The present study was conducted to investigate the comparative effects of organic and inorganic forms of sulfur, methyl sulfonyl methane (MSM) and sodium sulfate (SS), on laying performance, egg quality, ileal morphology, ileal volatile fatty acids, and antioxidant and stress markers in various biological samples in aged laying hens. A total of 144, 73-week-old Lohman Brown-Lite laying hens were randomly assigned to one of three experimental diets: basal diet (CONT), CONT + 0.2% MSM (MSM), and CONT + 0.3% SS (SS). The trial lasted for 12 weeks. MSM and SS diets contained 0.07% of sulfur, either organic or inorganic. Dietary MSM did not affect egg production or feed conversion ratio at 12 weeks compared with the CONT group. Dietary sulfur did not affect egg quality except for the Haugh unit at 4 weeks, which was lowered (p < 0.05) in the SS group. Compared with the CONT group, a higher (p < 0.05) villus height to crypt depth ratio was observed in the SS group. Dietary sulfur did not affect the percentages of short-chain fatty acids in the ileum. Total antioxidant capacity of the liver increased (p < 0.05) in laying hens fed MSM- and SS-added diets compared with the CONT group. The MSM and SS groups were found to have lowered (p < 0.05) malondialdehyde (MDA) concentration in serum samples compared with CONT. Finally, dietary MSM had the lowest (p < 0.05) MDA concentrations in yolk samples. Taken together, our study showed that dietary organic and inorganic sulfur have positive effects on ileal morphology and antioxidant capacity in laying hens. However, SS-mediated inhibition in laying performance needs to be clarified.

Abstract Background Kidney plays a central role in regulating salt-sensitivity of blood pressure (BP) to governs sodium excretion via several mechanisms including pressure natriuresis and the actions of renal sodium transporters. Purpose We clarified the effects of combination treatment of sodium-glucose cotransporter 2 (SGLT2) inhibitor and angiotensin receptor blocker (ARB) on BP and the pathogenesis of renocardiac injuries, and elucidated underlying molecular mechanisms involved in the regulation of renal sodium handling in the development of salt-sensitivity by comparing with each monotreatment in Dahl salt-sensitive (DSS) hypertensive rats. Methods DSS rats were treated orally for 8-weeks with normal salt diet (0.3% NaCl) (NS/Cont group), high salt diet (8% NaCl) (HS/Cont group), high salt diet with ipragliflozin (0.04%) (HS/Ipra group), high salt diet with losartan (0.05%) (HS/Los group), or high salt diet with combination of ipragliflozin and losartan (HS/Ipra+Los group). Results The combination group significantly reduced systolic BP compared with either high salt diet control group, losartan or ipragliflozin monotreatment groups (HS/Ipra+Los: 182.5±18.4mmHg vs HS/Cont: 227.7±26.1; HS/Ipra: 216.6±26.9; HS/Los: 208.6±21.6, at 8-weeks of treatment, P&lt;0.05, respectively) (Figure 1A). The slope of pressure-natriuresis curve was significantly increased in the HS/Ipra+Los group compared to that in the HS/Cont group (interaction P=0.024), HS/Ipra group (P=0.009), and HS/Los group (P=0.084) using the linear regression model (Figure 1B), which indicated that only the combination treatment of ipragliflozin and losartan improved salt-sensitivity. The combined treatment significantly improved creatinine clearance (HS/Ipra+Los: 3.3±0.9mL/min vs HS/Cont: 1.1±0.5; HS/Ipra: 1.7±0.6; HS/Los: 1.9±0.8, P&lt;0.05, respectively). The combination treatment also significantly ameliorated glomerulosclerosis, and improved cardiomyocyte hypertrophy and perivascular fibrosis (Figure 1C). Angiotensin II type 1 receptor (AT1R) protein expression level in the kidney was remarkably suppressed in the combination treatment group compared to the other high salt diet groups. The protein expression level of Na+/H+ exchanger isoform 3 (NHE3) and Na+-K+-Cl– cotransporter 2 (NKCC2), two of major sodium transports in the renal tubules, were significantly decreased with losartan monotreatment and combination treatment, but not with ipragliflozin monotreatment (Figure 2). Conclusions The dual inhibition of SGLT2 and AT1R effectively improved salt-sensitivity via reducing renal expression levels of the sodium transporters, which eventually lead to renocardiac protection. Thus, the combination treatment could be a novel and useful therapeutic strategy for treating salt-sensitive hypertension and renal injury in non-diabetic patients. AbstractEffects of combination therapy of dipeptidyl peptidase-4 (DPP-4) inhibitor and sodium-glucose co-transporter 2 (SGLT2) inhibitor on β-cells are still unclear, although combination agent of these two drugs has become common in clinical practice. Therefore, we aimed to elucidate the effects of DPP-4 inhibitor and/or SGLT2 inhibitor on β-cell mass and function and compared their effects between in an early and advanced phase of diabetes. We used 7-week-old db/db mice as an early phase and 16-week-old mice as an advanced phase and treated them for 2 weeks with oral administration of linagliptin, empagliflozin, linagliptin + empagliflozin (L + E group), and 0.5% carboxymethylcellulose (Cont group). Blood glucose levels in Empa and L + E group were significantly lower than Cont group after treatment. In addition, β-cell mass in L + E group was significantly larger than Cont group only in an early phase, accompanied by increased Ki67-positive β-cell ratio. In isolated islets, mRNA expression levels of insulin and its transcription factors were all significantly higher only in L + E group in an early phase. Furthermore, mRNA expression levels related to β-cell differentiation and proliferation were significantly increased only in L + E group in an early phase. In conclusion, combination of DPP-4 inhibitor and SGLT2 inhibitor exerts more beneficial effects on β-cell mass and function, especially in an early phase of diabetes rather than an advanced phase.

