1. The nitrogen requirement of the weanling kitten was determined in a series of three experiments. In each experiment, diets were formulated to provide the growing kitten with the essential amino acids at or above the level of requirement. Expt 1 utilized a 4 x 4 balanced Latin square design with two groups of kittens (four male and four female). The crystalline L-amino acid diets were presented at four levels of dietary crude protein (N x 6.25) of 140, 160, 180 and 200 g/kg diet. The design for Expts 2 and 3 was a 6 x 6 balanced Latin square. For each of these experiments, groups of six male and six female kittens were assigned to diets. The six levels of dietary crude protein were 120, 140, 160, 180, 200 and 220 g/kg diet; dietary N was supplied by crystalline L-amino acids for Expt 2 and casein plus a supplementary amino acid mix for Expt 3. Food intake, weight gain and N retention were determined in each experiment.2. A sigmoidal model y =P1+ P2/[1 +e(p2+p4. x)] was fitted to the response of weight gain and N retention to dietary N. The calculated requirement (95% of the upper asymptote, P1 + P2) for these experiments varied from 170 to 230 g protein/kg diet with the majority of these values falling between 180 and 200 g protein/kg.3. On the basis of these three experiments, the kitten's requirement for dietary crude protein is between 180 and 200 g/kg diet (28.8–32.0 gN/kg) for purified diets which provide a calculated 21 MJ metabolizable energ//kg diet.

1. The catabolism of L-[ l-14C]phenylalanine was used to indicate the effects of single amino acid supplementation of an inadequate protein diet (200 g crude protein (nitrogen x 6.25)/kg) on the utilization of dietary amino acids in pigs of 2.5 kg body-weight reared on an adequate protein diet (240 g crude protein/kg) containing skim milk and a mixture of free amino acids.2. The oxidation of phenylalanine was decreased by the addition of proline or arginine to the inadequate protein diet but not by the addition of threonine, methionine, lysine or a mixture of essential amino acids, indicating that proline and arginine were limiting the utilization of dietary amino acids in the inadequate protein diet.3. Dietary proline concentrations of 13.9 and 14.2 g/kg minimized phenylalanine oxidation in diets containing 200 or 260 g protein/kg. This indicates a dietary proline requirement of 14 g/kg.4. Increasing the dietary arginine concentration in a diet containing 240 g protein/kg showed that an arginine concentration of 5.1 g/kg minimized phenylalanine oxidation. However, increasing the arginine concentration in a diet containing 200 g protein/kg increased phenylalanine oxidation, suggesting an amino acid imbalance involving arginine at this lower level of protein.

The purpose of this experiment was to evaluate the effects of low protein corn-soybean meal-based diets on fecal CP, amino acid (AA) flow amount, AA digestibility and fecal and ileal microbial AA composition in growing pigs. Eighteen pigs (initial body weight = 30 ± 1.35) were randomly divided into three groups and fed with basal diets with CP levels of 12%, 15% and 18%, respectively. The Lys, Met + Cys, Thr and Trp level in the 12% CP and 15% CP groups is the same as 18% CP group by the addition of four crystalline Lys, Met + Cys, Thr and Trp to the diet. The results showed that with the decrease of dietary CP level from 18% to 12%, the fecal total nitrogen (N), CP and total AA (TAA) flow amount decreased linearly (p < 0.05). Dry matter (DM) digestibility, CP digestibility, TAA digestibility, essential amino acid (EAA) digestibility and non-essential amino acid (NEAA) digestibility increased linearly with the decrease of dietary CP concentration from 18% to 12%. Compared with 18% CP group, the flow amount of Asp, Ser, Glu, Gly, Tyr, Val, Leu and Phe in feces of pigs in the 15% CP group and 12% CP group decreased significantly, while the flow amount of Arg in the 15% CP group was lower than that in the 18% CP group and 12% CP group. The fecal microbial N and AA of the 15% CP group were higher than those of the 18% CP and 12% CP groups. Fecal TAA flow amount decreased linearly with the decrease of the dietary CP levels from 18% to 12%. Fecal TAA and NEAA flow amount also decreased linearly with the decrease of dietary CP level from 18% to 12%. Except for Glu, Gly, Met, Tyr, Thr and Phe, there were significant differences among the three groups in the composition of 17 kinds of AAs in fecal microorganisms. Among the 17 AA compositions of ileal microorganisms, except Tyr and Lys, the other AAs were significantly different among the three groups (p < 0.05)

