Riset dengan tujuan untuk mengevaluasi teknik pengendalian penyakit KHV pada ikan mas melalui penggunaan imunostimulan yaitu penambahan vitamin C pada pakan komersial dengan dosis yang berbeda telah dilakukan pada skala laboratorium. Perlakuan yang diterapkan adalah penambahan vitamin C jenis CFC-90 (microencapsulated vitamin C) pada pakan komersial dengan berbagai dosis yaitu: (A) tanpa vitamin C, sebagai kontrol, (B) vitamin C sebanyak 250 mg/kg pakan, (C) vitamin C sebanyak 500 mg/kg pakan, (D) vitamin C sebanyak 750 mg/kg pakan, dan (E) vitamin C sebanyak 1.000 mg/kg pakan. Masing-masing perlakuan dilakukan pengulangan sebanyak 3 (tiga) kali. Pada hari ke-15, seluruh kelompok perlakuan diinfeksi KHV secara buatan dengan teknik kohabitasi. Pengamatan dilakukan terhadap tingkah laku, gejala klinis, dan mortalitas ikan uji yang dilakukan setiap hari hingga akhir percobaan. Rataan persen sintasan ikan uji tertinggi pada akhir riset diperoleh pada penambahan vitamin C sebanyak 750 mg/kg pakan yaitu sebesar 82,22%; diikuti dengan penambahan vitamin C 250 mg/kg pakan sebesar 70,00%; vitamin C 1.000 mg/kg pakan sebesar 61,11%; vitamin C 500 mg/kg pakan sebesar 58,89%; sedangkan rataan sintasan terendah diperoleh pada kelompok kontrol yaitu sebesar 27,78%.Research with the aim to evaluate the technique of KHV disease control carp through the use of immunostimulatory namely the addition of vitamin C in commercial diet with different doses have been performed in a laboratorial scale. The treatment applied was the addition of vitamin C of CFC-90 (microencapsulated vitamin C) in the commercial diet with different doses, namely: (A) without vitamin C, as control, (B) Vitamin C 250 mg/kg feed, (C) vitamin C of 500 mg/kg feed, (D) Vitamin C 750 mg/kg feed, and (E) of vitamin C as much as 1,000 mg/kg feed. Each treatment was repeated three times. On day 15th, all treatment groups were artificially infected with KHV using cohabitation techniques. Observations conducted to monitor behavior, clinical symptoms and mortality of fish tests were done every day until the end of the experiment. The highest mean percent of survival rate was recorded in treatment D by adding vitamin C as much as 750 mg/kg of feed that was equal to 82.22%, followed by the addition of vitamin C 250 mg/kg diets at 70.00% (treatment B), Vitamin C 1,000 mg/kg feed amounted to 61.11% (treatment E), vitamin C 500 mg/kg diets at 58.89% (treatment C), while the lowest average of survival rate was obtained in the control group that was equal to 27.78%.

A safety study on ketoprofen 10% was carried out on pigs using a different dosing and treatment scheme. Forty healthy crossbreed pigs with similar age, weight, and body condition score were distributed into five treatment groups. The pigs were intramuscularly injected once with different doses of ketoprofen: 3 mg/kg (group 1X), 6 mg/kg (group 2X), 9 mg/kg (group 3X). In addition, the 3 mg/kg dosis was administered on three consecutive days (group 1X ext.). Intramuscular injections of saline solution were used in control group (CTL). The pigs were clinically examined throughout the trial and blood samples were taken for hematological and biochemical evaluation on days −4 (before treatment), +3, +7, and +14 (the end of the trial). Any unusual behaviour or clinical signs were reported as potential toxic effects of ketoprofen. Serum measurements showed that none of the ketoprofen doses produced changes in renal or hepatic biochemical parameters, liver enzymes, or total bilirubin. Likewise, hematological assessment indicated no altered parameters or hematocrit percentage in the study groups. These results demonstrate that ketoprofen has no adverse effects in pigs when the doses and scheme evaluated in this study are applied.

