The Effect of Algal Extract of Spirulina Platensis, Cladophora Glomerata on Some Positive and Negative Bacterial Isolates of Gram Stain

This study examines the efficacy of the algal alcoholic extract isolated from Cladophora Spirulina platensis and glomerata and testing their efficacy against positive bacteria of Gram stain Staphylococcus aureus and the negative bacteria for Escherichia coli and Klebislla sp. The organic solvent ethanol was used at 95%. The efficacy of the extract was tested against some strains of positive and negative bacteria of Gram stain. The results of the extract showed that the positive bacteria of Gram stain has sensitivity towards the extract higher than the negative bacteria. The highest inhibitory efficacy against Staphylococcus aureus by inhibitive diameter zone of 20 mm and less inhibitive against negative bacteria E.coli by inhibitive diameter of 16 mm.

Nudibranchs are marine invetebrates belongs to the class of Gastropoda that are able to camouflage and develop a self-defense systems. Marine bacteria contain chemicals compunds that have potentials on marine drugs discovery through the secondary metabolism. The purpose of this study was to isolate the symbiotic bacteria from nudibranches Phylidiella pustulosa and Thuridilla lineolate and to screen the antibacterial activity of these bacterial isolates against Escherichia coli and Staphylococcus aureus. Isolation and culture of the symbiotic bacteria was made on B1 (solid) media. The results of the antibacterial assay showed that the symbiotic bacterial isolates from Phylidiella pustulosa and Thuridilla lineolate were able to inhibit the growth of the Staphylococcus aureus and Escherichia coli test.  Ascidian is marine invertebrates in coral reef ecosystems that produce many bioactive compounds for pharmacology. The presence of symbiotic bacteria with marine organisms is protected the host biota by producing secondary metabolites. The purpose of this study is to obtain symbiotic bacterial isolates with Herdmania momus ascidian, then to observe the antibacterial activity of these bacterial isolates against Escherichia coli, and Staphylococcus aureus. Isolation and culture of the symbiotic bacteria were made on Nutrient Agar and Zobell Marine Broth media. A espécie Ptychopetalum olacoides Bentham, popularmente conhecida como “marapuama” ou “muirapuama”, é uma Olacaceae nativa da região norte do Brasil, há muito conhecida e utilizada por suas propriedades estimulantes e afrodisíacas, sendo inclusive exportada para diversos países. O estudo fitoquímico do lenho desta árvore, neste trabalho representado pelos galhos da planta, revelou a presença majoritária de vários ácidos graxos, esteróides e xantinas, sendo eles: ácido palmítico, ácido esteárico, -sitosterol, estigmasterol, lupeol, glutinol, a-amirina, cafeína, teobromina e adenina, sendo que as três últimas não haviam ainda sido reportadas na espécie. O estudo das atividades antimicrobianas revelou que os extratos da planta não apresentam atividade inibitória sobre o desenvolvimento de cepas de Escherichia coli, Staphylococcus aureus ou Staphylococcus epidermidis. Porém, o resíduo do extrato alcoólico apresenta uma significativa ação inibitória do crescimento micelial de Colletotrichum acutatum, e ação menos pronunciada sobre o crescimento de Fusarium oxysporum. The phytochemical study of the wood of this tree, represented by its stems, revealed the presence of several fatty acids, sterols and xanthines, including palmitic acid, stearic acid, -sitosterol, stigmasterol, lupeol, glutinol, a-amirin, caffeine, theobromine and adenine, and the latest three substances have not been reported in this specie yet. The study of antimicrobial properties revealed that the plant extracts don’t have any inhibitory activity against strains of Escherichia coli, Staphylococcus aureus or Staphylococcus epidermidis. However, the alcoholic extract residue does have a remarkable inhibitory action on the mycelial growth of Colletotrichum acutatum and Fusarium oxysporum.26

