This research is a study on the development of innovative clay mask mixed with the Acanthus ebracteatus Vahl extract product. The purposes of this research are as follows. Firstly, the research is to develop a clay mask product mixed with natural extracts from the Acanthus ebracteatus Vahl. Secondly, it is aimed to study on the antioxidant activity, anti-inflammatory effect, antimicrobial effect and total Phenolic contents of the Acanthus ebracteatus Vahl extracts. Thirdly, it is tostudy stability of the product. Finally, the research is to study on the calculation of suitable product formulations for clay products mixed with Acanthus ebracteatus Vahl extracts. By starting from the selection of raw materials, the extracts are prepared to determine the total phenolic content. Later, 2,2-diphenyl-1-picrylhydrazyl (DPPH) Assay method is conducted for antioxidant activity test, followed by anti-inflammatory effect test (Nitric oxide production inhibitory activity test, inhibition of LPS-induced TNF-α secretion test and Anti-inflammatory activity test (COX-2). Afterwards, a suitable formulation is developed. The test for the safety and physical properties of the product is then carried out followed by the skin irritation test in a group of 10 volunteers in order to ensure that the clay mask product containing Acanthus ebracteatus Vahl extracts is effective and safe. From the results of the product stability study, the findings indicated that the clay mask product containing Acanthus ebracteatus Vahl extracts did not show any phase separation. The irritation test in 10 volunteers after 12 hours of using the product showed no sign of allergic reaction, itching or irritation. Toxicity test results found no cytotoxicity. The anti-inflammatory effect test was found to have potent anti-inflammatory properties, when tested in the RAW 264.7 macrophage cell. This study is only for the development of an innovative product of skin clay mask mixed with the Acanthus ebracteatus Vahl extracts.

ABSTRACTNanamuha (Bridelia monoica Merr.) is a plant that was used as a cancer and tumor drug by the Sangihe Islands community. This plant contains phenol compounds, flavonoids, alkaloids and tannins. The purpose of this study was to determine the potential of nanamuha leaves that grow in the Sangihe Island to have antioxidant activity and determine the total phenolic ethanol extracts of nanamuha leaf. The extraction method used is maceration with ethanol solvent p.a. The extracts obtained was determined total phenolic and antioxidant activity test using DPPH method. Based on the results obtained, the ethanol extracts of nanamuha leaves has antioxidant activity with an IC50 value of 75,03 µg / mL and the result showed that the total phenol content the ethanol extracts of nanamuha leaves of  53,34 mg / L.  Keywords : Nanamuha leaf, total phenol, antioxidant activity, DPPH  ABSTRAKNanamuha (Bridelia monoica Merr.) merupakan tanaman yang dimanfaatkan sebagai obat kanker dan tumor oleh masyarakat Kepulauan Sangihe. Tujuan dari penelitian ini adalah unutuk mengetahui potensi dari daun nanamuha yang tumbuh di Kepulaun Sangihe ini memiliki aktivitas antioksidan dan menentukan total fenolik dari ekstrak etanol daun nanamuha. Metode ekstraksi yang digunakan adalah maserasi dengan pelarut etanol p.a. Ekstrak yang diperoleh ditentukan total fenolik dan uji aktivitas antioksidan dengan metode DPPH. Berdasarkan hasil yang didapatkan ekstrak etanol daun nanamuha memiliki aktivitas antioksidan dengan nilai IC50 75,03 µg/mL dan hasil penelitian menunjukan total kandungan fenolik ekstrak etanol daun nanamuha sebesar 53,34 mg/L. Kata kunci : Daun Nanamuha, Total fenol, Aktivitas antioksidan, DPPH

