Glioblastoma (GBM) is the most aggressive type of brain tumor. Microvascular proliferation and abnormal vasculature are the hallmarks of the GBM, aggravating disease progression and increasing patient morbidity. Here, we uncovered a key role of ETS1 on vascular abnormality in glioblastoma. ETS1 was upregulated in endothelial cells from human tumors compared to endothelial cells from paired control brain tissue. Knockdown of Ets1 in mouse brain endothelial cells inhibited cell migration and proliferation, and suppressed expression of genes associated with vascular abnormality in GBM. ETS1 upregulation in tumor ECs was dependent on TGFβ signaling, and targeting TGFβ signaling by inhibitor decreased tumor angiogenesis and vascular abnormality in CT-2A glioma model. Our results identified ETS1 as a key factor regulating tumor angiogenesis, and suggested that TGFβ inhibition may suppress the vascular abnormality driven by ETS1.

Abstract Background: Low-severity prescribed fire is a tool used for reducing fuel loads on public lands, particularly in dry conifer forests of the western United States characterized by historically mixed- and low-severity fire regimes. Understanding the ecological effects of prescribed fire treatments is important for predicting the impacts of these management actions on wildlife communities. But few studies have estimated small landbird responses to forest treatments at spatial scales relevant to their ecology or have examined potential differences in treatment effects applied within historically mixed- vs. low-severity fire regimes. Therefore, we evaluated prescribed fire treatment effects and relationships with burn severity for avian communities in dry conifer forests dominated by ponderosa pine (Pinus ponderosa) located on seven National Forests in the interior western United States. We surveyed birds for 1–4 years and 1–3 years before and after prescribed fire treatments at mixed- and low-severity fire regime locations, respectively, following a before-after, control-impact study design – 8 paired control-treatment units in mixed-severity locations (16 total study units with 320 survey points) and 4 paired control-treatment units in low-severity locations (8 total study units with 278 survey points). Using a Bayesian hierarchical multi-species occupancy model, we analyzed occupancy patterns for 95 species.Results: We found 33 species with statistically supported treatment effects and/or burn severity relationships primarily in mixed-severity locations. The data supported positive treatment effects at mixed severity locations for 9 species (American Robin [Turdus migratorius], Western Bluebird [Sialia mexicana], Hairy Woodpecker [Dryobates villosus], Black-backed Woodpecker [Picoides arcticus], American Three-toed Woodpecker [Picoides dorsalis], House Wren [Troglodytes aedon], Dusky Flycatcher [Empidonax oberholseri], Western Wood-peewee [Contopus sordidulus], Gray Flycatcher[Empidonax wrightii]), whose occupancy shifted towards more severely burned points after treatment, and a negative effect for one species (Ruby-crowned Kinglet [Corthylio calendula]), whose occupancy shifted away from burned points. At low severity locations, only two species exhibited treatment effects, both negative (Red-faced Warbler [Cardellina rubrifrons], and Lark Sparrow [Chondestes grammacus]). We also found supported occupancy relationships with burn severity post-treatment (i.e., regardless of species distribution before treatment) for 29 species, most of which were consistent with their life histories (e.g., patterns of positive relationships for cavity-nesting, bark insectivores and negative relationships for open-nesting, foliage insectivores). Stronger responses to prescribed fire treatments at mixed-severity locations were unexpected because prescribed fire applications are more similar to historical wildfires characterizing low-severity fire regimes.Conclusions: Bird populations in historically low-severity locations may be relatively unresponsive to prescribed fire because fire there is typically more frequent, expected, and regular. By comparison, fire events are relatively rare historically in mixed severity locations, potentially eliciting more responses to an infrequent opportunity, even by species that are strongly associated with recently burned forests by wildfire. Our results suggest that fire management activities intended to reduce fuels and lower the risk of high-severity wildfire can also be effective in creating habitat for some fire specialists at least in the short term.

Background: Psoriasis is a chronic skin disease in which lifelong treatment is required. Recently, we faced COVID-19 pandemic where adherence to a prescribed drug could have been affected. Objectives: We conduct a study to assess this statement. Methods: The study was performed in Dermatology Department with Pharmacy database in the Hospital Universitario de La Princesa. A retrospective observational database study was performed, including all psoriatic patients with a biological therapy prescription between March 13 and May 31, 2020, with a paired control group between March 13 and May 31, 2019. Medication possession ratio (MPR) was used to determine the adherence. Results: A total of 244 patients were included in the cohort and 228 in the control group. We observed a decrease in the percentage of adherent patients of 40.0% in COVID-19 period. Overall, MPR was lower in the 2020 period than in 2019. MPRs by treatment and drug family, with the exception of etanercept, certolizumab pegol, and guselkumab, were significantly lower between the 2019 and the 2020 period. Conclusions: Adherence to a prescribed drug can be affected by different factors such as age, sex, income quintile, or stress. We have demonstrated a deep impact on adherence because of the situation created by COVID-19 pandemic. These data are in contrast with previously reported results where adherence was slightly affected, probably due to the different epidemiological situation in different countries. Further efforts are needed to be done to mitigate COVID-19 pandemic influence on the therapeutic adherence.