Background. Drug resistant microorganisms lead to an increase in morbidity and mortality as they boost the risk of inappropriate therapy. Hence, data on antimicrobial resistance help define the best possible treatment for individual patients. Therefore, this study aimed to screen the antimicrobial resistant profile of 3rd generation cephalosporin drugs in Jimma University Specialized Teaching Hospital. Methods. A hospital based prospective cross-sectional study was conducted in Jimma University Specialized Hospital (JUSH) from April to August 2016. The clinical samples such as wound swab, urine, sputum, and stool were collected from hospitalized patients. Then, bacterial species were isolated and identified as per the standard microbiological methods. Antimicrobial susceptibility tests were carried out using various antimicrobial discs by Kirby–Bauer disc diffusion method. Results. Totally, 248 bacterial isolates were obtained from 154 (62.1%) male and 94 (37.9%) female patients. Escherichia coli (25.4%) and Staphylococcus aureus (19.0 %) were the predominant organisms isolated from specimens. About 140 (56.5%) and 149 (60.1%) of the total bacterial isolates were found to be resistant to ceftriaxone and ceftazidime, respectively. The majority of Escherichia coli isolates 46 (73%) were resistant to ceftriaxone and 41 (65%) of them were resistant to ceftazidime. Staphylococcus aureus, which accounted 19% of the total bacterial isolates, showed 23.4% and 34% resistance to ceftriaxone and ceftazidime, respectively. Among the bacterial strains revealing resistant to ceftriazone and ceftazidime, about 109 (44%) and 108 (43.5%) of them were resistant to two, three, or four other drugs, respectively. Conclusion. Bacterial resistance towards third-generation cephalosporin (ceftriaxone and ceftazidime) is escalating as more than half of the isolated strains demonstrated resistance to these drugs. Moreover, these strains also revealed multidrug resistance mainly against clinically used drugs which could render therapy unsuccessful. Therefore, in clinical use appropriate medications should be selected based on the data obtained from antimicrobial susceptibility tests.

Nosocomial infection is a rising problem in developing countries. Microorganisms Isolated from three private and public hospital environmental surfaces in Akure Metropolis, Ondo State, Nigeria were investigated in this study. Bacterial and fungal organisms were isolated and compared among the three hospitals. The study revealed that bacteria were the most predominant microorganisms found in the hospital environmental surfaces than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria but Aspergillus funmigatus was the predominant fungi. The result showed that the microbial loads of the public hospitals were higher than that of the private hospitals. The bacteria load of the male ward was found to be higher than that of the female ward while the fungal loads of each of the hospital environmental surfaces of female were higher than that of the male. The study revealed that bacteria were the most predominant microorganisms found in the hospital environment than fungi. Staphylococcus aureus, Streptococcus pyogenes, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae and Bacillus cereus were the bacterial isolates while fungi include Aspergillus fumigatus, Aspergillus niger and Candida albicans.  Staphylococcus aureus was found to be predominant bacteria. All the hospital environmental surfaces were contaminated with one or more microorganisms in the course of the research.

Abstract The study was conducted to obtain the alcoholic extract of the licorice roots plant and to test its effectiveness against diagnosed bacterial isolates. This research was conducted at the Food Contamination Research Center/Department of Environment and Water/Ministry of Science and Technology. The results showed the following: 1-80% ethyl alcohol was used to obtain the alcoholic extract using a saxolite device, and the activity of the extract was tested against four bacterial isolates diagnosed in the Food Contamination Research Center, which included two gram-negative isolates of Escherichia Coli and Pseudomonas aeruginosa, and two gram-positive isolates of Bacillus Cereus and Staphylococcus aeastureus by a method, in addition to yeast by Diffusion method. The study included two treatments of biscuits with alcoholic extract (A) at a concentration of 10% and compared to the control treatment (B) without any adding and for preservation periods (1, 4, 8, 15, 22) days, and microbial tests were conducted for the biscuits. The alcoholic extract showed a lethal activity to microorganisms and no bacterial cell appeared in the biscuit samples except for the incubation period (22) days for storing biscuits, which recorded the presence of fungi (2) cells/ml. Conducting sensory evaluation of the treatments (A, B) for biscuits where there were no significant differences (P<0.05) for sensory attributes except for flavor and flakes of biscuits.