Chronic heart failure (CHF) is a complex clinical syndrome caused by a variety of heart problems, with a high incidence. The 5-year survival rate of patients with clinical symptoms is similar to that of malignant tumors. Wenyang Zhenshuai granules are a safe and effective granule of traditional Chinese medicine components, including aconite, dried ginger, licorice, and red ginseng. In contemporary clinical applications, it is widely used in acute and chronic heart insufficiency, coronary heart disease, and arrhythmia. This research cultured H9C2 cardiomyocytes and divided them into the normal control group, LncRNA-MiR143HG overexpression group, LncRNA-MiR143HG silence group, Adriamycin (ADR) group, ADR + medicated serum group, ADR + LncRNA-MiR143HG overexpression + medicated serogroup, and ADR + LncRNA-MiR143HG silence + medicated serogroup. The cells of each group were treated differently, and the survival rate of each group of cells and the expression levels of LncRNA-MiR143HG/miR-143 and ERK5 were detected at the end of the experiment, and the expression of LncRNA-MiR143HG/miR-143 in H9C2 cardiomyocytes was regulated by Wenyang Zhenshuai granules’ impact. The results of this study showed that, in the doxorubicin-induced H9C2 cardiomyocyte injury model, the expression of miR-143 was upregulated, and the expression of LncRNA-MiR143HG and ERK5 was significantly downregulated. Wenyang Zhenshuai granules can downregulate the expression of miR-143 to promote ERK5 protein expression and phosphorylation. The process is regulated by LncRNA-MiR143HG/miR-143, which may be one of its important mechanisms for the treatment of chronic heart failure.

<p>Prevention of koi herpesvirus (KHV) infection using chemicals or medicines was ineffective way.  In this study garlic extract was used to prevent KHV infection.  Virus suspension of 0.1 ml and garlic extract of 0.1 ml in different dosage, i.e., 100, 200 and 300 ppt, was injected into common carp body.  Hemoglobin concentration, red and white blood cell numbers, and leukocyte number were counted.  The results of study showed that administration of 300 ppt of garlic extract could produce higher survival rate (67.5%), good blood parameters and clinical symptoms compared to other treatments.</p> <p>Keywords: garlic, KHV, common carp</p> <p> </p> <p>ABSTRAK</p> <p>Upaya penanggulangan wabah Koi Herpesvirus (KHV) menggunakan bahan-bahan kimia atau obat-obatan adalah tidak efektif. Pada penelitian ini dilakukan pemberian ekstrak bawang putih untuk menanggulangi infeksi KHV.  Suspensi virus sebanyak 0,1 ml ditambahkan dengan 0,1 ml ekstrak bawang putih dengan berbagai konsentrasi, yaitu 100, 200 dan 300 ppt, disuntikkan ke dalam tubuh ikan mas.  Kadar hemoglobin, jumlah sel darah merah dan sel darah putih jenis dan jumlah leukosit diamati. Hasil penelitian menunjukkan bahwa pemberian ekstrak bawang putih sebanyak 300 ppt menghasilkan kelangsungan hidup (67,5%) yang lebih baik dibandingkan dengan perlakuan lainnya, dan begitu pula dengan gambaran darah serta gejala klinisnya.  </p> <p>Kata kunci: bawang putih, KHV, ikan mas</p>

Although cisplatin is a potent anticancer drug, it instigates oxidative and pro-inflammatory reactions that pose significant and distressing clinical symptoms. Therefore, this study investigated the effects of vitamin C and (or) l-carnitine on cisplatin-induced gastric mucosa damage in rat. The rats were allocated into 6 groups (n = 5). The control group received distilled water, while the treatment groups received cisplatin alone (CIP), or cisplatin with vitamin C, l-carnitine, or their combination. Cisplatin caused disruption of the gastric mucosa histoarchitecture and altered the mucus barrier function. Moreover, the stomach tissue of the CIP-treated group showed increased levels of oxidative stress markers (malondialdehyde and H2O2) and decreased activities of antioxidant (superoxide dismutase, glutathione peroxidase, catalase, glutathione S-transferase) and non-antioxidant (reduced glutathione) enzymes. These deleterious events were accompanied with significant increases in pro-inflammatory cytokines and inflammatory infiltration markers, myeloperoxidase and inducible nitric oxide synthase. However, the administration of both vitamin C and l-carnitine, and not either of the two showed additive effects in attenuating the adverse effects of cisplatin. The histological results agreed with the biochemical assays. The study concluded that the combined administration of vitamin C and l-carnitine, but not the single therapy, could prevent the adverse effects of cisplatin on gastric tissue.