The objective of this study was to investigate the impacts of dietary crude protein (CP), fishmeal and sorghum on nutrient utilisation, digestibility coefficients and disappearance rates of starch and protein, amino acid concentrations in systemic plasma and their relevance to growth performance of broiler chickens using the Box-Behnken response surface design. The design consisted of three factors at three levels including dietary CP (190, 210, 230 g/kg), fishmeal (0, 50, 100 g/kg), and sorghum (0, 150, 300 g/kg). A total of 390 male, off-sex Ross 308 chicks were offered experimental diets from 14 to 35 days post-hatch. Growth performance, nutrient utilisation, starch and protein digestibilities and plasma free amino acids were determined. Dietary CP had a negative linear impact on weight gain where the transition from 230 to 190 g/kg CP increased weight gain by 9.43% (1835 versus 2008 g/bird, P = 0.006). Moreover, dietary CP linearly depressed feed intake (r = -0.486. P < 0.001). Fishmeal inclusions had negative linear impacts on weight gain (r = -0.751, P < 0.001) and feed intake (r = -0.495, P < 0.001). There was an interaction between dietary CP and fishmeal for FCR. However, growth performance was not influenced by dietary inclusions of sorghum. Total plasma amino acid concentrations were negatively related to weight gain (r = -0.519, P < 0.0001). The dietary transition from 0 to 100 g/kg fishmeal increased total amino acid concentrations in systemic plasma by 35% (771 versus 1037 μg/mL, P < 0.001). It may be deduced that optimal weight gain (2157 g/bird), optimal feed intake (3330 g/bird) and minimal FCR (1.544) were found in birds offered 190 g/kg CP diets without fishmeal inclusion, irrespective of sorghum inclusions. Both fishmeal and sorghum inclusions did not alter protein and starch digestion rate in broiler chickens; however, moderate reductions in dietary CP could advantage broiler growth performance.

1. Piglets were weaned at 3 d of age and reared to 2.5 kg on a liquid diet in which the protein was supplied by dried skim milk and a mixture of free amino acids. The oxidation of L-[l-14C]phenyIalanine was measured as an indication of the partition of amino acids between retention and catabolism in pigs (2.5 kg) offered meals containing vaned concentrations of crude protein (nitrogen x 6.25).2. The dietary protein concentration was varied either by increasing the inclusion of a mixture of free amino acids in a series of diets containing 100 g protein/kg from skim milk, or by increasing the level of inclusion of the skim milk in a series of diets containing the equivalent of 100 g protein/kg from the free amino acid mixture.3. The oxidation of phenylalanine was minimized by dietary protein concentrations of 240 and 258 g/kg for the diets containing increasing concentrations of free amino acids or skim milk respectively.4. These results show that a mixture of free amino acids is used more effectively than intact protein for promoting retention of essential amino acids.5. The recovery of radioactivity in expired carbon dioxide was inversely related to the recovery of radioactivity in liver tissue when the concentration of dietary crude protein was increased from deficient to adequate, demonstrating that the fractional oxidation of the indicator amino acid was inversely related to protein synthesis.

<p>This study assessed the effects of dietary crude protein on performance and nitrogen economy of broilers. Chicks were allotted to 22, 20, 18 and 16% crude protein corn-soy diets in a completely randomized design for forty-two days. Several essential amino acids were observed to be deficient compared to recommendations for broiler chicks as the dietary crude protein level reduced. Feed intake, weight gain, feed to gain ratio, faecal nitrogen, nitrogen retention, anthropogenic potential and dressing percentage were influenced (P &lt; 0.05) by the dietary treatments. However, mortality was not significantly different (P &gt; 0.05). Feed intake, weight gain, nitrogen intake and output were significantly (P &lt; 0.05) depressed as the CP level reduced. However, the best (P &lt; 0.05) nitrogen retention and percentage faecal nitrogen was recorded for broilers fed 20% crude protein diet. Serum total protein, albumin, uric acid, creatinine and glucose were affected (P &lt; 0.05) across treatments. It was concluded that there is a limit to which dietary crude protein of broilers could be reduced without any detrimental effects on the performance and nitrogen economy of the birds, even when the requirements for methionine and lysine has been met, as several other amino acids could be limiting. Consequently, to achieve significant improvement in nitrogen economy and reduction in amount of faecal nitrogen, 20% crude protein diets could be fed to broilers. However, there may be need to further manipulate the amino acid profile of the diet so as to improve its performance to be at par with higher crude protein diets.</p>

