Background and Aim: In the male reproductive system, the aging process can lead to infertility. Recently, placenta and its derivatives have been researched as regenerative agents. This study aimed to describe the basic components of dried bovine placenta powder and its potential effects as a regenerative agent in a rat model of male reproductive aging with D-galactose induction. Materials and Methods: We divided 15 male Wistar rats, 2 months of age, into three groups: A, the health control group; B, the D-galactose induction group, and C, the D-galactose induction and 10% dried bovine placenta supplementation group. We measured epididymal sperm concentration and testicular weight and volume and analyzed these using one-way analysis of variance. Results: Dried bovine placenta was rich in nutrients, with 61.98% protein, 21.25±2.07 carbohydrates, 8.58% water, 4.93% ash, and 3.27% fat. The mean epididymal spermatozoa concentration of the rats in Groups A, B, and C was 3026×106/mL, 1492.8×106/mL, and 2732.5×106/mL, respectively. The average total testicle weights were 2.44 g, 2.72 g, and 2.57 g, respectively. The average total testicle volumes were 2.29 cm3, 2.49 cm3, and 2.33cm3, respectively. Conclusion: Dried bovine placenta powder is rich in nutrients, especially protein. Supplementation with dried bovine placenta can improve epididymal spermatozoa concentration that is important in fertility.

AbstractCadmium (Cd) and diazinon (DZN) are known to be environmental risk factors for various bone diseases including osteoporosis. Selenium (Se), an essential constituent of many antioxidant enzymes, has in higher concentrations negative effects on the bone. The present study was aimed to investigate possible changes in femoral bone of adult male rats after their acute and subchronic exposures to Cd, DZN and Se. A total of 30 male Wistar rats were randomized into three experimental groups. The rats in the group A (4-months-old) were injected intraperitoneally with a mixture of 2 mg CdCl2 kg−1, 20 mg DZN kg−1 and 2 mg Na2SeO3 kg−1 body weight and killed 36 h after xenobiotics had been injected. In the group B, young males (1-month-old) were administered with a combination of 30 mg CdCl2 L−1, 40 mg DZN L−1 and 5 mg Na2SeO3 L−1 in their drinking water, for 90 days. Ten 4-months-old males without toxicant supplementation served as a control group (C). After treatment period, detailed histological analysis of femoral bone was performed in each group. Our results revealed apparent osteoporotic symptoms (resorption lacunae, osteoporotic fractures) in rats from groups A and B. Moreover, histomorphometrical evaluation showed reduced bone vascularization (constricted primary osteons’ vascular canals and Haversian canals) and weakness mechanical properties of bones (smaller size of the secondary osteons) in these rats in comparison with those of the control group. Our study demonstrates for the first time that acute and subchronic co-administrations to Cd, DZN and Se induce evident manifestation characteristics of osteoporosis in male rats.

Background: Obesity is a prevailing metabolic disorder that affects the functioning of the male reproductive system. Excessive adipose tissue enhances reactive oxygen species generation and is linked with male infertility. Spinach has demonstrated antioxidant effects. The present study was conducted to determine the antioxidant effects of spinach on sperm parameters in obese Sprague Dawley rats. Subjects and methods: This randomized control study was conducted at the animal house of the National Institute of Health Islamabad, Islamic International Medical College, Cosmesurge International Hospital, Rawalpindi, and Apollo lab, Islamabad, Pakistan from April 2016 to March 2017. Forty male Sprague Dawley rats having an age of 8 weeks and weight 160-200g were tagged from number 1 to 40. Every third rat was randomly allocated to control Group A (n=13) and remaining into the Experimental group (n=27). Rats of control Group A was given a standard diet while a high-fat diet was given to Experimental group rats to induce obesity for the duration of six weeks. Weight (g) was measured weekly and obesity was confirmed when rats attain more than 20% weight when compared with that of rats of control Group A. Then, after obesity induction, the experimental group was alienated into the obesity control group (Group B) and spinach treated group (Group C). For sample, rats of Group A and Group B were sacrificed, and the cauda epididymis of each rat was placed in a Petri dish containing normal saline and cut into pieces to allow the release of sperm and then sperm parameters (sperms concentration, motility, and morphology) were recorded under the microscope. Then, spinach (5% hot water extract) along with the persistence of fat diet was administered to Group C for 4 weeks and finally, sperm parameters were measured in this group. Results: Sperm concentration/ml, motility (%), and normal morphology (%) of Group B rats were significantly decreased as compared to Group A rats. However, sperm concentration/ml, motility (%), and normal morphology (%) of Group C (spinach treated group) rats was significantly increased (p<0.001) as compared to Group B (obesity control group) rats after administering spinach. Conclusion: The addition of Spinach in a normal diet regimen restores normal sperm morphology, improves sperm motility and concentration.