The aim of this study was to evaluate the total phenolic and flavonoid content, and the in vitro antioxidant, anti-inflammatory, antibacterial, antifungal, antimalarial, cytotoxicity, and antiprotozoal activities of the Algerian plant Cytisus villosus Pourr. (Syn. Cytisus triflorus L’Hérit.). Additionally, the radioligand displacement affinity on opioid and cannabinoid receptors was assessed for the extracts and isolated pure compounds. The hydro alcoholic extract of the aerial part of C. villosus was partitioned with chloroform (CHCl3), ethyl acetate (EtOAc), and n-butanol (n-BuOH). The phenolic content of the C. villosus extracts was evaluated using a modified Folin–Ciocalteau method. The total flavonoid content was measured spectrometrically using the aluminum chloride colorimetric assay. The known flavonoids genistein (1), chrysin (2), chrysin-7-O-β-d-glucopyranoside (3), and 2″-O-α-l-rhamnosylorientin (4) were isolated. The antioxidant activities of the extracts and isolated compounds were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DDPH) and cellular antioxidant activity (CAA) assays. The plant extracts showed moderate antioxidant activity. EtOAc and n-BuOH extracts showed moderate anti-inflammatory activity through the inhibition of induced nitric oxide synthase (iNOS) with IC50 values of 48 and 90 µg/mL, respectively. The isolated pure compounds 1 and 3 showed good inhibition of Inducible nitric oxide synthase (iNOS) with IC50 values of 9 and 20 µg/mL, respectively. Compounds 1 and 2 exhibited lower inhibition of Nuclear Factor kappa-light-chain-enhancer of activated B cells (NF-κB) with IC50 values of 28 and 38 µg/mL, respectively. Furthermore, the extracts and isolated pure compounds have been shown to exhibit low affinity for cannabinoid and opioid receptors. Finally, n-BuOH extract was a potent inhibitor of Trypanosoma brucei with IC50 value of 7.99 µg/mL and IC90 value of 12.61 µg/mL. The extracts and isolated compounds showed no antimicrobial, antimalarial nor antileishmanial activities. No cytotoxic effect was observed on cancer cell lines. The results highlight this species as a promising source of anti-inflammatory and antitrypanosomal agents.

AbstractBackgroundAsparagus horridus is an edible plant known as “Ayrelli” in North Cyprus. The scientific literature has not yet submitted a report about the antioxidant, anti-inflammatory and anti-cancer activities of A. horridus plant from North Cyprus until now. The purpose of the research was to determine the antioxidant, anti-inflammatory and anti-cancer activities of A. horridus.Materials and methodsSoxhlet extraction of A. horridus was performed using methanol. Antioxidant activity was determined by DPPH, TFC, FRAP and TPC assays. Protein-denaturation assay was performed to determine the anti-inflammatory effect. The anti-cancer effects of the extract on HepG2 and B-CPAP cell lines were determined with MTT assay.ResultsAntioxidant activity for A. horridus extract was determined by DPPH (50%), TFC (266.26 μg QUE/mg extract), FRAP (1.27 μg FeSO4/mg extract) and TPC (167.613 μg GAE/mg extract) assays at 25 mg/mL. Inhibition of protein-denaturation activity was found as 29.42% at 25 mg/mL. After 24 h of the extract treatment, cell proliferation of HepG2 and B-CPAP cancer cells were inhibited at IC50 values 63.24 μg/mL and 101.24 μg/mL, respectively.ConclusionThese results have shown that the methanol extract of A. horridus grows in North Cyprus has antioxidant, anti-inflammatory and anti-cancer activities.

