Abstract POTENSIAL DEVELOPMENT OF EXTRACT BELUNTAS (Plucea indica Less.) as BIOLARVACIDE TO MOSQUITO Aedes aegypti LARVAE. The eradication of Aedes aegyptY mosquito is difficult because they have the ability to adapt the environment which makes it very tough. Although, there are not disturbances due to natural phenomena or human intervention. Termination of the mosquito life cycle is an alternative to reduce the mosquito population. The antimicrobial of beluntas extract is expected to have the ability biolarvacide on mosquito larvae. The biolarvasicide of beluntas leaf extracts was determined LC50 values and strengthened by identification of the active compound. The biolarvacide tested was conducted on the larvae of Aedes aegypti with variations extract concentrations of 50, 100, 250, 500 and 1000 ppm for 24 hours observation. The experimental results found that yield of ethanol extract, extract n-hexane and ethyl acetate extract: 3.8742%, 1.2054% and 1.8627%. While the value of LC50 to extract n-hexane and ethyl acetate respectively amount to 46.09 ppm and 108.79 ppm. LC50 value obtained belong biolarvacide active and positive control using abate value LC100 Abate at a concentration of 100 ppm. The ability biolarvacide ethyl acetate fraction only make the mosquito larvae die, anwhile the fraction of n-hexane can degrade the cells larvae destroyed. The ability biolarvacide beluntas extract was corroborated by the results of GC-MS analysis which showed contains active compounds beluntas such as quinic acid, hydrazinecarboxamide, benzene acetic acid, and 1,2-benzendicarboxylic acid which is a compound of larvicides. Keywords: biolarvacide; beluntas; LC50, GC-MS  Abstrak Pemberantasan nyamuk Aedes aegypti sulit dilakukan karena mereka memiliki kemampuan adaptasi lingkungan yang membuat sangat tangguh, meski ada gangguan  akibat  fenomena  alam ataupun  intervensi manusia. Pemutusan siklus hidup nyamuk merupakan alternative dalam mengurangi populasi nyamuk. Sifat antimikroba ekstrak nyamuk diharapkan dapat memiliki kemampuan biolarvasida pada jentik nyamuk. Kemampuan biolarvasida ekstrak daun beluntas ditentukan melalui nilai LC50 dan diperkuat dengan identifikasi kandungan senyawa aktif. Uji biolarvasida ini dilakukan terhadap larva nyamuk Aedes aegypti dengan variasi konsentrasi ekstrak 50, 100, 250, 500 dan 1000 ppm selama 24 jam pengamatan. Hasil percobaan diketahui bahwa: rendemen untuk ekstrak etanol, ekstrak n-heksana dan ekstrak etil asetat masing-masing sebesar 3,8742 %, 1,2054 % dan 1,8627 % sementara nilai LC50 untuk ekstrak n-heksan dan etil asetat masing-masing sebesar 46,09 ppm dan 108,79 ppm. Nilai LC50 yang diperoleh termasuk golongan biolarvasida aktif dan kontrol positif menggunakan abate memiliki nilai LC100 Abate pada konsentrasi 100 ppm. Kemampuan biolarvasida fraksi etil asetat hanya membuat larva nyamuk mati sementara fraksi n-heksana dapat mendegradasi sel larva hingga hancur. Kemampuan biolarvasida aktif ekstrak beluntas ini dikuatkan dengan hasil analisa GC-MS yang menunjukkan bahwa kandungan senyawa aktif ekstrak daun beluntas seperti quinic acid, hydrazinecarboxamide, benzene acetic acid, dan 1,2-benzendicarboxylic acid yang merupakan senyawa larvasida.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Longan (Dimocarpus longan Lour.) belongs to the Sapindaceae family. We examined the antiproliferative activity of longan leaf extracts against cancer-derived cell lines in vitro. The tested samples were water extract, ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction of longan leaf. Cytotoxicity test is against brine shrimps that were screened using Brine Shrimp Lethality Test. Antiproliferative activity assay on WEHI-164 cells (mouse fibrosarcoma cancer cell), THP-1 cells (human peripheral blood acute monocyte cell), and Vero cells (non-cancer or normal cell) that was conducted using a hemocytometer with Trypan Blue Dye exclusion. The 50% lethality concentration (LC50) value of water extract, ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction were 854.64, 305.81, 446.55, 1313.44, and 1621.8 μg/ml. Ethanol extract exhibited significant cytotoxic due to the lowest LC50 value. The ethanol extract was then used for further examination. The highest antiproliferative activity was achieved 44.93% by 600 μg/ml ethanol extract on WEHI-164 and 57.45% by 500 μg/ml ethanol extract on THP-1. It was significantly equal to doxorubicin antiproliferative activity. Ethanol extract dose had a low effect on Vero cells. This present study confirmed that the longan leaf ethanol extract possesses marked antiproliferative activity on cancer-derived cell lines.

