Effect of Grape Seeds (Vitis vinifera L.) and Mandarin Peels (Citrus reticulate L.) Extracts on the Cardiotoxicity Induced by Cyclophosphamide in Rats

Aims: The current study was developed to investigate the influence of grape seeds (GS) and mandarin peels (MP) extracts as powerful antioxidants on the cardiotoxicity induced by cyclophosphamide (CP) in rats. Place of Study: Department of Biochemistry and Nutrition, Faculty of Women for Arts, Science and Education, Ain Shams University. Methodology: Sixty adult male Sprague-Dawley rats were divided into 6 groups. Group (1): Rats were received distilled water daily orally for 6 weeks and injected interperitoneally (i.p) with saline (0.9 %) (2.5 ml / kg BW) as single dose at the end of the sixth week of experiment. Group (2): Rats were received distilled water orally and injected with single dose of cyclophosphamide which dissolved in saline (200 mg/kg BW. i.p.) at the end of the sixth week of experiment. Groups (3 and 4):  Rats were received grape seeds extracts low and high doses (150 and 300 mg /kg BW), respectively daily orally for 6 weeks then injected with cyclophosphamide as group 2. Groups (5 and 6): Rats were received mandarin peels extracts low and high doses (150 and 300 mg /kg BW), respectively daily orally for 6 weeks then injected with cyclophosphamide as group 2. Results: Our results documented that CP caused a significant increase in serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALK-P), creatine kinase (CK-MB), lactate dehydrogenase (LDH), creatine kinase (CK) enzymes activity and serum malondialdehyde (MDA) level. While total antioxidant capacity level (TAC) showed a significant decrease. On the other hand cardiac catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GPx) activities and cardiac β cell lymphoma (Bcl-2) level showed a significant decrease in CP group while cardiac p53, caspase-3 and DNA fragmentation levels showed a significant increase in CP intoxicated group. Also, some histopathological changes were observed in liver and heart tissues in CP group. Oral administration of GS and MP caused an ameliorative effect in oxidative and apoptotic biomarkers, liver and heart function enzymes activity with an improvement of histopathological changes in liver and heart tissues. Conclusion: Our data proved that the protective effect of grape seeds and mandarin peels in cyclophosphamide intoxicated group may be due to their antioxidant, anti-inflammatory and anti- apoptotic properties.

Aims: The current study was developed to investigate the influence of ginger (G), ginger nanoparticles (GNPs) and ginger nano-base (GNB) on hepato-renal toxicity induced by carbon tetrachloride (CCl4) in rats in comparison with silymarin (SM). Place and Duration of Study: Department of Biochemistry and Nutrition, Faculty of Women for Arts, Science and Education, Ain Shams University. Methodology: Fifty-four adult male Sprague-Dawley rats were divided into 6 groups. Group (1): Rats received distilled water orally and injected intraperitoneally (i.p.) with single dose of corn oil (1 ml/kg b.wt). Group (2): Rats were injected with single dose of CCl4 diluted with corn oil (1:1) (1 ml/kg b.wt. i.p.) at the 4th week of experiment. Groups (3), (4) and (5): Rats were orally received 50 mg /kg b.wt./day of G, GNPs and GNB, respectively for 8 weeks and injected with CCl4 as group 2. Group (6): Rats were orally received 100 mg /kg b.wt /day of SM for 8 weeks and injected with CCl4 as group 2.  Results: Our results documented that injection with CCl4 caused significant increase (p<0.05) in liver function tests [serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP) activities], kidney function tests [serum creatinine, urea, uric acid and cystatin C] and serum levels of malondialdehyde (MDA), Nitric oxide (NO), tumor necrosis factor- alpha (TNF- α) and interleukin 1 beta (IL-1β). On the other hand, there was a significant decrease (p<0.05) in the serum total antioxidant capacity (TAC), Hepatic catalase (CAT) and superoxide dismutase (SOD) enzymes activity, with histopathological changes in liver and kidneys tissues. Oral administration of G, GNPs, GNB and SM caused an enhancement of liver and kidney function, decreasing serum oxidants and inflammatory markers levels while increasing the activities of antioxidant enzymes, also an improvement of organs histopathological changes was observed. Conclusion: Our data proved that using ginger in the form of GNPs and GNB are more efficient in ameliorating hepato-renal toxicity induced by CCl4 than using native ginger as evidenced by biochemical analysis and histological examination of liver and kidneys tissues.