Background: Beta-carotene (BETA) is a precursor of retinol (Vitamin A) and positively influenced on reproductive efficiency in cows. The aim of the study was to determine the effect of orally supplementation of beta-carotene on plasma carotene content and fertility rate of lactating crossbred Sahiwal cows. Methods: Twenty-four disease-free lactating crossbred Sahiwal cows with a close date of calving were randomly divided into two homogeneous groups for Control group (CONT; n = 12) and Beta-carotene group: 500 mg/cow/d (BETA; n = 12) from 7 days post partum (dpp) until 105 dpp thereafter on concentrations of beta-carotene and selected protein and energy parameters in plasma were determined. In addition, effects on fertility rate were studied. Result: Beta-carotene concentrations increased in plasma of beta-carotene (BETA-group) supplemented cows compared to control (CONT-group) cows (p less than 0.001). In BETA-group cows, urea concentration in blood plasma decreased significantly compared to cows of CONT-group (p less than 0.001) but did not differ in total protein, albumin, glucose and total cholesterol content in cows of both groups. Occurrence of first post partum estrus decreased (p less than 0.001) in beta-carotene (BETA-group: 74 dpp) cows compared to control (CONT-group: 81 dpp) cows. In this study, it was observed that the overall conception rate was higher (p greater than 0.001) in BETA-group: 83.33% (cows pregnant: 10/12) and lesser in the CONT-group: 50.00% (cows pregnant: 6/12). The results of this study indicated that oral supplementation with beta-carotene increased the concentration of beta-carotene and decreased the concentration of urea in plasma and cows with higher blood plasma beta-carotene content improved fertility over cows with lower blood plasma beta-carotene content.

The essential oil of Croton zehntneri (EOCZ) and its major compounds are known to have several biological activities. However, some evidence shows potential toxic effects of high doses of EOCZ (>300 mg/kg) in amphibian and human kidneys. The aim of the present work was to investigate the effects on renal function of EOCZ at 300 mg/kg/day in healthy Swiss mice and a subclinical acute kidney injury (subAKI) animal model, which presents tubule-interstitial injury (TII). Four experimental groups were generated: (1) CONT group (control); (2) EOCZ, mice treated with EOCZ; (3) subAKI; (4) subAKI+EOCZ, subAKI treated simultaneously with EOCZ. EOCZ treatment induced TII measured by increases in (1) proteinuria; (2) cortical tubule-interstitial space; (3) macrophage infiltration; (4) collagen deposition. A decrease in tubular sodium reabsorption was also observed. These results were similar and nonadditive to those observed in the subAKI group. These data suggest that treatment with EOCZ at higher concentrations induces TII in mice, which could be mediated by protein overload in the proximal tubule.

Abstract Background Although diabetes is one of the fastest increasing diseases in prevalence worldwide and demands significant medical resources, more than half of all patients with diabetes do not achieve the expected target level of blood glucose. As a potential cause of poor glycemic control, insufficient adherence to medication has long been discussed and variably studied. However, dropout from treatment as another plausible cause has not been fully examined. The aim of this study was to clarify profiles of patients with diabetes who discontinued pharmacotherapy (Discont group) by extracting reasons of their decisions and by comparing with those who continued (Cont group) in terms of the related factors to glycemic control. Methods A cross-sectional, internet-based survey was conducted among Japanese with diabetes registered in a database. A self-administered questionnaire consisting of the 8-item version of the Morisky Medication Adherence Scale (MMAS-8), glycosylated haemoglobin (HbA1c) level, and demographic and disease characteristics was completed by all participants. Reasons for medication discontinuation and resumption were also received retrospectively from participants in the Discont group. To examine the risk of uncontrolled HbA1c, logistic regression analysis was conducted in each group. Results In the Discont group (148 cases), older age at resumption of pharmacotherapy and current smoking habit increased the probability of uncontrolled HbA1c, whereas in the Cont group (146 cases), a familial history of diabetes and better medication adherence in MMAS-8 scores decreased the probability of uncontrolled HbA1c. A relationship between medication adherence and HbA1c level was seen in the Cont but not in the Discont group. About 70 % of those in the Discont group made their decision to terminate diabetes treatment without consulting physicians and half of them perceived their situations inappropriately. Conclusions Those who discontinued pharmacotherapy were less adherent to medication than those who did not discontinue. Risk factors for glycemic control also differed between those who discontinued and those who did not. More than one-third of participants with diabetes who discontinued pharmacotherapy had inappropriate perceptions of their disease, which medical professionals should be aware of for better interventions.