Oxidative stress is an imbalance between free radicals and antioxidants, and is an important etiological factor in the development of hypertension. Recent experimental evidence suggests that subpressor doses of angiotensin II elevate oxidative stress and blood pressure. We aimed to investigate the oxidative stress related mechanism by which a subpressor dose of angiotensin II induces hypertension in a normotensive rat model. Normotensive male Wistar rats were infused with a subpressor dose of angiotensin II for 28 days. The control group was sham operated and infused with saline only. Plasma angiotensin II and H2O2 levels, whole-blood glutathione peroxidase, and AT-1a, Cu/Zn SOD, and p22phox mRNA expression in the aorta was assessed. Systolic and diastolic blood pressures were elevated in the experimental group. There was no change in angiotensin II levels, but a significant increase in AT-1a mRNA expression was found in the experimental group. mRNA expression of p22phox was increased significantly and Cu/Zn SOD decreased significantly in the experimental group. There was no significant change to the H2O2 and GPx levels. Angiotensin II manipulates the free radical-antioxidant balance in the vasculature by selectively increasing O2− production and decreasing SOD activity and causes an oxidative stress induced elevation in blood pressure in the Wistar rat.

Introduction: Cigarette smoke causes oxidative stress which results in reduced sperm concentration, motility and morphology, also increased levels of 8-OHdG as a marker of DNA damage. Vitamin C and E have potential role in repairing spermatozoa damages. The aim of this study was to determine the effect of vitamin C and E combination on sperm quality and cement 8-OHdG level of smoke exposed rats.Methods: This study used a post test only control group design among 18 male Wistar rats subject, aged 8 week, 150-200 grams body weight (BW). The subject was randomly divided into 3 groups, K1: control, K2: cigarettes smoke exposed, K3: cigarettes smoke exposed and given a combination of 0.045 mg/gBW vitamin C and 0.036 IU/gBW vitamin E per oral. Analysis was done on day 21 using one-way ANOVA and post-hoc LSD for sperm concentration, motility and morphology; using Kruskal-Wallis and Mann-Whitney tests for cement 8- OHdG levels.Results: The lowest sperm concentration was found in   K2 (K2  32.59  million/mL,  K1 47.91 million/mL, K 339.43 million/mL); the lowest normal sperm motility was found in K2 (K 238.97%, K 164.57%, K3 51.43%); the lowest normal sperm morphology was found in K2 (K2 27.56%, K 138.36%, K 331.18%); and the highest cement 8- OHdG level was found in K2 (K2 20.18ng/mL, K1 3.43ng/mL, K3 5.28ng/mL).Conclusion: Combination of vitamin C and E can improve sperm concentration, motility and morphology and decrease cement 8-OHdG levels of smoke exposed rats.

Background: Consumption of a healthy diet abundant in antioxidant and anti-inflammatory phytochemicals, offers an effective and least expensive way to prevent neurodegeneration. Herein, the role of Tetracarpidium conophorum (African walnut) enriched diet in chronic stress-induced cerebellar neuropathology was investigated Methodology: Twenty-one male Wistar rats were used for this investigation. Rats were randomly assigned into three groups (A, B, and C), each consisting of 7 rats (n = 7). Group A (Control group) were fed control diet; group B rats were subjected to different chronic unpredictable stressors (CUS) + control diet for 21 days, while group C rats were subjected to CUS + Walnut-enriched diet for 21 days. Serum corticosterone levels, the expression level of antioxidant and inflammatory markers, and cytoarchitectural changes in cerebellum were assessed by enzyme-linked immunosorbent assay (ELISA) immunohistochemistry methods. Results: The walnut-enriched diet prevented astrogliosis, modulated serum corticosterone expression, and tumor necrotic factor-α in the cerebellum. The walnut-enriched diet also caused an improvement in the antioxidant profile, indicating that it suppressed chronic unpredictable stress-induced perturbations. Conclusion: Our results suggest that African walnut exerts protective effects against oxidative stress-driven dysfunction by reducing serum corticosterone levels, modulating oxidative stress pathways, and preventing neuronal morphological damage in the cerebellum.

Abstract The oxidative stress is one of the known and discussed culprits for development and progression of the disturbances in Metabolic syndrome (MetS). In our study, we are investigating the effects of antioxidant additives as additional treatment and prophylaxis in a model of MetS. We are using a fructose-induced metabolic syndrome rat model to examine the effects of antioxidant preparation ZellSchutz. Four experimental groups were formed using male Wistar rats (n = 60). The control group received standard rat chow, the other groups received either ZellSchutz (Z), fructose (F), or fructose and ZellSchutz (ZF). Our results showed an unexpected increase in the RBC indices in rats with dual treatment. There was a significant increase in hemoglobin concentration, MCHC, and MCV in those animals (p<0.001). The preliminary data shows the important role of antioxidants as supplementary therapy in metabolic syndrome. Further investigations will be carried on to reveal the molecular mechanisms of the observed changes.

