Cyperus rotondus tubers possess substances with allelopathic qualities. Some of these substances can act as synergists of plant hormones and be used for root induction in cuttings. This study aimed to shed light on the development and rooting of Eucalyptus urophylla cuttings treated with C. rotondus (nutgrass) tuber extract and IAA (indole-3-acetic acid). The experiments were conducted using a completely randomized design (0; 25; 50; 75 and 100% nutgrass extract and 0; 50; 250; 500; 750 and 1000 mg L-1 of IAA), with four repetitions per treatment and four cuttings per plot. Plantlet height, stem width and root length were measured. The greatest width and height values were recorded for the control (0% nutgrass extract and 0 mg L-1 of IAA), indicating that these substances did not contribute to the development of these variables. However, treatments of nutgrass extract up to 50% were associated with greater root length. C. rotundus extract concentrations of 53% can be used to improve root development in E. urophylla cuttings, and contribute to the production of higher quality plantlets.

In vitro rooting of micropropagated shoots were carried out with the aim of evaluating the root induction responses of two tomatoes (Lycopersicon esculentum MILL) varieties (Gelilema and Chali) using Indole -3- butyric acid (IBA). Seven levels of IBA (0.0, 0.25, 0.5, 0.75, 1.0, 1.25 and 1.5 mg L-1) were used in a completely randomized design (CRD) in factorial combinations (seven level of IBA*two varieties) with three replications.  After the plantlets were kept in the rooting media for three weeks, data on rooting percentage, number of roots/shoots and root length in cm were collected. The analysis of variance showed that the interaction of IBA*Var was highly significantly different for rooting percentage, a number of roots/shoot and root length at p<0.01. The highest rooting percentage (100.00±0.00), number of roots/shoot (14.20±0.35) and root length (10.7±0.29) were received from Chali on free Murashige and Skoog medium (MS). At the same time, the lowest percentage of rooting (11.11±0.00), number of roots/shoot (0.887±0.19) and root length (1.00±0.00 cm) were obtained from Gelilema on MS+1.5mg/l IBA. For acclimatization, the in vitro rooted shoots were transplanted into plastic pots containing a mixture of oven sterilized soil and sand at a ratio of 2:1. After three weeks, a survival rate of 67.7% for Chali and 58.1% for Gelilema was obtained. From the above result, it can be concluded that free MS medium was the best for in vitro rooting of the two tomato varieties. The optimized protocol will be useful for rapid in vitro multiplication of the two tomato varieties.

ABSTRACT The objective of this work was to evaluate the effect of six indole-3-acetic acid (IAA) concentrations and three immersion times in IAA solutions on the vegetative propagation of Spondias dulcis and Spondias tuberosa by stem cuttings. The experiment was conducted in a greenhouse, using a completely randomized design, with a 6×3 factorial arrangement consisting of six indole-3-acetic acid concentrations (0, 2, 4, 6, 8, and 10 g L-1) and three immersion times (8, 16, and 24 seconds), with ten cuttings of S. dulcis and S. tuberosa per plot, and three replications. Percentage of surviving cuttings, and percentage of rooted cuttings, number of leaf buds, number of roots per cutting, root length, and total fresh weight were evaluated at 180 days after planting. Data were subjected to analysis of variance by the F test and to regression analysis. Vegetative propagation from woody cuttings of S. dulcis is not viable to produce seedlings, thus, new studies on this technique for this species are needed. Vegetative propagation from woody cuttings of S. tuberosa is satisfactory, but with low efficiency for improvement and emergence of new shoots and roots; it is more effective when using a concentration of 10 g L-1 of indole-3-acetic acid and immersion time in the solution of 16 seconds.

The research of callus induction in potato (S. tuberosum  L.) from Dombu Village with indole-3-acetic-acid (IAA) was conducted from Januari to April 2019 in Laboratory of Tissue Culture, Biology Department, Faculty of Mathematics and Natural Science, Tadulako University. The research was aimed to determine of concentration IAA of the best to induce callus. This study was designed based on Completely Randomized Design (CRD) with 5 treatments, 3 replications and 3 explants on each unit. The treatments were P1= MS + 0 ppm, P2= MS + 0.5 ppm, P3= MS + 1.0 ppm, P4= MS + 2.0 ppm, P5= MS + 3.0 ppm. The results showed that  the best and the efficient treatments for induction of callus treatment was P4= MS + 2.0 ppm with to induce callus up to 100%, the callus colour was yellow to brown and intermediate texture.

