Introduction: Maize downy mildew (MDM) is considered as a major problem in all maize growing areas in Indonesia including the Province of Lampung. Objectives: The objectives of this study were (i) to identify the species of Peronosclerospora causing maize downy mildew (MDM) in Lampung, (ii) to determine the influence of varieties on the intensity of downy mildew and (iii) to determine the efficacy of metalaxyl to control MDM on some maize varieties. Methodology: To identify Peronosclerospora causing MDM, the pathogens were observed under light microscope and scanning electron microscope. Maize varieties response against pathogens and efficacy metalaxyl were studied in the field with the test plants exposed to plants showing MDM symptoms as the sources of inocula to mimic natural conditions. Results: Peronosclerospora sorghi, P. maydis, and P. philippinensis were found to cause MDM in Lampung. On both varieties Pioneer 27 (P-27) and NK-22, AUDPC on F1 plants was greater than that on F2 plants. On P-27, the production of F2 was higher than that of F1 plants, but there was not significant difference in production between F1 and F2 of NK-22 variety. Seed treatment using metalaxyl was not effective to control downy mildew of maize. Conclusion: Three species of Peronosclerospora were identified as disease-causing pathogen of MDM in Lampung, but these results were tentatively pending for further studies through molecular techniques. F1 plants were more susceptible than F2 plants to MDM in both P-27 and NK-22. Metalaxyl was not effective in controlling MDM.

Resistant varieties are more advisable for controlling maize downy mildew compared with fungicides which is not effective and not environmentally friendly. This study is aimed to determine resistance of some varieties of maize against downy mildew. The maize varieties used were BS 0114, BS 0214, BS 0314, PAC 105, Sweet Corn and BISI 2. The parameters measured were disease incidence and intensity, chlorophyll content of leaves, stomatal density and plants dry weight. Data were analyzed by using analysis of variance (ANOVA) and Duncan's Multiple Range Test (DMRT). The results indicated that PAC 105, BS 0214 and BS 0314 were resistant varieties, while BS 0114, Sweet Corn and BISI 2 were susceptible. PAC 105 variety has the lowest stomatal density (65.353/mm2), and Sweet Corn variety has the highest stomatal density (110.79/mm2). Stomatal density was positively correlated with the disease intensity. Higher disease intensity has lower chlorophyll content compared with the lower intensity. PAC 105 variety has the highest chlorophyll content and plant dry weight, while Sweet Corn variety has the lowest chlorophyll content and plant dry weight. Penggunaan varietas tahan bulai lebih dianjurkan digunakan dalam pengendalian penyakit bulai pada jagung dibandingkan dengan penggunaan fungisida karena tidak efektif dan tidak ramah lingkungan. Penelitian ini bertujuan untuk mengetahui ketahanan beberapa varietas jagung terhadap penyakit bulai. Varietas yang digunakan meliputi BS 0114, BS 0214, BS 0314, PAC 105, jagung manis, dan BISI 2. Parameter yang diamati adalah insidensi dan intensitas penyakit, kandungan klorofil setelah inokulasi, kerapatan stomata dan berat kering tanaman. Data yang diperoleh diuji dengan analisis varians (ANOVA) dan uji lanjut dengan Duncan’s Multiple Range Test (DMRT). Hasil penelitian rumah kaca menunjukkan bahwa varietas PAC 105, BS 0214 dan BS 0314 merupakan varietas tahan, sedangkan varietas BS 0114, jagung manis dan BISI 2 merupakan varietas yang rentan. Varietas tahan PAC 105 memiliki kerapatan stomata paling rendah yaitu 65,353/mm2, dan kerapatan stomata paling tinggi dimiliki oleh varietas rentan yaitu jagung manis dengan kerapatan 110,79/mm2. Kerapatan stomata berkorelasi positif dengan intensitas penyakit dengan koefisien korelasi (r) sebesar 0,72526. Semakin rendah intensitas penyakit bulai kandungan klorofil dan berat kering yang semakin tinggi. Varietas PAC 105 yang mempunyai intensitas penyakit paling rendah , mempunyai kandungan klorofil dan berat kering tertinggi, sedangkan varietas jagung manis mempunyai kandungan klorofil dan berat kering terendah.

