The aromatic heartwood of Santalum yasi has been harvested extensively in Fiji and Tonga over the past two centuries for international trade in the medicinal, perfume, and incense industry and other cultural purposes. Field surveys and a review of historic and modern documents reveal a sparse and scattered distribution and indicate that the natural distribution of S. yasi has fluctuated over time, even declining to local extinction in the wild in some areas, while S. album has been introduced and naturalisation of S. yasi × S. album hybrids is evident. Population data collected along transects in three in situ S. yasi populations show discontinuous size-class structures, indicating regenerative stress. The population densities at study sites ranged from 19 to 63 adult trees (≥5 cm dbh) per hectare and less than two heartwood-bearing trees (≥15 cm dbh) per hectare. Though S. yasi trees may attain up to 40 cm dbh, no trees greater than 23 cm dbh were found in any of the studied populations. Low density and small size of adult trees and human-induced bole damage are suggestive of frequent, premature, and defensive harvesting patterns and indicate the need for ongoing adaptive comanagement in recognition of underlying economic and sociocultural pressures.

To regenerate sandalwood (Santalum spicatum) stands in south-western Australia it is necessary to understand the complex relationship between woylies (Bettongia penicillata ogilbyi) and sandalwood. Sandalwood requires a seed disperser for successful recruitment and in the past the critically endangered woylie played an important role in dispersing and caching seeds, but it is not clear whether this mutualistic and antagonistic relationship is beneficial to regeneration efforts. An enclosure in a woodland and 46Scandium-labelled seeds, enabled study of the in situ predation of seeds, caching, the fate of cached seeds, the detection of cached seeds and predation of germinated seeds. Woylies preferentially cached sandalwood, then S. acuminatum seeds, before any interest was shown in Acacia acuminata and Gastrolobium microcarpum seeds, which were virtually all eaten in situ. Of a further 500 radiolabelled and individually numbered sandalwood seeds deployed, 42.2% were eaten in situ, 20.8% had an unknown fate and 37% were cached, with some seeds being recached up to four times. After nine months, only four cached seeds remained undisturbed. Olfaction appeared to be the primary method of cache detection. To examine the recruitment rate of cached seeds, the fate of 89 transplanted sandalwood seedlings at two study sites was followed. After one month 38% were intact and growing, but half of the transplanted seedlings were dug up and the remaining endosperm was eaten in situ or taken away. The results highlight the potential of providing seed supplies, including sandalwood seeds and seeds of their hosts, to seed-dispersal marsupials for passive ecosystem repair.

Preserving wild coffee in the natural forest, especially in southwest Ethiopia, is important for maintaining the genetic diversity of Arabica coffee and sustaining coffee production. To monitor the changes in wild coffee in the natural forest, we conducted an in situ baseline survey in 2015, and 30 of those survey sites were re-visited in 2019. Those surveys involved counting the number of mature mother trees, saplings and seedlings, as well as recording details of the sites, including disturbance, accessibility and forest conditions. Satellite imagery was combined with the site-specific in situ survey data to provide evidence of the forest condition around the study sites and therefore help more fully explore the causes for the changes in the wild coffee stock. The results show that, overall, the population of mother coffee trees was maintained during the 4-year period, and a slight increase in saplings occurred. Closer examination reveals considerable variations between sites, with some equally accessible sites showing a sharp decline in the number of mother trees while others show consistent increases. This study demonstrates the importance of systematic surveys, especially for the areas where forest cover and wild coffee plants are highly variable, and this may help explore community-specific approaches in managing wild coffee in the forest.

The distribution of the threatened fern Ophioglossum vulgatum L., a plant with extremely small populations (PSESPs) in Sardinia, is characterized by small disjunct populations with only a few individuals, and little is known about its status in the wild. To provide information for the conservation of O. vulgatum and with the aim to develop an in situ conservation strategy, we investigated its distribution, population size, and habitat. Field surveys confirmed that the species grows in only five localities. Two representative populations were selected for this study (Funtanamela and Gedili), and in each population, all plants were mapped and monitored monthly from April to August over an 8-year period. During the study, the populations had a very low number of reproductive plants and the populations appeared to be in decline, with the total number of plants per population slightly decreased in Gedili while a sharp reduction was recorded in Funtanamela due to wild boar threat. A fence was built in order to protect the site from further damage, but no noticeable signals of recovery were observed. The most urgent conservation requirement for this species is to preserve the threatened habitat of the remnant populations. Further field surveys and research are also required for an improved understanding of the species’ status.

This contribution presents a recent review of equipment and field techniques applicable to offshore seabed survey work and site investigation, with particular reference to Canadian interests.During the past decade many advances have taken place in equipment design and in under standing the behaviour of mechanical sampling devices and the inadequacies of data derived from samples physically removed from the site. There has been a significant improvement in the ability of both remote and contact sensory systems to characterize and resolve differences in the physical and mechanical properties of bottom sediment materials, and the use of even simplistic remotely sensed data, if rapidly obtained and processed, offers significant economic advantages. The trend towards greater use of in situ tests will continue.The application of facies concepts, in planning field surveys, may provide opportunities for improved sample recovery, reduced sampling, and greater confidence in data interpretation.Data, comments, and examples refer to a number of Canadian situations, for example, deep ocean studies, shelf studies (such as on the Labrador Shelf, the Scotian Shelf, and the Beaufort Sea), inter-island studies in the Canadian Arctic, and inland studies in the Great Lakes and smaller lake and river systems.

