The current study is focused on the biosynthesis of nutmeg oil/ polyurethane/ZnONPs bionanocomposite film for immunomodulatory and antioxidant activities. The fabricated film was prepared by using naturally extracted nutmeg oil functionalized with ZnONPs in the presence of polyutherane (PU) medium. The bionanocomposite film was obtained by incorporating dropwise 10 % (w/v) of nutmeg oil to the PU solution/ZnONPs blend. The active constituents of nutmeg oil were determined by gas chromatography coupled with mass spectrometry (GC-MS). The morphological characteristics of the resulting bionanocomposite film were confirmed using various microscopic and spectroscopic methods. Immunomodulatory potential of bionanocomposite was evaluated for RAW 264.7 macrophages. The results exhibited an excellent reduction in inflammatory cytokines (IL-6, IL-10, and TNFα) secretions after the treatment with bionanocomposite. The bionanocomposite exerted the highest inhibitory effects on certain cell signaling constituents that influence the initiation of expression of proinflammatory cytokines. The bionanocomposite was also tested for DPPH and ABTS free radicals scavenging assays and showed excellent antioxidant potential with IC50 values (0.28 ± 0.22 and 0.49 ± 0.36), respectively. The outcomes suggested promising immunomodulatory and antioxidant potentials for the biogenic synthesized nutmeg oil/PU/ZnONPs polymeric bionanocomposite.

Free radicals are reactive compounds, their excessive production is considered to be an important cause of oxidative damage in biomolecules causing degenerative diseases. Polyphenols are one of the most important groups of secondary metabolites of plants, which have an antioxidant activity depending on their properties as hydrogen donors. Echinops polyceras Boiss. (Asteraceae) is one of Echinops genus species that spread in Syria, Lebanon, and Palestine. Phytochemicals found in this species leaves have been extracted with gradient polarity solvents, and primary screening of the secondary metabolites was established. The phenolic compounds and flavonoids contents were determined. The free radicals scavenging activity was evaluated for all extracts with DPPH• in a 96-well microplate. The specificity study indicates that ascorbic acid was absent, and reducing sugars were exist in the aqueous extract. The identification tests showed the presence of polyphenols like flavonoids and coumarins. The methanolic extract of the E. polyceras leaves was the most effective scavengers of free radicals (90.22% in 30 min) with phenolic compounds content 682.5 mg GAE/g of dried extract (DE) and flavonoids content 194.5 mg QE/ g DE. The chloroform extract was the least effective as free radical scavenging (60% in 30 min) as the phenolic compounds content was 278.5 mg GAE/g DE and flavonoids content 94 mg QE/ g DE.  In conclusion, the phenolic compounds and flavonoids from Echinops polyceras Boiss. are effective in free radicals scavenging and protecting from diseases caused by oxidative stress.

Cerium oxide nanoparticles (CeO2 NPs) and zinc oxide nanoparticles (ZnO NPs) are emerging pollutants that are likely to occur in the contemporary environment. So far, their combined effects on terrestrial plants have not been thoroughly investigated. Obviously, this subject is a challenge for modern ecotoxicology. In this study, Pisum sativum L. plants were exposed to either CeO2 NPs or ZnO NPs alone, or mixtures of these nano-oxides (at two concentrations: 100 and 200 mg/L). The plants were cultivated in hydroponic system for twelve days. The combined effect of NPs was proved by 1D ANOVA augmented by Tukey’s post hoc test at p = 0.95. It affected all major plant growth and photosynthesis parameters. Additionally, HR-CS AAS and ICP-OES were used to determine concentrations of Cu, Mn, Fe, Mg, Ca, K, Zn, and Ce in roots and shoots. Treatment of the pea plants with the NPs, either alone or in combination affected the homeostasis of these metals in the plants. CeO2 NPs stimulated the photosynthesis rate, while ZnO NPs prompted stomatal and biochemical limitations. In the mixed ZnO and CeO2 treatments, the latter effects were decreased by CeO2 NPs. These results indicate that free radicals scavenging properties of CeO2 NPs mitigate the toxicity symptoms induced in the plants by ZnO NPs.

Abstract Background Bauhinia strychnifolia Craib is an herb in Thai traditional medicine. Its decoction is traditionally used as an anticancer, antidiarrheal, and hangover remedy for centuries. Several studies described bioactivities of its organic solvent extracts, however, only few demonstrated the usefulness of the decoction. Here, we aimed to determine the bioactivities of Bauhinia strychnifolia Craib root and stem aqueous extracts in gut and liver perspective. Methods To achieve the goal, we performed MTT test, microscopic analyses, disc diffusion assay, broth microdilution assay, free radicals scavenging assays, and LC-MS analysis. Results We found that the extracts inhibited the growth of human hepatocellular carcinoma (HepG2) and colon adenocarcinoma (HT-29) cell lines. Moreover, they also inhibited the growth of gram-positive bacteria Staphylococcus aureus and Bacillus cereus but not inhibited the growth of gram-negative bacteria Escherichia coli and Pseudomonas aeruginosa. Furthermore, the extracts exhibited moderate antioxidant activity and increased GSH production in HepG2 cell line when compared with untreated. Our LC-MS analysis confirmed the existence of anticancer and antioxidant; 3,5,7,3′,5′-pentahydroxyflavanonol-3-O-α-L-rhamnopyranoside and β-sitosterol, in the extracts. Conclusion The results from our study supported that the administration of Bauhinia strychnifolia Craib root and stem decoction would really aid colon or liver cancer patients and detoxify the alcoholic drunkard as it is claimed in Thai traditional medicine.