Abstract Background Inflammation is a symptom associated with many diseases. This symptom is treated with steroidal and non-steroidal anti-inflammatory drugs, which can cause severe side effects when used as long-term treatments. Natural products are an alternative source of new compounds with anti-inflammatory activity. Jefea gnaphalioides (Astereaceae) (A. Gray) is a plant species used to treat inflammatory problems, in Mexico. This study determined the anti-inflammatory activity and the composition of the methanol extract of Jefea gnaphalioides (MEJG). Methods The extract was obtained by heating the plant in methanol at boiling point for 4 h, and then the solvent was evaporated under vacuum (MEJG). The derivatization of the extract was performed using Bis-(trimethylsilyl) trifluoroacetamide, and the composition was determined by GC-MS. Total Phenols and flavonoids were determined by Folin-Ciocalteu AlCl3 reaction respectively. The antioxidant activity of MEJG was determined by DPPH method. The acute and chronic anti-inflammatory effects were evaluated on a mouse ear edema induced with 12-O-Tetradecanoylphorbol-13-acetate (TPA). Acute oral toxicity was tested in mice at doses of MEJG of 5000, 2500 and 1250 mg/kg. The levels of NO, TNF-α, IL-1β and IL-6 were determinate in J774A.1 macrophages stimulated by Lipopolysaccharide. The production of inflammatory interleukins was measured using commercial kits, and nitric oxide was measured by the Griess reaction. Results The anti-inflammatory activity of MEJG in acute TPA-induced ear edema was 80.7 ± 2.8%. This result was similar to the value obtained with indomethacin (IND) at the same dose (74.3 ± 2.8%). In chronic TPA-induced edema at doses of 200 mg/kg, the inhibition was 45.7%, which was similar to that obtained with IND (47.4%). MEJG have not toxic effects even at a dose of 5000 mg/kg. MEJG at 25, 50, 100 and 200 μg/mL decreased NO, TNF-α, IL-1β and IL-6 production in macrophages stimulated with LPS. The major compounds in MEJG were α-D-Glucopyranose (6.71%), Palmitic acid (5.59%), D-(+)-Trehalose (11.91%), Quininic acid (4.29%) and Aucubin (1.17%). Total phenolic content was 57.01 mg GAE/g and total flavonoid content was 35.26 mg QE/g MEJG had antioxidant activity. Conclusions MEJG has anti-inflammatory activity.

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.

Pyrazoline compounds are reported to have many useful biological activities such as antimicrobials, antihyperglycemia, antioxidants, anti-inflammatory, anticancer and anticonvulsants. The pyrazoline analogues of 3- (2-methoxy-phenyl) -5-naphthalene-1-yl-4,5-dihydro-1H-pyrazol were synthesized usingglacial acetic acid as catalyst and assisted by microwave irradiation. The structures of the compounds were obtainedby UV, IR and HRMS spectroscopy data. Antioxidant activity test using DPPH method showed that pyrazoline compound had IC50 value of 27.41 μg/mL.

Antioxidant activity, total phenolic and total flavonoid contents of some traditional medicinal plants grown in East Kalimantan, Indonesia, were investigated. Plant sample are Macaranga gigantea leaf, Ceiba pentandra leaf, Ceiba pentandra bark, Blumea balsamifera leaf and Artocarpus altilis leaf. Natural antioxidants from plant species are considered safe. Therefore, people are now more interest in finding out the treatment through natural remedies. The plant's total phenolic content was determined by the Folin-Ciocalteu method, while total flavonoid content was determined by the Colorimetric assay method. Antioxidant activity was evaluated by DPPH radical scavenging assay. The results showed that plant samples tested contained 22.02 to 214.88 µg of gallic acid equivalents (GAE)/mg extract represented the total phenolic content. The flavonoid content of the samples was in range from 50.00 to 896.67 µg of catechin equivalents (CE)/mg extract. The plant samples displayed ability to inhibit DPPH free radical formation by 59-81% at 50 ppm concentration. These finding suggest that these plants could possess natural antioxidants and give a scientific basis to the traditional uses of the investigated plants.

Teucrium polium leaf is one of the folkloric medicinal plants used traditionally to treat many diseases in the Palestinian community. It has antibacterial antioxidant and anti-inflammatory consequences. Present study aims the evaluating the in vitro inhibitory effect of Teucrium polium leaf extracts on tumor necrosis factor-α (TNF-α) using polymorphonuclear cells (PMNCs), in addition to determine its antioxidant and total phenolic and flavonoids contents. Polymorphonuclear cells were withdrawn from whole blood according to Histopaque (Ficol-1077) method. Blood cells were cultured in an enriched Roswell Park Memorial Institute (RBMI) medium. The levels of tumor necrosis factor (TNF-α) were determined after 24 h using LPS stimulation. Total phenolic contents, flavonoids contents and antioxidant activity were measured using spectrophotometric method. The TNF-α concentrations were compared using paired-samples t test. The leaf extracts of Teucrium polium revealed significant reduction in terms of TNF-α levels. The extract contained high phenolic and flavonoids contents and its antioxidant activities were remarkable. The reduced values in the TNF-α levels as affected by Teucrium polium leaf extracts indicate its effect in anti-inflammation. The plant is rich with polyphenolic compounds and flavonoids and has strong antioxidant activity. The observed anti-inflammatory effect of the extracts under study may be discussed as the influence of the significant presence of the phenolic compounds and flavonoids.