This study aimed to evaluate the liver, kidney damage caused by S. typhimurium and to estimate the oxidative damage in association with this bacteria. A highly virulent isolates of S. typhimurium were obtained from the department of internal and preventive medicine/ College of Veterinary Medicine/ University of Baghdad. A twenty five local rabbits of both genders with age range (2-4 months) weeks old were used for this study, the rabbits were divided randomly into five groups each group contains 5 rabbits :- group 1: drenched orally with 5 ml of normal saline and consider as control group, group 2: were drenched orally with (5 ml) suspension which contain (5��109 CFU) of Salmonella typhimurium and regarded as infected group, group 3 were drenched orally with (5 ml) suspension which have (5��109 CFU) of Salmonella typhimurium then treated with a single dose of gentamicin alone at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation), group 4 were drenched (5 ml) suspension having (5��109 CFU) of Salmonella typhimurium then treated with a single dose of Ca-EDTA alone at 40mg/kg orally after presence of signs (after 24hrs. post inoculation) and group 5 were drenched (5 ml) suspension that contain (5��109 CFU) of Salmonella typhimurium then treated with a single dose of combined gentamicin at 0.05ml/kg (5mg/ml) orally after presence of signs (after 24hrs. post inoculation) and Ca-EDTA 40mg/kg after presence of signs (after 24hrs. post inoculation).The results of biochemical profile showed a significant increase (p less than 0.05) in ALT, creatinine and urea levels in infected group as compared with control group, while, the treated groups especially group 5 showed a significant improvement in ALT, Urea and creatinine levels which returned to relative normal levels as compared with infected group after 96hrs. post treatment. Also, the results of oxidative stress showed a significant increase in the levels of MDA in G2, G3, G4 and G5 after 48 hrs. post treatment, while the level of GSH showed a significant decrease in the level at 48hrs., both were returned to relative normal levels after 96hrs.post treatment especially in group 5.In conclusion, S. typhimurium can causing liver and kidney damage which is manifested by increase ALT, Urea and Creatinine. Also, MDA and GSH is increased due to salmonellosis.

Objective: Oronasal fistula (ONF) is a known complication after primary palatoplasty (PP). Studies investigating the effect of perioperative antibiotics on fistula rates after PP are limited by inadequate sample size or reliance on self-reporting through national databases. In this study, the authors evaluated the association between single-dose perioperative antibiotics and postoperative fistula rates after PP at a single institution. Design: A retrospective study. Participants: Children younger than 2 years who underwent PP from April 2009 to September 2019 were included. Interventions: Patients were divided into 2 categories: Group 1 received a single intraoperative dose of IV antibiotic, while group 2 did not. Main Outcome Measure(s): Outcome measures included ONF formation, length of stay (LOS), and 30-day readmission rates. Multivariable firth logistic regression, quantile regression, and χ2 tests were performed. Results: Of the 424 patients, 215 and 209 patients were in groups 1 and 2, respectively. The overall ONF rate was 1.9% among all patients. Patients in group 1 experienced an ONF rate of 3.3%, while patients in group 2 had an ONF rate of 0.5%. After correcting for confounding variables, the difference in ONF rates was not statistically different ( P = .68). Median LOS was 35.7 hours and 35.5 hours ( P = .17), while the rate of readmission within 30 days was 4.7% and 2.4% for group 1 and 2, respectively ( P = .96). Conclusions: Administration of a single-dose perioperative antibiotic did not decrease fistula formation after PP, nor did it affect the patient’s LOS or 30-day readmission rate.

Background The rationale of this study was to compare high-dose epidural clonidine with a more commonly used agent, such as bupivacaine. This was performed to give a more objective idea of the relative analgesic potency of epidural clonidine. Methods Sixty patients undergoing intestinal surgery during propofol anesthesia were studied. At induction, the patients received epidurally a dose of 10 micrograms/kg [corrected] clonidine in 7 ml saline followed by an infusion of 6 micrograms [corrected] x kg(-1) x h(-1) (7 ml/h) (group 1, n = 20), a dose of 7 ml bupivacaine, 0.5%, followed by 7 ml/h bupivacaine, 0.25% (group 2, n = 20), or a dose of 7 ml bupivacaine, 0.25%, followed by 7 ml/h bupivacaine, 0.125% (group 3, n = 20). Intraoperatively, increases in arterial blood pressure or heart rate not responding to propofol (0.5 mg/kg) were treated with intravenous alfentanil (0.05 mg/kg). Additional doses of propofol were given to maintain an adequate bispectral index. The epidural infusions were maintained for 12 h. In cases of subjective visual analogue pain scores up to 5 cm at rest or up to 8 cm during coughing, the patients were given access to a patient-controlled analgesia device. Results During anesthesia, patients in group 1 required less propofol than those in groups 2 and 3 (78 [36-142] mg vs. 229 [184-252] mg and 362 [295-458] mg; P &lt; 0.05) and less alfentanil than patients in group 3 (0 [0-0] mg vs. 11 [6-20] mg; P &lt; 0.05). Analgesia lasted 380 min (range, 180-645 min) in group 1 versus 30 min (range, 25-40 min) in group 2 and 22 min (range, 12.5-42 min) in group 3 (P &lt; 0.05). There was no suggestion of a hemodynamic difference among the three groups except for heart rates that were significantly reduced in patients in group 1. Sedation scores were significantly higher in this group during the first 2 h postoperatively. Conclusion Our results show that high doses of epidural clonidine potentiate general anesthetics and provide more efficient postoperative analgesia than the two bupivacaine dosage regimens investigated.

