Цель исследования - изучение динамики морфологических изменений роговицы и заднего отдела глаза крыс при длительной гипергликемии. Методика. Исследование выполнено на 36 самцах крыс Wistar. Сахарный диабет индуцировали внутрибрюшинной инъекцией стрептозотоцина (65 мг/кг), после чего ежедневно вводили подкожно малые дозы инсулина (2 ЕД/кг). На 50-е, 58-е и 66-е сут эксперимента производили энуклеацию глаз у глубоко наркотизированных животных. Гистологические срезы фрагментов глаз окрашивали гематоксилин-эозином, проводили морфометрию параметров роговицы и сетчатки. Результаты. Средняя концентрация глюкозы и кетоновых тел крови в группе сахарного диабета составила 29,8 ммоль/л и 0,889 ммоль/л, в контрольной группе - 6,2 ммоль/л и 0,847 ммоль/л соответственно. Анализ гистологических срезов глаз выявил признаки отека роговицы, хориоидеи и наружных слоев центральных отделов сетчатки до появления других качественных и количественных морфологических изменений. Заключение. Оценка толщины роговицы, хориоидеи и наружных слоев центральных отделов сетчатки может служить предиктором развития диабетической ретинопатии. The aim was to study morphological changes in the cornea and the posterior part of rat eye during prolonged hyperglycemia. Methods. The study was performed on 36 Wistar male rats. Diabetes mellitus was induced by an injection of streptozotocin (65 mg/kg, i.p.) followed by daily injections of low doses of insulin (2 U/kg, s.c.). Eyes were enucleated from deeply anesthetized rats on days 50, 58, and 66 of the experiment. Histologic sections were stained with hematoxylin-eosin, and morphometry of the cornea and the retina was performed. Results. Mean blood concentrations of glucose and ketone bodies were 29.8 mmol/L and 0.889 mmol/L, respectively, in the diabetic group and 6.2 mmol/L and 0.847 mmol/L, respectively, in the control group. The histological analysis revealed signs of edema in the cornea, choroid and outer layers of the central retina, which preceded other morphological changes. Conclusion. Evaluating thickness of the cornea, choroid and outer layers of the central retina may serve for prediction of diabetic retinopathy.

There is a significant prevalence of dental diseases in patients with diabetes. Temporomandibular joint (TMJ) disorders occupy one of the first places among them. However, there is no data on the diabetes effect on the disc of the TMJ. The aim of the study – to establish structural changes of TMJ disc in rats with hyperglycemia. Materials and Methods. Sixty white mature male rats were divided into 4 equal groups. Animals of first and second groups were simulated with streptozotocin-induced diabetes. Rats of third and fourth groups were used as controls at appropriate period to the experimental. Slaughtering of animals was carried out after 30 and 60 days from the start of the experiment, after which the TMJ complexes and adjacent soft tissues were collected. Biological material was prepared according to conventional methods. Histological sections were examined with the help of microscopes. Results and Discussion. The structure of the disc of TMJ of control rats did not change. Its’ significant remodeling was visualized in animals with diabetes. Thickening of the walls of arteries and widening of venous lumen and swelling of the collagen fibers were observed after 30 days of the experiment. Thickening of the fibers of collagen and predominance of histiocytes were visualized after 2 months of the diabetes. Lumen of arteries were narrowed and plethora was observed in the veins of the hemomicrocirculatory bed. Conclusions. Restructuring of the disk of TMJ of rats with diabetes are manifested by mucoid swelling of collagen fibers and morphological changes in the hemomicrocirculatory vascular bed that can lead to dystrophy of the disc.

In this work, we studied the pharmacological activity of zinc and magnesium salts of 2-aminoethanesulfonic acid in white non-linear male rats with amyotrophic lateral sclerosis, which was modeled by neurotoxicantsimplication into the pelvic part of spinal cord. After the reproduction of the pathology in animals, the indices of motor activity were recorded in the Rotarod test, and morphological studies of spinal cord sections stained according to Nisl in the Belshovsky modification were carried out. It was shown that the magnesium salt of 2-aminoethanesulfonic acid (compound LHT-317) to a greater extent reduces the development of motor disorders in experimental animals compared with the control group on the 4th day of observation. The course of intravenous administration of the studied compounds of 2-aminoethanesulfonic acid did not inhibit morphological changes in the spinal cord that develop in degenerative-dystrophic pathology of the central nervous system: connections. Moreover, if, against the background of treatment with zinc salt, the total area of motor zones in animals of the experimental group exceeded that of control rats, then the number of motoneurons did not differ from the control.