<p>Telah dilakukan penelitian tentang formulasi dan uji aktivitas krim pengkelat merkuri berbahan dasar ekstrak etanol alga coklat (<em>Sargassum sp.</em>) telah dilakukan. Krim diformulasikan dengan beberapa bahan seperti vaselin, gliserol, asam askorbat, karbomer, setil alkohol, dan NaOH. Parameter yang dievaluasi diantaranya organoleptik krim, homogenitas, pH, daya sebar, tipe krim, iritasi kulit, dan daya tercuci krim. Aktivitas pengkelatan diamati dengan menganalisis kandungan merkuri pada kulit tikus yang sebelumnya diberikan krim bermerkuri dibandingkan dengan kulit tikus yang telah dioleskan krim pengkelat merkuri berbahan dasar ekstrak etanol alga coklat. Spektrum <em>Fourier Transform Infrared Spectroscopy</em> (FT-IR) menunjukkan bahwa ekstrak etanol alga coklat diindikasikan mengandung glutation. Berdasarkan parameter evaluasi krim, formulasi terbaik adalah krim dengan komposisi ekstrak alga coklat sebanyak 0,035% w/v terhadap volume krim, yang ditandai dengan krim yang berbentuk gel kental dan tidak berbau serta memiliki tekstur seperti krim wajah pada umumnya, yaitu lembut dan sedikit berminyak. Krim bersifat homogen dan stabil hingga hari ke 21 percobaan dengan pH=5. Krim juga tidak menimbulkan iritasi dan dapat tercuci habis tanpa bekas. Hasil analisis AAS membuktikan bahwa penggunaan krim pengkelat merkuri berbahan dasar esktrak etanol alga coklat selama kurang dari 2 minggu dengan dosis pemberian 1 kali sehari mampu menyerap merkuri pada tikus dengan aktivitas penyerapan sebesar 50%.</p><p><strong>Formulation and Activity Test of Mercury Chelator Cream of Ethanol Extract of Brown Algae (<em>Sargassum Sp.</em>). </strong>Research on the study of formulation an activity test of mercury chelator cream from ethanol extract has successfully done. The cream was formulated with ingredients such as vaseline, glycerol, ascorbic acid, carbomer, cetyl alcohol, and sodium hydroxide. Parameters were evaluated against organoleptic, homogenity, pH, dispersion type of cream, skin irritation test and washing test. The mercury chelating activity was observed by analyzing mercury concentration in mercury induced mouse skin. The FTIR spectrum shows that ethanol extract of brown algae was dominated by glutathione functional group. The best cream formulation was composed by 0,035% (w/v) brown algae, formed as a creamy gel that was thick and odorless and had a texture like soft facial cream in general with slightly oily.The cream was homogeneous and stable until the 21<sup>st</sup> day of the experiment with pH = 5. The cream produced no skin irritation and could be washed out without a trace. The results of AAS analysis proved that the use of brown algae ethanol extract-based mercury chelating cream for less than 2 weeks with a dose of 1 time a day could absorb mercury in mouse skin with absorption activity of 50%.</p>