AbstractAbdominal angiostrongyliasis (AA) is caused by Angiostrongylus costaricensis, which inhabits mesenteric arteries. There is no drug treatment for AA, and since intestinal infarction due to thrombi is one of the main complications of the disease, the use of anticoagulants may be a treatment option. Thus, we aimed to assess the effect of high doses of enoxaparin on the prevention of ischaemic intestinal lesions and on the survival of mice infected with A. costaricensis. Twenty-four mice were infected with L3 of A. costaricensis and divided equally into two groups: Group 1, control treated with placebo, and Group 2, treated daily with enoxaparin (2.5 mg/kg) for 50 days. All mice were subjected to necropsy and histological analysis. The results from gross and microscopic assessments showed no variation in the prevalence of lesions between the groups. An analysis was also performed among survivors and non-survivors, showing that animals that died often presented lesions, such as granulation tissue in the serosa, and intestinal infarction and adhesion. The mortality rate did not vary between the enoxaparin-treated and control groups. Thus, we showed that high doses of enoxaparin have no protective effect against AA, as the survival rates and lesions of mice did not vary between the treated and control groups. Considering that the use of prophylactic doses was also shown to be ineffective in a previous study, we do not recommend the use of enoxaparin for AA treatment.

Aim: To evaluate the nephroprotective property of Cnidoscolus chayamansa aqueous leaf extract in diabetic rats. Study Design: Rats were randomly divided into five groups with group 1 as the normal control. Diabetic was induced in groups 2-4. Group 2 was used as the test control while groups 3 and 4 were treated with different concentrations of the leaf extract; group 5 was treated with the standard drug, glipizide. Place and Duration of Study: Departments of Biochemistry and Anatomy, University of Jos, Nigeria, between August to November, 2019. Methodology: Fourty (40) male albino Wistar rats were grouped into five groups. The rats were treated for fourteen days and then sacrificed by decapitation after anaesthesia. Blood was collected for biochemical parameters; kidney was excised and stored in formaldehyde until required for histopathological study. Serum urea, creatinine, uric acid, sodium, potassium, chloride and bicarbonate were determined using appropriate methods. Results: The test control had a significant (P < .05) decrease in the concentrations of Na+, Cl- and HCO3- ions; significant (P < .05) increases in serum K+ ion, urea, uric acid and creatinine. Treatments of the test groups with the different doses of the leaf extract and the standard drug increased the concentration of Na+ ion which was not significantly (P < .05) different from the test control. On the other hand, the concentration of Cl- and HCO- ions  were significantly (P < .05) increased; the concentrations of K+, urea, uric acid and creatinine were significantly (P < .05) decreased. The histochemistry of the kidneys revealed that the injury brought about under diabetic condition was ameliorated with the treatments with the low and high doses of the leaf extract, and the standard drug. Conclusion: The results show that the aqueous leaf extract has nephroprotective property.

PURPOSE: To evaluate the shear bond strength (SBS) of metallic brackets bonded with Transbond Plus Self-Etching Primer (TPSEP) and Transbond Plus Color Change (TPCC) under contamination with a mixture of saliva and blood. MATERIALS AND METHODS: 42 human premolars were randomly divided into 2 groups (n=21). Group 1 (G1) were bonded under no contamination, and Group 2 (G2) was contaminated with saliva/blood before bonding. Both groups were bonded according to the manufacturer’s instructions and were cleaned with pumice before bonding. The shear bond strength (SBS) tests were performed after 24 hours in distilled water at 37°C and after thermocycling. RESULTS: Both groups showed a homogeneous distribution in the Levene’s test (p>0.05). The main shear bond strength value of G1 was 8.89 MPa with a standard deviation of 2.27; the value for G2 was 6.00 MPa with a standard deviation of 2.62 MPa. There was a significant difference between G1 and G2 (t-student test p<0.05). IRA scores indicated that the main value was IRA 0, and no significant difference (α=0.05) was found between the groups. CONCLUSION: Contamination of blood mixed with saliva significantly decreases the shear bond strength, but even so, the performance is clinically acceptable.