Downy mildew disease, caused by Peronosclerospora sorghi, is one of the most serious diseases of maize. The disease is currently managed by seed treatment with metalaxyl fungicides. However, problems regarding environmental pollution resulting from the use of fungicides and development of fungicide resistance within populations of P. sorghi are of increasing concern. Assuming that biological control by means of using antagonistic microorganisms may be an alternative for the management of this disease, the efficacy of biocontrol agents viz., Bacillus subtilis G1, Bacillus amyloliquefaciens B2, Brevibacillus brevis 57 and Pseudomonas fluorescens Pf1 for the management of downy mildew of maize and for promoting plant growth was evaluated. The results indicated that seed treatment with B. subtilis G1 and B. amyloliquefaciens B2 significantly (P = 0.05) increased the germination percentage and seedling vigour of maize as assessed by roll towel method. Among them, B. subtilis G1 was the most effective and recorded 9% and 31% increases in germination percentage and seedling vigour of maize respectively, as compared to the control. A talc- based powder formulation of B. subtilis G1 when applied through seed at the rate of 10 g/kg reduced the downy mildew incidence up to 54% under greenhouse conditions. Results of this study suggest that B. subtilis G1 is a promising bioagent for the management of downy mildew of maize and for promoting plant growth. This antagonist could be further exploited for commercial scale up for ecofriendly management of downy mildew of maize under localized climatic conditions.

Globally, Ralstonia solanacearum (Smith) is ranked one of the most destructive bacterial pathogens inducing rapid and fatal wilting symptoms on tomatoes. Yield losses on tomatoes vary from 0 to 91% and most control measures are unaffordable to resource-poor farmers. This study investigated the antimicrobial activities of chitin and chitosan extracted from black soldier fly (BSF) pupal exuviae against R. solanacearum. Morphological, biochemical, and molecular techniques were used to isolate and characterize R. solanacearum for in vitro pathogenicity test using disc diffusion technique. Our results revealed that BSF chitosan significantly inhibited the growth of R. solanacearum when compared to treatments without chitosan. However, there was no significant difference in the antibacterial activities between BSF and commercial chitosan against R. solanacearum. Soil amended with BSF-chitin and chitosan demonstrated a reduction in bacterial wilt disease incidence by 30.31% and 34.95%, respectively. Whereas, disease severity was reduced by 22.57% and 23.66%, when inoculated tomato plants were subjected to soil amended with BSF chitin and chitosan, respectively. These findings have demonstrated that BSF pupal shells are an attractive renewable raw material for the recovery of valuable products (chitin and chitosan) with promising ability as a new type of eco-friendly control measure against bacterial wilt caused by R. solanacearum. Further studies should explore integrated pest management options that integrate multiple components including insect-based chitin and chitosan to manage bacterial wilt diseases, contributing significantly to increased tomato production worldwide.

A series of experiments were conducted to test the effect of the presence of Fusarium verticillioides in the maize plant on subsequent infestation by coleopteran and lepidopteran pests. The effect of percent internodes 1 to 5 infected with F. verticillioides, time after planting, and maize variety on attacks of stem and ears by lepidopterous and coleopteran pests was assessed in field experiments in early and late season 1998 and early season 1999 in Benin Republic. Artificial inoculation of the first internode with fungal-treated toothpicks was compared with a hot-water-fungicide seed treatment and a control. In 1998, two varieties that differed in husk tightness, the improved DMRLSR-W and the local Gbogbe, were used. Percentage of node 1 to 5 and plants infected was highest with the inoculation treatment but tended to be similar in the seed treatment and the control. The infection rate tended to increase with time and, within sampling date, decreased with node level. Ear infection was strongly correlated with percent infected nodes, indicating that F. verticillioides in the stem predisposed kernel infection. F. verticillioides incidence was higher in Gbogbe than in DMRLSR-W. Stem and ear infestations by the pyralid Eldana saccharina, the major pest in the area, tended to be highest in inoculation and lowest in the protection treatment. The same trends were found for the pyralid Chilo spp., the tortricid Cryptophlebia leucotreta, and beetles pooled across species. Significant positive correlations were found between ear/stem F. verticillioides infection and E. saccharina, Cryptophlebia leucotreta, Mussidia nigrivenella, and the noctuid Sesamia calamistis, but the latter three pest species were only significantly correlated with fungal infection of the upper nodes of the plant. Similar to disease incidence, E. saccharina numbers in stem and ear were higher in Gbogbe than DMRLSR-W in late 1998, whereas for the pyralid ear feeder M. nigrivenella, it was reversed. It was suggested that some lepidopterous and coleopteran pests are attracted by and survive longer (or have lower mortality) on plants infected with F. verticillioides.

