Vancomycin (VCM) is indicated in combat against Gram-positive infections, but it is not considered a first-choice drug because of its adverse effects. It is believed that oxidative stress is the primary mechanism of endothelial injury and the consequent VCM toxicity, which varies from phlebitis to nephrotoxicity. Moreover, dose recommendations, dilution, rates and types of infusion are still controversial. The aim of this study was to determine the effect of different VCM dilutions in endothelial, liver and kidney injuries by biochemical parameters and histopathological analysis. Wistar rats were randomly divided into six groups and subjected to femoral vein cannulation for drug administration. Control groups received 0.9 ml of saline and the others received VCM (10mg/Kg/day) at dilutions of 5.0 and 10.0 mg/mL for 3 and 7 days. Homocysteine, hs-CRP, AST, ALT, GGT, urea, creatinine, lycopene, alpha-tocopherol, beta-carotene and retinol were analyzed. Kidney, liver and cannulated femoral vein fragments were collected.This study showed alterations in ALT which featured hepatotoxicity. However, drug dilutions were not able to show changes in other biochemical parameters. In contrast, kidney and endothelium pathological changes were observed. More studies are needed to characterize VCM induced kidney and endothelium toxicity and biochemical markers able to show such morphological modifications.

Rudgea viburnoides leaves are widely used in popular Brazilian medicine as a diuretic, antirheumatic, hypotensive and blood depurative tea. The present study was carried out to investigate the effects of this infusion on the liver and on the plasma biochemical parameters of Wistar rats. Two groups received the R. viburnoides leaf infusion at a daily dose of 10 or 20g dry-leaves/L water, during 40 days. The histopathological analysis did not show degenerated areas or infiltration of leucocytes. Hepatic morphometry showed accumulation of fat in the hepatocytes of the treated groups. There was no significant change in the plasma levels of urea, creatinin, uric acid, direct bilirubin, cholesterol, total proteins, albumin, gamma glutamyl tranferase (gamma-GT), alanine transaminase (ALT), aspartate transaminase (AST), chlorine, phosphate and calcium. A significant reduction in the plasma levels of triacylglycerol (TAG) occurred in the group that received the higher dose.

Aim This study is the second report from a single experiment planned to determine if acupuncture at LI4 and SP6, or at the sacral points, could produce any harm in the pregnancy outcome of Wistar rats, as has been hypothesised in traditional knowledge. Here, we observe if these points can be harmful to the health of pregnant Wistar rats. Methods A total of 48 pregnant Wistar rats were randomly divided into 4 groups: total control, where rats were left in cages without manipulation; anesthetised control, where rats were manipulated and anesthetised but did not receive electroacupuncture; and peripheral point and sacral point groups, where rats were anesthetised and received 6 sessions of electroacupuncture at 4 acupuncture points: LI4 and SP6, and BL27 and BL28, respectively. In the 19th day of pregnancy the rats were killed and examined. The primary endpoints were levels of glucose, aspartate aminotransferase (AST), alanine aminotransferase (ALT) creatinine and histological changes of liver and kidney. Results There were no differences between the levels of biochemical parameters. No differences were also seen regarding the histopathological analysis. Conclusions We found no evidence that acupuncture at the LI4, SP6 and sacral points could be harmful to the Wistar rat dams.

Ricinodendron rautanenii is a plant, used in traditional medecine to treat fever, eczema, back pain, cancer and stomacal disorders. This work was carried out to evaluate the safety of the aqueous extract from the stem bark of Ricinodendron rautanenii by determining its potential toxicity after acute and subacute administration in Wistar rats. In Acute toxicity assay, the animals received the extract at the single dose of 5000 mg/Kg and were observed during 48h for mortality and any toxicity manifestations. General behavior, adverse effects and mortality were determined for up to 14 days post treatment. In subacute toxicity assay, extract was given orally to rats at doses of 6, 20, 40 and 80 mg/kg/day for 28 days respectively. Animal body weight, water and food intake, biochemical and hematological parameters were determined. Liver and kidney were examined histologically for any signs of organ damage. No behavioral changes or mortality were recorded in the treated groups. No significant hematological changes were observed in the both sex. The biochemical analysis indicated a significant increase of high density lipoprotein levels in both male and female rats. Histopathological analysis of the liver and kidney did not show any observable cellular damages. The overall finding of this study suggest that, extract of Ricinodendron rautanenii did not cause any death up to a dose of 5000 mg/kg and can be considered non-toxic. Biochemical and histological studies of the extract did not revealed major signs in subacute toxicity.

