The efficacy of steroids in the treatment of acute aspiration pneumonia, following intratracheal kerosene instillation in primates, was examined. Forty Chacma baboons were utilized in three groups: group I (normal animals), group II (animals receiving 0.3 ml/kg intratracheal kerosene alone, control group), and group III (animals receiving 0.3 ml/kg intratracheal kerosene plus pre- and posttreatment with 5 mg/kg IMI dexamethasone). The lungs from all animals were examined macroscopically and microscopically and their lung wet weight-dry weight and lung weight-body weight ratios determined. On all parameters utilized, the lungs from groups II and III were different from group I. No statistical difference, however, could be detected between values obtained from groups II and III. These data tend to support the contention that steroids, even when given early, and in large doses, in the course of kerosene pneumonitis, do not alter the acute inflammatory process that occurs.

Olanzapine is an antipsychotic drug effective in the treatment of stress-associated psychiatric illnesses, but its effect on the spleen remains unclear. Vitamin C is essential for the optimum function of the immune system. We aim to investigate the effect of Olanzapine on spleen structures and to assess the protective effect of vitamin C. Forty adult male albino rats were divided into four groups: group (I), a control; group (II), rats were given vitamin C at 40 mg/kg body weight; group (III), rats were given Olanzapine at 2 mg/kg body weight; and group (IV), rats were given vitamin C and Olanzapine at the same dose of group (II) and group (III) for one month. The hematoxylin and eosin (H&E) of the olanzapine treated group showed focal areas of cellular depletion and a decrease in the size of the white pulp. The red pulp was expanded and showed marked congestion and dilatation of blood sinusoids. Cluster of differentiation 3 (CD3) was significantly reduced, however both tumor necrosis factor alpha (TNF-α), and vascular endothelial growth factor (VEGF) were significantly higher. The administration of vitamin C repaired structural and immunohistochemical changes via increased CD3 and decreased TNF-α and VEGF. Therefore, the oxidative and the inflammatory pathways may be the possible mechanisms underlying olanzapine immunotoxicity. Vitamin C exerted immune modulator and antioxidant effects against olanzapine.

The study was conducted to determine the effects of vitamin E on reproductive performance in mice treated with 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The 75 female and 40 male mice were randomly assigned into five groups. The levels of both TCDD and vitamin E given by gavage were 0 and 0 (Control group), 100 and 0 (experimental group I), 100 and 20 (experimental group II), 100 and 100 (experimental group III), and 100 ng/kg/day and 500 mg/kg/day (experimental group IV), respectively. Males and females were mixed to mate at the ratio of 1:2 after 4-week experiment. The gavage treatments were continued until the end of gestation in female mice after mating. The results showed that the litter number, survival rate, and body weight at birth of offspring in experimental group I had significantly decreased, and the females’ pregnancy rate and pup sex ratio in experimental group I had the decreasing tendencies when compared with the control group. The litter number in experimental group III, survival rate in experimental group II and III, body weight at birth in experimental group III and IV exhibited significant increase compared with experimental group I. The female pregnancy rate in both experimental group III and IV recovered to 100%, but there were no significant differences when compared with experimental group I. The pups’ sex ratio had a gradually increasing tendency with increase of vitamin E level, but there was no significant difference among experimental group I–IV. The results suggest that TCDD could induce reproductive toxicity in mice, whereas vitamin E alleviated adverse effects on reproductive performance in mice caused by TCDD.