Penelitian ini bertujuan untuk mengetahui aktivitas dari ekstrak daun beluntas terhadap mortalitas larva nyamuk Aedes aegypti. Ekstrak daun beluntas diperoleh dengan menggunakan metode ekstraksi pada pelarut etanol. Setelah didapatkan ekstrak kental, maka ekstrak tersebut difraksinasi dengan pelarut n-heksana dan etil asetat. Kemampuan biolarvasida ekstrak daun beluntas ditentukan melalui nilai LC50 dan diperkuat dengan identifikasi kandungan senyawa aktif. Uji biolarvasida ini dilakukan terhadap larva nyamuk Aedes aegypti dengan variasi konsentrasi ekstrak 50, 100, 250, 500, dan 1000 ppm selama 24 jam pengamatan. Identifikasi kandungan senyawa dilakukan dengan pengujian sampel dengan analisa GC-MS. Hasil penelitian menjunjukkan bahwa rendemen ekstrak etil asetat sebesar 1,86 %. Nilai LC50 ekstrak etil asetat ini adalah 105,79 ppm. Nilai LC50 yang diperoleh termasuk golongan biolarvasida aktif dan kontrol positif menggunakan abate memiliki nilai LC100 pada konsentrasi 100 ppm. Sementara itu, nilai LC50 pada granul dengan kandungan ektrak daun beluntas yang terbaik diperoleh nilai 96,34 ppm dan nilai LC90 adalah 905.1 ppm. Kemampuan biolarvasida aktif ekstrak beluntas ini dikuatkan dengan hasil analisa GC-MS yang menunjukkan bahwa kandungan senyawa aktif biolarvasida yang terkandung dalam ekstrak etil asetat diduga asam quanat.(THE EFFICATION OF AEDES AEGYPTI BIOLARVASIDA GRANUL FROM ETIL ACETAT EXTRACT OF MARSH FLEABANE LEAVES)This study was aimed at determining the activity of Marsh Fleabane leaves extract against Aedes aegypti mosquito larvae mortality. Marsh Fleabane leaves extract was obtained using ethanol solvent. After obtaining the viscous extract, the extract was fractionated with solvent n-hexan and ethyl acetate. The ability of this biolarvaside was determined by LC50 value and reinforced by identifying of active compound content. This biolarvaside test was conducted on Aedes aegypti mosquito larvae with variation of extract concentration 50, 100, 250, 500, and 1000 ppm during 24 hours observation. Identification of compound content was done by testing the sample with GC-MS analysis. The results show that the yield of ethyl acetate extract equal to 1.86%. The LC value of this ethyl acetate extract was 105.79 ppm. The obtained LC50 values including active biolarvacidal groups and positive controls using abate had LC100 values at 100 ppm concentrations. Meanwhile, the LC50 value on granules using marsh Fleabane leaves extract content was obtained at 96.34ppm and LC90 was 905.1 ppm. The ability of active biolarvaside is corroborated by GC-MS analysis results indicating that the biolarvacid content of active compounds contained in ethyl acetate extract expected as quinic acid

BACKGROUND: Nutmeg (Myristica fragrans H.), one of native plants of Maluku Indonesia, has long been used as traditional medicines especially to treat tumors, externally to treat skin infections. M. fragrans also has important biological activities as anticancer. However, antimelanoma activity of M. fragrans remains unknown. The aim of this study is to compare M. fragrans extracts as anticancer on melanoma B16-F10 cells by inducing apoptosis.METHODS: M. fragrans seed was extracted with ethanol then fractionated with n-hexane, ethyl acetate, and n-butanol. B16-F10 melanoma cells were cultured and treated with various doses and tested using resazurin reduction assay. Apoptosis signalling via caspase-3 was measured by using western blot.RESULTS: The extract and fractions of M. fragrans reduced viability of cells with IC50 value for ethanol extract 21.83 µg/mL, ethyl acetate fraction 21.66 µg/mL, n-hexane