AbstractThese studies were conducted to determine whether dietary homoarginine, derived from guanidination of synthetic lysine, affects the measurement of apparent and true ileal amino acid digestibility in the pig and to determine the optimal ratio of o-methylisourea (OMIU) to free synthetic lysine in the guanidination reaction that produces homoarginine. In two preliminary experiments, specific amounts of lysine were weighed and added to a solution of 0·6 mol/l OMIU to produce OMIU to free synthetic lysine ratios of 2:1, 1·5:1, 1·4:1, 1·3:1, 1·2:1, 1·1:1 and 1·0:1. The mixture was incubated at a pH of 10·5 and a temperature of 20±1°C for 6 days. The highest yield of homoarginine (proportionately 0·995) was produced with an OMIU to free synthetic lysine ratio of 1·5:1. The serum concentrations of homoarginine and lysine were measured in two test pigs at various times after consuming a single meal containing either homoarginine or lysine. Blood samples were taken before and 0·5, 1·5, 2·5, 4·5, 6·0, 7·5, and 11·5 h after feeding. Thirty minutes after feeding the homoarginine-containing diet, the homoarginine concentration in serum was 0·013 mmol/dl, and increased to a maximum of 0·031 mmol/dl 1·5 h after feeding. Thirty minutes after feeding the lysine-containing diet, the serum lysine concentration was 0·011 mmol/dl and also reached its maximum concentration of 0·025 mmol/dl 1·5 h after feeding. The timing of the changes in concentration of homoarginine and lysine related to feeding were similar, which suggests that homoarginine is absorbed and metabolized in a similar manner to lysine. Six Large White×Landrace barrows, weighing 68·5±3·5 kg and fitted with a simple T-cannula at the distal ileum as well as a jugular vein catheter, were used to determine the effects of homoarginine on endogenous amino acid flows and apparent and true ileal amino acid digestibility. The experimental design was a doubly replicated 3×3 Latin-square design with the three treatments consisting of a lysine-free, zein-based diet and jugular infusion of a lysine solution; a lysine-free, zein-based diet supplemented with homoarginine; and a lysine-free, zein-based diet supplemented with l-lysine. There were no significant differences in endogenous ileal lysine flow for pigs receiving the different treatments (396·6, 407·6, and 360·4 mg/kg DMI, respectively, P=0·71). In addition, with the exception of phenylalanine (P=0·04), there were no significant differences (P>0·10) in the apparent and true ileal digestibility of crude protein and amino acids among the three treatments. Therefore, it would appear that the presence of dietary homoarginine, derived from guanidination of synthetic lysine, does not affect endogenous amino acid losses or the digestibility of crude protein as well as most amino acids. These results are therefore supportive of the use of the homoarginine method as a means of measuring true ileal lysine digestibility.

AbstractThe prime purpose of this review is to explore the pathways whereby progress towards reduced-crude protein (CP) diets and sustainable chicken-meat production may be best achieved. Reduced-CP broiler diets have the potential to attenuate environmental pollution from nitrogen and ammonia emissions; moreover, they have the capacity to diminish the global chicken-meat industry’s dependence on soybean meal to tangible extents. The variable impacts of reduced-CP broiler diets on apparent amino acid digestibility coefficients are addressed. The more accurate identification of amino acid requirements for broiler chickens offered reduced-CP diets is essential as this would diminish amino acid imbalances and the deamination of surplus amino acids. Deamination of amino acids increases the synthesis and excretion of uric acid for which there is a requirement for glycine, this emphasises the value of so-called “non-essential” amino acids. Starch digestive dynamics and their possible impact of glucose on pancreatic secretions of insulin are discussed, although the functions of insulin in avian species require clarification. Maize is probably a superior feed grain to wheat as the basis of reduced-CP diets; if so, the identification of the underlying reasons for this difference should be instructive. Moderating increases in starch concentrations and condensing dietary starch:protein ratios in reduced-CP diets may prove to be advantageous as expanding ratios appear to be aligned to inferior broiler performance. Threonine is specifically examined because elevated free threonine plasma concentrations in birds offered reduced-CP diets may be indicative of compromised performance. If progress in these directions can be realised, then the prospects of reduced-CP diets contributing to sustainable chicken-meat production are promising.