ABSTRACT The concentration of sex hormones and their binding in the plasma were measured in male patients (20–40 years old), who weighed 140–170 % (I), 170–200 % (II) and > 200 % (III) of their ideal weight. Correlated to body weight, there is a reduction in the total concentration of testosterone, which, in the very obese patients, amounts to 41 % (in group I: 85 % in group II: 68 % P < 0.001) of that found in an age-matched healthy control group of subjects of "normal" body weight (90–115 % of the ideal body weight; n: 20). Androstenedione values show only a trend downwards (from 0.94 to 0.72 ng/ml plasma), while the oestrogen values increase significantly; oestrone increases by a factor of 1.09 (I), 1.43 (II; P < 0.001) and 1.69 (III; P < 0.001) and oestradiol by 1.13, 1.43 P < 0.001) and 1.76 (P < 0.001), respectively. Despite the fall in testosterone there are no clinical signs of hypogonadism, as SHBG (from 5.1 ± 0.8 in the controls to 2.4 ± 0.6 ×10−8 Mol/l in the very obese patients of group III) and the protein-bound fraction of testosterone also decrease. As a result the concentration of free testosterone remains constant (120 pg/ml), except in the very obese (93 pg/ml). Because of the different affinity of the binding proteins for testosterone and oestradiol the ratio of free oestradiol: free testosterone shifts less strongly in favour of the feminizing hormone (11.1 × 10−3 in group III as compared to 4.1 × 10−3 in the controls), than is suggested by the total hormone concentrations. A disturbance in the gonadal function of the pituitary gland or the testes is not present, since the concentration of LH is normal and the testicular response to HCG in very obese patients adequate (increase of testosterone by a factor of 3.11 as compared to 2.23 in the controls). The cause of the decrease in testosterone and SHBG is unknown, while the increase of plasma oestrogens is likely to be due to the increased conversion of androgens to oestrogens in the adipose tissue, which clearly plays an important role for plasma concentrations of sex hormones in obese patients.

Introdution: Crataegus oxycanta (hawthorn) is used in herbal and homeopathic medicine as a cardiotonic.The present study was done to investigate the effect of the Crataegus oxycanta on antioxidant status in induced myocardial infarction in rat. Methods: In this experimental study, four groups of wistar rats (200-220g) each comprising 10 animals, were selected for this study. Group I, rats served as control. Group II rats were given isoperternol (85mg/kg body weight) subcutaneously on 15th and 16th days. Group III rats were given Crataegus oxycanta (100mg/kg/day), orally for 30 days.  Group IV rats were given Crataegus oxycanta (100mg/kg/day), orally for 30 days and isoperternol (85mg/kg body weight, subcutaneously) was given on 15th and 16th days. At the end of the experimental period, the rats were anaesthetized and blood obtained from the heart then rats were sacrificed and the hearts were removed for biochemical and histological analysis. The activity of malondialdehyde (MDA), catalase, superoxide dismutase (SOD), glutathione peroxidase (GPX) and total antioxidants was studied. Descriptive one-way analysis of variance (ANOVA) was used in different group. Significance was defined as P ≤ 0.05. Statistical analysis was performed using SPSS software version 16. Results: Crataegus significantly reduced plasma and heart tissue MDA levels (p<0.05) and significantly increased catalase, SOD, GPX and total antioxidant levels versus the group that received only isoperternol (p<0.05 ). Crataegus also decreased the rate of edema, inflammatory cell infiltration and heart tissue necrosis compared to the group that received only isoperternol. Conclusion: The study confirms the protective effect of Crataegus oxycanta against tissue damage and  oxidative stress caused by  isoperternol induced myocardial infarction

We used the isolated-perfused rat lung model to study the influence of pulmonary ventilation and surfactant instillation on the development of postreperfusion lung microvascular injury. We hypothesized that the state of lung inflation during ischemia contributes to the development of the injury during reperfusion. Pulmonary microvascular injury was assessed by continuously monitoring the wet lung weight and measuring the vessel wall125I-labeled albumin (125I-albumin) permeability-surface area product ( PS). Sprague-Dawley rats ( n = 24) were divided into one control group and five experimental groups ( n = 4 rats per group). Control lungs were continuously ventilated with 20% O2and perfused for 120 min. All lung preparations were ventilated with 20% O2before the ischemia period and during the reperfusion period. The various groups differed only in the ventilatory gas mixtures used during the flow cessation: group I, ventilated with 20% O2; group II, ventilated with 100% N2; group III, lungs remained collapsed and unventilated; group IV, same as group IIIbut pretreated with surfactant (4 ml/kg) instilled into the airway; and group V, same as group III but saline (4 ml/kg) was instilled into the airway. Control lungs remained isogravimetric with baseline125I-albumin PS value of 4.9 ± 0.3 × 10−3ml ⋅ min−1⋅ g wet lung wt−1. Lung wet weight in group III increased by 1.45 ± 0.35 g and albumin PSincreased to 17.7 ± 2.3 × 10−3, indicating development of vascular injury during the reperfusion period. Lung wet weight and albumin PS did not increase in groups I and II, indicating that ventilation by either 20% O2or 100% N2prevented vascular injury. Pretreatment of collapsed lungs with surfactant before cessation of flow also prevented the vascular injury, whereas pretreatment with saline vehicle had no effect. These results indicate that the state of lung inflation during ischemia (irrespective of gas mixture used) and supplementation of surfactant prevent reperfusion-induced lung microvascular injury.

