Influence of parasitism on plasma concentrations of growth hormone, somatomedin-C and somatomedin-binding proteins in calves

ABSTRACT A parasitic disease model (sarcocystosis) was used to study the effects of infection and associated plane of nutrition on GH and somatomedin-C (SM-C) patterns in plasma, and SM-C binding protein patterns in plasma from 4-month-old male Holstein calves. Calves, matched by age and rate of growth before the experiment, were divided into three treatment groups (n = 7). In the first (control), animals were uninfected and food was available ad libitum; in the second, animals were infected with Sarcocystis cruzi and food was available ad libitum. The third group consisted of uninfected animals pair-fed to the level of feed intake of the infected animals. Blood samples were obtained at various times after infection for analysis of the secretory patterns of GH (day 27 after infection, samples every 10 min for 6 h), SM-C (days 27, 35 and 58 after infection) or binding protein (day 42 after infection). Samples were analysed for GH and SM-C by radioimmunoassay. Relative molecular weights of binding proteins were assessed by elution patterns from gel permeation columns. Clinical signs of infection were manifest abruptly on day 26 after infection. Voluntary feed intakes of infected calves as a per cent of control calves were 18, 46 and 78 on days 27, 35 and 58 after infection respectively. Plasma GH concentrations were lower in infected and pair-fed than in control calves (P < 0·05). Plasma SM-C concentrations were reduced in calves with diminished feed intakes and lower still in infected calves (P < 0·05). Plasma SM-C was positively correlated with nitrogen retention across treatment groups (r = 0·81). Two classes of binding proteins differing in molecular weight were identified. The relative amounts of each binding protein in plasma were reduced during low feed intake with some differences in the endogenous saturation affected by infection. These data suggest that altered growth and metabolism in parasitized calves may arise in part from both nutritional and infection-mediated effects on the regulation of GH, SM-C and SM-C binding proteins. Improving the productivity and profitability of smallholder cattle enterprises in Indonesia requires greater and more efficient utilisation of underutilised feed resources such as rice straw. The experiment tested the hypothesis that an Ongole cow with low energy requirements can maintain weight (W) on a rice straw-based diet with the addition of a small amount of tree legumes. Thirty-two Ongole cross (Bos indicus) cows were allocated to one of four treatments in a randomised block design with eight cows per treatment. Cows were offered untreated rice straw ad libitum with four levels of tree legumes (0, 11, 21, and 42 g DM/kg W0.75.day) for 20 weeks. Feed intake was determined daily and liveweight was measured every second week. There was no difference in total feed intake between the treatment groups (P > 0.05). Intake of tree legumes was higher when more was offered (P < 0.05), but cows did not consume all of the legumes offered to them. The inclusion of tree legumes in the diet had no effect on organic matter digestibility, ME content of the diet, liveweight gain or estimated energy balance of the cows (P > 0.05). Rice straw alone contained insufficient ME and rumen-degradable N to meet the maintenance requirements of the cows. From the regression relating liveweight change and ME intake for all cows across all diets, the inclusion of tree legumes in the diet at ~12 g DM/kg W0.75.day or 2.8 g DM/kg W.day was enough to meet the energy requirements for maintenance of Ongole cows fed rice straw ad libitum.

A specific radioimmunoassay procedure was developed to monitor the plasma concentrations of thiamin-binding protein, a minor yolk constituent of the chicken egg. By using this sensitive assay, the kinetics of oestrogen-induced elaboration of this specific protein in immature chicks was investigated. After a single injection of the steroid hormone, with an initial lag period of 4–5h the thiamin-binding protein rapidly accumulated in the plasma, attaining peak concentrations around 75h and declining thereafter. A 4-fold amplification of the response was noticed during the secondary stimulation, and this increased to 9-fold during the tertiary stimulation with the steroid hormone. The magnitude of the response was dependent on the hormone dose, and the initial latent period and the duration of the ascending phase of induction were unchanged for the hormonal doses tested during both the primary and secondary stimulations. The circulatory half-life of the protein was 6h as calculated from the measurement of the rate of disappearance of the exogenously administered 125I-labelled protein. Simultaneous administration of progesterone, dihydrotestosterone or corticosterone did not alter the pattern of induction. On the other hand, hyperthyroidism markedly decreased the oestrogenic response, whereas propylthiouracil-induced hypothyroidism had the opposite effect. The anti-oestrogen E- and Z-clomiphene citrates, administered 30min before oestrogen, effectively blocked the hormonal induction. α-Amanitin and cycloheximide administered along with or shortly after the sex steroid severely curtailed the protein elaboration. A comparison of the kinetics of induction of thiamin- and riboflavin-binding proteins by oestrogen revealed that, beneath an apparent similarity, a clear-cut difference exists between the two vitamin-binding proteins, particularly with regard to hormonal dose-dependent sensitivity of induction and the half-life in circulation. The steroid-mediated elaboration of the two yolk proteins thus appears to be not strictly co-ordinated, despite several common regulatory features underlying their induction.