Abstract Although the antidiabetic effects of leptin require intact neuronal melanocortin signaling in rodents with uncontrolled diabetes (uDM), increased melanocortin signaling is not sufficient to mimic leptin's glucose-lowering effects. The current studies were undertaken to clarify the role of melanocortin signaling in leptin's ability to correct metabolic and neuroendocrine disturbances associated with uDM. To accomplish this, bilateral cannulae were implanted in the lateral ventricle of rats with streptozotocin-induced diabetes, and leptin was coinfused with varying doses of the melanocortin 3/4 receptor (MC3/4R) antagonist, SHU9119. An additional cohort of streptozotocin-induced diabetes rats received intracerebroventricular administration of either the MC3/4R agonist, melanotan-II, or its vehicle. Consistent with previous findings, leptin's glucose-lowering effects were blocked by intracerebroventricular SHU9119. In contrast, leptin-mediated suppression of hyperglucagonemia involves both melanocortin dependent and independent mechanisms, and the degree of glucagon inhibition was associated with reduced plasma ketone body levels. Increased central nervous system melanocortin signaling alone fails to mimic leptin's ability to correct any of the metabolic or neuroendocrine disturbances associated with uDM. Moreover, the inability of increased melanocortin signaling to lower diabetic hyperglycemia does not appear to be secondary to release of the endogenous MC3/4R inverse agonist, Agouti-related peptide (AgRP), because AgRP knockout mice did not show increased susceptibility to the antidiabetic effects of increased MC3/4R signaling. Overall, these data suggest that 1) AgRP is not a major driver of diabetic hyperglycemia, 2) mechanisms independent of melanocortin signaling contribute to leptin's antidiabetic effects, and 3) melanocortin receptor blockade dissociates leptin's glucose-lowering effect from its action on other features of uDM, including reversal of hyperglucagonemia and ketosis, suggesting that brain control of ketosis, but not blood glucose levels, is glucagon dependent.

Objective: to investigate the effect of administration of supraphysiological âcaroteno on biological parameters (ectoscopy and blood pressure), laboratory (malondialdehyde) and histological (liver and carotid arteries) of spontaneously hypertensive rats prone to stroke (SHR-sp).Methods: we used 36 male rats were divided into three groups, each containing 12 rats Wistar, SHR and SHR-sp, subdivided into six control animals and six animals treated with supraphysiological doses of âcaroteno for two periods of ten weeks interspersed with one week interruption. In the experiment were assessed daily physical examination and blood pressure (plethysmography). At sacrifice, blood was collected for measurement of serum malondialdehyde, liver and carotid arteries for histological examination.Results: temporary change in color of the fur, decreased significantly (p<0.0001) blood pressure (20mg supplementation âcaroteno) and serum levels of malondialdehyde (p<0.05) and increased amount of elastic fibers in the carotid wall of SHR and SHR-sp.Conclusion: supplementation of supraphysiological âcaroteno caused no toxic effects, showed positive response in the modulation of blood pressure and lower serum malondialdehyde. No significant morphological changes were found in both groups, except an increase in the number of elastic fibers in the muscle layer carotid suggesting elastosis in SHR and SHR-sp.

ОСОБЛИВОСТІ МАСОМЕТРИЧНИХ ПОКАЗНИКІВ ТА МОРФОЛОГІЧНИХ ЗМІН ГОЛОВНОГО МОЗКУ СТАТЕВОЗРІЛИХ ЩУРІВ В УМОВАХ ВПЛИВУ НА ОРГАНІЗМ СУЛЬФАТІВ МІДІ, ЦИНКУ ТА ЗАЛІЗА - З метою вивчення масометричних показників щурів та їх головного мозку за умов довготривалої дії (упродовж 90 діб) на організм сульфатів міді, цинку та заліза було проведено експеримент на 48 білих статевозрілих щурах-самцях масою 200-250 г віком 5-7 місяців. Застосовували анатомічні, статистичні та загальноприйняті методики мікроанатомічного методу дослідження. Встановлено, що комбінований вплив на організм сульфатів міді цинку та заліза чинить на головний мозок досить виражений токсичний ефект, що негативно позначається на масометричних показниках загальної маси щурів та маси головного мозку. Це свідчить про розвиток у головному мозку явищ гострого набряку з ознаками геморагічної інфільтрації. Применялись анатомические, статистические и общепринятые методики микроанатомического метода исследования. Установлено, что комбинированное воздействие на организм сульфатов меди и цинка и железа оказывает на мозг достаточно выразительный токсический эффект, что отрицательно сказывается на массометрических показателях общего веса крыс и веса головного мозга. Это свидетельствует о развитии в головном мозге явлений острого отека с признаками геморрагической инфильтрации, степень выраженности которого максимальна в конце эксперимента.<br />FEATURES OF MASS INDICES AND MORPHOLOGICAL CHANGES IN ADULT RAT BRAIN UNDER THE INFLUENCE ON THE BODY OF COPPER SULFATE, ZINC AND IRON - To study the performance of rats and their mass brain in long acting (for 90 days) on the body of copper sulfate, zinc and iron, an experiment was conducted on 48 white adult male rats weighing 200-250 gram, aged 5-7 months. There was applied anatomy, statistics and conventional techniques microanatomical research method. It was established that the combined effect on the body of copper and zinc sulphates and iron in the brain has enough expressive toxicity, which affects performance on the total weight of the rats and brain weight. This testifies to the development of brain edema, acute phenomena with signs of hemorrhagic infiltration. This study aims to determine the content of flavonoid and the effect of ethanol extract of moringa seeds on the regeneration of pancreatic β cells in male white rats streptozotocin induced diabetes. This study method used has total flavonoid equivalent quercetin by spectrophotometry uv-vis and to regeneration of pancreatic β cells in male white rats used 30 test animals,namely male white rats divided into 6 groups, each group consisted of 5 male white rats with details of group I as normal control, Group II as negative control given 0.5% Na-CMC suspension, Group III as positive control given glibenclamide suspension and in Groups IV, V, and VI were given with each dose of 100 mg/kg BW, 200 mg/kg BW and 400 mg/kg BB. Histopathological damage picture of the pancreas was observed by staining HE using a 400x magnification olympus Cx21 microscope. The results showed that the ethanol extract of moringa seeds contained secondary metabolites, namely flavonoids, alkaloids, saponins and tannins. The results showed has total flavonoid equivalent quercetin of moringa seeds is 1,26% and regeneration of pancreatic β cells in male white rats streptozotocin induced diabetes of Moringa seed ethanol extract at a dose of 400 mg/kg BB can have an effect on the regeneration of β cells in the pancreas of white diabetic male rats.