The effect of environmental conditions on hatching indices of poultry eggs is sufficiently important that new methods for their improvement are sought, among others through exposing the eggs during hatching to an artificially generated magnetic field of variable frequency. Hatching eggs in this study came from Pharaoh quail in the 4th month of laying. The flock was kept under optimum microclimate conditions and fed a complete feed mix containing 21% total protein and 11.7 MJ ME. The eggs (n = 150), after weighing, were divided into 3 groups equal in respect of numbers: control (I) and two experimental (II and III). The eggs of group II and III were exposed to the action of variable magnetic field of the same intensity but different times of application. The highest percentage of dead embryos during incubation in relation to fertilised eggs was found in control group (11.36%), while the smallest was from group II (4.17%). The highest value of hatching indices calculated in relation to fertilised eggs was found in group II (91.6%), while the smallest was in group III (83.7%) with 86.3% in control group. The results point to the possibility of increasing egg hatchability indices through the use of additional variable magnetic field. The chicks hatched from eggs exposed to the action of this experimental agent had similar body weight. The average weight of one-day-old chicks ranged from 7.82 g (group II) to 8.05 g (group III). In the last week of rearing, mean body weight in both sexes was similar and ranged from 168 (group I) to 172 g (group III) in males, and from 186 g (group I) to 199 g (group III) in females; these differences were not statistically significant. The females of group III reached sexual maturity at 41 days, this being one and three days (non-significantly) earlier than birds in group I and II. &nbsp;

