Infectious bronchitis virus (IBV), a gammacoronavirus, is an economically important virus to the poultry industry, as well as a significant welfare issue for chickens. As for all positive strand RNA viruses, IBV infection causes rearrangements of the host cell intracellular membranes to form replication organelles. Replication organelle formation is a highly conserved and vital step in the viral life cycle. Here, we investigate the localization of viral RNA synthesis and the link with replication organelles in host cells. We have shown that sites of viral RNA synthesis and virus-related dsRNA are associated with one another and, significantly, that they are located within a membrane-bound compartment within the cell. We have also shown that some viral RNA produced early in infection remains within these membranes throughout infection, while a proportion is trafficked to the cytoplasm. Importantly, we demonstrate conservation across all four coronavirus genera, including SARS-CoV-2. Understanding more about the replication of these viruses is imperative in order to effectively find ways to control them.

Lameness is an animal welfare issue that incurs substantial financial and environmental costs. This condition is commonly caused by digital dermatitis (DD), sole ulcers (SU), and white line disease (WLD). Susceptibility to these three foot disorders is due in part to genetics, indicating that genomic selection against these foot lesions can be used to reduce lameness prevalence. It is unclear whether selection against foot lesions will lead to increased susceptibility to other common diseases such as mastitis and metritis. Thus, the aim of this study was to determine the genetic correlation between causes of lameness and other common health disorders to identify loci contributing to the correlation. Genetic correlation estimates between SU and DD and between SU and WLD were significantly different from zero (p < 0.05), whereas estimates between DD and mastitis, DD and milk fever, and SU and metritis were suggestive (p < 0.1). All five of these genetic correlation estimates were positive. Two-trait genome-wide association studies (GWAS) for each of these five pairs of traits revealed common regions of association on BTA1 and BTA8 for pairs that included DD or SU as one of the traits, respectively. Other regions of association were unique to the pair of traits and not observed in GWAS for other pairs of traits. The positive genetic correlation estimates between foot disorders and other health disorders imply that selection against foot disorders may also decrease susceptibility to other health disorders. Linkage disequilibrium blocks defined around significant and suggestive SNPs from the two-trait GWAS included genes and QTL that were functionally relevant, supporting that these regions included pleiotropic loci.

Objective: To review how transport and stunning of cattle affect animal welfare. Approach: During the transport of beef cattle to slaughter plants, several factors affect animal welfare, such as travel time, stress, and load density. Additionally, the correct stunning of cattle helps comply with the animal welfare guidelines established by different protocols such as Welfare Quality®. Study limitations/Implications: Meat quality is affected by several factors, being of utmost importance the way animals are transported to the slaughterhouse, and they are stunned. Therefore, it is critical to perform these stages properly to obtain good quality meat; besides, it is a welfare issue. Conclusions: It is critical to comply with transport and slaughter procedures that guarantee good beef meat quality and ensure animal welfare to avoid stress in cattle as possible.

Specific anatomy of the porcine stomach results in a high risk of ulceration in the pars oesophagea. A fully developed ulcer leads to intermittent gastrointestinal bleeding followed by clinical symptoms of chronic pain; thus, gastric ulceration in pigs causes significant production losses and serious welfare problems. Nowadays, gastric ulceration is common in domestic pigs throughout the world, but a great deal of attention has been directed at finishers. This paper was intended to provide new data on the prevalence of the disease in commercially reared highly prolific sows. The study was carried out on 329 culled sows from four commercial farms located in Northern Poland. The severity of gross pathological lesions of pars oesophagea was assigned to grade 0 (no lesions), 1 (parakeratosis), 2 (erosion), or 3 (gastric ulceration with or without stenosis) by a veterinary practitioner. Abattoir analysis revealed 66.6% of sows’ stomachs with some degree of alterations. Gastric ulceration was detected in 45.5% of examined organs. Parakeratosis and erosion were found in 1.2 and 15.8% of stomachs, respectively. The investigation did not find any oesphageal strictures. Our investigation of the problem in high health status hyper-prolific sows showed the lowest prevalence of gastric lesions (i.e., ulcerative and preulcerative alterations collectively) compared to earlier studies but the highest percentage of gastric ulcers. The results indicate that gastric ulcers may be a highly prevalent and underestimated welfare issue in sows in modern pig production, worth further studies focusing on particular risk factors related to feeding, genetics, management issues, and sows longevity.

Overweight is common in cats and has health and welfare implications. This study aimed to assess potential predictive/protective factors for feline overweight associated with owner management and their relationship with cat behavior and welfare. A questionnaire was administered to 197 owners to collect information about cat demographics, management, environment, dietary habits and behavior. The feline Body Mass Index was recorded for each cat. Univariate logistic regression was used to evaluate the association of parameters with overweight cats. Variables with a p-value < 0.10 at univariable analyses were selected for the multivariable model. Most cats were mixed- breed, 1–7 years old and neutered; 51.3% were overweight. Age higher than 7 years, neutering, being alone all day and being stressed were predictive for overweight (p < 0.05). Conversely, the presence of other animals had a protective effect (p < 0.05). A general disagreement between owners’ perception and clinical evaluation of cats’ body condition was a common and significant risk factor for overweight (OR = 8.532, 95% CI = 4.073–17.875; p < 0.001). According to the owners, many veterinarians did not inform them about their cat being overweight nor about the risks (p < 0.001). This study provides helpful information on the influence of management and environment on cats’ body condition and its relationship with cat welfare.