Background and Purpose: Hepatorenal toxicity is a side effect of the anthracycline cytotoxic antibiotics, doxorubicin that is used in cancer treatment. The study investigated the ameliorative potential of Clerodendrum volubile ethanol leaf extract (CVE) on doxorubicin (DOX)-induced hepatorenal toxicities. Methods: Male Wistar rats were pretreated with Clerodendrum volubile ethanol leaf extract (50 - 400 mg/kg/day, p.o) followed by intraperitoneal injection of 2.5 mg/kg of DOX on alternate days for 14 days. Hepatorenal toxicity was assessed using renal function parameters (serum electrolytes, blood urea and creatinine), hepatic function endpoints [aspartate aminotransferase (AST), alanine aminotransferase (ALT) and alkaline phosphatase (ALP), total protein (TP), albumin (ALB) and total bilirubin (TB)]. In addition, the antioxidant activity in the kidney and liver tissues were assayed and histological studies of these tissues were also conducted. Results: Oral pretreatment with 50 mg/kg/day, 100 mg/kg/day, 200 mg/kg/day and 400 mg/kg/day of CVE remarkably ameliorated DOX-induced liver and kidney injury by lowering the serum ALT, AST, ALP, Cr and BUN levels. CVE pretreatment remarkably ameliorated DOX-induced increases in the CAT, SOD and GPx activities and MDA levels compared to the DOX-treated rats. The biochemical changes were corroborated by improvements in the DOX-induced histological lesions seen in the hepatic and renal tissues examined. Conclusions: Overall, these findings suggest that Clerodendrum volubile ethanol leaf extract elicits protective effect against DOX-induced hepatorenal toxicities mediated primarily via oxidative stress suppression and improvement in the free radicals scavenging activities of CVE.

Phenolic compounds and thiamine may serve as therapies against oxidative stress-related neurodegenerative diseases. However, it is important to note that these components show high instability under changing conditions. The study’s aim was to determine the impact of the thiamine concentration (hydrochloride—TH and pyrophosphate—TP; in the range 0.02 to 20 mg/100 g on the indices of the chelating properties and reducing power, and free radicals scavenging indices of EGCG, EGC, ECG and caffeine added from 0.04 to 6.0 mg/100 g. Our research confirmed that higher concentrations of TH and TP can exhibit significant activity against the test antioxidant indices of all components. When above 5.0 mg/100 g of thiamine was used, the radical scavenging abilities of the compound decreased in the following order: EGCG > ECG > EGC > caffeine. The highest correlation was found for the concentration of thiamine pyrophosphate to 20.0 mg/100 g and EGCG. Knowledge of the impact of factors associated with the concentration of both EGCG, EGC, ECG or caffeine and thiamine on their activity could carry weight in regulating the quality supplemented foods, especially of nutrition support for people of all ages were oral, enteral tube feeding and parenteral nutrition).

AbstractNatural extracts and compounds from marine resources have gained intensive scientific and industry attention for radioprotective activities in the past ten years. However, the marine-derived radioprotectants have been studied against UV-rays, gamma (γ)-rays and X-rays for more than 30 years. This review aims to identify key marine-derived extracts/compounds and their modes of action studied for radioprotective activities from 1986 to 2019. A comprehensive survey was conducted to establish the trend in terms of the publications each year and the countries of origin. A total of 40 extracts and 34 natural compounds showing radioprotective activities against UV-rays, gamma (γ)-rays and X-rays were identified from a range of marine plants and animals. These extracts and compounds are broadly categorized into polysaccharides, phlorotannins, carotenoids and mycosporine-like amino acids (MAAs). Macroalgae and microalgae were found to be the dominant sources of polysaccharides, phlorotannins and carotenoids. MAAs were mainly identified in algae, sponges, sea cucumber and corals that showed significant UV-absorbing activities. A number of radioprotective mechanisms were shown by these compounds, predominantly free radicals scavenging, inhibition of apoptosis, UV-ray absorption and DNA damage-repair signaling pathways. While these bio-discoveries warrant further investigation and development of radioprotective therapeutics, however, the lack of clinical studies is a major obstacle to be tackled in the future.