Abstract The cortical masticatory area (CMA) regulates masticatory movements and controls rhythmic jaw movements. However, information regarding the effect of respiratory disturbances on the functional development of the CMA remains limited. This study aimed to examine the effect of chronic unilateral nasal obstruction (UNO) on the motor representation of the anterior digastric (AD) muscle during the growth period. Forty-eight 8-day-old male Wistar albino rats were randomized into control (CONT, n=24) and UNO (n=24) groups. Both groups underwent intracortical microstimulation (ICMS) at ages 5, 7, and 9 weeks. Arterial oxygen saturation (SpO2) and the number of positive ICMS sites for the AD muscle were measured throughout the experiment. The SpO2 values were significantly lower in the UNO group than in the CONT group at each age. Further, the number of positive ICMS sites for the AD muscle was significantly higher in the UNO group than in the CONT group at each age. Intragroup comparisons revealed that the number of positive ICMS sites increased with age. The onset latencies of the evoked AD potential significantly differed between the right and left sides. However, no differences were found between the CONT and UNO groups at any age. These findings suggest that UNO during development induces changes in the motor representation in the CMA.

Background Maternal obesity may disrupt the developmental process of the fetus during gestation in rats. Recent evidence suggests that taurine can exert protective role against detrimental influence of obesogenic diets. This study aimed to examine the effect of maternal cafeteria diet and/or taurine supplementation on maternal dietary intake, plasma metabolites, fetal growth and development. Methods Female Wistar rats were fed a control diet (CON), CON supplemented with 1.5% taurine in drinking water (CONT), cafeteria diet (CAF) or CAF supplemented with taurine (CAFT) from weaning. After 8 weeks all animals were mated and maintained on the same diets during pregnancy and lactation. Results Dietary intakes were significantly different between the groups. Both CAF and CAFT fed dams consumed less water in comparison to CON and CONT dams. Taurine supplementation only increased plasma taurine concentrations in CONT group. Maternal plasma adiponectin concentrations increased in CAF and CAFT fed dams compared to CON and CONT fed dams and there was no effect of taurine. Hyperleptinemia was observed in CAF fed dams but not in CAFT fed dams. Malondialdehyde was significantly increased only in CAF fed dams. Litter size, sex ratio and birth weight were similar between the groups. There was an increase in neonatal mortality in CONT group. Discussion This study showed that maternal taurine supplementation exerted modest protective effects on cafeteria diet induced maternal obesity. The increased neonatal mortality in CONT neonates indicates possible detrimental effects of taurine supplementation in the setting of normal pregnancy. Therefore, future studies should investigate the optimal dose of taurine supplementation and long term potential effects on the offspring.

Abstract Background: The effect of nighttime aromatherapy on sleep has been explored. However, its effect on mood states has not been established. This study aimed to determine the effects of nighttime aromatherapy, using skin patches with lavender aroma, on mood states of young women. Methods: Twenty-four young women slept as usual with a skin patch placed under their clothing for seven consecutive nights. One group of subjects (AROM group) used skin patches with lavender aroma, whereas the other group (CONT group) used skin patches without the aroma. The primary outcome measure was mood states assessed with the Profile of Mood States 2nd Edition (POMS 2). The secondary outcome measures were physiological indices of stress assessed by salivary α-amylase activity, cortisol concentration, and pulse rate measured immediately after waking up. Assessments were performed pre-intervention, the morning after the first intervention night, and post-intervention.Results: Skin patches with lavender aroma improved total mood disturbance and two negative subscales scores of the POMS 2 in the AROM group over time. There was an interaction between time and treatment condition (with/without aromatherapy) for the fatigue-inertia subscale, although no interaction was detected between time and condition for total mood disturbance and the anger-hostility subscale. The positive subscales scores of POMS 2 did not change in both groups. In the AROM group, salivary α-amylase level measured the morning after the first intervention night was significantly higher than the pre-intervention level; however, there was no significant difference between pre- and post-intervention. Moreover, there were no significant changes in salivary cortisol concentration and pulse rate in both groups. Conclusions: Unlike the AROM group, no improvement in mood states was observed in the CONT group. Low-cost and easily applicable aromatherapy using skin patches are potentially useful in stabilizing mood states in various populations.Trial registration: UMIN-CTR, UMIN000035051.