Abstract The current research was performed to evaluate the potential protective effect of Lactobacillus paracasei ssp paracasei, Pediococcus acidi-lactis and Lactoccus lactis ssp lactis, Sylimarin in the alleviation of health (hepatic and renal) complications caused by carbon tetrachloride (CCl4) in rats. Healthy sixty albino rats were divided into six groups, the first group was control (negative), the second group (control positive) was injected CCl4 (1 ml/kg, 1:1 v/v paraffin oil mixture, i.p. every third day for 8 weeks, The third group (CCl4 + silymarin group) receiving both CCl4 and daily silymarin therapy (50 mg/kg, oral), the fourth group: CCl4+ (Lactobacillus paracasei 1ml orally). The fifth group (CCl4+ Pediococcus acidilactis 1ml orally) and the six group (CCl4+ Lactococcus lactis (1ml orally) for eight weeks per day. Biochemical markers were tested for blood, liver and kidney tissue. Histopathological tests on liver and kidney tissues were performed. The findings obtained have shown that Lactobacillus paracasei ssp paracasei, Pediococcus acidilactis and Lactoccus lactis ssp lactis improved the disrupted biochemical parameters caused by CCl4 therapy. Besides, the findings of the histopathological analysis are in consistent with biochemical parameters and the protective ability of lactic acid bacteria suggesting that the best lactic acid bacteria was Pediococcus acidilactis that helped strengthen liver fibrosis caused by CCl4 therapy, while the best bacterium for improving renal damage was Lactoccus lactis.

The biochemical assessment in animals is of capital interest to define the diagnosis of many diseases. The variation of biochemical parameters (urea, glycemia, creatinemia, transaminases: GPT and GOT, total cholesterol, HDL-c, LDL-c, triglycerides and total lipids) is an indicator of the state of health of certain vital organs such as the kidney, liver, pancreas, veins, heart, etc... . For this study, to be able to determine the normal values of the rats, a set of batches of male and female Wistar rats was formed and acclimatized for 2 months. Each batch set consisted of thirty-nine (39) rats. Each rat was weighed and then sampled by venipuncture at the caudal level to collect their blood in a dry tube (red). The red tube was used to determine the biochemical parameters. The results of this study showed that some animals already had certain pathologies before the use of any beneficial or harmful substance on them. However, most of the animals used have normal biochemical parameters revealing the good state of their various vital functions (kidney, liver, heart, veins, pancreas, etc.). These values of determining standard biochemical parameters can form a basis for future tests.

Background: Paracetamol toxicity is currently the single most important cause for acute liver failure and is associated with significant number of deaths. Nilitanduliyadi leha is one among the formulation explained in the context of Vishahara yogas (anti-poisonous formulations) in the text Vishavaidya Jyotsnika. Objectives: To experimentally evaluate the hepatoprotective activity of nilitanduliyadi leha in paracetamol induced hepatotoxicity in Wistar albino rats. Methods: Albino Wistar rats of either sex weighing 200 – 250, g were selected and divided into four groups of six animals in each group (n = 6). Treatment was given for 7 days. Blood was drawn and sent for tests and important organs like liver and kidney were dissected out, cleaned to remove extraneous tissues, blotted to remove blood stain and weighed. A piece of liver tissue was preserved in 10% formalin for histopathological processing. Results: The formulation has helped in balancing the biochemical parameters studied almost as efficiently as the standard drug. In the antioxidant study also, the drug has given good results and shows even slightly more effective than the standard drug. The histopathology study also reveals mild protection and regeneration of tissues by the effect of test drug. Conclusion: This present study proves that the formulation is having a comparable hepato protective activity with that of silymarin.