Downy mildew caused by Peronosclerospora maydis is an important disease in the centers of corn cultivation in Java. The aim of this study was to determine the effectiveness of a combination of metalaxyl fungicide and varieties that have a high sustainability of downy mildew caused by P. maydis. The study was conducted in Kediri, East Java, which is an endemic area of downy mildew caused by P. maydis. The Split Plot Design with 3 replications was used in this study. The main plots were 5 corn varieties (1) Bima-3 Bantimurung, (2) Bima-20 URI, (3) Lagaligo, (4) Bima-15 Sayang, and (5) Anoman as a susceptible check. The subplots were 5 levels of seed treatment dose with metalaxyl fungicide (0 g/kg, 2 g/kg, 3 g/kg, 5 g/kg, and 7 g/kg seeds). The combination of resistant varieties with metalaxyl at a dose level of 5 g and 7 g/kg of corn seeds was effective in controlling downy mildew caused by P. maydis. In Bima-3 varieties Bantimurung and Lagaligo showed low infection reactions of 7.7-8.1%, and 10.4–11.2% respectively. In a combination of treatment conditions of susceptible varieties (Anoman) with 2, 3, 5 and 7 g/kg seeds, disease incidence reach 100% or most of the plants die. The lower incidence was also followed by yield, ear length and weight of a 1000 seeds that higher than other treatments. The combination of the use of susceptible variety with 2 g to 7 g/kg of metallaxyl doses was not effective in controlling downy mildew caused by P. maydis.

ABSTRACT: Prohexadione-calcium (ProCa) is a gibberellin biosynthesis inhibitor and the objective of this study was to evaluate the effect of ProCa on the downy mildew (DM) and Botrytis bunch rot (BBR) epidemiology, and the yield and technological and phenolic parameters of grape Merlot cultivar. Experiments were carried out in a commercial vineyard in São Joaquim Municipality/SC, Southern Brazil, during the 2017-2018 and 2018-2019 growing seasons. ProCa was applied at four doses and three phenological stages: A) 0 g ha−1 (control); B) 1000 g ha−1 (inflorescence fully developed); C) 500 + 500 g ha−1 (inflorescence fully developeted and full flowering); D) 500 + 500 + 500 g ha−1 (inflorescence fully developed, full flowering, and berries pea-sized). DM and BBR incidence and severity were quantified weekly from the first symptom appearance until harvest, and their epidemiology was compared according to: a) the beginning of symptom appearance; b) the time to reach the maximum disease incidence and severity; c) the maximum value of disease incidence and severity; d) the area under the disease progress curve. In general, there were significant differences in the ProCa doses for all epidemiologic parameters of DM and BBR compared with the control plot; however, there was no significant difference among the ProCa doses. The principal epidemiological variables that differentiated the effect of ProCa on the DM and BBR control were the Smax and AUSDPC. Some yield and technological and phenolic parameters were negatively affected by different doses of ProCa, but it was still a good option for DM and BBR control in highland region of southern Brazil during the 2017-2018 and 2018-2019 growing seasons.