Ardacin is an antibacterial glycopeptide effective against gram positive bacteria. Research elsewhere has shown that in growing calves, it increased the efficiency of microbial protein synthesis and protein flow to the duodenum. In goats it reduced methane production but did not affect the site or extent of organic matter digestion. Results on its effects on feed intake are equivocal but it has been shown to increase liveweight gain in finishing cattle. The objective of the present study was to ascertain the effects of Ardacin on feed intake and growth rate of growing and finishing steers.Two experiments were carried out simultaneously using the same feeds and Ardacin dose levels. In Experiment 1, 72 Friesian and Charolais x Friesian growing steers (8 months old and 240 kg initial weight) were blocked to 4 treatment groups, tied in individual stalls, and offered grass silage ad libitum plus 1 kg ground barley containing Ardacin per head daily.

Male Hartley guinea pigs and male rhesus macaques were used to determine an efficacious dose of 1,1′-methylenebis{4-[(hydroxyimino)methyl] pyridinium} dimethanesulfonate (MMB4 DMS) that would result in 80% survival, 24 hours following a single exposure to cyclosarin (GF). The pharmacokinetic/pharmacodynamic relationship between acetylcholinesterase activity and MMB4 plasma concentrations relative to survival was evaluated. Guinea pigs and non-human primates (NHPs) were concurrently administered MMB4 DMS (guinea pigs: 0, 10, 30, or 40 mg/kg, intramuscular [IM] and NHPs: 0.1, 1, 5, 10, or 20 mg/kg, IM), atropine, and diazepam following a 3 × median lethal dose (LD50) GF challenge. Clinical observations were evaluated using a quality-of-life (QOL) scoring system. All GF-exposed animals exhibited typical signs of nerve agent poisoning immediately following challenge. In guinea pigs, 24-hour survival was 0%, 50%, 90%, and 90% for 0, 10, 30, and 40 mg/kg MMB4 DMS groups, respectively. In addition, nearly all animals surviving to 24 hours were clinically normal, with many in the 30 and 40 mg/kg MMB4 DMS dose group observed as normal by 4 hours post-challenge. In NHPs, survival was 100% for all treatment groups, with all animals noted as clinically normal by 48 hours. Following treatment with atropine/MMB4 DMS/diazepam, NHPs exhibited dose- and temporal-related decreases in incidence and duration of the clinical signs of toxicity. The QOL scores improved with increasing MMB4 DMS dose in both species. The estimated ED80s were 25.5 mg/kg MMB4 DMS (human equivalent dose [HED] of 5.5 mg/kg) and ≤0.1 mg/kg (HED of 0.03 mg/kg) in guinea pigs and NHPs, respectively.