One of the most important problems faced by microbiologists is to preserve bacterial isolates in the best state to study and further diagnosis. The current study aims to provide a summary of experimental results to maintain two species of bacteria alive after being stored by using some additives. This study found that the best temperature to preserve Staphylococcus aureus was -20°C for a year, while for Escherichia coli it was the same temperature except in using Glycerol (G) 100% and Food oil (FO) methods. The optimum method to preserve S. aureus was by using Normal Saline (NS), while Distilled Water (DW) was the optimum method to preserve E. coli at temperatures (4, 25 and -20)°C for a year, the phenotypic patterns for examining bacteria were maintained except in NS at 4°C for S. aureus after a year ago. Glycerol was used alone at concentrations (100, 50, 30 and 15)%, and another group used G+NS in the same volumes, good results were achieved when it used alone or with NS to preserve bacteria for six months at 4°C except for methods of G100% and (G100% + NS) for examining bacteria. FO has never been used as preservation liquid, it is successful to survive S. aureus at -20°C for a year, and when it was added to NS, E. coli survived for a year at three temperatures (4, 25 and -20)°C, while S. aureus didn’t survive for a year when FO+NS method used at room temperature. The precipitation method was used for bacterial suspension, then added the preserving liquid, but the results were not effective compared to the First method.

This research was conducted to find out the capabilities of endophytic bacteria isolated from flowers of the clove plants in inhibiting the growth of bacteria Streptococcus mutans, Staphylococcus aureus, Klebsiella pneumoniae and Escherichia coli and to identify endophytic bacteria that potensial to produce an antibacterial. Stages of this research include the isolation of endophytic bacteria from flowers of the clove plants, antibacterial test, and molecular identification based on 16S rRNA. Isolates of endophytic bacterial of clove plants flower produce 5 isolates, 4 isolate inhibited the bacteria S. aureus. Based on 16S rRNA molecular identification, endophytic bacterial isolates of clove plants flower which have inhibitory closely related to Bacillus amyloliquefasiens, Staphylococcus epidermidis 1034 MPA and Bacillus cereus JL.

Aims: The study was carried out to determine the phytochemical constituents and antibacterial activity of aqueous and ethanolic extracts of fresh leaves of Moringa oleifera Lam., Pterocarpus santalinoides L’Herit DC and Ceiba pentandra L. on bacterial isolates; Salmonella typhi, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Methodology: The plant leaves were dried, pulverized and phytochemical tests were done according to standard laboratory procedure. Aqueous and ethanolic extracts were obtained from 20 g of the of the ground leaves. Antibacterial assay was carried out with Disc diffusion method on seven concentrations of the extracts ;100,50,25,12.5, 6.25,3.125,1.5625 mg/ml and compared with standard antibiotics. Isolated bacterial pathogens; Salmonella typhi, Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa (1.0 x 105 cfu /ml) were used as test organisms. Results: Alkaloids, steroidal aglycones, glycosides, proteins, carbohydrates, reducing sugars, tannins, saponins, vitamins A and E were present in all the plant samples. Flavonoids and cardiac glycosides were not detected in Pterocarpus santalinoides and Ceiba pentandra, respectively. Anthracene glycoside was absent in all samples. Aqueous and ethanolic extracts of M. oleifera showed antibacterial activities against all the bacterial isolates at minimum inhibitory concentration (MIC) of 3.125 mg/ml and 1.5625 mg/ml respectively. Pterocarpus santalinoides showed inhibitory activity only on Salmonella typhi at 3.125 mg/ml and Escherichia coli 1.5625 mg/ml MIC. Ceiba pentandra showed spectrum of antibacterial activity against all the bacterial isolates at 1.56 mg/ml MIC with exception of Salmonella typhi. E. coli was the most susceptible to the leaf extracts. Salmonella typhi was not sensitive to the leaf extracts of Ceiba pentandra, while Staphylococcus aureus and Pseudomonas aeruginosa were not sensitive to the leaf extracts of Pterocarpus santalinoides. Conclusion: It can be concluded that both aqueous and ethanolic leaf extracts had antibacterial activity against the test organism, thus justifying their use in folklore medicine.