Background: Funcionalized multi-walled carbon nanotubes (ox-MWCNTs) were used for the preparation of therapeutic nanoparticles for delivery of some bioactive compounds. Consequently, this work deals with the preparation of grafted MWCNTs with n-vinyl caprolactam in the presence of pomegranate peel extract (P. granatum), titanium dioxide (TiO2) and/or silver nanoparticeles and their toxic effects on male mice using in vivo biological examination (liver and kidney dysfunction biomarkers) and the histopathological analysis. Methods: P. granatum extract was immobilized onto functionalized MWCNTs using simple adsorption technique. Moreover, The prepared materials were analyzed by Fourier transform infrared spectroscopy (FTIR), scanning electron microscope (SEM). In vivo examination using liver and kidney dysfunction biomarkers was investigated. In addition, the histopathological study was carried out. Results: The ox-MWCNTs induced significant elevation in the liver enzymes including AST, ALT and ALP relative to the control group. While, the treatment with P. granatum extract only did not induce any change in the liver and kidney biomarkers. In other words, P. granatum extract loaded onto functionalized MWCNTs showed low effects on liver enzymes and kidney function biomarkers in the treated mice in comparison with ox-MWCNTs and extract separately. Moreover, histopathological analysis revealed that the P. granatum extract functionalized MWCNTs exhibited normal renal tissue with no histopathological alteration. Conclusion: The grafted MWCNTs with n-vinyl caprolactam in the presence of pomegranate peel extract (P. granatum), titanium dioxide (TiO2) and/or silver nanoparticeles were successfully prepared. SEM-micrographs showed complete coating of MWCNTs fiber with the extract. The prepared materials resulted in no toxic effects and the histopathological findings were confirmed by inflammation of the liver and kidney tissues.

Background: Insulin, like most peptides, is classified as a hydrophilic and macromolecular drug that is considered as a low permeable and unstable compound in the gastrointestinal (GI) tract. The acidic condition of the stomach can degrade insulin molecules. Moreover, the presence of proteolytic activities of some enzymes such as trypsin and chymotrypsin can hydrolyze amide-bonds between various amino-acids in the structures of peptides and proteins. However, due to its simplicity and high patient compliance, oral administration is the most preferred route of systemic drug delivery, and for the development of an oral delivery system, some obstacles in oral administration of peptides and proteins including low permeability and low stability of the proteins in GI should be overcome. Objective: In this study, the effects of orally insulin nanoparticles (INPs) prepared from quaternerized N-aryl derivatives of chitosan on the biochemical factors of the liver in diabetic rats were studied. Methods: INPs composed of methylated (amino benzyl) chitosan were prepared by the PEC method. Lyophilized INPs were filled in pre-clinical capsules, and the capsules were enteric-coated with Eudragit L100. Twenty Male Wistar rats were randomly divided into four groups: group1: normal control rats, group 2: diabetic rats, group 3: diabetic rats received capsules INPs(30 U/kg/day, orally), group 4: the diabetic rats received regular insulin (5 U/kg/day, subcutaneously). At the end of the treatment, serum, liver and kidney tissues were collected. Biochemical parameters in serum were measured using spectrophotometric methods. Also, oxidative stress was measured in plasma, liver and kidney. Histological studies were performed using H and E staining . Results: Biochemical parameters, and liver and kidney injury markers in serum of the diabetic rats that received INPs improved significantly compared with the diabetic group. INPs reduced oxidative toxic stress biomarkers in serum, liver and kidney of the diabetic treated group. Furthermore, a histopathological change was developed in the treated groups. Conclusion: Capsulated INPs can prevent diabetic liver and oxidative kidney damages (similar regular insulin). Therefore oral administration of INPs appears to be safe. Lay Summary: Although oral route is the most preferred route of administration, but oral delivery of peptides and proteins is still a challenging issue. Diabetes Mellitus may lead to severe complications, which most of them are life-threatening. In this study, we are testing the toxicity of oral insulin nanoparticles in kidney and liver of rats. For this investigation, we will prepare insulin nanoparticles composed of a quaternized derivative of chitosan. The nanoparticles will be administered orally to rats and the level of oxidative stress in their liver and kidney will be determined. The data will be compared to the subcutaneous injection of insulin.

Abstract Background A novel goose-origin astrovirus (GoAstV) has broken out across China in recent years, causing gout in goslings with a mortality rate of around 50%. However, our understanding of the dynamic distribution, tissue tropism and pathogenesis of GoAstV is incomplete. In order to assess its pathogenicity, one-day-old goslings were inoculated separately with GoAstV via oral and subcutaneous injection routes. Results Clinical symptoms, gross and microscopic lesions, blood biochemical parameters and viral loads were detected and recorded for 20 days after infection. Typical gout was observed in experimental goslings. GoAstV can be replicated in tissues and cause pathological damage, especially in the kidney, liver, heart and spleen. Virus-specific genomic RNA was detected in blood, cloacal swabs and all representative tissues, and virus shedding was detected up to 20 days after inoculation, suggesting that GoAstV has a wide tissue tropism and spread systematically after inoculation. The viral copy numbers examined in kidney were the highest, followed by spleen and liver. Conclusion This experiment determined the accurate value of viral loads and biochemical indicators of GoAstV-induced goslings. These findings increase our understanding of the pathogenicity of GoAstV in goslings and provide more reference for future research.