This experiment was conducted to examine the effect of various legume herbs supplementation on feed intake, digestibility and liveweight gain of fat-tailed lambs given Mulato (Brachiaria mulato) grass. Twenty four male fat-tailed lambs (8 months of age and 12.73± 0.56 kg (SE) of initial liveweight) were randomly allotted to four treatment groups based on unfasted liveweight.  Animals were housed in individual metabolism crates. The experiment was designed in a completely randomized block design, with four treatments and six replicates. The dietary treatment tested included Mulato grass ad libitum (M), M + Centrosema pascuorum  (MCP), M + Dolichos lab lab (MDL) and M+ Clitoria ternatea (MCT). All supplement was offered at 1.5% body weight (W) per day.  The experiment lasted for 10 weeks, with 2 and 8 weeks for adaptation and measurement period, respectively. The main parameter measured included feed intake, feed digestibility, and average daily liveweight gain (ADG).  Results showed that total DM intakes (DMI) were not affected (P>0.05) by legume herbs supplementation. Total DMI was 2.75, 2.79, 2.84 and 2.87 %W/d for lamb treated with M MCP, MDL and MCT respectively. In contrast, supplementation with various legume herbs significantly increased (P <0.05) DM digestibility (DMD) and ADG of lamb received Mulato grass, with no significant difference (P>0.05) between legume herbs.  Feed DMD was 58.17, 67.48, 64.91; 65.03% and ADG were 35.67; 58.39; 54.31 and 54.41 g/day, for lamb treated with M, MCP, MDL and MCT respectively. It was concluded that supplementation of legume herbs to fat-tailed lamb fed Mulato grass significantly increased DMD and ADG, but no significant differences across the legume herbs as feed supplements were observed.

Grazing saltbush reduces productivity in sheep mostly because the high salt intake decreases feed intake and challenges the metabolism of the animal. However, little is known of the effect of salt load on the endocrine control systems that regulate voluntary feed intake and metabolism. Plasma concentrations of leptin, insulin and cortisol and blood glucose were monitored in wethers fed for 2 weeks with either a control diet (adequate salt) fed ad libitum, a high salt diet (20% of dry matter) fed ad libitum or a group fed the control diet with an intake restricted to that of the high salt ad libitum group (control pair-fed). High salt intakes reduced voluntary feed intake within 1 day and circulating concentrations of insulin and glucose within 2 weeks. Liveweight and leptin concentrations were not specifically affected by the high intake of salt but decreased in response to the decrease in intake. Cortisol secretion was not affected. Although salt intake had a specific effect on insulin and glucose (over and above the effect of reduced feed intake alone), the reduction in insulin would be expected to increase rather than decrease appetite and feed intake. Therefore, insulin, leptin and cortisol do not appear to play major roles in the control of feed intake in sheep consuming high levels of salt.

A feedlot trial and a digestion and nitrogen balance trial were carried out to evaluate the effect of feeding one (1 ×) or four times (4 ×) daily on the utilization of two high-forage diets. On a dry matter basis, diet 1 contained 76% alfalfa haylage and 22% high-moisture corn; diet 2 contained 93% corn silage and 4% soybean meal; the balance was composed of mineral and Rumensin premixes. In the feedlot trial, feed was offered ad libitum, and feeding frequency resulted in small differences in eating patterns within diets. Thus, rate of gain, feed intake and conversion were not significantly affected. In the digestion trial, feed intake was restricted to 90% of each individual's maximum intake, which resulted in larger differences in eating patterns between 1 × and 4 × feeding. Under these conditions, feeding frequently increased (P < 0.05) dry matter and organic matter digestibility, as well as N retention (percent of N intake). Increased feeding frequency had no beneficial effects under ad libitum feeding of high-forage diets. However, under restricted feeding, feeding more frequently improved total diet digestibility and nitrogen retention. Key words: Cattle, feeding frequency, digestibility, growth, nitrogen retention