fraction 47.53 µg/mL, and n-butanol fraction >1,000 µg/mL. The active fraction of ethyl acetate induced apoptosis via caspase-3 proteins similar with cisplatin as positive control in B16-F10 cells at 10 hours treatment.CONCLUSION: Taken together, M. fragrans ethyl acetate fraction has the highest IC50 than n-hexane and n-butanol fractions that significantly inhibited B16-F10 cell proliferation by inducing apoptosis via caspase-3. It provides the insight that it has the most potential activity as a chemopreventive agent for addressing melanoma skin cancer.KEYWORDS: M. fragrans, apoptosis, fraction, melanoma

Penelitian ini bertujuan untuk mengukur kadar ekstrak daun tumih dan menguji aktivitas larvasidanya terhadap Aedes aegypti. Daun tumih dimaserasi dan difraksinasi dengan pelarut metanol, n-heksana, etil asetat, dan etanol. Aktivitas larvasida ekstrak diuji dengan konsentrasi 0, 5, 10, 25, 50, 75, dan 100 ppm. Hasil penelitian menunjukkan rendemen dari ekstraksi daun tumih pada berbagai larutan adalah sebagai berikut ekstrak metanol sebesar 15%, n-heksana 51%, etil asetat 35% dan etanol 85%. Hasil uji fitokimia menunjukkan bahwa ekstrak metanol daun tumih positif mengandung senyawa alkaloid, flavonoid, saponin, dan tanin. Pengujian larvasida ekstrak daun tumih menunjukkan pengaruh terhadap mortalitas larva Aedes aegypti. Aktivitas larvasida ekstrak daun tumih optimum pada ekstrak etil asetat dengan LC(50) = 24,54 ppm, ekstrak metanol LC(50) = 45,65 ppm, ekstrak etanol LC(50) = 46,77 ppm dan ekstrak n-heksana LC(50) = 48,97 ppm. Ekstrak etil asetat daun tumih merupakan ekstrak teraktif dalam aktivitas larvasidanya. Selanjutnya, analisis FT-IR menunjukkan adanya gugus fungsi C-H alkana dan C = C aromatik. Gugus fungsi tersebut diduga penyusun senyawa alkaloid, flavanoid, saponin, dan tanin. Hasil analisis LCMS mengindikasikan adanya 7 senyawa bioaktif yaitu hexadecyl-ferulate, 21-o-methyl toosendanopentaol, 23-acetate alismaketone, dehydroxy-24-acetate alisol, physanol, prosapogenin 2, dan stigmastan-3,6-dione Larvacide Activity of Tumih (Combretocarpus rotundatus (Miq.)Danser) Leaf Extracts against Aedes aegyptiAbstractThis research aimed to measure the content of Combretocarpus Rotundatus (Miq.) Danser leaf extracts and to test its larvicidal activity against Aedes aegypti. The leaves were macerated and fractionated using methanol, n-hexane, ethyl acetate, and ethanol. The extract contents from the leaves extraction were metanol extract of 15%, n-hexane extract of 51%, ethyl acetate extract of 35% and ethanol extract of 85%. The larvicidal activity of extracts was tested with concentration of 0, 5, 10, 25, 50, 75, and 100 ppm. Phytochemicals test exhibited that the methanol extract of Combretocarpus rotundatus (Miq.) Danser leaves contained alkaloids, flavonoids, saponins, and tannins. Larvicidal test conducted on the extracts exhibited an effect on the mortality levels against Aedes aegypti larvae. The larvicidal activity of leaf extracts was optimum in the ethyl acetate extract at LC(50) = 24.54 ppm, methanol extract at LC(50) = 45.65 ppm, ethanol extract at LC(50) = 46.77 ppm, and n-hexane extract at LC(50 = 48.97 ppm. It was found that the ethyl acetate extract was the most active larvicide. FT-IR analysis showed existing functional groups of C-H alkanes and C=C aromatics. Those functional groups were assumed to be flavanoid, alkaloid, saponin, and tannin constituents. Results of LC-MS analysis indicated 7 bioactive compounds i.e.hexadecyl-ferulate, 21-o-methyl toosendanopentaol, 23-acetate alismaketone, dehydroxy-24-acetate alisol, prosapogenin 2, and stigmastan-3,6- dione.