Studies of the nature of a surface, either metallic or nonmetallic, in the past, have been limited to the instrumentation available for these measurements. In the past, optical microscopy, replica transmission electron microscopy, electron or X-ray diffraction and optical or X-ray spectroscopy have provided the means of surface characterization. Actually, some of these techniques are not purely surface; the depth of penetration may be a few thousands of an inch. Within the last five years, instrumentation has been made available which now makes it practical for use to study the outer few 100A of layers and characterize it completely from a chemical, physical, and crystallographic standpoint. The scanning electron microscope (SEM) provides a means of viewing the surface of a material in situ to magnifications as high as 250,000X.

Inorganic particulate material of diverse types is present in the ambient and occupational environment, and exposure to such materials is a well recognized cause of some lung disease. To investigate the interaction of inhaled inorganic particulates with the lung it is necessary to obtain quantitative information on the particulate burden of lung tissue in a wide variety of situations. The vast majority of diagnostic and experimental tissue samples (biopsies and autopsies) are fixed with formaldehyde solutions, dehydrated with organic solvents and embedded in paraffin wax. Over the past 16 years, I have attempted to obtain maximal analytical use of such tissue with minimal preparative steps. Unique diagnostic and research data result from both qualitative and quantitative analyses of sections. Most of the data has been related to inhaled inorganic particulates in lungs, but the basic methods are applicable to any tissues. The preparations are primarily designed for SEM use, but they are stable for storage and transport to other laboratories and several other instruments (e.g., for SIMS techniques).

The properties of metal/semiconductor interfaces have received considerable attention over the past few years, and the Al/GaAs system is of special interest because of its potential use in high-speed logic integrated optics, and microwave applications. For such materials a detailed knowledge of the geometric and electronic structure of the interface is fundamental to an understanding of the electrical properties of the contact. It is well known that the properties of Schottky contacts are established within a few atomic layers of the deposited metal. Therefore surface contamination can play a significant role. A method for fabricating contamination-free interfaces is absolutely necessary for reproducible properties, and molecularbeam epitaxy (MBE) offers such advantages for in-situ metal deposition under UHV conditions

Way Kambas National Park (WKNP) is home of five protected big mammals including sumatran elephants.  It shares its border with 22 of 37 villages surrounding the national park.  Understanding their existence in the wild is a priority, and  wildlife genetics is a crucially needed. Besides poaching and habitat fragmentation, wildlife-human conflict is one big issue.  Elephant Training Center (ETC) in WKNP is built for semi in-situ conservation effort on captive sumatran elephants that mainly have conflict histories with local people.  Participative observation and bio-molecular analysis were conducted to learn the importance of captive Sumatran elephant for conservation effort.  Through captive sumatran elephants, database and applicable methods are expected to be developed supporting the conservation of their population in the wild.  Participative observation and molecular identification was carried on captive sumatran elephants in ETC, WKNP under multiple year Terapan grant of Ministry of Research and Technology Higher Education, Indonesia. Gene sequence and cytological analyses showed that the captive sumatran elephants are closely related and tend to be domesticated.  Translocation among ETC to avoid inbreeding, and maintaining the captive sumatran elephant as natural as possible are highly recommended. Developing genetic database can be a reference for both captive and wild sumatran elephants.

Over the past decades, a number of bone tissue engineering (BTE) approaches have been developed to address substantial challenges in the management of critical size bone defects. Although the majority of BTE strategies developed in the laboratory have been limited due to lack of clinical relevance in translation, primary prerequisites for the construction of vascularized functional bone grafts have gained confidence owing to the accumulated knowledge of the osteogenic, osteoinductive, and osteoconductive properties of mesenchymal stem cells and bone-relevant biomaterials that reflect bone-healing mechanisms. In this review, we summarize the current knowledge of bone-healing mechanisms focusing on the details that should be embodied in the development of vascularized BTE, and discuss promising strategies based on 3D-bioprinting technologies that efficiently coalesce the abovementioned main features in bone-healing systems, which comprehensively interact during the bone regeneration processes.

The aim of this study is to develop a relocatable modelling system able to describe the microbial contamination that affects the quality of coastal bathing waters. Pollution events are mainly triggered by urban sewer outflows during massive rainy events, with relevant negative consequences on the marine environment and tourism and related activities of coastal towns. A finite element hydrodynamic model was applied to five study areas in the Adriatic Sea, which differ for urban, oceanographic and morphological conditions. With the help of transport-diffusion and microbial decay modules, the distribution of Escherichia coli was investigated during significant events. The numerical investigation was supported by detailed in situ observational datasets. The model results were evaluated against water level, sea temperature, salinity and E. coli concentrations acquired in situ, demonstrating the capacity of the modelling suite in simulating the circulation in the coastal areas of the Adriatic Sea, as well as several main transport and diffusion dynamics, such as riverine and polluted waters dispersion. Moreover, the results of the simulations were used to perform a comparative analysis among the different study sites, demonstrating that dilution and mixing, mostly induced by the tidal action, had a stronger effect on bacteria reduction with respect to microbial decay. Stratification and estuarine dynamics also play an important role in governing microbial concentration. The modelling suite can be used as a beach management tool for improving protection of public health, as required by the EU Bathing Water Directive.