Sunflower (Helianthus annuus L.) plants showing systemic downy mildew (Plasmopara helianthi Novot.) symptoms (stunting and leaf chlorosis) were collected during May and June 2001-2003 in fields located in four regions of central Italy (Umbria, Tuscany, The Marches, and Emilia-Romagna). Generally restricted, a low disease incidence (2 to 12%) was observed in fields planted with commercial hybrids. In the surveyed areas, higher field incidences (20 to 30%) of downy mildew were only observed where suitable climatic conditions occurred. To determine pathogen race variability of P. helianthi, 18 isolates (one for each field) were recovered directly from infected leaves showing sporulation or after 24 h of incubation in a humid chamber at room temperature. After increasing initial inocula on the susceptible cv. Ala, race identification of all isolates was determined by the whole-seedling immersion technique (2) on three sets of nine differential sunflower lines (three lines per each set): HA-304, RHA 265, RHA 274; PMI-3, PMI-17, 803-1; and HAR-4, QHP-1, HA 335 using triplet coding (1). All differential lines were tested twice with 40 seeds per replicate (three replicates per line). Twelve days after inoculation, plants were placed in a chamber maintained at 20°C and 100% relative humidity for 24 h and then evaluated for a susceptible (sporulation on cotyledons and/or first true leaves) or resistance (absence of sporulation) reaction. Races 700 and 703 were identified during the 3-year survey. Race 700 was recovered frequently (56% of isolates) from the Italian regions except Emilia-Romagna. Race 703 was not detected in Tuscany. Races 700 and 703 have been previously isolated in other European countries (3,4), but to our knowledge, this is the first report of the occurrence of either race in Italy. Our results confirm previous investigations and suggest that the restricted presence of downy mildew should be attributed to several factors: (i) lack of any source of resistance to both races in commercial hybrids; (ii) insufficient seed dressing with metalaxyl as observed on remnants of commercial hybrid treated seeds tested in laboratory analyses; and (iii) short rotation of cereals (often only 2- to 3-year interval) with sunflower crops. Although greenhouse tests showed most P. helianthi isolates were not controlled by metalaxyl seed treatment at the registered rate (2.1 g kg-1) loss of fungicide efficiency in fields has not yet been observed indicating that appropriate phytosanitary measures and proper seed treatment can provide control of sunflower downy mildew. Introdução: A osteoporose é considerada um grave problema de saúde pública em todo o mundo. No Brasil ainda não há dados estatísticos sobre a real incidência desta doença. Os glicocorticóides representam uma classe de drogas muito usada, sendo que podem influenciar significativamente a massa óssea. Entretanto, após tratamento com glicocorticóides em ratos, esses podem ou não apresentar alterações na massa óssea. Objetivo: Avaliar a eficácia da administração de dexametasona ou metilprednisolona em produzir osteoporose em ratas. Métodos: Vinte e quatro ratas, oriundas do biotério da Faculdade de Medicina de Itajubá, foram utilizadas e divididas em três grupos: controle, metilprednisolona e dexametasona. Foram avaliados íons e proteínas totais sanguíneos e feita a análise histopatológica de tecido ósseo. Resultados e conclusão: Pela análise histológica da espessura trabecular óssea, houve redução da massa óssea dos grupos tratados com glicocorticóides em comparação com o grupo controle, evidenciando o sucesso da indução de osteoporose. Além disso, houve uma maior redução da massa óssea no grupo tratado com dexametasona, em comparação com o grupo tratado com metilprednisolona. A análise bioquímica de cálcio, fósforo, magnésio e proteínas totais não sofreram diferença significativa entres os grupos, não sendo um bom marcador para remodelamento ósseo e para diagnóstico de osteoporose.Evaluating the Effectiveness of Dexamethasone or Methylprednisolone to Produce Osteoporosis in RatsIntroduction: Osteoporosis is considered a serious public health problem in worldwide. In Brazil we do not have statistics on the real disease incidence. Glucocorticoids are a class of drugs widely used, and can significantly influence bone mass. However, after treatment with glucocorticoids in rats, these may or may not show changes in bone mass. Objective: Evaluating the effectiveness of dexamethasone or methylprednisolone to produce osteoporosis in rats. Methods: Twenty-four rats, originated from the vivarium of the Faculty of Medicine Itajubá were used and divided into three groups: control, methylprednisolone and dexamethasone. Ions was evaluated blood and histopathology of bone tissue. Results and conclusion: by histologic analysis of trabecular bone thickness, a reduction of the mass bone in the groups treated with glucocorticoids compared with the group control, demonstrating the successful induction of osteoporosis. Furthermore, there was a greater reduction in bone mass in the group treated with dexamethasone compared with the group treated with methylprednisolone. The analysis biochemistry of calcium, phosphorus, magnesium and total protein did not undergo significant difference among the groups, there is a good marker for bone remodeling and osteoporosis diagnosis.

Purpose. To compare the capsule edges ultrastructure obtained by two femtosecond laser-assisted cataract surgery (FLACS) platforms and manual continuous curvilinear capsulorhexis (CCC) using scanning electron microscopy (SEM). Setting. Eye Clinic, University of Trieste, Italy. Design. Experimental comparative study. Methods. 150 anterior capsules were collected and divided into three groups as follows: Group 1 (50 capsules) obtained with manual CCC, Groups 2 and 3 (each with 50 capsules) obtained with the Catalys Laser and the LenSx Laser, respectively. All samples were imaged by means of SEM and regularity of the cut surface, and thickness of the capsule edge were evaluated and compared. Results. All femtosecond laser (FSL) capsules were perfectly circular, whereas some alteration of the circular shape was observed in the manual ones. Group 1 showed a smooth and regular capsule edge without any surface irregularity, conversely Groups 2 and 3 showed postage-stamp perforations on the capsule edge. The cut surface irregularity value in Group 2 was 1.4 ± 0.63, while it was 0.7 ± 0.49 in Group 3 (p<0.05). Group 1 had a significantly lower thickness of the capsule edge than the FSL groups (p<0.05). No statistically significant difference in the capsule edge thickness between the FSL groups was found (p=0.244). Conclusions. Despite the presence of slight cut surface irregularities, both FSL capsulotomies showed a better geometry and circularity than the manual ones. Capsulotomy specimens obtained using both FSL capsulotomies showed laser-induced alterations of the capsule edge when compared with smooth and regular edges obtained using manual CCC.