National data shows the prevalence of coronary heart disease is 3,6%. The prevalence of dyslipidemia is around 35,9%. Dyslipidemia has a causal relationship with cardiovascular disease. Based on previous research, 70% ethanolic extract of Ridge Gourd has been shown to have antihyperlipidemic activity. This study aimed to study the activity of 70% ethanolic extract fraction of Ridge Gourd in decreasing the levels of total cholesterol, low-density lipoprotein (LDL-C), and triglycerides in hyperlipidemic hamsters. A total of 32 hamster were divided into 8 groups. Negative control, normal control, positive control (using atorvastatin dose 1,24 mg/kg body weight), positive control (fenofibrate dose 12,35 mg/kg body weight), ethyl acetate fraction, n-hexane fraction, water fraction (fraction doses 36,75 mg/kg body weight), and ethanol extract group (extract dose of 240 mg/kg body weight). The results showed that fraction group (n-hexane, ethyl acetate, water) and ethanol extract group experienced a decrease in total cholesterol, LDL-C, and triglycerides levels. Statistical analysis with one-way ANOVA showed an effect on the treatment given (p = 0,005). Tukey's test showed that ethyl acetate fraction group had an effect comparable to positive control with a decrease in total cholesterol,  LDL-C, and triglycerides level by 56,03%, 52,14%, and 59,51% consecutively. Ethyl acetate fraction (doses 36,75 mg/kg body weight) was the most effective fraction in reducing total cholesterol, LDL-C, and triglycerides.

BACKGROUND: Gastritis is one of the most common diseases found in internal medicine clinics and daily life. AIM: The purpose of this study was to determine the histopathological changes of gastric rat model of aspirin-induced gastritis. MATERIALS AND METHODS: Group K(−) is a negative control (normal) not given any treatment, Group K(+) is a positive control induced by aspirin 200 mg/kgBW, Group 1 (P1) is a treatment group induced by aspirin 200 mg/kgBW with ethanol extract of chayote 100 mg/kgBW, po, Group 2 (P2) was the treatment group induced by aspirin 200 mg/kgBW with ethanol extract of chayote 200 mg/kgBW, po, Group 3 (P3) was the treatment group induced by aspirin 200 mg/kgBW with ethyl acetate fraction chayote 100 mg/kgBW, PO, Group 4 (P4) was the treatment group induced by aspirin 200 mg/kgBW with ethyl acetate fraction chayote 200 mg/kgBW, PO, and Group 5 (P5) was the treatment group induced by aspirin 200 mg/kgBW with omeprazole 20 mg. RESULTS: Analysis of the results of inflammatory infiltration in gastric histopathology in Groups P1, P2, P3, and P4 showed mild inflammatory infiltration compared to Groups P(5) and K(+). The description of the acinar glands on gastric histopathology showed that the P2 group gave a better picture of the acinar gland repair than the K(+), P2, P3, P4, and P5 groups. CONCLUSION: This study showed that ethanol extracts of squash 200 mg/KgBW own effectiveness as an anti-inflammatory on the improvement of infiltration of inflammatory and picture histopathological gastric mice compared with the ethanol extract of squash 100 mg/kg, the fraction of ethyl acetate chayote 100 mg/KgBW and 200 mg/KgBW and omeprazole 20 mg.