Abstract Background: Although donkey contributed vital role in human livelihoods through direct and indirect contributions to financial, human and social capital, emphasis has not been given to study the welfare issue which include health problems and management requirement. A retrospective study and questionnaire survey was conducted to assess the health and the welfare problem of donkeys coming to the Haro Sabbu Veterinary clinic for treatment at the Dale Sadi district, Haro SabbuVeterinary clinic, Oromia, Ethiopia. Data on 448 donkeys from 2018 to 2020 were analyzed from the data record sheet kept by the clinic. Results: The most common case for wound occurrence were back sore 135(30.1%), chest wound 37(8.3%), bite 27(6.0%) and combined wound of both back and bite 14(3.1%). The finding indicated that the occurrence of wound in donkeys having poor 170 (64.4%), medium 41(33.6%) and good 7(11.6%) body condition and this was found to be statistically significantly associated (p=0.000). Among the common health problems and abnormalities encountered in donkeys back sore wound 135(30.1%), lameness 92(20.5%), parasitic infestation 60(13.6%), chest wound 37(8.3) and signs of colic 34 (7.6%) were the major problems encountered in donkeys coming to clinic for treatment with there was no a statistically significant difference among different years. In addition to body condition, the occurrence of wounds was found to be statistically significantly associated with age category and work type. In addition, pack donkeys experienced higher lameness occurrences as compared to cart donkeys and it was statistically significant (P>0.05). In addition to health related problems, the questionnaire survey conducted also revealed welfare problems such as management condition, overloading, working hour, housing problems, and wounds due to harnessing were the major welfare problems of donkeys in the study area.Conclusion: This retrospective study and questionnaire survey revealed the significant health and welfare problems of donkeys that need to be addressed in order to improve welfare status, their health and working efficiency. Therefore, there should be immediate awareness creation for community, government, community, local service providers and policy makers on welfare issue of donkey and health management in the study area in particular and in the country in general.

Flystrike is a major cost and a welfare issue for the New Zealand sheep industry. There are several factors that can predispose sheep to flystrike, such as having fleecerot, a urine-stained breech, and “dags” (an accumulation of fecal matter in the wool of the breech). The FABP4 gene (FABP4) has been associated with variation in ovine fleecerot resistance, with a strong genetic correlation existing between fleecerot and flystrike occurrence. In this study, blood samples were collected from sheep with and without flystrike for DNA typing. PCR-SSCP analyses were used to genotype two regions of ovine FABP4. Sheep with the A1 variant of FABP4 were found to be less likely (odds ratio 0.689, P = 0.014) to have flystrike than those without A1. The likelihood of flystrike occurrence decreased as copy number of A1 increased (odds ratio 0.695, P = 0.006). This suggests that FABP4 might be a candidate gene for flystrike resilience in sheep, although further research is required to verify this association.

Lameness is 3rd most prevalent disease after mastitis and infertility, in the dairy industry today and is an important welfare issue causing significant impact on dairy economics. Lameness leads to involuntary culling of lactating dairy cattle due to significant effect on production and reproduction.Mostly lameness occurs due to foot lesions irrespective of breed, management and of animal. Two hundred and four Sahiwal dairy cows were evaluated for lameness and associated claw lesionsin the study from local dairy farms of Punjab. Clinical lameness (lameness score 2 & 3) was evident in 6 per cent of animals. After lameness scoring, each animal was restrained in chute and hooves were examined before and after paring 1 mm of hoof tissue from weight bearing surface. Seventy three per cent animals had at least one lesion in their hooves. On animal level, under run soles (45%) were found to be most prevalent claw lesion followed by white line lesions (29%), overgrown soles (7%), cork screw hooves (5%), sole avulsions (5%), sole hemorrhages (3%), heel erosions (3%), double soles (2%) and wall cracks (1%). At claw level, prevalence of lesions was 35.3 per cent, among which under run soles were the most prevalent lesion (19.3%) followed by white line lesions (16.3%), overgrown soles (2.82%), cork screw hooves (1.84%), sole avulsions (1.47%), sole hemorrhages (1.04%), heel horn erosions (0.61%), double soles (0.49%), wall cracks (0.25%) and toe hemorrhages (0.12%). Lactation length and parity had no effect on lameness. Comparatively more lesions were seen in hind limb hooves (56.8%) as compared to forelimb hooves (43.2%). Fore lateral and hind medial claws had more lesions (58.8% and 76.47%, respectively) as compared to contralateral claws 52.9% and 70.5%, respectively). Overall, a low prevalence of clinical lameness was seen in Sahiwal dairy cows.

The elevation of ambient temperature beyond the thermoneutral zone leads to heat stress, which is a growing health and welfare issue for homeothermic animals aiming to maintain relatively constant reproducibility and survivability. Particularly, global warming over the past decades has resulted in more hot days with more intense, frequent, and long-lasting heat waves, resulting in a global surge in animals suffering from heat stress. Heat stress causes pathophysiological changes in animals, increasing stress sensitivity and immunosuppression, consequently leading to increased intestinal permeability (leaky gut) and related neuroinflammation. Probiotics, as well as prebiotics and synbiotics, have been used to prevent or reduce stress-induced negative effects on physiological and behavioral homeostasis in humans and various animals. The current data indicate dietary supplementation with a Bacillus subtilis-based probiotic has similar functions in poultry. This review highlights the recent findings on the effects of the probiotic Bacillus subtilis on skeletal health of broiler chickens exposed to heat stress. It provides insights to aid in the development of practical strategies for improving health and performance in poultry.