The aim of this research was to investigate the effect of mobile learning on students’ academic achievements and their opinions about the course. This research was a mixed study where both quantitative and qualitative methods were used together. The quantitative part of the study used quasi-experimental models in a pre-test/post-test design according to the patterned paired control group. In the qualitative part, a semi-structured interview method was used from descriptive models. The study group consisted of 28 students studying in the 8th grade of a state secondary school in the Pasinler district of Erzurum province of Turkey in the 2018-2019 academic year. The study lasted approximately six weeks. As a result of the study, it was observed that there was a positive increase in academic achievement in favor of the experimental group of students. Moreover, although there were differences of opinion among the students, a large majority declared that the mobile supported learning increased their interest towards the course. Recommendations are made based on this research.

Abstract Background Circular RNA (circRNA) have been reported to play important roles in cardiovascular diseases including myocardial infarction and heart failure. However, the role of circRNA in atrial fibrillation (AF) has rarely been investigated. We recently found a circRNA hsa_circ_0099734 was significantly differentially expressed in the AF patients atrial tissues compared to paired control. We aim to investigate the functional role and molecular mechanisms of mmu_circ_0005019 which is the homologous circRNA in mice of hsa_circ_0099734 in AF. Methods In order to investigate the effect of mmu_circ_0005019 on the proliferation, migration, differentiation into myofibroblasts and expression of collagen of cardiac fibroblasts, and the effect of mmu_circ_0005019 on the apoptosis and expression of Ito, INA and SK3 of cardiomyocytes, gain- and loss-of-function of cell models were established in mice cardiac fibroblasts and HL-1 atrial myocytes. Dual-luciferase reporter assays and RIP were performed to verify the binding effects between mmu_circ_0005019 and its target microRNA (miRNA). Results In cardiac fibroblasts, mmu_circ_0005019 showed inhibitory effects on cell proliferation and migration. In cardiomyocytes, overexpression of mmu_circ_0005019 promoted Kcnd1, Scn5a and Kcnn3 expression. Knockdown of mmu_circ_0005019 inhibited the expression of Kcnd1, Kcnd3, Scn5a and Kcnn3. Mechanistically, mmu_circ_0005019 exerted biological functions by acting as a miR-499-5p sponge to regulate the expression of its target gene Kcnn3. Conclusions Our findings highlight mmu_circ_0005019 played a protective role in AF development and might serve as an attractive candidate target for AF treatment.

Cities around the world are transitioning to more efficient lighting schemes, especially retrofitting traditional, high-pressure sodium (HPS) streetlights with light-emitting diode (LED) lights. Although these initiatives aim to address the problems of urban sustainability and save money, the ecological impacts of these retrofits remain poorly understood, especially in brightly lit cities and in the tropics, where urbanisation is most rapid. We performed an experimental study of the retrofit in Singapore–focusing on insectivorous bats, whose activity we monitored acoustically along paired control (HPS-lit) and treatment (LED-lit) streets. We recorded seven species along these streets, but only obtained enough recordings to measure the effect of light type for three of them–all of which can reasonably be described as urban adapters. The strongest predictor of bat activity (an index of habitat use) was rainfall–it has a positive effect. Light type did not influence bat activity or species composition of the bat assemblage along these streets, though it did interact with the effects of rainfall and traffic noise for one bat species. Ultimately, the retrofit may be ecologically meaningless to urban-adapted, tropical insectivores that already experience high levels of light pollution as they do in Singapore. However, while our findings may appear reassuring to those concerned with such retrofits in other tropical and/or brightly-lit cities, they also highlight the contextual nature of ecological impacts. We point out that they should not be prematurely generalised to other locales and systems. In particular, they do not imply no impact on species that are less urban-adapted, and there is a clear need for further studies, for example, on responses of other foraging guilds and of bats (and insects) throughout the tropics.