Abstract The objective was to determine the influence of amino acid (AA) supplementation during the adaptation period on the ileal digestibility of crude protein and AA in corn and soybean meal (SBM). Six barrows with an initial body weight of 30.9 ± 2.6 kg fitted with a T-cannula in the distal ileum were assigned to a 6 × 6 Latin square design with 6 dietary treatments and 6 periods. Two experimental diets contained corn or SBM as the sole source of AA and an N-free diet was additionally prepared. For AA supplementation groups, an AA mixture consisted of Gly, Lys, Met, Thr, Trp, Ile, Val, His, and Phe was added to the corn diet and the N-free diet at the expense of cornstarch, and an AA mixture of Lys, Met, and Thr was added to the SBM diet. All diets contained 0.5% of chromic oxide. The 6 experimental diets were fed to the pigs for 4 and half days, and the 3 diets containing AA mixture were switched to the respective diets without AA mixture during the following 2 and half days. Ileal digesta were collected during the last 2 days. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Arg and Trp in corn (P &lt; 0.05), but did not affect that in SBM. The addition of AA mixture during the adaptation period caused increased apparent ileal digestibility of Pro and Gly regardless of feed ingredient (P &lt; 0.05), but did not affect that of other AA. All AA except Pro in corn and SBM were unaffected by the addition of AA mixture during the adaptation period. In conclusion, the addition of amino acid during the adaptation period does not affect the standardized ileal digestibility of indispensable amino acids in feed ingredients.

Four dairy steers were allotted to four lysine infusion levels in a 4 × 4 latin square design and fed an 11.5% crude protein (90% dry matter (DM)) diet continuously (10-min intervals every 24 hr). Lysine hydrochloride equivalent to 0.0, 3.0, 6.0, and 9.0 g lysine per day was infused per abomasum. When 9 g lysine were infused per day, the percent of absorbed nitrogen (N) retained was significantly (P < 0.05) reduced; urinary N excretion as a percentage of N intake and plasma-free lysine were increased significantly compared with the other three infusion treatments. The infusion of 9 g lysine per day apparently exceeded the body tissue requirements for this amino acid and the excess N was excreted in the urine. A possibility of lysine being limiting (0.28% lysine of a 100% DM diet) was apparently offset by the synthesis of lysine by rumen microorganisms, which increased the dietary lysine two- to threefold. Increased levels of infused lysine did not result in a linear increase of lysine in the abomasum. With 3 g per day lysine infusion rumen ammonia and N retentions were high. However, a smaller amount of N reached the abomasum with steers on this treatment.

Six barrows, average initial body weight 32.5 kg, fitted with a simple T-can-nula at the distal ileum, were used to investigate factors responsible for the variability of apparent ileal amino acid (AA) digestibility values among six pea samples. The barrows were fed six corn-starch-based diets, formulated to contain 16.5% crude protein (CP) (as-fed) from six different pea samples, according to a 6 × 6 Latin square design. Chromic oxide (0.3%) was used as the digestibility marker. The pigs were fed twice daily, at 0800 and 2000 h. The dietary allowance was 1600 g−1. Each experimental period lasted 7 d. Ileal digesta were collected at 2-h intervals for a total of 24 h from 0800 h on day 6 to 0800 h on day 8. With the exception of arginine, cysteine, histidine, and methionine, there were differences (P < 0.05) in the apparent ileal digestibility values of the indispensable including semi-essential (+semi-) AA among the pea samples. Of the indispensable AA (+semi-) within each pea sample, the digestibility values of arginine and lysine were relatively high, ranging from 88.3 to 91.3% and from 78.7 to 85.2%, respectively, while the digestibility values of cysteine, methionine, and tryptophan were relatively low, ranging from 53.8 to 62.7%, from 69.4 to 75.4%, and from 53.1 to 70.4%, respectively. With the exception of arginine, cysteine, and tryptophan, the ileal digestibility values of the indispensable AA (+semi-) were negatively correlated (P < 0.05) with the neu-tral-detergent fiber content in the pea samples. Of all the AA, only the digestibility of tryptophan was negatively correlated (P < 0.05) with the trypsin inhibitor activity in the pea samples. The significant finding of this study was that with the exception of arginine, cysteine, and proline, differences in the digestibility values of all other AA in the pea samples were positively correlated (P < 0.05) with their respective dietary AA contents. This suggests that differences in AA levels in the assay diets were responsible for the variability of apparent ileal digestibility values of AA between pea samples. Therefore, to eliminate this methodological effect, true rather than apparent ileal AA digestibility values for peas should be determined. Key words: Amino acids, ileal digestibility, peas, pigs