The exposure to high doses of ionizing radiation during radiotherapy results in severe morphological and functional alterations of the salivary glands, such as xerostomia. In the present study we investigated the chronic effect of a single radiation dose of 15 Gray (Gy) limited to head and neck on rat salivary gland function (salivary secretion and gland mass) and histology. Results indicate that norepinephrine (NE)-induced salivary secretion was reduced significantly at 30, 90, 180 and 365 days after the administration of a single dose of 15 Gy of ionizing radiation compared to non-irradiated animals. The maximal secretory response was reduced by 33% at 30 and 90 days post irradiation. Interestingly, a new fall in the salivary response to NE was observed at 180 days and was maintained at 365 days post irradiation, showing a 75% reduction in the maximal response. The functional fall of the salivary secretion observed at 180 days post irradiation was not only associated with a reduction of gland mass but also to an alteration of the epithelial architecture exhibiting a changed proportion of ducts and acini, loss of eosinophilic secretor granular material, and glandular vacuolization and fibrosis. On the basis of the presented results, we conclude that ionizing radiation produces irreversible and progressive alterations of submandibular gland (SMG) function and morphology that leads to a severe salivary hypo-function.

ABSTRACT. The effects of a signaling anti-CD28 mAb (JJ319), which interferes with the CD28-B7 T cell costimulation pathway thought to be involved in the development of chronic rejection of organ transplants, was investigated. Functional, morphologic, and molecular changes in rat renal allografts were examined up to 24 wk after placement. Control Lewis rats, recipients of F344 kidneys, received a single dose of a nonspecific mouse mAb intravenously on the day of transplantation (group 1). Group 2 animals were given anti-CD28 mAb in similar fashion. Group 3 animals were treated with a short course of cyclosporin A (CsA), and group 4 received both anti-CD 28 mAb and CsA. The majority (>95%) of animals in groups 2, 3, and 4 survived throughout the follow-up, compared with 28% in group 1 (P < 0.001). Group 2 and 4 recipients produced negligible proteinuria, whereas group 1 controls developed progressively increasing proteinuria after 4 wk and group 3 animals developed proteinuria by 24 wk. Allografts in groups 2 and 4 were morphologically unremarkable at 24 wk. Kidneys of group 1 animals rapidly developed changes of acute rejection, and those that survived long-term showed extensive glomerulosclerosis and interstitial fibrosis. Changes of early chronic rejection were noted in group 3 grafts. By reverse transcriptase–PCR, expression of representative inflammatory factors interferon-γ and interleukin-10 were significantly elevated at 24 wk only in the surviving group 1 animals. A single dose of a signaling anti-CD28 mAb administered at transplantation or in combination with a short course of CsA significantly prolonged recipient survival, normalized function, and preserved the morphology of renal allografts in an established model of chronic rejection. These data support an important role for T cell costimulation in the evolution of the chronic process.

Aluminum is recognized as a public health concern because of its potential toxic effects on human health. Therefore, the present experiment was undertaken to determine the effectiveness of curcumin (CUR) in reducing the hepatotoxicity induced by aluminum chloride (AlCl3) in animals. In this experimental study, forty male rats were allocated to five groups (N = 8), viz. no treatment (control), solvent (DMSO or distilled water), CUR (10 mg/kg B.W.), AlCl3 (10 mg/kg B.W.), and CUR+AlCl3 (each with 10 mg/kg B.W.). Treatments were performed by intra-peritoneal injections for 28 days. On the final day, animals were sacrificed, and liver function markers in blood plasma, hepatic antioxidants, and lipid peroxidation index in liver homogenate were estimated. AlCl3 treatment resulted in a significant increase in plasma AST, ALT, ALP and LDH activities with decreased total protein compared to control. AlCl3 significantly reduced superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels but increased malondialdehyde (MDA) level in the liver compared to control. AlCl3 also caused various histopathological changes in the livers of rats. Curcumin could normalize nearly all these parameters. CUR improved levels of changes in different parameters when was combined with AlCl3. It is concluded that CUR has beneficial effects being able to antagonize AlCl3 toxicity.