Using alternative feedstuffs can be a feasible measure to reduce swine production cost. GuarPro F-71 (GP71), a newly developed guar meal product, is considered as a potential protein source for pigs. This preliminary study was conducted to evaluate the effects of GP71 inclusion in feed on growth performance and nutrient metabolism of growing pigs. Sixty crossbred barrows (31.9±1.84 kg body weight (BW)) were allotted to one of five dietary treatments in a completely randomised experiment design, with four pens/treatment and three pigs/pen. Diet 1 was a maize- and soybean-based basal control diet formulated to meet NRC recommendations. GP71 was used to replace 25, 50, 75, and 100% soybean meal in diet 1 (on an equivalent crude protein (CP) basis) to generate diets 2, 3, 4, and 5, respectively. Crystalline amino acids were used to balance the essential amino acid concentrations across the five diets. Inclusion of GP71 quadratically reduced average daily feed intake (ADFI) (P=0.01) with no observed clinical signs of unhealthiness of pigs. The blood plasma concentrations of six metabolites, including urea nitrogen, total protein, albumin, glucose, total cholesterol and total triglycerides, were not affected (P>0.30), indicating that the pigs’ metabolic utilisation of nutrients was not compromised. The cubic effect (P<0.05) of dietary GP71 inclusion on G:F ratio suggested that the level of soybean meal replacement (up to 75% soybean meal) did not negatively affect feed efficiency in pigs. In short, the feed intake reduction due to the dietary GP71 inclusion was, if not solely, responsible for the average daily gain (ADG) and final BW reduction. This study suggested that using GP71 to replace soybean meal up to 75% (on an equivalent CP basis) can negatively affect ADFI and ADG, but not feed efficiency, in growing pigs. The reasons for the ADFI and ADG reduction need to be understood in future research.

Abstract Porcine reproductive and respiratory syndrome (PRRS) virus is one of the most economically significant pig pathogens worldwide. The virus infects pigs at all production stages and significantly reduces the tissue accretion of growing pigs. However, the metabolic explanation for these reductions in tissue accretion remain poorly defined. Additionally, PRRS virus infection is often accompanied by reductions in feed intake, making it difficult to discern which effects are virus versus feed intake driven. To account for this, a pair-fed model was employed to examine the effects of PRRS infection and nutrient restriction on skeletal muscle and liver metabolism. Forty-eight pigs were randomly selected (11.34 ± 1.54 kg BW) and allotted to 3 treatments (n = 16 pigs/treatment): 1) PRRS naïve, ad libitum fed (Ad), 2) PRRS-inoculated, ad libitum fed (PRRS+), and 3) PRRS naïve, pair-fed to the PRRS-inoculated pigs’ daily feed intake (PF). At dpi 10 and dpi 17, 8 pigs per treatment were euthanized and tissues collected. Tissues were assayed for markers of proteolysis [longissimus skeletal muscle (LM) only], oxidative stress (LM only), and glycogen levels (LM and liver). Markers of LM proteolysis (calpain, 20s proteasome, caspase 3/7 activities) and mitochondrial reactive oxygen species production did not differ (P > 0.10) between treatment groups at either timepoint. Liver glycogen stores were reduced (P < 0.001) in PRRS+ pigs compared with both Ad and PF pigs, which did not differ from each other. Muscle glycogen did not differ (P > 0.10) between treatment groups. However, liver glycogen stores were completely depleted due to PRRS+, but not pair-feeding, indicating that liver glycogen stores and glucose are preferentially utilized by pigs to produce and support immune components. These data suggest that even under severe viral challenge and feed restriction, the pig does not upregulate LM proteolysis to re-allocate nutrients to fuel the immune response.

Abstract. Title of the paper: Investigations on feed intake behaviour of ad libitum fed group housed pregnant sows at tube feeders Investigations took place in 3 farms with group sizes of 8 to 32 sows and ad libitum feeding at tube feeders with energy reduced feed (9.3 to 9.6 MJ ME/kg) using unmolassed sugar beet chips (10 %), straw meal (25 %) resp in ration. Behavioural observations have been made with infrared video technique and time lapse video recording always during 24 hours. Individual duration of staying at trough in 24 hours and percentage of time feeding places were used by different number of sows per hour during 24 hours were analysed by QBSERVER/Video-Tape-Anatysis-System. Large individual differences in duration of staying at trough in 24 hours occurred between sows of a group with means from 1.8 to 18.0 % although all sows had free access to always available food. Behavioural investigations have shown that several sows did not come to trough over 24 hours without showing clinical signs of a disease At the same time this is a hint at the problem of disease control of ad lib fed sows especially in large groups Analysing the percentage of time feeding places were used by different number of sows a biphasic dynamics of feed intake behaviour of ad lib fed sows was found with a peak in the morning (7 to 10 a.m.) and a more distinct peak in the afternoon (3 to 7 p.m.).