Elevated ambient temperature has been known to be deleterious to the developmental competence of mammalian oocytes and embryos, although the mechanism is still unclear. The objective of this study was to determine the effect of heat shock (HS) on the alteration of intracellular calcium concentrations ([Ca2+]i) of matured pig oocytes by two different calcium releasing agents. Porcine cumulus–oocyte complexes were aspirated from the follicles (3–6 mm) and subjected to standard in vitro maturation procedure for 42 h. Matured oocytes were then randomly allocated to different heat treatments at 41.5°C for 0 (Control, C0h), 1 (HS1h), 2 (HS2h), or 4 h (HS4h). An additional control group was cultured for 4 h without heat shock (C4h). Oocytes were incubated with 2 µM fura-2 acetoxymethyl ester (AM) and 0.02% pluronic F-127 in Ca2+-free PBS (40 min) following heat shock, and then washed with Ca2+-free PBS (30 min) for detection of [Ca2+]i. Fluorescent images were captured with alternative excitation wavelengths at 340/380 nm by a rotating chopper disk equipped with an Axon imaging system. Data from both experiments were analyzed by ANOVA using the General Linear Model (GLM) of the SAS (SAS Institute, Inc., Cary, NC, USA). In Experiment 1, matured oocytes were activated by 200 mM thimerosal (10 min) following heat treatment. The maximal [Ca2+]i in the HS2h group was the highest among all treatment groups. The lowest maximal peak of [Ca2+]i was observed in the HS4h group, but it was still higher than that in the C4h group (P < 0.05). The total amount of Ca2+ release represented by the total area of the peaks in C4h was lower than in any other groups except HS4h (P < 0.05). In Experiment 2, each matured oocyte was injected with approximately 10 pL of inositol 1,4,5-triphosphate (IP3, 0.5 mM); the Ca2+ transient was recorded as described in the previous experiment. The maximal value of [Ca2+]i in the C4h group was still the lowest among the heat-shocked and C0h groups (P < 0.05). The total Ca2+ release in the HS2h group was the highest among all treatment groups, but only significantly higher than the HS1h and C4h groups (P < 0.05). A similar pattern of Ca2+ release in HS-oocytes was induced by thimerosal and IP3 stimulations. These results indicate that Ca2+ releasing capacity of matured pig oocytes is enhanced by a shorter duration of heat shock, but declines after prolonged exposure of heat shock and/or in vitro culture. The differential Ca2+ releasing capacity of heat-shocked oocytes prior to fertilization revealed physiological changes of pig oocytes after heat shock. This finding provides further insight for the low fertilization and developmental competence that occurs in farm species during hot seasons.

This clinical study aimed to clinically and radiographically compare the implant survival rate and peri-implant tissue response between immediate and delayed loading protocols for unsplinted implant retained mandibular overdentures. Twenty patients were enrolled to participate in this study. Each subject was randomly assigned to 1 of 2 treatment groups: test group patients' implants (n = 10), which were immediately loaded, and control group patients' implants (n = 10), which were conventionally loaded. Locator abutments were torqued to 15 Ncm at delivery. Attachments were picked up intraorally immediately after implant placement for the test group and at 3 months for the control group, and 3-lb retention inserts were placed. Marginal bone levels based on cone beam computed tomography fixed reference points were recorded at baseline and 12 months. Modified plaque index, gingival index, and implant stability quotients were recorded at baseline, 3 months, and 12 months. After 12 months, implant survival rate was 100% in both groups. Marginal bone levels, keratinized mucosa, modified plaque index, and gingival index were significantly different among the groups at 3- and 12-month intervals, whereas no significant differences were found in implant stability quotients between the groups. The fact that implant survival rate was 100% in both treatment groups suggests that, within the limitations of this study, immediate loading protocol for unsplinted implant retained mandibular complete overdenture is as predictable, safe, and successful as the delayed loading protocol. Implementing the immediate loading protocol for mandibular implant retained overdentures could shorten treatment time, which could lead to better patient's satisfaction.

