) of T lymphocytes, thus enabling its application as a test antigen when evaluating development of adaptive vaccine immunity against brucella.Experimental testing of brucellosis antigen for carrying out the in vitro antigen-stimulated cellular reactions, aiming for evaluation of post-vaccinal immunity development against brucellosis, showed that the usage of BrAg promotes increase in diagnostic sensitivity of cellular reactions under in vitro experimental conditions. The applied experimental antigen is a quite promising tool for development of laboratory algorithms for brucellosis diagnostics, and assessment of actual vaccination efficiency in cohorts previously vaccinated against brucellosis.

Bronchial Asthma generally starts from childhood which is condition where respiration channel experiences constriction because of hyperactivity with some specific stimulation which cause inflammation. Some risk factors are smoking exposure of cigarette smoke, weather changes, mite on house dirt, pet and history of family sickness. The purpose of this research is to know Relation of mite on house dirt, exposure of cigarette smoke  and history of family sickness with bronchial asthma incident to child 5 – 10  years old on working area of Puskesmas Lempake Samarinda City in 2016. Method which used was analytic survey with Case Control approaching. The total sample was 36 children which consisted of 18 case group and 18 control group with matching by using age and gender which submitted with Purposive Sampling technique. Data Analysis used Chi Square with wrong degree α = 0,005. The result showed that there was relation of mite of house dirt (p = 0,006), history of family sickness (p = 0,001) and no relation with exposure of cigarette smoke (p = 0,370) with bronchial asthma incident to child 5 – 10 years old on working area of Puskesmas Lempake Samarinda City in 2016.

Exposure to mobile phone radiation causes deleterious health effects on biological systems. The objects of this study were to investigate the effect of 900-MHz radiofrequency waves (RFW) emitted from base transceiver station antenna on intrapancreatic homocysteine (Hcy), tumor necrosis factor-α (TNF-α), and nerve growth factor (NGF) as predisposing factors involved in pancreatic beta cell damage. Thirty male rats (Sprague-Dawley, 200 ± 10 g) were randomly divided into the control (without any exposure) and exposed groups: short time (2 h/day), long time (4 h/day), and exposed to 900-MHz RFW for 30 consecutive days. On the last days of the experiment, animals were killed and pancreas tissue was dissected out for evaluation of serotonin, Hcy, TNF-α, and NGF. There was a significant decrease in the serotonin and NGF levels in the pancreatic tissue of exposed groups compared to the control group ( p < 0.05). Also, the levels of serotonin and NGF in the long-time exposure were significantly lower than the short-time exposure ( p < 0.05). However, levels of Hcy and TNF-α were significantly increased in the pancreas of exposed groups compared to the control groups ( p < 0.05). Exposure to 900-MHz RFW decreased pancreatic NGF and serotonin levels and increased the proinflammatory markers (Hcy and TNF-α), which can be a predisposing factor for type 2 diabetes.

Abstract Background This study investigates the effect of autologous bone marrow transfusion (BMT) on the reconstruction of both bone marrow and the immune system in patients with AIDS-related lymphoma (ARL). Methods A total of 32 patients with ARL participated in this study. Among them, 16 participants were treated with conventional surgery and chemotherapy (control group) and the remaining 16 patients were treated with chemotherapy followed by autologous bone marrow transfusion via a mesenteric vein (8 patients, ABM-MVI group) or a peripheral vein (8 patients, ABM-PI group). Subsequently, peripheral blood and lymphocyte data subsets were detected and documented in all patients. Results Before chemotherapy, no significant difference in indicators was observed between three groups of ARL patients. Unexpectedly, 2 weeks after the end of 6 courses of chemotherapy, the ABM-MVI group, and the ABM-PI group yielded an increased level of CD8+T lymphocytes, white blood cells (WBC), and platelet (PLT) in peripheral blood in comparison to the control group. Notably, the number of CD4+T lymphocytes in the ABM-PI group was significantly higher than that in the other two groups. Additionally, no significant difference in haemoglobin levels was observed before and after chemotherapy in both the ABM-MVI and ABM-PI groups, while haemoglobin levels in the control group decreased significantly following chemotherapy. Conclusions Autologous bone marrow transfusion after chemotherapy can promote the reconstruction of both bone marrow and the immune system. There was no significant difference in bone marrow recovery and reconstruction between the mesenteric vein transfusion group and the peripheral vein transfusion group.