The sulfation of bile acids is hormone dependent, being increased in females and ethynylestradiol (EE)-treated males compared with normal males. Diabetes causes significant alterations in estrogen metabolism and uterine estrogen receptor kinetics. Male rats were given streptozotocin (90 mg/kg) and diabetes was verified. An increase in hepatic bile acid sulfotransferase (BAST) activity was significant by 6 days and continued to increase to 29 days. This increase was prevented by insulin replacement. Administration of EE (6.0-600 micrograms X kg-1 X day-1) to normal male rats resulted in a significant increase in hepatic BAST activity; however, administration of similar doses of EE to diabetic males failed to further increase activity levels over the already-elevated levels in the diabetic controls. This increase in in vitro specific activity was accompanied by an increase in the biliary excretion of lithocholate 3-sulfate and taurolithocholate 3-sulfate in 21-day-diabetic animals. Bile flow and total bile acid excretion were also markedly increased in the diabetic animals. The data indicate that streptozotocin-induced diabetes causes a significant increase in hepatic BAST activity. These findings are consistent with an alteration in hepatic estrogen action in streptozotocin-induced diabetes.

This study investigated the reproductive toxicity of methylmercury (MeHg) and Aroclor (Sigma-Aldrich), alone or in combination, following exposure of prepubertal male rats considering the chromatoid body (CB) as a potential target. The CB is an important molecular regulator of mammalian spermatogenesis, primarily during spermatid cytodifferentiation. Male Wistar rats were exposed to MeHg and/or Aroclor , according the following experimental design: control group, which was administered in corn oil (vehicle) only; MeHg-treated group, which was administered 0.5mg kg−1 day−1 MeHg; Aroclor-treated group, which was administered 1mg kg−1 day−1 Aroclor; Mix-LD, group which was administered a low-dose mixture of MeHg (0.05mg kg−1 day−1) and Aroclor (0.1mg kg−1 day−1); and Mix-HD group, which was administered a high-dose mixture of MeHg (0.5mg kg−1 day−1) and Aroclor (1.0mg kg−1 day−1). MeHg was diluted in distilled water and Aroclor was made up in corn oil (volume 1mL kg−1). Rats were administered the different treatments from PND23 to PND53 by gavage, . The morphophysiology of CBs was analysed, together with aspects of steroid hormones status and regulation, just after the last treatment on PND53. In addition, the long-term effects on sperm parameters were assessed in adult animals. MeHg exposure increased mouse VASA homologue (MVH) protein levels in seminiferous tubules, possibly affecting the epigenetic status of germ cells. Aroclor produced morphological changes to CB assembly, which may explain the observed morphological defects to the sperm flagellum and the consequent decrease in sperm motility. There were no clear additive or synergistic effects between MeHg and Aroclor when administered in combination. In conclusion, this study demonstrates that MeHg and Aroclor have independent deleterious effects on the developing testis, causing molecular and morphological changes in CBs. To the best of our knowledge, this is the first study to show that CBs are targets for toxic agents.