Змееголовник молдавский (Dracocephalum moldavica L.) в народной медицине используется в качестве противовоспалительного, ранозаживляющего, отхаркивающего и седативного средства. В ФГБНУ ВИЛАР разработан змееголовника молдавского травы экстракт сухой под условным названием «Люкатил» (сумма фенольных соединений 64,12% в пересчёте на цинарозид). Цель работы - изучение острой токсичности и противовоспалительной активности экстракта змееголовника для разработки на его основе лекарственного препарата. Методика. Проведено определение параметров острой токсичности и противовоспалительной активности экстракта. При изучении острой токсичности экстракта по методу Кербера использованы белые нелинейные мыши-самцы в количестве 30 особей. «Люкатил» вводили животным внутрижелудочно в дозах 500, 1000, 1500 и 2000 мг/кг. Для выявления противовоспалительной активности экстракта змееголовника молдавского использована in vitro ферментная биотест-система на основе индуцибельной NO-синтазы. Для выявления противовоспалительной активности экстракта in vivo использованы нелинейные мыши-самцы. Оценку влияния экстракта в дозе 200 мг/кг на экссудативную стадию воспаления проводили на модели 1% формалинового отёка. В качества препарата сравнения использовали индометацин (5 мг/кг). Формалиновый отёк вызывали однократным субплантарным введением под апоневроз задней правой лапки мыши 0,05 мл 1% формалина в качестве флогогенного агента. Величину отёка определяли по разнице в массе лапок контрольных и опытных животных и рассчитывали процент снижения степени отёка. Результаты. При однократном введении экстракт «Люкатил» не приводил к гибели животных, изменения внешнего вида и поведенческих реакций мышей не наблюдалось. В соответствии с классификацией токсичности химических веществ по ГОСТ 12.1.007-76 «Люкатил» является малотоксичным веществом. In vitro установлена высокая противовоспалительная активность экстракта, при этом остаточная активность iNOS снижалась до 25%. Экстракт в дозе 200 мг/кг in vivo обладал статистически значимым противовоспалительным эффектом. Он подавлял развитие экссудативной фазы воспаления на 33,7%, по сравнению с контрольной группой животных, уступая противовоспалительному эффекту индометацина. Заключение. Змееголовника молдавского травы экстракт сухой под условным названием «Люкатил» является малотоксичным веществом, обладает выраженным противовоспалительным эффектом в опытах in vitro, in vivo и является перспективным объектом для дальнейшего фармакологического изучения в качестве противовоспалительного лекарственного средства.Moldavian dragonhead (Dracocephalum moldavica L.) is used in traditional medicine as an anti-inflammatory, wound-healing, expectorant, and sedative means. In our Institute, a Moldavian dragonhead herb dry extract (total phenolic content, 64.12% in cynaroside equivalent) was developed and conventionally named Lyukatil. Objective. To study acute toxicity and anti-inflammatory activity of the dragonhead extract for developing a drug based on this extract. Method. Parameters of acute toxicity and anti-inflammatory activity of the extract were assessed. The study of acute toxicity of the extract was performed using the Kerber method on male white mongrel mice (n=30). Lyukatil was administered to the animals intragastrically at doses of 500 mg/kg, 1000 mg/kg, 1500 mg/kg, and 2000 mg/kg. Anti-inflammatory activity of the Moldavian dragonhead extract was determined in vitro using an enzyme Biotest system based on inducible NO synthase. Mongrel male mice were used to study the anti-inflammatory activity of the extract in vivo. The effect of the extract at a dose of 200 mg/kg on the exudative phase of inflammation was evaluated on a model of 1% formalin-induced edema. Indomethacin 5 mg/kg was used as a reference drug. Formalin edema was induced by a single subplantar injection of 0.05 ml of 1% formalin as a phlogogenic agent under the aponeurosis of the right hind leg. The degree of edema was determined by the difference in leg weights in control and experimental animals; then the decrease in edema was calculated in per cent. Results. A single administration of the extract Lyukatil did not cause death of animals, changes in the appearance or in behavioral responses, shortness of breath, or drowsiness. In accordance with the toxicity classification for chemical substances as per GOST Standard 12.1.007-76, Lyukatil is a low-toxic substance. The extract at a dose of 200 mg/kg exerted a significant anti-inflammatory effect as shown by suppression of the exudative phase of formalin-induced inflammation by 33.7% compared to the control group. However, this effect was inferior to the anti-inflammatory effect of indomethacin. Conclusions.