Background: Insecticides such as lambda-cyhalothrin(LCT), dibenzalacetone (DBA) and heptanone HD are teratogenic substances. Skeleton is one of the organs sensitive to teratogens during the period of organogenesis. Material and methods:This study was conducted on fifty adult female albino mice, divided randomly into four groups: Group I (control group, n= 5 received corn oil orally in adose of 10 mg /kg body weight), group II (LCT group, n=10 received LCT (nano 8%)10, 20 mg /kg body weight for each subgroup), group III (DBA group, n=20, received DBA (main) 10, 20 mg /kg body weight and DBA (nano 8%)) 10,20mg /kg body weight for each subgroup) and group IV (HD group, n=15, received HD (main) 10 mg /kg body weight and HD (nano 8%)) 10,50mg /kg body weight for each subgroup),The calculated treatment dissolved in 1 ml of corn oil by orogastric tube was given to the pregnant mice during the first week of pregnancy (1st, 3rd and 5th day).The delivered live and dead pups were examined externally, then double stained and prepared for skeletal examination by dissecting microscope. Results: All experimental groups showed various anomalies that can be categorized as the following: 1) presence of dead pups and resorption sites 2) macroscopic anomalies in the form of anencephaly, extended forelimb, cranioshisis, macroglossia, lowset ears, lage eye bulge, half body, amelia, internal rotation of forelimb and encephalocele. 3) skeletal anomalies by double staining of fetal skeleton showed incomplete ossification of nasal and interparietal bones, open arch of atlas, incomplete ossification of vertebrae, supernumerary ribs, wavy ribs, incomplete ossification of ribs and sternebrae, incomplete ossification of sternum, incomplete ossification of 5th metacarpal and terminal phalangal bones and incomplete ossification of tarsal , and phalangal bones. Conclusion: Lambda-cyhalothrin, dibenzalacetone and derivatives of heptanone result in numbers of congenital gross and skeletal abnormalities that indicate their teratogenic effect. Introduction: Taking into account the patho-immune mechanisms of formation of inflammatory process in periodontium, there is a necessity for in-depth study of the pathogenesis of periodontal diseases from the position of changes in the reactivity of the organism. The aim: Carrying out a comparative estimation of phagocytic activity of leukocytes of animals with inflammation in periodontium with altered reactivity. Materials and methods: Experiments were performed on 30 white rats: Group I - 10 white rats with hypoergic reaction; Group ІІ - 10 white rats with hyperergic reaction; Group III - 10 white rats with normergic reaction - control group. Slaughter and blood sampling under thiopental anesthesia was performed 7 days after the beginning of the experiment. The percentage of phagocytic leukocytes - phagocytic index, phagocytic number were determined and the index of phagocytic activity was calculated. Results: The phagocytic index (Fi,%) decreased by 2,09 times (p <0,05) at hypoergic, it increased by 1,37 times (p <0,05) at hyperergic; the index of phagocytic activity (IFA) decreased by 1.96 times at hypoergic (p <0.05); growth was 1.94 times (p <0.05) in the hyperergic group of animals. In both experimental groups, the phagocyte number (Fu) increased by 6.25% and 41.7%, with hypoergic and hyperergic, respectively. Conclusions: Different directions of changes of the phagocytic activity were observed: increase of these parameters at hyperergic and decrease at hypoergic. An increase in phagocyte number was likely to indicate some autonomy of the process of phagocytosis and independence from the reactivity of the organism.

Background: Cypermethrin is a well know agricultural pesticide used in the developing countries. It is associated with significant toxic potential on human health. Hence the present study was aimed to evaluate the protective role of Amomum subulatum against cypermethrin induced haematalogical changes in Wistar albino rats.Methods: The albino rats were divided into five different groups of six rats each. Group I considered as normal control, group II cypermethrin control (25mg/kg body weight p.o.), group III only test drug and group IV and V administered with cypermethrin 25mg/kg body weight along test drug 1.08 and 2.16mg/kg body weight for 28 consecutive days. At the end of 28th day blood was withdrawn and total haematalogical parameters were estimated.Results: In the cypermethrin control there was significant reduction in the WBC, Platelet, MCHC and considerable reduction in the haemoglobulin concentration in comparison to normal control. The test drug administered at both dose levels was significantly reversed the cypermethrin induced changes in haematalogical parameters.Conclusions: Authors can conclude that the Amomum subulatum has potency to reverse the cypermethrin induced haematalogical changes.

 Objective: The compound, 2,4-dichlorophenoxyacetic acid (2,4-D), is a systemic phenoxy herbicide and a plant growth regulator. Histopathological effects of 2,4-D alone and in combination with melatonin on the kidney in adult albino male mice of the Swiss strain were evaluated in the present study.Methods: Twenty adult male albino mice weighing between 30 and 40 g were assigned into four groups with five mice in each group. Group I served as control; Group II - melatonin alone (10 mg/kg body weight); Group III - 2,4-D alone (50 mg/kg body weight); and Group IV - 2,4-D + melatonin was given orally for 45 days.Results: It revealed a significant reduction in the body and kidney weights (p<0.001 and p<0.01 respectively) of mice treated with 2,4-D, while the body and kidney weights after treatment of melatonin and 2,4-D + melatonin were comparable to control. Histological study revealed that 2,4-D induced nephrotoxicity evidenced by various histological changes including shrinkage of the glomerulus, tubular dilation, hypertrophy of tubular epithelium, and degeneration of renal tubules.Conclusion: Treating animals with the melatonin along with 2,4-D showed mitigative effects of melatonin and minimized the structural alteration in the kidney of mice appeared due to 2,4-D toxicity.