ABSTRACTIntroductionLeprosy can cause different lesions in peripheral nerves and inervatory structures.ObjectivesTo analyse the effectiveness of evaluation protocols used to identify neural lesions in leprosy such as Degree of Physical Disability (DPD), Simplified Neurological Assessment (SNA), and Neurodynamic Assessment (NDA).MethodDescriptive analytical study, associative, with 27 individuals treated in two outpatient leprosy clinics in São Paulo State, between 2017 and 2019, and 27 individuals from the paired control group. The Mann-Whitney, Multivariate Linear Regression and association between variables and P<0.05 values were used.ResultsThe test that most captured the neurological alterations was the SNA, with 22 (81.5%) in the upper limbs (ULs) and 25 (92.6%) in the lower limbs (LLs), followed by the NDA, with 20 (74.1%) in the ULs and 11 (40.7%) in the LLs. The DPD showed handicap in the hands of 16 (59.2%) individuals and in the feet of 17 (62.9%) individuals, and they have expressed sensitivity. DPD showed agreement with SNA in 21 (77.8%) of the cases in ULs (p=0.010) and 19 (70,4%) of the cases in LLs (p=0.060). DPD and NDA showed that in 19 (70.4%) of the patients evaluated there was concordance of results in ULs (p=0.143); 9 (33.3%) in LLs (p=0.125). SNA and the NDA in the ULs found agreement in 21 (77.8%); 11 (40.7%) (p=0.786) in the LLs.ConclusionThe three assessment instruments are specific and will hardly produce false positive tests. DPD can produce more false negatives than SNA. If there is an instrument to be chosen, it should be the SNA, since it is more sensitive, more accurate and has a less negative likelihood ratio. Neurodynamic tests were positive in 7.4% of individuals while there were still no changes in the SNA; afterwards, these changes appeared.

Multiple symmetric lipomatosis (MSL) is a rare disorder characterized by overgrowing lipomatous tissue (LT) in the subcutaneous adipose tissue (SAT). What LT is and how it expands are not completely understood; previous data suggested that it could derive from brown AT precursors. In six MSL type I patients, we compared LT morphology by histological and immunohistochemistry (IHC) analysis, gene expression, by qPCR, kinase activity, by Western Blot and in vitro assay to paired-control SAT using AT from patients with pheochromocytoma as a human browning reference. In the stromal vascular fraction (SVF), we quantified adipose stem cells (ASCs) by flow cytometry, the proliferation rate, white and beige adipogenic potential and clonogenicity and adipogenicity by a limiting dilution assay. LT displayed white AT morphology and expression pattern and did not show increased levels of the brown-specific marker UCP1. In LT, we evidenced AKT, CK2 and ERK1/2 hyperactivation. LT-SVF contained increased ASCs, proliferated faster, sprouted clones and differentiated into adipocytes better than the control, displaying enhanced white adipogenic potential but not increased browning compared to SAT. In conclusion, LT is a white AT depot expanding by hyperplasia through increased stemness and enhanced white adipogenesis upregulating AKT, CK2 and ERK1/2, which could represent new targets to counteract MSL.

Category: Trauma; Ankle Introduction/Purpose: Fibular nailing (FN) is a method of fixation that has proven to be useful for the treatment of distal fibular fractures (DF). FN minimizes soft tissue complications, provides similar stability compared to plating with less hardware related symptoms. Nevertheless, FN has been associated with syndesmotic malreduction and incapacity of restoring length and rotation of the fibula. We aimed to evaluate the fibular and syndesmotic reduction after fixation with FN compared to the uninjured ankle. Methods: Prospective cohort, cross-sectional study with a paired control group. Patients with DF fractures treated with FN between January 2017 and July 2019 were included. Immediate postoperative bilateral Ankle CT was obtained in all cases. Two independent radiologists performed all the measurements on both ankles (fibular rotation, length, translation, and syndesmotic diastasis. Statistical analysis was made using the Wilcoxon Test and Intraclass correlation coefficient (ICC). Considering a medium size sample effect, 95% confidence with .05 alpha error, a total sample of 26 subjects were needed to achieve a .80 of statistical power (G-Power 3.1). All analyses were performed using SPSS V20. Results: Twenty-six patients were included (14 Women). The mean age was 47 years (18-91). No statistically significant differences were identified considering fibular rotation (p:0,694), fibular length (p:0,585) and syndesmotic diastasis (p:0,078) between the injured and uninjured ankle. Fibular translation has statistical differences (p:0,043). The ICC shows an excellent concordance between radiologists except for Fibular translation (ICC 0,47) Conclusion: In this cohort, fixation of DF fractures with FN allows restoration of anatomical parameters of the ankle in terms of fibular rotation, length, and syndesmotic diastasis. However, Fibular translation had significant differences compared with the uninjured ankle.