<b><i>Objective:</i></b> Cigarette smoking is a life-threatening habit that has rapidly spread in every socioeconomic part of the public worldwide. There exist mechanisms of nicotine delivery available to use in the hope of halting cigarette smoking, and the electronic cigarette (EC) is one of the common methods used for tobacco smoking replacement. This study aimed to investigate experimentally the oxidative effects of tobacco smoke and EC smoke which contain nicotine. <b><i>Method:</i></b> We constructed smoke circuit rooms for exposing the rats to EC or tobacco smoke. Three groups were created, the control group (<i>N</i> = 8); the electronic cigarette group (<i>N</i> = 8), exposure to electronic cigarette smoke for 2 h per day; and the tobacco group (<i>N</i> = 8), exposure to traditional cigarette smoke for 2 h per day. After the first and second week of exposure, blood samples were obtained, and serum oxidative stress index (OSI), paraoxonase 1 (PON1) activity, and prolidase levels were evaluated. <b><i>Results:</i></b> Higher values of OSI and prolidase levels were detected in the first week of EC or tobacco smoke exposure in both study groups (<i>p</i> &#x3c; 0.001) when compared with the control group, and partial decrements were observed in the second week. By contrast, elevated PON1 levels were observed in the second week after EC or tobacco smoke exposure. The highest OSI levels were observed in the tobacco smoke group (<i>p</i> &#x3c; 0.001). The lowest values of PON1 levels were detected in the first week of the electronic cigarette smoke group, and this decremental value was statistically different than normal, the second week of the electronic cigarette smoke group, the first week of the traditional cigarette smoke exposure group, and the second week of the traditional cigarette smoke exposure group values (<i>p</i> &#x3c; 0.000). <b><i>Conclusion:</i></b> Our results indicate that EC smoke induced oxidative stress. Therefore, ECs are potentially risky for human health and can lead to important health problems.

Background: Infertility has been more common problems among couple of reproductive age. One of the factors causing this disorder is unhealthy environmental factors including exposure to cigarette smoke. Polynuclear aromatic hydrocarbons (PAHs) in cigarette smoke can cause testicular atrophy, while the free radicals can inhibit the stages of spermatogenesis, and nicotine in cigarettes affects the brain dopamine levels affecting the levels of GnRH, and subsequently affect the levels of FSH and LH needed in spermatogenesis. The use of Mucuna pruriens seed extract containing antioxidants and L-dopa is expected to improve the quality of sperm after exposure to cigarette smoke. Objective: This study aimed to determine the effect of Mucuna pruriens seed extract on the sperm quality in mice exposed to cigarette smoke. Methods: This study was an experimental study with a post test only control group design. A total of 20 mice were divided into 4 groups of five mice each. All groups were exposed to cigarette smoke. Group 1 was the negative control exposed to cigarette smoke. Groups 2, 3, 4 were exposed to cigarettes smoke and given Mucuna pruriens seed extracts at the dose of 250; 300; and 350 mg/Kg BW/day. Parameters of sperm quality included concentration, morphology, motility and viability. Results: Post hoc tests showed there were significant differences among treatment groups. Conclusion: the administration of Mucuna pruriens seed extract affects the sperm quality of BALB/c mice exposed to cigarettes smoke. Bangladesh Journal of Medical Science Vol.20(4) 2021 p.768-773

Abstract Clinical observations recognized the co-existence and interactions of pain and depression a long time, ago. The aim of this work was to study the effect of ibuprofen and fluoxetine on BCGinduced depressive-like behaviour, on formalin-induced pain, as well as on mechanical allodynia after planter incision in mice. BCG induced a depressive behaviour that was seen in the forced swim test (FST) and the tail suspension test (TST). It also induced a decrease in pain-related behaviour in the formalin test, and an increase in the baseline in mechanical allodynia test compared to the control group. Fluoxetine (80 mg/L of drinking water) showed a significant decrease in the immobility time in the FST and TST and enhanced pain related behaviour in formalin test in the BCG-inoculated group. However, it did not affect the increase in the pain threshold in the planter incision allodynia model. Adding ibuprofen to drinking water (0.2 g/L of drinking water), reversed the depressive like behaviour induced by BCG and enhanced pain-related behaviour in formalin test, in both the total pain-related behaviour and phase 2. It also prevented the increase in the base line induced by BCG. On the other hand, the incisional pain model was not affected by BCG inoculation except at the 2-hour time point, where it showed hypoalgesia, as well.

Objective This study aimed to determine if prolonged antibiotic use at birth in neonates with a negative blood culture increases the total cost of hospital stay. Study design This was a retrospective study performed at a 60-bed level IV neonatal intensive care unit. Neonates born <30 weeks of gestation or <1,500 g between 2016 and 2018 who received antibiotics were included. A multivariate linear regression analysis was conducted to determine if clinical factors contributed to increased hospital cost or length of stay. Results In total, 190 patients met inclusion criteria with 94 infants in the prolonged antibiotic group and 96 in the control group. Prolonged antibiotic use was associated with an increase length of hospital stay of approximately 31.87 days, resulting in a $69,946 increase in total cost of hospitalization. Conclusion Prolonged antibiotics in neonates with negative blood culture were associated with significantly longer hospital length of stay and increased total cost of hospitalization. Key Points