ABSTRACTRifampin monotherapy was compared to the combination of linezolid or vancomycin with rifampin in an experimental rat model of methicillin-resistantStaphylococcus aureus(MRSA) chronic foreign body osteomyelitis. MRSA was inoculated into the proximal tibia, and a titanium wire was implanted. Four weeks after infection, rats were treated intraperitoneally for 21 days with rifampin alone (n= 16), linezolid plus rifampin (n= 14), or vancomycin plus rifampin (n= 13). Thirteen animals received no treatment. At completion of treatment, qualitative cultures of the wire and quantitative cultures of the bone (reported as median values) were performed. Quantitative cultures from the control, rifampin monotherapy, linezolid-plus-rifampin, and vancomycin-plus-rifampin groups revealed 4.54, 0.71, 0.10, and 0.50 log10CFU/gram of bone, respectively. The bacterial load was significantly reduced in all treatment groups compared to that in the control group. Rifampin resistance was detected in isolates from 10, 2, and 1 animal in the rifampin, linezolid-plus-rifampin, and vancomycin-plus-rifampin groups, respectively. Cultures of the removed wire revealed bacterial growth in 1 and 2 animals in the rifampin and linezolid-plus-rifampin groups, respectively, with no growth in the vancomycin-plus-rifampin group and growth from all wires in the untreated group. In conclusion, we demonstrated that combination treatment with linezolid plus rifampin or vancomycin plus rifampin is effective in an animal model of MRSA foreign body osteomyelitis in the context of retention of the infected foreign body.

The aim of this study, the effect of addition to salep at different concentrations (2.5, 5, 7.5 and 10%) into sucuk on physicochemical, textural and sensorial properties were investigated at d fermentation and storage periods (5., 10., 20. ve 30. days). The results indicated that TBARS values gradually increased during fermentation and storage periods in all treatment groups. However, addition of 10% salep decreased TBARS values compared to control group at the end of fermentation and storage period. pH values of sucuks were decreased with addition of salep. Higher L* values were observed in sucuk with 10% salep at the end of the fermentation and storage period. Increasing levels of salep in sucuk formulation caused an increase in hardness and ash values, a decrease in moisture values in sucuk. The results of this study indicated that salep may be used in sucuk manufacture by the meat industry to improve shelf-life and quality parameters of the product.

The research with the aim to know the optimal feeding frequency of supplemented ascorbic acid (microencapsulated vitamin C CFC-90) on the dose of 750 mg/kg feed to control Koi Herpesvirus (KHV) disease infecting common carp has been done in field condition. Fish were reared in floating cages with the size of 3.5 m x 3.5 m x 2.0 m and stocking density of 1,250 fish/cage with the size range of ± 10 g/fish. The treatments applied in the research were: (A) daily application, (B) every 3 days application, and (C) without vitamin C addition as the control. Fish test were challenged to KHV infection on the mid cultivation by cohabitation method in the laboratory scale for 2 weeks. Examination on behavior, clinical sign, and mortality of fish test conducted daily. The results showed that the highest survival rate was found on the application of vitamin C every 3 days (60.16%); and followed by every day (52.00%), and the lowest was found on the control group (47.36%).

Background The tiger frog (Hoplobatrachus rugulosus) is widely raised by many farms in southern region of China as an economically edible frog. The growth, development, and sexual differentiation of amphibians are influenced by temperature and steroid hormone level. However, the problem of hormone residues is caused by the addition of exogenous hormones in frog breeding, it is worth considering whether non-sterol aromatase inhibitors can be used instead of hormones. Methods In our study, H. rugulosus tadpoles were subjected to two water temperatures (29 °C and 34 °C) and three letrozole concentrations in the feed (0, 0.1 and 1 mg/g) to examine the effects of temperature, aromatase inhibitor and their interaction on metamorphosis, locomotion, and sex ratios. A G-test and contingency table were used to analyze the metamorphosis rate of tadpoles and the survival rate of froglets after feeding for 90 days. A G-test was also used to analyze sex ratios in different treatment groups. Results Metamorphosis time and body size (snout–vent length, body mass and condition factor) were significantly different between the two temperature treatments. Metamorphosis time was longer and body size was increased at 29 °C compared to those at 34 °C. Letrozole concentration and the temperature × letrozole interaction did not affect these variables. The jumping distance of froglets following metamorphosis was positively associated with the condition factor; when controlling for condition factor, jumping distance was not affected by temperature, letrozole concentration and their interaction. Temperature and letrozole concentration also did not affect metamorphosis and survival rate. Sex ratio of the control group (0 mg/g letrozole) was 1:1 at 29 °C, but there were more males at 34 °C. The sex ratios of H. rugulosus treated with letrozole at 29 °C and 34 °C were significantly biased toward males, and male ratio increased as letrozole concentration increased. Furthermore, more males were produced at 34 °C than at 29 °C at each letrozole concentration.