One chemichal component that is often reported to have pharmacological activity against treatment or prevention of disease is flavonoids which can be found in gedi hijau leaves. Staphylococcus aureus is the main cause of infection in humans which can cause various severe infections. This study aims to determine the total flavonoids content, minimum inhibitory concentration (MIC). To determine the correlation between total flavonoids and antibacterial activity of extract and fractions (Abelmoschus manihot L.), The method of this study was done by obtaining the results of total flavonoid content, testing the antibacterial activity by dilution using a positive control of ciprofloxacin. The results showed the highest total flavonoid content in ethyl acetate fraction, and the best antibacterial activity in ethyl acetate fraction. Correlation between total flavonoid content from extract and fraction (Abelmoschus manihot L.) with antibacterial activity against Staphylococcus aureus bacteria on ethyl acetate fraction, n-hexane fraction, and ethanol extract at a concentration of 1000 µg / mL had good values with kneeling significance values according to 0.012 (p <0.05), 0.034 (p <0.05), and 0.023 (p <0.05) with total flavonoid content affecting how much antibacterial activity is 40% for ethyl acetate fraction, 35% fraction n -heksane and 33% ethanol extract. Keywords         : Antibacterial, Total Flavonoids, Staphylococcus aureus, Abelmoschus manihot L. ABSTRAK Salah satu komponen kimia yang sering dilaporkan memiliki aktivitas farmakologi terhadap pengobatan atau pencegahan penyakit adalah flavonoid yang dapat ditemukan pada daun gedi hijau. Staphylococcus aureus merupakan kuman penyebab utama infeksi pada manusia yang dapat menyebabkan berbagai infeksi berat. Penelitian ini bertujuan menentukan kandungan flavonoid total, aktivitas antibakteri dan korelasi antara flavonoid total dengan aktivitas antibakteri dari ekstrak dan fraksi (Abelmoschus manihot L.). Metode penelitian ini dilakukan dengan mendapatkan hasil kandungan flavonoid total, uji aktivitas antibakteri dengan cara dilusi menggunakan kontrol positif ciprofloxacin. Hasil penelitian didapatkan kandungan flavonoid total tertinggi pada fraksi etil asetat, dan aktivitas antibakteri paling bagus pada fraksi etil asetat. Korelasi antara kandungan flavonoid total dari ekstrak dan fraksi (Abelmoschus manihot L.) dengan aktivitas antibakteri terhadap bakteri Staphylococcus aureus pada fraksi etil asetat, fraksi n-heksane, dan ekstrak etanol pada konsentrasi 1000 µg/mL memiliki nilai yang baik baik dengan nilai signifikansi bertutut-turut 0,012 (p <0,05), 0,034  (p <0,05), dan 0,023 (p <0,05) dengan kandungan flavonoid total mempengaruhi seberapa besar aktivitas antibakteri yaitu 40% untuk fraksi etil asetat, 35% fraksi n-heksane dan 33% ekstrak etanol.Kata kunci          : Antibakteri, Flavonoid Total, Staphylococcus aureus, Abelmoschus manihot L.

Background: Staphylococcus aureus is a gram-positive bacteria that can cause infection. One of the plants that has antibacterial activity is jatropha leaves which contain flavonoids, saponins, tannins, alkaloids, steroids and polyphenols. Purpose: To determine the antibacterial activity of ethanol extract and ethyl acetate fraction with concentrations of 30%, 60% and 100% against Staphylococcus aureus bacteria. Method: The method for extracting jatropha leaves is maceration with 96% ethanol solvent and the fractionation method, namely liquid-liquid fractionation with ethyl acetate solvent. Antibacterial activity test was carried out in vitro with the disc diffusion method and compared the mean zone of inhibition of each treatment with a positive control (gentamicin 10 μg). Result: The results showed that the ethanol extract and ethyl acetate fraction of jatropha leaves had a strong resistance response, while the positive control gave a very strong inhibitory response to the growth of S. aureus bacteria. Based on the one way ANOVA test, ethanol extract and ethyl acetate fraction showed a significant difference from each treatment with a significant value (P=<0.05). Conclusion: The ethanol extract of Jatropha leaves can inhibit the growth of S. aureus bacteria at a concentration of 100% (18.28 ± 0.50 mm), 100% concentration of ethyl acetate fraction (15.10 ± 0.12 mm). The ethanol extract provided the best inhibition power, namely 18.28 ± 0.50 mm and a positive control 21.82 ± 0.092 mm.

This study aims to find the treatment of diabetes using natural materials by exploring plants in the province of East Nusa Tenggara. his research was conducted out by extracting the Strychnos nitida G.Don stem using a method of maceration by ethanol 70%. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Simplicia from maceration and fractionation results were then tested for antioxidant activity, α-glucosidase inhibition activity and identification of active compounds. The results showed that ethyl acetate fraction had the lowest IC50 value of 86.83 μg / ml. Results of the α-glucosidase activity test showed that ethyl acetate fraction and n-heksan fraction at 900 ppm had the highest percentage of inhibition of 34.23% and 33.89%. Identification using LCMS/MS method showed that ethyl acetate fraction consist of Benzenemethamine, N, N-dioctyl- as an antioxidantcompound and compound 24-methyl-5-cholestone-hexol as an antidiabetic compound. From the results of this study, we concluded that the extract of kayu ular Strychnos nitida G.Don stem has inhibition activity toward α-glucosidase enzyme.