Provision of Vanilla (Vanilla planifolia Andrews) with traditional cultivation often has problems, including the availability of unhealthy seeds due to disease caused by Fusarium oxysporum. This disease can thwart vanilla plantations up to 85% because the pathogen can infect all parts of the vanilla plant making it difficult to control efforts. Alternative efforts were made to overcome this problem, namely through the initiation process in tissue culture by combining the hormones IAA (indole-3-acetic acid) and BAP (6-benzylaminopurine). This study aims to determine the effect and the maximum concentration of the combination of IAA and BAP on the initiation of vanilla plants. This study was conducted based on a completely randomized design (CRD) with 5 treatments and 3 repetitions. This treatment consisted of: A (MS + 0.1 ppm IAA + 1 ppm BAP), B (MS + 0.2 ppm IAA + 1 ppm BAP), C (MS + 0.3 ppm IAA + 1 ppm BAP), D (MS + 0.4 ppm IAA + 1 ppm BAP), and E (MS + 0.5 ppm IAA + 1 ppm BAP). Observations were made after 40 days from the planting process. The results showed that the concentration of the combination of IAA and BAP can affect the growth and organogenesis of the initiation of vanilla plants (Vanilla planifolia Andrews). Treatment C (MS + 0.3 ppm IAA + 1 ppm BAP) was the best concentration from this study, with average values: number of shoots (1), shoot length (1.73 cm), number of roots (1), and number of leaves (1.33).

The cajá-manga (Spondias dulcis Parkinson) is an exotic fruit originally from the Polynesian Islands. It is a climacteric fruit, and the postharvest life of its fruits is very short, due to its fast ripening. Thus, this study aimed to evaluate the effect of auxin concentrations on delaying the ripening process of cajá-manga fruits. One hundred twenty cajá-manga fruits obtained from, Hidrolândia-GO, Brazil, at 16°55'32.35" S and 49°21'39.76" W). A completely randomized design in a 5×2+2 factorial scheme was used. Five indole-3-acetic acid (IAA) concentrations (0, 50, 100, 150 and 200 mg L-1), two moments of evaluations (at 5 and 10 days after IAA applications), and two additional treatments (ripe and unripe fruits) were evaluated. Evaluations were carried out every 5 days, in triplicates. The physicochemical variables, pulp firmness, titratable acidity (TA), soluble solids (SS), ascorbic acid content, and color of pulp and peel were evaluated. The concentrations of 50 and 100 mg L-1 of IAA presented the best results, causing a delay in ripening and the lowest sensory losses in the fruits of cajá-manga.

The purpose of this research is to determine the concentration of young coconut water that is appropriate for the growth of orchid plantlets (Dendrobium sp.) In vitro. This study used an experimental method, descriptive data analysis to compare several different concentrations of coconut water. The design of this study uses a completely randomized design (CRD). The treatments consist of 0% coconut water concentration (as a control), 15%, 30% and 60%. Each concentration was carried out 5 replications and each repetition consisted of 5 Dendrobium sp plantlets in each culture bottle conducted for 40 HST, for observing the root length carried out for 50 HST. The highest number of shoots and leaves were produced at the same concentration, namely 150 ml / L coconut water treatment (15% concentration) with an average of 2.8 shoots and the average number of leaves 10.8 leaves. The average number of roots and the longest root length was produced at a concentration of 600 ml / L coconut water (60% concentration) with an average of 6 roots, and the longest root length was 0.5 cm.Keywords: Young coconut water, (Cocos nucifera L.), Dendrobium sp., in vitro, growth.ABSTRAKTujuan penelitian ini ialah menentukan konsentrasi air kelapa muda yang tepat untuk pertumbuhan planlet anggrek (Dendrobium sp.) secara in vitro. Penelitian ini menggunakan metode eksperimen, analisis data secara deskriptif untuk membandingan beberapa konsentrasi air kelapa yang berbeda. Rancangan penelitian ini menggunakan Rancangan Acak Lengkap (RAL). Perlakukan terdiri dari konsentrasi air kelapa 0 % (sebagai kontrol), 15% , 30% dan 60%. Masing-masing konsentrasi dilakukan 5 kali ulangan dan setiap ulangan terdiri dari 5 planlet Dendrobium sp dalam setiap botol kultur yang dilakukan selama 40 HST, untuk pengamatan panjang akar dilakukan selama 50 HST. Jumlah tunas dan jumlah daun terbanyak dihasilkan pada konsentrasi yang sama, yaitu perlakuan air kelapa 150 ml/L (konsentrasi 15%)  dengan rata-rata jumlah tunas terbanyak 2,8 tunas dan rata-rata jumlah daun terbanyak 10,8 helai daun. Rata-rata jumlah akar terbanyak dan panjang akar terpanjang dihasilkan pada konsentrasi air kelapa 600 ml/L (Konsentrasi 60%) dengan rata-rata jumlah akar terbanyak sebanyak 6 akar, dan rata-rata panjang akar terpanjang 0,5 cm.Kata kunci : Air kelapa Muda (Cocos nucifera L.), Dendrobium sp., in vitro, pertumbuhan

This study aimed to determine the best dosage of goat manure as a natural fertilizer on Beha grass(Brachiaria humidicola). This study used 64 poles of Beha grass grown in polybags divided into 16plots, and it used a completely randomized design (CRD) to analyze the data. This study consist of 4treatments that is P0 (0 ton/ha of goat manure), P1 (10 ton/ha of goat manure), P2 (15 ton/ha of goatmanure), and P3 (20 ton/ha of goat manure). This study's variables were plant height, number of leaves,numbers of tillers, new production, and root length. Data analysis used ANOVA variance continued withreal honest difference test. The result of this study showed that averages of plant height (cm) wereP0=26, P1=50.5, P2=51.5, and P3=52.75. Averages of the number of leaves were P0=22, P1=110,P2=160, and P3=185. Averages of numbers of tillers were P0=7, P1=25, P2=37, and P3=38. Averages ofnew production (g) were P0=17.75, P1=123.5, P2=178, and P3=192.25. Averages of root length (cm)were P0=37.8, P1=39.5, P2=41.775, and P3=49.5. Fertilization with natural fertilizer from goat manurehas a genuine effect (P<0.01) on the growth of Beha grass. The best dosage of fertilization with goatmanure was 15 ton/ha.