Background: Diabetes mellitus is a chronic metabolic disorder that develops due to insulin deficiency or insulin resistance. Recent animal and human studies have reported bromocriptine to be effective in the management of type 2 diabetes mellitus. The present study was done to evaluate the antihyperglycemic effect of bromocriptine in dexamethasone induced hyperglycemic rats.Methods: Male wistar rats were used and divided into 5 groups. Dexamethosone was used to induce hyperglycemia in group B-E. Group A was the untreated control group, group B was the standard control group, group C was the oral 10 mg/kg of bromocriptine dissolved in 0.9% normal saline, group D was the oral 20 mg/kg metformin dissolved in 0.9% normal saline, group E was the oral 10 mg/kg bromocriptine+20 mg/kg metformin dissolved in 0.9% normal saline. Fasting blood glucose, post prandial blood glucose and body weight was estimated on day 1, 15, 30.Results: It was seen that dexamethasone induced hyperglycemia and increase in body weight in male wistar rats, which were significantly controlled by oral bromocriptine and bromocriptine with metformin combination.Conclusions: Results obtained from this study showed that bromocriptine can be a promising drug with novel mechanism to treat type 2 diabetes mellitus.

Male infertility is increasingly worrisome and has been linked to increase rate of hormonal toxicity and imbalance. Sufferers now result to using medicinal plants due to its availability and cost effectiveness. This study was to assess ameliorating effects of Phoenix dactylifera (date), Cyperus esculentus (tigernut) and Cocos nucifera (coconut)mixture on pesticides-induced hormonal toxicity in male Wistar rats. Aqueous extraction of crops - date (maceration apparatus), tiger nut (rotary evaporator) and coconut (spray drying process) wereconducted after which, 5g of each extract were mixed in equal proportion. Hormonal toxicity was induced in rats using Ochratoxin A and endosulfan before oral treatment of the mix at graded doses (250 to 1000 mg/kg/day) for 28 days. Aseptic bloodsample was obtained before and after treatment for hormonal assay. At termination, animals were euthanized via decapitation; organs were excised, processed histologically, stained with H&E, and testes examined microscopically. Hormonal toxicity in rats was successfully induced, body weight of highly treated rats reduced (P ≤ 0.02) while testicular weight increased insignificantly (P ≥ 0.05). Mixed extract ameliorated the effect of pesticides on testosterone levels in group: B (1.8 ± 0.5ng/dL), C (2.4 ± 0.4ng/dL), D (3.8 ± 0.6 ng/dL) and E (4.1 ± 0.3 ng/dL) including FSH and LH values (P ≤ 0.05). Stained sections of testis were in keeping with normal histology after investigations. High doses of extract mix successfully reversed pesticides-induced hormonal toxicity in rats without harming the testes. Aim Angiogenesis plays an important role in the pathogenesis of endometriosis. Thus, the inhibition of angiogenesis may prevent endometriosis. Bevacizumab is a recombinant humanized monoclonal antibody against VEGF. The aim of this study is to investigate the efficacy of anti-VEGF therapy on the prophylaxis and treatment of endometriotic foci in a rat model. Methods This experimental study is prospective, randomized, and placebo-controlled. Thirty-six Wistar-Albino female rats were divided into 3 groups. Experimental endometriosis was induced by the implantation of autologous endometrial tissue. The bevacizumab administration route was intraperitoneal. Group A was the prophylaxis group; Group B was the treatment group, and Group C was the control group. The volumes of the implants as well as their VEGF and Ki-67 immunohistochemical staining are main outcome measures. Results The volumes of the lesions were smaller in Group A than Group C [P<.05]. The volumes of endometriotic foci in Group B were smaller than in Group C [P<.05]. Bevacizumab caused regression and atrophy of the endometriotic lesions. After the treatment the histopathologic and immunohistochemical scores in Group B were less than before treatment and less than the scores in Group C. Conclusions Bevacizumab treatment had a regressive effect on the endometriotic implants. As an anti-VEGF agent, bevacizumab has beneficial effects on the prophylaxis and treatment of endometriosis.

To evaluate the effect of gingko biloba (EGb) on diethylstilbestrol (DES) induced testicle injury in mice. Fifty male mice were divided into a control group (A), DES group (B), and 3 EGb groups (C, D, E). The EGb-treated groups received peritoneal EGb at 8.75 (C), 17.5 (D), 35 mg/kg (E) BW daily for 7 days. The control group was given equivalent amount of normal saline. The mice in groups B, C, D and E were injected hypodermically with DES at 40 mg/kg BW daily 4 hours after the first herbal administration, while the control was given olive oil. Compared with DES group, the testis coefficients-relative testicular weight increased in the three EGb-treated groups. No significant difference was observed in epididymis coefficients. Lipid peroxidation status and antioxidant enzyme activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) were significantly elevated in testes of EGb-treated groups. Lactate dehydrogenase (LDH) activities and malonaldehyde (MDA) contents were significantly decreased in testes of the EGb groups. The results indicate that EGb protects the testis from diethylstilbestrol-induced injury.