Shoot tips of newly emerging mango shoots were used as an explant and cultured on MS media for direct shooting. Different plant growth regulators i.e. BAP, NAA and IAA in varying concentrations were added to basal MS media. Experiment was laid out according to Completely Randomized Design (CRD) with four treatments and each treatment was replicated thrice. Maximum shoot induction (55.56%) was observed with 1 mg/L of BAP. Maximum mortality (83.30%) was observed when maximum concentration of BAP (3 mg/L) was used. Minimum days (12.633) to induce shoots were observed on MS media supplemented with 1 mg/L of BAP. Maximum number of shoots (2.50) were recorded in MS media supplemented with 1 mg/L of BAP. Low concentration of NAA initiated roots earlier in regenerated shoots as compare to high concentrations of NAA and IAA. MS media supplemented with NAA (1 mg/L) took less days (21 days) to induce roots. The combination of auxins (MS+ NAA 3 mg /L + IAA 1 mg /L) proved the best for root induction (38.87%). Plants that can be used as pesticides are Gadung tubers. Gadung tubers contain cyanide acid to kill armyworm pests because they contain dioscorin toxins which can cause nervous disorders. The purpose of this study was to determine the effect of Gadung tuber extract (Dioscorea hispida) on Caterpillar mortality (Spodoptera exigua) in Shallot plants. This type of research is experimental research. This study used a completely randomized design (CRD). The sample used was 5 caterpillars (Spodoptera exigua) in each polybag with 5 repetitions for each concentration of Gadung tuber extract, namely 0%, 0.5%, 1.0%, 1.5%, 2.0% and 2.5%, so the total Caterpillar used was 150 Caterpillars. Gadung tuber extraction was carried out by maceration method with methanol solvent. Data were analyzed using a one-way analysis of variance at the 95% significance level, followed by DMRT (Duncans Multiple Range Test). The results showed that the Gadung tuber extract had a significant effect on the mortality of caterpillars (Spodoptera exigua) because the F count was greater than the F table (45.40>2.62), and the DMRT test results showed that the optimal concentration of Gadung tuber extract was 2.5%. Therefore, it can be concluded that the extract of Gadung tuber (Dioscorea hispida) affects the mortality of Caterpillars (Spodoptera exigua) in Shallots (Allium cepa).

Abstrak. Umbi bit merupakan salah satu bahan pangan yang dapat digunakan sebagai pewarna alami pada pembuatan soft candy seperti rshmallow. Penelitian ini dilakukan dengan menggunakan Rancangan Acak Lengkap (RAL) dengan pola faktorial yang terdiri atas dua faktor. Faktor pertama yaitu konsentrasi ekstrak umbi bit (B) yang terdiri dari 3 taraf yaitu B1 = 5% B2 = 10% dan B3 = 15%. Faktor kedua adalah konsentrasi gelatin (G) yang terdiri dari 3 taraf yaitu G1 = 3% G2 = 5% dan G3 = 8%. Ulangan dilakukan sebanyak 3 kali sehingga diperoleh 27 satuan percobaan. Hasil penelitian menunjukkan bawa konsentrasi ekstrak umbi bit berpengaruh sangat nyata terhadap kadar air, vitamin C dan aktivitas antioksidan serta uji deskrpsi aroma, rasa, warna buram, tekstur, dan mouthfeel marshmallow umbi bit. Adapun konsentasi gelatin berengaruh nyata terhadap kadar vitamin C dan uji deskripsi tekstur marshmallow umbi bit. Marshmallow umbi bit memiliki kadar air yaitu sebesar 22,06%, kadar vitmin C 80,67 Mg dan aktivitas antioksidan sebesar 30%.Abstract: Beetroot is one of food that can be used as natural dyes in making of soft candy like marshmallows. This research was conducted using a Completely Randomized Design (CRD) with a factorial pattern consisting of two factors. The first factor is the concentration of beet tuber extract (B) which consists of 3 levels, namely B1 = 5% B2 = 10% and B3 = 15%. The second factor is the concentration of gelatin (G) which consists of 3 levels, namely G1 = 3% G2 = 5% and G3 = 8%. Tests were carried out 3 replications so that 27 trial units were obtained. The results showed that the concentration of coffee extract had a very significant effect on water content, vitamin C and antioxidant activity, as well as the description of aroma, flavor, opaque color, texture, and mouthfeel marshmallow tuber content. The concentration of gelatin significantly affected vitamin C levels and test description of marshmallow tuber tuber texture. Marshmallow beetroot has a water content of 22.06%, vitamin C content of 80.67 Mg and antioxidant activity of 30%.
